Аннотация: Karyotypes of three Larix species (L. sibirica, L. gmelinii, and L. cajanderi) were analyzed using fluorescence in situ hybridization (FISH) with 45S and 5S ribosomal RNA gene probes and 4',6-diamidino-2-phenylindole (DAPI) staining. Two major 45S ribosomal DNA (rDNA) loci (per haploid genome) have been observed in the intercalary regions of two metacentric chromosomes, III and IV, of L. sibirica; in addition to them, minor nucleolus organizing regions (NORs) were mapped in pericentromeric regions of chromosomes I, II, VI, and XII. Two closely related species, L. gmelinii and L. cajanderi, showed similar hybridization patterns; both species possessed an additional major locus of 45S rDNA in the distal region of the long arm of submetacentric chromosome VII that is absent in L. sibirica. Only one locus of the 5S rDNA was found in all larch species we studied; it was located in the distal region of the chromosome III short arm, which also carried the major NOR in the opposite arm. This chromosome containing major loci of the two ribosomal RNA gene families can serve as a marker of the genus Larix. The intra- and interspecific karyotype diversity in the genus Larix is discussed.

Аннотация: The cytogenetic study of Abies sibirica seed progenies in declining fir stands of West Sayan High Mountains has been carried out. The increased amount of chromosome and genome mutations, pathologies of mitotic cycle and cells with micronuclei were found at studying. The revealed irregularities may be the result of a long-term exposure to extreme environmental factors and probably reflect the high degree of disturbance of the ecosystem in this region.

Аннотация: The results of a karyological study on Siberian stone pines growing in the bog demonstrated differences from the populations of this species studied earlier with respect to sizes of chromosomes and location of secondary constrictions. The number of nucleolar organizer regions in the chromosomes of Siberian stone pine trees growing in the bog was larger than in other populations. A wide spectrum of chromosome aberrations was revealed, which had not been observed in this species before. These were genome and chromosome aberrations of various types, as well as structural aberrations accompanied by changes in the number of chromosomes. It is assumed that the aberrations in the karyotype of Siberian stone pine resulted from stressful conditions.

Аннотация: Data are presented on the karyotype structure of the Siberian fir (Abies sibirica Ledeb.) from five populations of the Central Siberia. The chromosome set (2n = 24) contains seven pairs of metacentric (I-VII), four pairs of submetacentric (VIII and X-XII), and one pair of intercentric (IX) chromosomes. The morphometric parameters of the identified chromosome groups had close values in the studied populations. The variation coefficients of chromosomal parameters of the Siberian fir correspond to very low and low variability. The intraspecific chromosome polymorphism of the Siberian fir is connected mainly to variations in the number and peculiarities of the nucleolar organizing regions of chromosomes. In the territory with technogenic loading, a wide spectrum of genomic mutations of the mixoploidy type was observed in the seed off-spring of the Siberian fir; triploid seedlings, as well as rare cases of somatic reduction of chromosomes, were revealed. Аннотация: We present the results of cytogenetic analysis of seven young and long-term cultured embryogenic cell lines (CLs) of Larix sibirica obtained by somatic embryogenesis. All CLs were found to be mixoploid. Three CLs (1, 2 and 6 years of cultivation) were the most stable cytogenetically with 91-96% cells having diploid chromosome number 2n=24, typical for the species. These CLs can be successfully used for the development of planting material and in clonal plantation forestry. At the same time, some embryogenic CLs were shown to possess various chromosome and genome mutations. We found two aneuploidy proliferating CLs (2n=25 and 28) capable of somatic embryo formation. One of the CLs analyzed in a current study was found to be cytogenetically unstable with chromosome numbers ranging from 24 to 35 and containing one-two micronuclei in the interphase cells. Fluorescent in situ hybridization (FISH) and morphometric analyses of karyotypes of these embryogenic CLs allow an assumption that the trisomy for one or several chromosomes is the most widespread type of genomic disturbances in embryogenic CLs of L. sibirica.

