Аннотация: The paper focuses on investigating the densities of different ecological trophic groups of soil microorganisms, cellulose decomposition rates, and respiration of cryogenic forest soils in the north of Central Siberia and those of the southern taiga subzone, including those affected by surface wildfires. Psychrotolerant bacteria and micromycetes were found to dominate these soil complexes. The biological activity of the upper soil horizon in the north appeared to be close to that in the south, although the growing season in the former area is shorter compared to the latter.

Аннотация: The introduction of Trichoderma viride spores (10(8) CFU per 1 cm(2)) essentially changed the structure of micromycetes in the soils of tree nurseries in Krasnoyarsk region. During the first 20 days, in the variants with dark gray forest soils and podzolized chernozems, the total number of fungi decreased by 3-4 and 1.5 times, respectively, as compared to that in the control plots. During the intense development of the introduced microbes, the species composition of the soil fungi changed considerably. The treatment of Scots pine seeds with metabolites of Trichoderma fungi, as well as Pseudomonas and Bacillus bacteria, in the form of water suspensions, biopreparations, and dry spores promoted an increase in the yield of seedlings and improve their morphometric parameters. At the end of the growing period, the treatment with Trichoderma and the biopreparation on its basis increased these parameters, on average, by 18-70%, and the treatment with bacteria increased the same parameters by 13-15%. The application of microbial preparations improved the phytosanitary state of the soils in the studied tree nurseries. The use of the strains of indigenous microorganisms might be feasible for solving bioremediation problems more successfully in particular regions.

Аннотация: Soils in the tree nurseries studied were characterized by a lower species diversity of fungi than adjacent virgin soils. In particular, the relative abundances of representatives of the genera Mucor Chaetomium, and Trichoderma in the nursery soil were two times lower than in adjacent virgin soils. On the other hand, the nursery soil exhibited greater abundances of fungi of the genus Fusarium, which are causative agents of many diseases of conifer seedlings. To appreciate the efficiency of biocontrol of the infectious diseases of conifer seedlings, we introduced several indigenous Trichoderma strains into the nursery soil and found that this affected the species composition of soil microflora considerably. Changes in the species composition of mycobiota beneficially influenced the phytosanitary state of soils and reduced the infectious lodging of conifer seedlings.

Аннотация: The quantitative composition of various trophic groups of microorganisms and the enzyme activity of forest cryogenic soils in Central Evenkia was studied. The preferential development of complexes of psychrotolerant bacteria and micromycetes was found. During a short growing period, the biological activity of upper organic horizons in northern soils is comparable with that in southern taiga soils of Siberia. The microflora and biological activity of southern taiga soils in Central Siberia is understood rather completely. At the same time, information on the biogenic properties of forest soils in northern Siberian regions is scarce, though there are representative data on sylvicultural, soil, physiological, and biochemical studies of forest biogeocenoses.

Аннотация: Two types of bogs were studied in Tomsk oblast-Maloe Zhukovskoe (an eutrophic peat low-moor bog) and Ozernoe (an oligotrophic peat high-moor bog). The gram-negative forms of Proteobacteria were found to be dominant and amounted to more than 40% of the total population of the microorganisms investigated. In the peat bogs, the population and diversity of the hydrolytic microbial complex, especially of the number of micromycetes, were lower than those in the mineral soils. The changes in the quantitative indices of the total microbiological activity of the bogs were established. The microbial biomass and the intensity of its respiration differed and were also related to the depth of the sampling. In the Zhukovskoe peat low-moor bog, the maximal biomass of heterotrophic microorganisms (154 mu g of C/g of peat) was found in the aerobic zone at a depth of 0 to 10 cm. In the Ozernoe bog, the maximal biomass was determined in the zone of anaerobiosis at a depth of 300 cm (1947 mu g of C/g of peat). The molecular-genetic method was used for the determination of the spectrum of the methanogens. Seven unidentified dominant forms were revealed. The species diversity of the methanogens was higher in the oligotrophic high-moor bog than in the eutrophic low-moor bog.

    Agrochemical and microbiological properties of soil within a forest nursery in the south of krasnoyarsk region

Аннотация: Morphological, physical, and chemical characteristics of the studied soil are given; its type is classified as loamy podzolized chernozem. The investigations show that the main nutrients necessary for growth and development of seedlings of conifers are in balance. Microbiological processes in the soil of the nursery proceed intensively owing to the activity of different physiological groups of microorganisms that take part in destruction and synthesis of organic matter. Phytopathogenic micromycetes with a predominance of species from the Fusarium, Alternaria, and Verticillium genera are the main factor interfering with the production of high-quality seedlings of conifers. Аннотация: The rate of accumulation and location of proanthocyanidins and lignin was studied in fir (Abies sibirica Ledeb.) trunk phloem inoculated with two species of pathogenic micromycetes Verticillium sp. and Thysanophora sp., the usual associates of the sawyer Monochamus urussovi Fisch. Greater accumulation of the phenolic substances and more rapid localization of pathogen as a result of a hypersensitivity reaction were detected in firs infected with Verticillium sp. It was concluded that accumulation of proanthocyanidins and lignin is a part of nonspecific immune response of fir to the pathogen action and that the pathogen specific of fir can inhibit induced synthesis of defensive substances by the host plant.

