    Genetic diversity, structure and differentiation of Gmelin Larch (Larix gmelinii (Rupr.) Rupr.) populations from Central Evenkia and Eastern Zabaikalje

Аннотация: As a results of studies conducted it was established that genetic diversity and structure of L. gmelinii populations in Central Evenkia and in Eastern Zabaikalje are significantly distinguished. The mean genetic distance D between populations from these regions calculated on the basis of allelic frequencies 17 isoenzyme loci is equal to 0.03. The review of data at genetic diversity and intraspesific differentiation of conifer species showed that such extent of genetic differentiation is characteristic of geographical races of single species. On this basis we concluded that L. gmelinii populations from Central Evenkia and Eastern Zabaikalje are genetically isolated and can be regarded as geographical races. The data obtained confirm the supposition of Abaimov and Milyutin that L. gmelinii, growing in various on natural conditions regions within the limits of its vast area, has to subdivide into more small intraspecific taxons. Results of our studies represent only a part of the total work that is necessary to fully assess the variability of this species.

Аннотация: The genetic diversity and population structure of Siberian fir (Abies sibirica LEDEB.) in Middle Siberia were studied on the basis of allozyme variation analysis at 20 loci. The vegetatives buds collected from 260 trees in nine populations representing different geographical localities and altitudinal sites were used as materials for study. Horizsontal starch gel electrophoresis was applied to analyze the isozyme patterns of 11 enzyme systems. 20% of the gene loci assayed were revealed to be polymorphic at 95%, and 35% at a 99% criterion. The mean number of alleles per locus, the mean observed heterozygosity, the mean expected heterozygosity, and the effective number of alleles were equal. The obtained data indicates a low level of genetic diversity and a weak differentiation among the A. sibirica populations studied in Middle Siberia. The most significant difference in structure was that between Kozulka and Western Sayan.

Аннотация: Genetic diversity and defferentiation of Scotch pine populations situated on eutrophic, mesetrophic, oligotrophic swamps and on dry land in south-taiga subzone of Western Siberia lowland (Tomsk region) were srudied on the basis of allozyme variation analysis at 20 loci. The most essential distinctions in genetic structure were revealed between populations of Scotch pine growing under contrast conditions of water-mineral nutrition on eutrophic and oligotrophic swamps as well as between populations situated on the various oligotrophic swamps. The obtained results testify to considerable genetic heterogeneity studied in Tomsk region of marsh populations of Scotch pine.

Аннотация: The genetic diversity of SSR markers was studied in six populations of Siberian larch, two popu-lations of Gmelin larch, and four populations of Cajander larch. Seven pairs of nuclear microsatellite loci were used for this analysis. 103 allelic variants were detected in 365 individuals of three species of larch. According to the AMOVA results, the variability proportion that characterizes the differences between three Larix species was twice as higher (15%) than the proportion that accounts for among-population differences within the species (6%). The differentiation of the populations of three species of larch based on SSR markers exceeded 12% (F (ST) = 0.121). A significant correlation of the genetic distances with the geographic distances between populations was found (r = 0.835, P = 0.01).

Аннотация: Using AFLP analysis, the genetic variation in four population samples of Rhododendron ledebourii, three such samples of Rh. dauricum, two samples of Rh. sichotense, and one sample of Rh. mucronulatum was examined. Application of three selective primer pairs provided genotyping of 87 accessions from the four species at 213 loci. An analysis of molecular variance revealed a high level of differences between all four species (F (ST) = 0.172). Populations within the species were also differentiated: for Rh. dauricum F (ST) = 0.245; Rh. ledebourii F (ST) = 0.264; Rh. sichotense F (ST) = 0.034. The highest Nei distance was observed for Rh. mucronulatum with Rh. dauricum and Rh. ledebourii (D = 0.057 and 0.069, respectively). Similarity was observed between the species pairs of Rh. ledebourii-Rh. dauricum (0.032) and Rh. sichotense-Rh. mucronulatum (0.034). The Far Eastern species Rh. sichotense and Rh. mucronulatum were characterized by an elevated level of intrapopulation variation. The data from the AFLP-based analysis of differentiation and variation that were thus obtained were compared with the literature data on the morphological criteria that differentiate the species of interest and those regarding the history of the formation of their ranges. The independence of all the species we examined was confirmed.

