    Применение грибных метаболитов для оценки состояния сосновых древостоев в условиях промышленного загрязнения

Аннотация: На постоянных пробных площадях в сосняках разной степени загрязнения близ Красноярска испытали 2 метода оценки состояния древостоев: глазомерный и воздействия грибными метаболитами. В первом случае состояние древостоев оценили по 6-бальной шкале действующих в РФ санитарных правил как среднее значение баллов, присвоенных каждому из 200 деревьев на пробных площадях. Во втором, их состояние определяли по размеру некротического пятна на лубе ствола случайно выбранных 22-25 сосен после инокулирования деревьев экстрактом гриба Ceratocystis laricicola Redf. et Minter. Размер некрозов у сосен загрязненного бора был достоверно больше по сравнению с таковым у деревьев фонового массива. Это указывает на снижение активности противоинфекцион-ной защиты деревьев, ослабленных загрязнением, хотя состояние загрязненных и фоновых древостоев по результатам глазомерного метода не различалось.

Аннотация: Ophiostomatoid fungi colonize the conducting tissues of conifer stems, the phloem and the xylem. These pathogenic fungi penetrate into the stem through injuries made by xylophagous insects vectoring these pathogens. In this study the response of the phloem of Scotch pine (Pinus sylvestris L.) to wounding (treatment 1) was compared with the response to wounding combined with application of high-molecular-weight compounds isolated from the mycelium of the ophiostomatoid fungus Ceratocystis laricicola Redfern & Minter (treatment 2). Both treatments induced the appearance of necrosis in the inner bark, the formation of periderm separating living and dead tissues, and formation of the callus alongside the wound perimeter. In addition, the bark accumulated lignin, bound proanthocyanidins, and resins, with a parallel decrease in the content of free proanthocyanidins, low-molecular-weight carbohydrates, and non-lignin components of the cell wall (P > 0.95). The size of necrotic spots, as well as changes in the content of most substances, were significantly higher in the treatment 2 than in the treatment 1 (P > 0.95). The accumulation of lignin in cell walls of phloem sieve cells was delayed in the treatment 2 as compared with that in the treatment 1. This suggested that the mycelial extract temporarily inhibited lignification at the early stage of the wound response. This disturbance of the cell wall protective transformation led to the hypothesis that the fungal suppressors retard the repair of inner bark injured by insects, thereby favoring the invasion of conifer tissues by ophiostomatoid fungi.

Аннотация: Two methods for evaluation of pine stand vigor were tested on permanent sample plots near Krasnoyarsk City: visual evaluation and exposure to fungal metabolites. In the first case, forest vigor was estimated using the six-point system of the Sanitary Regulations of the Russian Federation as the mean score for 200 trees on the sample plots. In the second case, the vigor was evaluated from the size of the necrotic spot on inner bark of the tree after inoculation of 22-25 randomly chosen pines with the extract of fungus Ceratocystis laricicola Redfern and Minter. The necrotic spot size was significantly larger in pines from polluted forest as compared to background one. This points to the decrease in infection protection of trees affected by pollution, although visual evaluation of the stand vigor could not distinguish the polluted and background stands.

Аннотация: Callus cultures of Siberian larch (Lar ix sibirica Ledeb.) and Siberian spruce (Picea obovata Ledeb.) were used to demonstrate the elicitor activity of two ophiostomatoid fungal species, Ceratocystis laricicola and Ceratocystis polonica, as the pioneer settlers on larch and spruce, respectively. The extract from C. laricicola mycelium stimulated the accumulation of lignin in larch cells by 37% and that of hound proanthocyanidins by 25%. In spruce callus cultures, C. laricicola and C. polonica increased the bound PA content by 25 and 46%, respectively. In the callus cultures of larch and spruce, the addition of extract of C. laricicola increased the concentration of p-hydroxybenzoic acid 13-fold and nearly 4-fold, respectively. The metabolic characteristics of accumulation of phenolic compounds in conifer cells are discussed.

Аннотация: Effect of tannin and non-volatile components of lesion resin on the growth of blue-stain fungi - Ceratocystis laricicola, C. polonica, Ophiostoma minus, Leptographium sp. and Ophiostoma sp. - the primary invaders of conifers in Siberia (Russia), has been studied. The fungi under study exhibited tolerance to plant's defensive substances in bioassays. The cultures of Leptographium sp., C. polonica, and Ophiostoma sp. proved to be the most tolerant to tannin. In the range between 0.05 and 0.15% tannin concentration their growth tended to stabilize. Less tolerant to tannin were C. laricicola and O. minus. With an increase in tannin concentration their biomass gradually decreased. The fungi more tolerant to tannin regulated the medium pH about 5.0 and higher, white the species which were less tolerant preferred lower pH values. Lesion resins of conifers differed in their inhibitory effect on fungal growth. Fir resin had the greatest inhibitory effect of all the conifer resins tested, causing reduction to 50-60% in fungal growth. When fungi were grown on the resin of any other conifer, the reduction in the linear growth rate did not exceed 20 to 30%. The assumption that fungi are more tolerant to the resin of a host plant than to that of a non-host plant has not been confirmed.