Аннотация: It has been found that the total productivity of bacteria and micromycetes in the 0- to 50-cm layer of homogeneous cryozems (Cryosols) on slopes of northern and southern exposures varies from 1.2 to 1.4 t/ha, respectively, and the calculated content of microbial carbon varies in the range 0.7-0.9 t/ha. The respiratory activity of the upper soil layer is 2.5-2.6 mu g C-CO2/(g h); the potential methane formation capacity reaches 0.13 nmol CH4/(m(2) day) for soils on slopes of northern exposure and 0.16 nmol CH4/(m(2) day) for slopes of southern exposure. Accumulation of sorbed ammonium is recorded in the range 15-17 mg NH4/100 g soil in summer. The increase of temperature in the upper horizons of soils on slopes of southern exposure by 5A degrees C compared to the northern slopes results in only an insignificant increase in the emission of CO2 and CH4. The accumulation of sorbed ammonium and nitrate nitrogen in homogeneous cryozems during the vegetation period is comparable to that in gray forest soils of the southern taiga subzone of the Middle Siberia.

Аннотация: The microbiological method is applied for the purpose of artificial forest regeneration, as the most effective method of protecting forest planting material grown in forest nurseries. At present, literature data contain many examples of using species and genera of microorganisms belonging to different taxa in order to protect plants. The aim of the research was to establish biological (antagonistic, enzymatic and growth-stimulating) activity of the museum microorganism cultures belonging to different taxonomic groups (bacteria, fungi), and to assess their influence on the growth and development of Scots pine seeds in vitro and a decrease in the number of phytopathogenic fungi. We isolated previously selected microorganisms from the nursery soils; these microorganisms belong to different taxonomic groups, namely, Trichoderma micromycetes (T. harzianum, T. longibrachaitum, and T. lignorum), Streptomyces lateritius bacteria, Bacillus amyloliquefaciens, as well as phytopathogenic Fusarium fungi (F. oxysporum, F. moniliforme, F. proliferatum, F. moniliforme var annullatum, and F. oxysporum B3). Antagonistic activity of microbial strains was determined by the dual culture method, and the presence of enzymatic activity (lipase, proteinase and chitinase) of the tested strains was observed by qualitative express tests. We studied the growth-promoting activity by soaking pine seeds in aqueous suspensions of antagonists (106 spores/ml) (Pegalado, 2000; Cullimore, 2001; Montealegre, 2003; Asaturova, 2012). The results of the research showed that the investigated microorganisms (fungi, actinobacteria and bacteria) are biologically active. The most powerful antagonists were micromycetes of T. harzianum, T. lignorum, and T. longibrachiatum, which are also capable of exhibiting mycophilic properties (hyperparasitism). Thus, T. longibrachiatum showed mycophilia against three strains: F. moniliforme, F. moniliforme var annulatum, and F. oxysporum B3, whereas T. harzianum and T. lignorum did against two: F. moniliforme and F. proliferatum; the degree of phytopathogen inhibition (ID) varied from 30 to 100% (See Table 1). The strain of B. amiloliquefaciens bacterium was less active, the DI was 41.4%, on the average, and the slowest antagonistic properties were exhibited by actinobacterium S. lateritius - 14.8%, on the average. The investigation of the presence of the main hydrolytic enzymes (a hitinaze, a lipase, protease) showed that Trichoderma micromycetes had the average and strong hydrolytic activity (T. harzianum and T. longibrachiatum), and bacteria (S. lateritius, B. amyloliquefaciens) had the average and weak hydrolytic activity (See Table 2). Also, all the investigated strains improved Scots pine seed germination, while the strains of B. amyloliquefaciens and T. longibrachiatum showed the greatest growth-promoting activity (See Figures). Thus, we found that the investigated strains (T. harzianum, T. lignorum, T. longibrachiatum, S. lateritius, and B. amyloliquefaciens) had a high antagonistic activity, and Trichoderma micromycetes revealed the ability for mycoparasitism. The high biological (enzymatic, antagonistic, growth-stimulating) activity of the studied strains of microorganisms makes them effective agents for biological control in forest nurseries. © 2018 Tomsk State University. Аннотация: In vitro cultivation of embryogenic cultures of Siberian larch on AI medium supplemented with different concentrations of antimicrobial peptides of two Trichoderma species, T.citrinoviride (strain TYVI 4/11) and T. viride (strain 346), has been performed to achieve a direct antimicrobial effect and initiate the mechanisms of induced resistance (regulatory function of embryogenic cultures) as well as to study the morphogenesis and growth activity of regenerant plants. The experiment was arranged using four cell lines, CL4, CL5, CL6, and CL12, differing in their embryogenic activity. The effect of treatment of cell cultures with Trichoderma peptides was manifested via significant growth stimulation of proliferating embryogenic cell lines, root growth stimulation, and formation of callus excrescences on regenerant plants. No difference between the treated and untreated variants was observed in seedlings. Conceivably, such peptide treatment may provide an increased immunity of soil-grown seedlings in forest nurseries. Study of the effect of biocontrol strains' use in a plantation reforestation to stimulate the growth and development of calluses and embryos and to obtain disease-resistant regenerant plants of conifer species will provide the development of a clonal silviculture, a new trend that recently appeared abroad (Park 2002, 2014).