Аннотация: The variability of the masticatory surface of the first inferior molar tooth in the narrow-sculled vole was studied in four partly isolated populations in Central Siberia. The morphological differentiation consisting in the specific quantitative relation between the structural variants of teeth was revealed. The main trend of reducing the frequency of occurrence of complicated structural tooth variants from the north to the south was revealed.

Аннотация: The seasonal development of phloem in the stems of Siberian larch (Larix sibirica Ldb.) was studied over two seasons on 50-60-year-old trees growing in a natural stand in the Siberian forest-steppe zone. Trees at the age of 20-25 years were used to study metabolites in differentiating and mature phloem elements, cambial zone, and radially growing xylem cells in the periods of early and late wood formation. The development of the current-year phloem in the stems of 50-60-year-old trees started, depending on climatic conditions, in the second-third decades of May, 10-20 days before the xylem formation, and ended together with the shoot growth cessation in late July. Monitoring of the seasonal activity of cambium producing phloem sieve cells and the duration of their differentiation compared to the xylem derivatives in the cambium demonstrated that the top production of phloem and xylem cells could coincide or not coincide during the season, while their differentiation activity was always in antiphase. Sieve cells in the early phloem are separated from those in the late phloem by a layer of tannin-containing cells, which are formed in the period when late xylem formation starts. The starch content in the structural elements of phloem depends on the state of annual xylem layer development. The content of low molecular weight carbohydrates, amino acids, organic acids, and phenols in phloem cells, cambial zone, and xylem derivatives of the cambium depends on the cell type and developmental stage as well as on the type of forming wood (early or late) differing by the cell wall parameters and, hence, by the requirement for assimilates. Significant differences in the dynamics of substances per dry weight and cell were observed during cell development.

Аннотация: Based on the analysis of 22 loci controlling allozyme variation of MDH, SKDH, 6-PGD, IDH, PEPCA, GOT, FDH, LAP, PGI, PGM, SOD, and GDH, the data on within-and among-population variation for nine cenopopulations of Siberian spruce (Picea obovata Ledeb.) located along the Yenisei mieridian, from 65 degrees 50' NL to 52 degrees 14' NL, were obtained. It was demonstrated that 86.36% of the loci, tested in the species analyzed, were polymorphic. The mean number of alleles per locus was 2.91, and the observed and expected heterozygosity constituted 0.161 and 0.168, respectively. More than 97% of total variation occurred within the populations, while the proportion of the among-population variation constituted only 2.3% (F-st =0.0230). Genetic distance (D-N) between the populations examined varied from 0.0019 to 0.0115, averaging at 0.0051. It was shown that in the part of the Siberian spruce range examined there was no close association between the level of the genetic differentiation of the populations and the geographic distance between them. It seems likely, that this finding can be associated with the fact that Siberian spruce, growing on this territory, is the intrazonal species, i.e., it is not attached to a certain forest zone, and its distribution is mostly determined by local ecological conditions.

Аннотация: Multivariate analysis unambiguously demonstrated the differentiation of oxidoreductase activity (catalase, peroxidase, and dehydrogenase) in peat soils after a 20-25-year period of bog drainage and afforestation. The enzyme activity depended on the drainage depth. A statistical model has been developed to predict the degree of humification of peat organic matter from peroxidase activity and moisture of drained soils. Soil peroxidase activity can be an important indicator of the degree of biochemical transformation of drained and forested bogs.

