Аннотация: Karyotypes of three Larix species (L. sibirica, L. gmelinii, and L. cajanderi) were analyzed using fluorescence in situ hybridization (FISH) with 45S and 5S ribosomal RNA gene probes and 4',6-diamidino-2-phenylindole (DAPI) staining. Two major 45S ribosomal DNA (rDNA) loci (per haploid genome) have been observed in the intercalary regions of two metacentric chromosomes, III and IV, of L. sibirica; in addition to them, minor nucleolus organizing regions (NORs) were mapped in pericentromeric regions of chromosomes I, II, VI, and XII. Two closely related species, L. gmelinii and L. cajanderi, showed similar hybridization patterns; both species possessed an additional major locus of 45S rDNA in the distal region of the long arm of submetacentric chromosome VII that is absent in L. sibirica. Only one locus of the 5S rDNA was found in all larch species we studied; it was located in the distal region of the chromosome III short arm, which also carried the major NOR in the opposite arm. This chromosome containing major loci of the two ribosomal RNA gene families can serve as a marker of the genus Larix. The intra- and interspecific karyotype diversity in the genus Larix is discussed.

Аннотация: The cytogenetic study of Abies sibirica seed progenies in declining fir stands of West Sayan High Mountains has been carried out. The increased amount of chromosome and genome mutations, pathologies of mitotic cycle and cells with micronuclei were found at studying. The revealed irregularities may be the result of a long-term exposure to extreme environmental factors and probably reflect the high degree of disturbance of the ecosystem in this region.

Аннотация: The karyological data on populations of Siberian fir (Abies sibirica Ledeb.) growing in lowland swamp and dry valleys are given. The diploid set of both populations included 24 chromosomes (2n = 24). Polykaryogram analysis revealed seven pairs of metacentric and five pairs of submetacentric chromosomes. The revealed differences between the populations included the absolute length of chromosomes, number of nucleolar organizer regions, and number of nucleoli. Changed chromosome numbers (mixoploidy and aneuploidy) as well as chromosomal aberrations were recorded. For the first time, mitosis was studied in this species and anaphase/telophase aberrations were revealed. The population of Siberian fir growing under extreme conditions of lowland swamp featured the widest range of mutations.

Siberian stone pine -- bog -- karyotype -- secondary constrictions -- chromosome and genome aberrations

Аннотация: The results of a karyological study on Siberian stone pines growing in the bog demonstrated differences from the populations of this species studied earlier with respect to sizes of chromosomes and location of secondary constrictions. The number of nucleolar organizer regions in the chromosomes of Siberian stone pine trees growing in the bog was larger than in other populations. A wide spectrum of chromosome aberrations was revealed, which had not been observed in this species before. These were genome and chromosome aberrations of various types, as well as structural aberrations accompanied by changes in the number of chromosomes. It is assumed that the aberrations in the karyotype of Siberian stone pine resulted from stressful conditions.

The role of polyploidy in adaptation and settling of steppe shrubs in Central Asia
/ N. V. Ekimova, E. N. Muratova, P. P. Silkin // Russian Journal of Genetics: Applied Research. - 2012. - Vol. 2, Is. 2. - P105-109, DOI 10.1134/S2079059712020037 . - ISSN 2079-0597
Аннотация: Chromosome numbers for some species of Central Asia steppe shrubs are given: Rhamnus erythroxylon Pall. (Rhamnaceae), Caragana buriatica Peschk. (Fabaceae), Amygdalus pedunculata Pall., Armeniaca sibirica (L.) Lam. (Rosaceae), Atraphaxis pungens (Bieb.) Jaub. et Spach, and A. frutescens (L.) C. Koch (Polygonaceae). Chromosome numbers of some species were determined for the first time. A comparative analysis of adaptive properties of polyploidy and diploid species has been conducted. It was established that natural polyploids are more adaptive to existence in extreme conditions. They possess a high potential for survival and are characterized by a higher level of intraspecific polymorphism, abundant flowering and fruitification, ability to propagate both via generative and vegetative means, and high seed germination. Аннотация: Data are presented on the karyotype structure of the Siberian fir (Abies sibirica Ledeb.) from five populations of the Central Siberia. The chromosome set (2n = 24) contains seven pairs of metacentric (I-VII), four pairs of submetacentric (VIII and X-XII), and one pair of intercentric (IX) chromosomes. The morphometric parameters of the identified chromosome groups had close values in the studied populations. The variation coefficients of chromosomal parameters of the Siberian fir correspond to very low and low variability. The intraspecific chromosome polymorphism of the Siberian fir is connected mainly to variations in the number and peculiarities of the nucleolar organizing regions of chromosomes. In the territory with technogenic loading, a wide spectrum of genomic mutations of the mixoploidy type was observed in the seed off-spring of the Siberian fir; triploid seedlings, as well as rare cases of somatic reduction of chromosomes, were revealed. Аннотация: Results of karyological study of the Siberian spruce from different provenances are presented. A comparative idiogramm for the Siberian spruce from Krasnoyarsk Territory and the Khakass Autonomous Region was constructed. In addition to the typical chromosome number (2n = 24), the Siberian spruce has one or two B-chromosomes (2n = 24 + 1B; 2n = 24 + 2B). Hereditary transmission of B-chromosomes and their behaviour in mitosis have been studied. It is supposed that B-chromosomes of the Siberian spruce have a nucleolar-organizing ability.

Аннотация: The authors studied karyotypes of common pine from three ecologically different habitats (raised bog, drained lowland bog, upland). It was established that the diploid set of all of the studied populations contains 24 chromosomes. When their karyotypes were compared, differences were noted in the total length of the chromosome set, and also in the number, localization, and frequency of occurrence of secondary constrictions and nucleoli. In the pine population on a raised bog, two pairs of chromosomes with four secondary constrictions were found for the first time. Chromosome mutations were discovered in the seed progeny of the investigated populations. The peculiarities that were revealed may be connected with different environmental conditions.