    Влияние обработки семян сосны обыкновенной микробными и фитопрепаратами на сохранность сеянцев и свойства почвы в лесном питомнике

Аннотация: Исследования проводили на территории опытного питомника экспериментально-опытного хозяйства “Погорельский бор” Института леса им. В.Н. Сукачева СО РАН. В модельных полевых экспериментах изучали влияние предпосевной обработки семян сосны обыкновенной (<i>Pinus sylvestris</i> L.) водными суспензиями аборигенных микроорганизмов, обладающих антагонистической активностью к фитопатогенам, и фитопрепаратами, полученными из растительного сырья, на микробную биомассу, ферментативную активность почвы и сохранность сеянцев хвойных. Обработку семян сосны проводили штаммами микромицетов <i>Trichoderma harzianum</i>, <i>T. longibrachiatum</i>, <i>T. lignorum</i>, их смесью; бактериями <i>Bacillus amyloliquefaciens</i>, смесью бактерий (<i>B. amyloliquefaciens</i>, <i>B.</i> <i>subtilis</i>, <i>Pseudomonas</i> sp.) и фитопрепаратами (пять вариантов). Показано, что предпосевная обработка семян сосны способствовала улучшению их всхожести, сохранности сеянцев к концу сезона вегетации, морфометрических параметров сеянцев по сравнению с контролем в среднем на 25%. Интродуцированные с семенами популяции бактерий и микромицетов повышали продуктивность, содержание микробной биомассы и ферментативную активность почвы лесного питомника в 1.3–2.0 раза. Установлено, что в контрольной почве на протяжении всего периода вегетации преобладала олиготрофная группа микроорганизмов, а при обработке семян сосны (микроорганизмы и фитопрепараты) уже через два месяца после посева семян доминировала гидролитико-копиотрофная группа. Внесение микробов-антагонистов (<i>B. amyloliquefaciens</i>, <i>T. longibrachiatum</i>, Смесь бактерий) и фитопрепаратов также благоприятно влияло на функционирование почвенного микробного сообщества, что проиллюстрировано уменьшением значений микробного метаболического коэффициента в среднем почти в 2 раза по сравнению с контролем. Результаты исследования можно рекомендовать для использования в практике лесного хозяйства при выращивании сеянцев хвойных.
The studies were carried out on the experimental farm “Pogorelsky bor” of the Sukachеv Institute of Forest, SB RAS. The effect of pre-sowing treatment of Scots pine seeds (<i>Pinus sylvestris</i> L.) by aqueous suspensions of indigenous microorganisms with antagonistic activity towards phytopathogens and phytopreparations obtained from plant materials on soil biogenicity (microbial biomass and enzymatic activity) and safety of coniferous seedlings was studied in model field experiments. Strains of micromycetes <i>Trichoderma harzianum</i>, <i>Trichoderma longibrachiatum</i>, <i>Trichoderma lignorum</i> and their mixture (<i>T. harzianum, T. longibrachiatum, T. lignorum</i>); Bacteria <i>Bacillus amyloliquefaciens</i>, their mixture (<i>B. amyloliquefaciens</i>, <i>B. subtilis</i>, <i>Pseudomonas</i> sp.), five phytopreparations were variants of Scots pine seed treatments. It was shown that the pre-sowing treatment of Scots pine seeds contributed to the improvement of their germination, preservation of seedlings by the end of the vegetation season, and improved morphometric parameters of the seedlings compared with the control by 24.5% on the average. The bacteria and micromycetes introduced with the seeds increased the productivity and biogenicity of the forest nursery soil by 1.3–2.0 times. It was established that the oligotrophic group of microorganisms prevailed in the control soil throughout the vegetation season, while the hydrolytic-copyotrophic group of microorganisms prevailed after the treatment of Scots pine seeds with microorganisms and phytopreparations two months after sowing seeds. The introduction of antagonist microbes (<i>B.amyloliquefaciens</i>, <i>T. longibrachiatum</i>, and a mixture of bacteria) and phytopreparations also favorably affected the restoration of the ecophysiological norm of functioning of the soil microbial community (returning the value of the microbial metabolic coefficient to the background ones). A 2-fold decrease in the values of the microbial metabolic coefficient (<i>q</i>CO<sub>2</sub>) was observed in the soil of the studied areas. The obtained results can be recommended for using in forestry practice when growing coniferous seedlings.
