    The difference in the lignification of earlywood and latewood in larch (Larix sibirica Ldb.)

Аннотация: The rate of lignin deposition, its content and composition, and molecular weight distribution of different stages of secondary wall thickening during tracheid development of both earlywood and latewood in Siberian larch (Larix sibirica Ldb.) were studied in the stems of 25-year-old trees. The cells of early and late xylem at different stages of secondary wall development were obtaint in late June and early in August, respectively. Lignification of the two types of wood was found to involve different dynamics. The intensity of lignin synthesis during earlywood formation increases gradually, reaching the maximum at the last stage of tracheid maturation. In contrast, lignin deposition in the course of latewood development is the highest only in the first stage of lignification and declines by the end of tracheid maturation. There were differences in the composition of alkaline oxidation products of lignin preparations at different development stages of early and late xylem. The amount of cell wall substances deposited before lignification in earlywood is larger than that in latewood.

Аннотация: Karyotypes of three Larix species (L. sibirica, L. gmelinii, and L. cajanderi) were analyzed using fluorescence in situ hybridization (FISH) with 45S and 5S ribosomal RNA gene probes and 4',6-diamidino-2-phenylindole (DAPI) staining. Two major 45S ribosomal DNA (rDNA) loci (per haploid genome) have been observed in the intercalary regions of two metacentric chromosomes, III and IV, of L. sibirica; in addition to them, minor nucleolus organizing regions (NORs) were mapped in pericentromeric regions of chromosomes I, II, VI, and XII. Two closely related species, L. gmelinii and L. cajanderi, showed similar hybridization patterns; both species possessed an additional major locus of 45S rDNA in the distal region of the long arm of submetacentric chromosome VII that is absent in L. sibirica. Only one locus of the 5S rDNA was found in all larch species we studied; it was located in the distal region of the chromosome III short arm, which also carried the major NOR in the opposite arm. This chromosome containing major loci of the two ribosomal RNA gene families can serve as a marker of the genus Larix. The intra- and interspecific karyotype diversity in the genus Larix is discussed.

Аннотация: Using AFLP analysis, the genetic variation in four population samples of Rhododendron ledebourii, three such samples of Rh. dauricum, two samples of Rh. sichotense, and one sample of Rh. mucronulatum was examined. Application of three selective primer pairs provided genotyping of 87 accessions from the four species at 213 loci. An analysis of molecular variance revealed a high level of differences between all four species (F (ST) = 0.172). Populations within the species were also differentiated: for Rh. dauricum F (ST) = 0.245; Rh. ledebourii F (ST) = 0.264; Rh. sichotense F (ST) = 0.034. The highest Nei distance was observed for Rh. mucronulatum with Rh. dauricum and Rh. ledebourii (D = 0.057 and 0.069, respectively). Similarity was observed between the species pairs of Rh. ledebourii-Rh. dauricum (0.032) and Rh. sichotense-Rh. mucronulatum (0.034). The Far Eastern species Rh. sichotense and Rh. mucronulatum were characterized by an elevated level of intrapopulation variation. The data from the AFLP-based analysis of differentiation and variation that were thus obtained were compared with the literature data on the morphological criteria that differentiate the species of interest and those regarding the history of the formation of their ranges. The independence of all the species we examined was confirmed.

Аннотация: Ophiostomatoid fungi colonize the conducting tissues of conifer stems, the phloem and the xylem. These pathogenic fungi penetrate into the stem through injuries made by xylophagous insects vectoring these pathogens. In this study the response of the phloem of Scotch pine (Pinus sylvestris L.) to wounding (treatment 1) was compared with the response to wounding combined with application of high-molecular-weight compounds isolated from the mycelium of the ophiostomatoid fungus Ceratocystis laricicola Redfern & Minter (treatment 2). Both treatments induced the appearance of necrosis in the inner bark, the formation of periderm separating living and dead tissues, and formation of the callus alongside the wound perimeter. In addition, the bark accumulated lignin, bound proanthocyanidins, and resins, with a parallel decrease in the content of free proanthocyanidins, low-molecular-weight carbohydrates, and non-lignin components of the cell wall (P > 0.95). The size of necrotic spots, as well as changes in the content of most substances, were significantly higher in the treatment 2 than in the treatment 1 (P > 0.95). The accumulation of lignin in cell walls of phloem sieve cells was delayed in the treatment 2 as compared with that in the treatment 1. This suggested that the mycelial extract temporarily inhibited lignification at the early stage of the wound response. This disturbance of the cell wall protective transformation led to the hypothesis that the fungal suppressors retard the repair of inner bark injured by insects, thereby favoring the invasion of conifer tissues by ophiostomatoid fungi.