Radioecological monitoring of the Yenisei River and citological characterization of a submerged aquatic plant Elodea canadensis
/ A. I. Bolsunovskii [и др.] // Radiats Biol Radioecol. - 2007. - Vol. 47, Is. 1. - С. 63-73 . - ISSN 0869-8031
Аннотация: The study was devoted to investigation of the contents of radionuclides and of heavy metals and to evaluate the frequency of chromosomal aberrations in samples of Elodea canadensis, a submerged plant, collected in different parts of the Yenisei River. The samples were collected in the area subjected to radioactive impact of the Mining-and-Chemical Combine (MCC) at Zheleznogorsk and in the control area, upstream of the MCC. The investigations shown that elodea biomass in the area affected by MCC operation contained a long inventory of artificial radionuclides typical for the MCC discharges. The upstream of the MCC, in the control sampling area, the sediments and the elodea biomass contained only one artificial radionuclide--137Cs. Thus, the exposure doses to elodea shoots and roots upstream of the MCC are small (not more than 8 microGy/d) and the main contribution info the dose is made by natural radionuclides. At the MCC discharge site (the village of Atamanovo) and at the downstream of it, the total dose rate increases almost an order of magnitude, reaching its maximal values--72 microGy/d for elodea shoots and 58 microGy/d for its roots. Cytogenetic investigations of elodea roots shown that at the MCC discharge site (the village of Atamanovo) and at downstream of it the occurrence of chromosomal aberrations in ana-telophase and in metaphase cells of elodea was considerably higher than in the control area. It is highly probable that this simultaneous dramatic increase in the total exposure rate and the occurrence of chromosomal aberrations in elodea is associated with the radiation factor. It is suggested that elodea is affected not only by the radiation factor but also by the chemical factor--toxicity of heavy metals.

Аннотация: Investigations on B chromosomes found for the first time for Picea abies (L.) H. Karst. have been conducted. Seeds of Picea abies from two populations of Western Rhodopes (Bulgaria) located at the southern border of species range, and protected according to Bern Convention and EC Habitat Directive were collected for this study. Mixoploidy was detected in some germinating seeds of Picea abies. It was found that metaphase cells of germinating seeds contain 0-4 B chromosomes of both metacentric and submetacentric types. The variability of B chromosomes number and their occurrence was observed. Along with B chromosomes, some chromosome aberrations such as fragments and ring chromosomes were revealed in metaphase cells of Picea abies from studied populations. The possible adaptive role of B chromosomes presence for Picea spp. is discussed.

Аннотация: Cytogenetic studies on three species of submerged plants from different parts of the Yenisei River area subjected to radioactive impact of the Krasnoyarsk Mining-and-Chemical Plant and the Electrochemical Factory have been conducted. A high level of irregularities in ana-telophase and metaphase of mitoses has been revealed in test samples compared to the control: agglutination and fragmentation of chromosomes, lagging chromosomes, bridges, fragments, misdivisions, and others. The nature of the disorders indicates that they are related in part to the direct damage to the chromosome structure and in part to damage to the spindle.

Аннотация: We present the results of cytogenetic analysis of seven young and long-term cultured embryogenic cell lines (CLs) of Larix sibirica obtained by somatic embryogenesis. All CLs were found to be mixoploid. Three CLs (1, 2 and 6 years of cultivation) were the most stable cytogenetically with 91-96% cells having diploid chromosome number 2n=24, typical for the species. These CLs can be successfully used for the development of planting material and in clonal plantation forestry. At the same time, some embryogenic CLs were shown to possess various chromosome and genome mutations. We found two aneuploidy proliferating CLs (2n=25 and 28) capable of somatic embryo formation. One of the CLs analyzed in a current study was found to be cytogenetically unstable with chromosome numbers ranging from 24 to 35 and containing one-two micronuclei in the interphase cells. Fluorescent in situ hybridization (FISH) and morphometric analyses of karyotypes of these embryogenic CLs allow an assumption that the trisomy for one or several chromosomes is the most widespread type of genomic disturbances in embryogenic CLs of L. sibirica.

The main criteria of chromosomal polymorphism, namely the variability of the chromosome number, the level and spectrum of chromosomal rearrangements, were studied in natural populations and artificial plantations of Scots pine (Pinus sylvestris) growing in the south of its range in the steppe provenances of the Lower Volga and Southern Siberia (Volgograd Region, Republic of Tyva, Republic of Khakassia). The analysis of chromosomes was performed in the cells of the root meristems of seedlings at the stage of metaphase of mi-tosis. In the seed progeny of P. sylvestris trees from all the studied provenances, a disturbance of the chromosome number, myxoploidy, was detected, the level of its distribution increasing with increase of aridity and deterioration of edaphic growth conditions. In the seed progeny of P. sylvestris in the Volgograd Region and Khakassia, chromosome rearrangements represented by ring chromosomes, polycentric chromosomes, frag-ments, and multiple disturbances were found. A significant increase in the occurrence of chromosomal rearrangements and the expansion of their set is noted in the seedlings from an isolated provenance of P. sylvestris in rocky steppe of Khakassia. The frequency of mixoploidy and chromosomal rearrangements occurrence in the intraspecific forms of P. sylvestris trees is not the same, which is associated with differences in the conditions of their growth. The revealed cytological instability of seed progeny of trees in the plantations of P. syl-vestris growing in steppe ecotopes may indicate the activation of microevolutionary processes, apparently ac-companied by natural selection of resistant genotypes and forms adapted to arid conditions. © 2021, Komarov Botanical Institute.