Аннотация: A comparative study of karyotypes of two Siberian species of larchL. sibirica and L. gmeliniigrowing in the zone of forest-tundra on the Taimyr Peninsula was carried out. The number of chromosomes has been analyzed and the increase in chromosomal instability in larches in forest-tundra conditions has been shown. The size and morphology of chromosomes and the localization and frequency of secondary constrictions in chromosomes of L. sibirica and L. gmelinii were estimated. The parameters of chromosomes in polyploid cells of L. gmelinii were studied. The asymmetry of karyotypes of L. sibirica and L. gmelinii was analyzed. The spectrum and frequency of chromosome rearrangements in L. sibirica and L. gmelinii were studied. Using the data of the karyological analysis, the issues of differentiation and adaptation of the investigated larch species in Taimyr are discussed.

Аннотация: Обобщены результаты кариологических и цитогенетических исследований в популяциях видов хвойных семейств Pinaceae и Cupressaceae из экстремальных условий произрастания. У хвойных, произрастающих у южной и северной границ ареалов на экологическом пределе распространения, в антропогенно нарушенных экосистемах, в условиях интродукции, выявлена хромосомная изменчивость, обнаружены геномные и хромосомные мутации различного типа. Отмечены высокая частота изменений числа хромосом (миксоплоидия, анеуплоидия и полиплоидия) и их морфологии. Найдены различные типы хромосомных аномалий (кольцевые и полицентрические хромосомы, фрагменты, множественные хромосомные аномалии), агглютинация хромосом, появление В-хромосом, нарушения митоза и мейоза. С использованием классических методов и флуоресцентной гибридизации in situ (FISH) проведены исследования полиморфизма нуклеолярных районов хромосом, выявлены особенности локусов генов 5S и 45S рибосомной РНК.
Results of karyological and cytogenetical studies in populations of conifer species of Pinaceae and Cupressaceae families from extreme conditions are summarized. In conifers growing in southern and northern borders of areas, on ecological limits of distribution, in anthropogenically disturbed ecosystems, at the introduction, chromosome variability is revealed. A high frequency of changes in the number of chromosomes (mixoploidy, aneuploidy and polyploidy) and their morphology were noted. Different types of chromosome anomalies (ring and polycentric chromosomes, fragments, multiple chromosome irregularities), B-chromosomes occurrence, irregularities of mitosis and meiosis are found. Using classical methods and fluorescent in situ hybridization (FISH), studies of the polymorphism of the nucleolar regions of chromosomes were carried out, and features of the loci of the 5S and 45S ribosomal RNA genes were revealed.

Аннотация: Представлены результаты цитогенетического исследования четырех эмбриогенных клеточных линий (КЛ) лиственницы сибирской (Larix sibirica Ledeb.) разной продолжительности культивирования: молодые (две четырехмесячные и 16-месячная КЛ) и длительно-пролиферирующая (11-летняя КЛ). Три клеточные линии являются диплоидными и содержат в кариотипе нормальное для данного вида число хромосом (2n = 24). Клеточная линия, которая культивировалась 16 месяцев, способная формировать соматические зародыши, оказалась анеуплоидной (2n = 25). Для данной клеточной линии, вероятно, характерна трисомия по одной из пар субметацентрических хромосом. Длительно пролиферирующая КЛ, от которой получены жизнеспособные плодоносящие клоны, в течение 11 лет культивирования сохраняет диплоидное число хромосом и является генетически стабильной. Кариотип хвойных растений характеризуется высокой степенью стабильности и большая часть видов семейства Pinaceae, к которому относится род Larix Mill., содержит 24 хромосомы (2n = 24). В связи с этим обнаружение цитогенетически устойчивых клеточных линий с измененным хромосомным набором представляет большой интерес для генетики этой группы растений. Выявление стабильно диплоидных клеточных линий актуально для генетико-селекционных работ, направленных на создание высокопродуктивных плантаций с заданными наследственными свойствами.
The results of cytogenetic analysis of four embryogenic cell lines of Siberian larch (Larix sibirica Ledeb.) of different ages: young (two four-month and 16-month CLs) and long-proliferating (11-year CL) are presented. Three cell lines are diploid and contain in the karyotype the normal number of chromosomes for this species (2n = 24). A 16-month-old CL capable of forming somatic embryos was aneuploid (2n = 25). This cell line is probably characterized by trisomy on one of the pairs of submetacentric chromosomes. The long-term proliferating CL, from which regenerants and clones were obtained, retains a diploid number of chromosomes during 11 years of cultivation and remains genetically stable. The karyotype of conifers is characterized by high degree of stability and most of the species contain 24 chromosomes (2n = 24). Therefore, the detection of cytogenetically stable cell lines is of great interest for genetic work of this group of plants and creation of highly productive plantations with specified hereditary properties.