Аннотация: The seasonal development of phloem in the stems of Siberian larch (Larix sibirica Ldb.) was studied over two seasons on 50-60-year-old trees growing in a natural stand in the Siberian forest-steppe zone. Trees at the age of 20-25 years were used to study metabolites in differentiating and mature phloem elements, cambial zone, and radially growing xylem cells in the periods of early and late wood formation. The development of the current-year phloem in the stems of 50-60-year-old trees started, depending on climatic conditions, in the second-third decades of May, 10-20 days before the xylem formation, and ended together with the shoot growth cessation in late July. Monitoring of the seasonal activity of cambium producing phloem sieve cells and the duration of their differentiation compared to the xylem derivatives in the cambium demonstrated that the top production of phloem and xylem cells could coincide or not coincide during the season, while their differentiation activity was always in antiphase. Sieve cells in the early phloem are separated from those in the late phloem by a layer of tannin-containing cells, which are formed in the period when late xylem formation starts. The starch content in the structural elements of phloem depends on the state of annual xylem layer development. The content of low molecular weight carbohydrates, amino acids, organic acids, and phenols in phloem cells, cambial zone, and xylem derivatives of the cambium depends on the cell type and developmental stage as well as on the type of forming wood (early or late) differing by the cell wall parameters and, hence, by the requirement for assimilates. Significant differences in the dynamics of substances per dry weight and cell were observed during cell development.

Аннотация: The swelling of larch wood in 1-butanol, dimethyl sulfoxide, N,N-dimethyl formamide, ethylene glycol, ethanol, methyl cellosolve, 1-propanol and toluene was investigated at room temperature. The volumetric, tangential and radial swelling completion was lagging behind the liquid absorption process. The kinetic indices of swelling of larch wood were determined based on the equation of Kolmogorov-Erofeev. The volumetric swelling was related to molecular mass, molar volume, dielectric permittivity and integral heat of wood wetting. The anisotropy of wood swelling was characterized by the ratio of equilibrium tangential swelling to radial swelling (alpha(t)/alpha(r)). It was demonstrated that alpha(t)/alpha(r) is related to the dipole component of Hansen's parameter and cannot exceed the value 3.

Аннотация: In this article, we report observations made during thirteen years on foliar fungal pathogens attacking European and Eurasian woody broadleaved species in Siberian arboreta and cities and discuss the possibility of using such data for detecting exotic pathogens that may represent a danger for European tree and shrub species, should these pathogens be introduced into Europe. A total of 102 cases of symptomatic infections (fungus-host plant associations) involving 67 fungal species were recorded on 50 of the 52 European and Eurasian woody plant species. All but four of the fungi found during the surveys were previously reported in Europe. However, 29 fungus-host plant associations are apparently new to science, suggesting that complexes of cryptic species differing in their host range and geographic range may occur. Seventeen percentage of associations were given a high damage score, that is, more than 50% of plant area was attacked, for at least some localities. In nearly half of the cases, fungus-host plant associations were found to be very frequent, that is, occurring every year and at all locations where the plant was inspected. A list of pathogen-host associations in Siberia deserving further investigation is provided, either because the pathogen is not yet recorded in Europe or because the pathogen-host association has not yet been reported, and the damage is high or, finally, because the damage and infestation level is unusually high in known associations. Further studies should involve molecular characterization of these foliar pathogens and their host range testing.