Аннотация: A method for estimation of tree's condition on activity of defence response in phloem was checked in Pinus sylvestris L. stands located near Krasnoyarsk (Siberia, Russia). The length of necrosis caused by inoculation of phloem of living tree by Ceratocystis laricicola Redf. et Minter was used as the parameter being measured. The field experiments were carried out in two even-aged (about 60 years) pine stands that were approximately equal on structure, productivity, density and recreation loading, but differentiated on degree of industrial pollution. The two permanent sample plots (SP) were founded in the severely polluted stand. The other two SP were placed in the unpolluted pine forest. The amount of pine trees within each SP varied from 200 to 250. From 22 up to 37 pine-trees selected randomly within every SP were inoculated with C. laricicola mycelium (test 1) and its extract (test 2). One inoculation hole per one tree (diameter 7 mm) was made in stem at a height of 1.3 m. Application of the both agents caused necrosis in phloem around the place of inoculation. In the case of fungal inoculation (test 1), the average length of necrosis in the unpolluted forest exceeded significantly the same parameter in the polluted stand: 51.7-79.4mm and 39.4-41.3mm, correspondingly. The action of the fungal metabolites caused the opposite results: in the unpolluted stand the average size of necroses was smaller in comparison with this parameter in the polluted stands 44.5-15.3 mm and 57.9-61.8 mm. The reasons of this difference are discussed. The both agents (C. laricicola mycelium and its extract) were suitable to reveal the difference of tree's condition in polluted and unpolluted pine stands. The application of fungal extract is more preferable in comparison with fungal mycelium because of smaller variability of necrosis size. Besides, the application of extract allows controlling inoculum dose and excludes the dangerous of spreading infection in forests.

Аннотация: Ceratocystis laricicola and C. polonica are fungal symbionts of bark beetle species of the genus lps that attack species of Larix and Picea, respectively, across Eurasia. Earlier studies found that these fungal species were morphologically identical, had similar isozymes patterns, and had identical internal transcribed spacer (ITS) sequences of the rDNA operon. We analyzed 27 isolates from Europe, southwestern Siberia (Russia) and Japan, representing the known geographic ranges of the two species. Phylogenetic analysis of the DNA sequences of a portion of the MAT-2 idiomorph showed these species to be distinct, with the Japanese isolates of C. laricicola having a sequence slightly different (5 bp) from those of the Russian and European isolates of C. laricicola. Sexual compatibility tests showed full interfertility among isolates of C. polonica from Europe, Russia and Japan, but isolates of C. polonica were not fully interfertile with isolates of C. laricicola. A Russian and a European isolate of C. laricicola mated with each other but not with the Japanese isolates of C. laricicola. Mature L. sibirica and P. obovata were inoculated with isolates of C. laricicola and C. polonica from Europe, Russia, and Japan, and measurement of lesions in the inner bark/cambium region demonstrated strong host specialization. The data suggest that the two fungal species are very closely related and are distinguished primarily by their physiological specialization to the hosts of their bark beetle vectors.

Аннотация: В 2002–2013 гг. на постоянных пробных площадях (ПП) исследовали состояние пригородных средневозрастных сосняков Красноярска. Была проведена ежегодная оценка параметров иммунного некротического ответа луба ствола у выборочных деревьев на действие экстрактивных веществ из мицелия Ceratocystis laricicola (Redfern&Minter). Измеряли размер некроза и его смещение вдоль ствола относительно инокуляционного отверстия. Выбранные сосняки (загрязненный и условно-фоновый) – удобные объекты для поиска критериев изменения их состояния на ранней стадии повреждения. Эти древостои подвергаются действию разных антропогенных факторов, но не различаются по категории состояния, визуально оцененной по 6-балльной шкале Санитарных правил РФ. ПП имеют близкие таксационные характеристики, за исключением ПП на опушке загрязняемого бора, где бонитет снижен. Зарегистрировано достоверное смещение некроза в лубе вверх по стволу в течение двух лет, следующих за годом, когда случился низовой весенний пожар. Это доказывает изменение нормального нисходящего транспорта сахаров на апикальный (в сторону вершины) и возможность использования асимметрии некроза для оценки воздействия пожара на физиологическое состояние древостоев. Увеличение некрозов (снижение иммунитета) после пожара отмечено в более поздний период по сравнению изменением транспорта ассимилятов в стволе. Влияние химического ожога хвои на апикальное смещение некроза выражено слабее по сравнению с пожаром.

Аннотация: Five permanent sample plots (SPs; 200-250 trees per plot) were established in middle-aged high-grade suburban pine stands near the industrial city of Krasnoyarsk, Siberia, Russia. Needle damage, inventory parameters of the stands, and the defense response of the stem phloem were evaluated annually for the years 2002-2019 and attributed to acute or chronic toxic exposures (creeping fire or industrial pollutants, respectively). The results form a basis for using trees as bioindicators. A newly elaborated stem lesion test was formed from a hypothesis on the upward sugar transport for the regeneration of an injured crown, based on Eschrich's model of bidirectional sugar transport in the phloem. The formation of a phloem lesion was induced by inoculation of the stem with a mycelial extract of the ophiostomatoid fungus Ceratocystis laricicola. The lesion length and its shift relative to the inoculation hole were measured. An increase in the length of needles at early stages of stand weakening by pollutants was found to correspond to the hormesis model (Selye's adaptation syndrome). A possibility of assessing the chronology of pollutant toxicity and the duration of the recovery period after creeping fire was shown.

Needle damage, inventory parameters of the stands, and the defense response of the stem phloem were evaluated annually for the years 2002-2019 and attributed to acute or chronic toxic exposures (creeping fire or industrial pollutants, respectively). The results form a basis for using trees as bioindicators. A newly elaborated stem lesion test was formed from a hypothesis on the upward sugar transport for the regeneration of an injured crown, based on Eschrich‘s model of bidirectional sugar transport in the phloem. The formation of a phloem lesion was induced by inoculation of the stem with a mycelial extract of the ophiostomatoid fungus Ceratocystis laricicola. The lesion length and its shift relative to the inoculation hole were measured. An increase in the length of needles at early stages of stand weakening by pollutants was found to correspond to the hormesis model (Selye‘s adaptation syndrome). A possibility of assessing the chronology of pollutant toxicity and the duration of the recovery period after creeping fire was shown. © 2020 by the authors.
