Using AFLP analysis, the genetic variation in four population samples of Rhododendron ledebourii, three such samples of Rh. dauricum, two samples of Rh. sichotense, and one sample of Rh. mucronulatum was examined. Application of three selective primer pairs provided genotyping of 87 accessions from the four species at 213 loci. An analysis of molecular variance revealed a high level of differences between all four species (F (ST) = 0.172). Populations within the species were also differentiated: for Rh. dauricum F (ST) = 0.245; Rh. ledebourii F (ST) = 0.264; Rh. sichotense F (ST) = 0.034. The highest Nei distance was observed for Rh. mucronulatum with Rh. dauricum and Rh. ledebourii (D = 0.057 and 0.069, respectively). Similarity was observed between the species pairs of Rh. ledebourii-Rh. dauricum (0.032) and Rh. sichotense-Rh. mucronulatum (0.034). The Far Eastern species Rh. sichotense and Rh. mucronulatum were characterized by an elevated level of intrapopulation variation. The data from the AFLP-based analysis of differentiation and variation that were thus obtained were compared with the literature data on the morphological criteria that differentiate the species of interest and those regarding the history of the formation of their ranges. The independence of all the species we examined was confirmed.

Genetic diversity -- Genetic structure -- Glacial refugia -- Northeast Asia -- Phylogeography -- Plastid DNA -- Rhododendron dauricum s.l

Аннотация: Genetic variation in 45 populations (267 plants) of Rhododendron dauricum s.l. across its range in Northeast Asia was assessed with four regions of plastid DNA (ptDNA). A total of 14 haplotypes were detected. The highest diversity was observed in the south of West Siberia (the Altai and Western Sayan Mountains) and the southern Russian Far East (the Sikhote-Alin Mountains). In contrast, only one haplotype occurred in populations from East Siberia located from Baikal to the Sikhote-Alin Mountains. In general, distribution of haplotypes showed a strong phylogeographical structure (GST = 0.897; NST = 0.985) and evidence of isolation by distance, supporting the independence of four species: R. ledebourii Pojark. and R. dauricum L. s.s. in Siberia, and R. sichotense Pojark. and R. mucronulatum Turcz. in the southern part of the Far East. © 2018 Springer-Verlag GmbH Austria, part of Springer Nature

SSR markers -- population structure -- genetic variability -- microsatellites

Аннотация: SSR analysis of Rhododendron aureum Georgi in eight populations (63 samples) from Siberia (West and East Sayans) and Kamchatka was performed. Polymorphism of five SSR loci showed a high level of genetic diversity in all the populations (mean values H-o = 0.667, H-e = 0.638). The analysis of molecular variation (AMOVA) revealed that about 16% of the genetic diversity of R. aureum occurs in interpopulation differences (F-st = 0.161, p = 0.001). The Bayesian approach in the STRUCTURE program revealed that the species has a heterogeneous genetic structure-populations from Kamchatka form a separate cluster and show high homogeneity, and populations from Western and Eastern Siberia are of mixed origin. The obtained results indicate the prospects of using this type of markers for population-genetic studies of R. aureum.

mountains of southern Siberia -- population variation -- rhododendrons -- Ericaceae -- Rhododendron -- Rhododendron dauricum

Аннотация: Abstract—: Twenty cenopopulations of Rhododendron dauricum and Rh. ledebourii, two closely related and morphologically hardly distinguishable species, have been studied with respect to one of the most significant taxonomic features, the life span of leaves on the shoot. The correlation of this feature with other morphological characters of plants and climatic conditions in the species habitats has been evaluated, and its intra- and interpopulation variation has been analyzed. In both species, leaf life span has been found to increase with an increase in elevation above sea level and annual precipitation. Significant differences in leaf life span between conspecific populations and the absence of significant differences between species have been revealed. © 2019, Pleiades Publishing, Ltd.

Alpine plants -- Biogeography -- Microsatellites -- Refugium -- Rhododendron -- Siberia

Аннотация: The vast territory of East Asia, including southwestern Beringia, is considered to have been almost ice free during the Pleistocene. Cold-resistant flora may have persisted in this region expanding or contracting its range during the climate cooling. Only a few plant genera have been studied with a sampling area across their entire geographic range in East Asia; therefore, the understanding of the biogeographic history of alpine flora in this region remains limited. In the present study, genetic variation and population structure in 21 populations of the alpine shrub Rhododendron aureum across its range in East Asia were assessed using 18 microsatellite loci. Phylogenetic analyses revealed three main genetic groups: Siberia, Northeast, and North Pacific. According to the geographical pattern of genetic diversity, the North Pacific group includes populations from Kamchatka, south of Russian Far East, and territories close to central Japan. This group is the most diverse and likely diverged earlier than the Siberia and Northeast groups. Ecological niche modeling predicts range expansion of this species during the period of cooling and, together with demographic history, suggests that the divergence between the three main genetic groups predated the Last Glacial Maximum. Similar to other cold-resistant species such as Larix sibirica and Juniperus communis, the pattern of genetic diversity of R. aureum supports the survival of the species at high latitudes during the Pleistocene with limited contribution of the southern populations to expansion of the species range to the Northeast region and Siberia. © 2021, The Botanical Society of Japan.

A study of morphological traits in populations of R. aureum from the Western and Eastern Sayan and Kamchatka was carried out. The variability of genetic nuclear DNA microsatellite markers (nSSRs) was studied in the same samples. A total of 15 absolute and relative morphological traits of shoots and leaves, as well as 18 polymorphic microsatellite markers in 8 natural populations of R. aureum were observed. It was found that the following parameters were the most variable within species populations: plant size, shoot length, length of the foliated part of shoots and number of leaves relative to shoot length; more stable parameters were relative leaf shape parameters. Reliable division of populations into 3 geographical groups on all set of morphological characters is established. A great contribution to the division of geographical groups of populations and individual populations of an important taxonomic character - the duration of leaf life on the shoot was noted. The revealed structure of morphological differentiation of populations is confirmed by the results of the analysis of genetic variability of the same populations of R.aureum by nuclear microsatellite markers. The greatest genetic distances from other populations are established for Kamchatka populations. There is a tendency to differentiation of populations within the Altai-Sayan mountain country. The results of the study agree well with the information on geography, ecology and biology of the species. The study of intra- and interpopulation morphological differentiation of R. aureum populations in Siberia was conducted for the first time, following the recently initiated studies on genetic variability of the species in Northern Eurasia.