Аннотация: The contents of ascorbic acid (AA) and its oxidized form, dehydroascorbic acid (DHA), were assessed as related to the tracheid differentiation in the course of early and late wood development in the Siberianlarch (Larix sibirica Ldb.) trees. The samples of the cambium, cell enlargement zone and mature cells were collected at the successive developmental stages by scraping tissues off layer by layer from trunk segments of the 20-year-old trees according to anatomical and histochemical criteria. While cambium initials were rapidly dividing, the AA contents per dry weight and per cell considerably exceeded the corresponding values characteristic of the late xylem development; such difference corresponded to the higher number of early tracheids per annual ring, as compared to the late tracheids. The AA content decreased as cells enlarged. The radial growth of the early wood tracheids, as compared to the late wood tracheids, was accompanied with a threefold increase in the AA and a decline in the DHA contents. The AA/DHA ratio was in line with the early tracheid enlargement. The maximum AA content was observed at the early stage of the secondary cell wall thickening in the tracheids of early and late xylem preceding lignification. During this stage of early wood development, the DHA content exceeded sixfold the corresponding value in the late xylem; as a result, the initial rates of lignification were different in two tissues. The rate of lignification in a newly developing layer of the early xylem increased gradually and was the highest in the completely differentiated tracheids. In the late xylem, the lignification rate was at its highest at the very beginning and then declined in the course of tracheid maturation. The dissimilar patterns of lignification in the early and late xylem were primarily associated with the DHA content, which increased in the early xylem and decreased in the maturing late xylem. Thus, the AA content and its accessibility to oxidation in the growing and mature xylem cells exhibited the diverse developmental patterns in the early and late xylem: two tissues differed in the tracheid number and radial diameter as well as in the rate of lignification.

Аннотация: Within- and among-population diversity of Gmelin larch Larix gmelinii (Rupr.) Rupr. from Evenkia was inferred from data on 17 genes determining allozyme diversity of ten enzymes. More than 50% of the genes proved to be polymorphic. On average, each tree was heterozygous at 9.2% genes. Heterozygosity expected from the Hardy-Weinberg proportions was higher, 12.5%. A deficit of heterozygous genotypes was observed in all populations under study and attributed to inbreeding. With Wright's F statistics, average individual inbreeding was estimated at 26.6% relative to the population (F-IS) and at 27.8% relative to the species (F-IT). The greatest deficit of heterozygosity was observed for the youngest population II. Within- population variation accounted for more than 98% of the total variation, while the contribution of among-population variation was 1.66%. Genetic distance between populations varied from 0.0025 to 0.0042, averaging 0.0035.

Аннотация: it has been determined that the sex structure of the population is of great importance for pine adaptation to extreme environmental conditions. As environmental conditions deteriorate, differentiation of populations with respect to sex becomes more apparent, the proportion of trees of mixed type decreases, and the number of male trees increases. A tendency toward correlation between tree habitus, needle and shoot sizes, and the sex structure of population is noted. The prevalence of male trees in populations exposed to increasing stress reflects the rearrangement of such populations aimed at the more efficient use of natural resources.

Аннотация: The main parameters of genetic variability have been determined in an isolated natural Scotch pine population from Chita oblast (Tsasuchei Forest) by analysis of 19 genes coding for nine enzymes: GDH, IDH, LAP, PGM, AAT, ADH, MDH, 6-PGD, and DIA. Polymorphic genes constituted 63.2% of all structural genes studied in the population at the 99% polymorphism criterion. The mean number of alleles per locus was 1.63. The observed and expected heterozygosities were 0.237 and 0.251, respectively. These estimates are close to the corresponding mean values for Scotch pine according to the data on 18 or more structural genes.

Аннотация: Statistical analysis of high-frequency variations in the radial dimensions of tracheids was carried out using precise measurements of five radial rows in each annual tree ring for several coniferous species: Larix gmelinii, L. leptolepis, L. sibirica, Picea abies, Pinus sylvestris is, P. rigida, and P. densiflora. More than 25 tree rings with a varying number of cells and width were measured for each species. High-frequency variations in radial dimensions were found to have a regular (cyclic) characteristic and accumulated 5 to 11% of the total variability. The cyclic pattern was identified using Fourier analysis of time-related or successive series statistical procedure. The first order autocorrelations (-0.45 to -0.74) and the mean cycle of high-frequency variations (2.1-2.5) did not depend on the number of cells in the annual ring, i.e., they are determined by internal causes. The cyclic pattern of high-frequency variation in tracheid dimensions is used to interpret the seasonal mechanism of xylem formation. Specifically, (1) high-frequency variations in the tracheid radial size take place during the last asymmetrical division of the xylem mother cell before transition to the elongation zone, (2) the final tracheid radial size is mostly determined within the zone of division due to the last asymmetrical division, and (3) acceleration of cell cycle from the initial to the periphery of the cambial zone. The results obtained are also discussed with respect to the mechanisms of xylem differentiation.