Аннотация: Обобщены сведения, полученные авторами при кариологическом изучении хвойных, произрастающих в дендрариях, парках, ботанических садах различных стран. В семействе сосновые (Pinaceae) изучены представители 7 видов и один межвидовой гибрид рода сосна ( Pinus L.) и 8 видов рода ель ( Picea A. Dietr . ), которые в диплоидном наборе имеют 24 хромосомы (2 n = 24), в семействе кипарисовые (Cupressaceae) - 5 интродуцированных видов родов туя ( Thuja L.), кипарис ( Cupressus L.), кипарисовик ( Chamaecyparis Spach) и секвойядендрон ( Sequoiadendron J. Buchholz), кариотип которых включает 22 хромосомы (2 n = 22). У видов, форм и культиваров хвойных из семейств сосновые и кипарисовые, находящихся в условиях интродукции, а также являющихся компонентами зеленых насаждений в населенных пунктах различных географических регионов, обнаружена большая изменчивость кариологических признаков. Отмечаются вариабельность числа хромосом (миксоплоидия), изменение морфологии хромосом, увеличение числа нуклеолярных локусов в хромосомах, появление добавочных хромосом, высокая встречаемость и широкий спектр хромосомных аномалий. Изменения, вероятно, вызваны акклиматизацией растений в новых условиях произрастания и адаптацией к повышенной рекреационной нагрузке в парковых насаждениях. Они могут вызывать повышение фенотипического и генетического разнообразия, что необходимо учитывать при интродукции и проведении мероприятий по селекции различных видов хвойных. На основе полученных данных могут быть решены задачи научного, учебно-просветительского, культурного и оздоровительного значения, разработаны рекомендации по отбору видов и внутривидовых таксонов хвойных, пригодных для озеленения населенных пунктов конкретного региона.
The information obtained by the authors during the karyological study on conifers growing in arboretums, parks, and botanical gardens from different countries is summarized. Representatives of two genera, pine ( Pinus L.) and spruce ( Picea A. Dietr . ), have been studied in the Pinaceae family; these species have 24 chromosomes in diploid complement (2 n = 24). Seven species and one interspecific hybrid of pine and eight species of spruce were analyzed. In Cupressaceae family, five introduced species were studied, which are representatives of the genera thuja ( Thuja L.), cypress ( Cupressus L.), false cypress ( Chamaecyparis Spach) and Sequoiadendron ( Sequoiadendron J. Buchholz); karyotype of Cupressaceae representatives includes 22 chromosomes (2 n = 22). In species, forms and cultivars of conifers from Pinaceae and Cupressaceae families, which are under introduction, as well as being components of green plantations in settlements of different geographical regions, great variability of karyological features. There area variability of chromosome numbers (mixoploidy), changes in the morphology of chromosomes, an increase of the number of nucleolar loci, appearance of supernumerary chromosomes, a high occurrence and a wide range of chromosomes anomalies. The changes are probably caused by the acclimatization of plants in new growing conditions and adaptation to increased recreational load in park planting. They can cause an increase in phenotypic and genetic diversity. It should be taken into account during introduction and carrying out activities on breeding of conifer species. On the basis of the data obtained, problems of scientific, experimental production, educational and cultural significance can be solved, recommendations for the selection of species and intraspecific taxa suitable for landscaping settlements in a particular region can be developed.