Аннотация: Data on the bulk content of microelements in thixotropic and homogeneous cryozems of autonomous (eluvial and transeluvial) landscapes in the northern taiga and forest-tundra subzones in the Yenisei River basin are presented. The main associations of elements and their correlative relationships with soil properties were determined for genetic horizons. Weak differentiation of most microelements in the soil profile is typical for these soils, which can be attributed to severe hydrothermic conditions of the region and to the regional peculiarities of chemical composition of parent rocks.

Аннотация: Changes in the levels of ascorbic acid (AA), its oxidized form, dehydroascorbic acid (DHA), and uronic acids as initial precursors for the AA synthesis were studied as related to the degree of xylem and phloem cell development in the course of early and late wood formation in the trunks of Scots pine (Pinus sylvestris L.). The cells of mature and conducting phloem, cambial zone, differently developed cells in the zones of cell enlargement and maturation were obtained by successive scraping tissue layers from trunk segments of 20-25-year-old trees; tissue identification was checked anatomically and histochemically. The contents of compounds tested were calculated per dry weight and per cell basis. We found great differences in the contents of AA and DHA and also in their ratio in dependence of the wood type developing in the pine trunks during growth period and on the stage of differentiation of xylem and phloem cells. Changes in the AA content during xylem cell differentiation were accompanied by changes in the content of uronic acids. The amounts of AA, DHA, and uronic acids were the highest at the stage of early lignification and reduced with tracheid maturation. The AA to DHA ratio changed differently in the course of early and late xylem lignification. It reduced from the start of lignification to the formation of early mature xylem and, in contrast, increased in mature late wood; this indicates a difference in the level of redox processes in these tissues.

Аннотация: Natural and human-induced changes in the composition of boreal forests will likely alter soil properties, but predicting these effects requires a better understanding of how individual forest species alter soils. We show that 30 years of experimental afforestation in Siberia caused species-specific changes in soil chemical properties, including pH, DOC, DON, Na+,NH4+, total C, C/N, Mn2+, and SO42-. Some of these properties-pH, total C, C/N, DOC, DON, Na+-also differed by soil depth, but we found no strong evidence for species-dependent effects on vertical differentiation of soil properties (i.e., no species x depth interaction). A number of soil properties-NO3-, N, Al3+, Ca2+, Fe3+, K+, Mg2+ and Cl- -responded to neither species nor depth. The six studied species may be clustered into three groups based on their effects on the soil properties. Scots pine and spruce had the lowest pH, highest C/N ratio and intermediate C content in soil. The other two coniferous species, Arolla pine and larch, had the highest soil C contents, highest pH values, and intermediate C/N ratios. Finally, the two deciduous hardwood species, aspen and birch, had the lowest C/N ratio, intermediate pH values, and lowest C content. These tree-mediated soil chemical changes are important for their likely effects on soil microbiological activities, including C and N mineralization and the production and consumption of greenhouse gases.

Аннотация: The analysis of the anatomical characteristics of tree rings of the Pinus sibirica scions and Pinus sylvestris rootstocks is presented. The main anatomical features maintain the seasonal dynamics characteristic of the reference (ungrafted) trees of scion and rootstock, pointing to the stability of the genetic control of the xylem differentiation. However, various anatomical characteristics are reduced for rootstocks and increased for scions relative to the reference trees. A mechanism consistent with our results is that the graft union zone creates a barrier for the ascending and descending transport of substances, including phytohormones. The reaction of the scion and rootstock to climatic factors is weakened relative to the reaction of the reference trees. Thus, the presumed shift towards phytohormone disbalance reduces the influence of external factors on xylem differentiation. The interannual variation of the individual characteristics of the scions and rootstocks is increased relative to the reference trees, reflecting either the influence of the graft partners on each other or the non-uniform distribution of the growth regulators across the graft union. Thus, heterografts can be used to evaluate the interaction of the internal and external mechanisms of xylem differentiation.

Аннотация: An ecologo-phytocenotic classification of forest types has been worked out for the mountainous areas of the Western Transbaikalia. Using the floristic and ecologo-geographical criteria and the DCA ordination method, we identified seven groups of forest types belonging to two altitudinal-belt complexes. The resulting classification refines the existing understanding of the phytocenotic structure of forest cover under sharply continental climate conditions in the contact zone with island steppes of the Transbaikalia and serves the vegetation cover monitoring and modeling purposes.