Аннотация: Leaf quality of the mountain birch (Betula pubescens ssp. tortuosa) for herbivores was studied at several hierarchical levels: among trees, among ramets within trees, among branches within ramets, and among short shoots within branches. The experimental units at each level were chosen randomly. The indices of leaf quality were the growth rate of the larvae of a geometrid, Epirrita autumnata, and certain biochemical traits of the leaves (total phenolics and individual phenolic compounds, total carbohydrates and individual sugars, free and protein-bound amino acids). We also discuss relationships between larval growth rate and biochemical foliage traits. Larval growth rates during two successive years correlated positively at the level of tree, the ramet, and the branch, indicating that the relationships in leaf quality remained constant between seasons both among and within trees. The distribution of variation at different hierarchical levels depended on the trait in question. In the case of larval growth rate, ramets and short shoots accounted for most of the explained variation. In the case of biochemical compounds, trees accounted for most of the variance in the content of total phenolics and individual low-molecular-weight phenolics. In the content of carbohydrates (total carbohydrates, starch, fructose, glucose, and sucrose) and amino acids, variation among branches was generally larger than variation among trees. Variation among ramets was low for most compounds. No single leaf trait played a paramount role in larval growth. Secondary compounds, represented by phenolic compounds, or primary metabolites, particularly sugars, may both be important in determining the suitability of birch leaves for larvae. If phenols are causally more important, genet-specific analyses of foliage chemistry are needed. If sugars are of primary importance, within-genet sampling and analysis of foliage chemistry are necessary.

Аннотация: The following major phenolics with non-flavonoid structure were identified from leaves of Betula pubescens (white birch) and Betula pendula (silver birch): 1-O-galloyl-beta-d-(2-O-acetyl)-glucopyranose, 1-(4 ''-hydroxyphenyl)-3'-oxopropyl-beta-d-glucopyranose, gallic, chlorogenic, neo-chlorogenic, cis- and trans-forms of 3- and 5-p-coumaroylquinic acids. Chromatographic (analytical and preparative HPLC), chemical (hydrolysis) and spectroscopic (UV,H-1 and C-13 NMR, MS) techniques were applied for separation, isolation, purification and identification of these phenolics. Moreover, 33 low-molecular-mass phenolics were detected and quantitated and their occurrence was compared in leaves of white and silver birches.

Аннотация: Two types of bogs were studied in Tomsk oblast-Maloe Zhukovskoe (an eutrophic peat low-moor bog) and Ozernoe (an oligotrophic peat high-moor bog). The gram-negative forms of Proteobacteria were found to be dominant and amounted to more than 40% of the total population of the microorganisms investigated. In the peat bogs, the population and diversity of the hydrolytic microbial complex, especially of the number of micromycetes, were lower than those in the mineral soils. The changes in the quantitative indices of the total microbiological activity of the bogs were established. The microbial biomass and the intensity of its respiration differed and were also related to the depth of the sampling. In the Zhukovskoe peat low-moor bog, the maximal biomass of heterotrophic microorganisms (154 mu g of C/g of peat) was found in the aerobic zone at a depth of 0 to 10 cm. In the Ozernoe bog, the maximal biomass was determined in the zone of anaerobiosis at a depth of 300 cm (1947 mu g of C/g of peat). The molecular-genetic method was used for the determination of the spectrum of the methanogens. Seven unidentified dominant forms were revealed. The species diversity of the methanogens was higher in the oligotrophic high-moor bog than in the eutrophic low-moor bog.

Аннотация: The formation of phloem was studied for two years in stems of 50 to 60 year old trees of Scots pine (Pinus sylvestris L.) growing in nature. The sieve cells of early phloem were separated from those of late phloem by a layer of tannin-containing cells, which are formed simultaneously with the formation of late xylem cells by the cambium. Seasonal dynamics of accumulation of starch grain in structural elements of the phloem is related to the xylem development. The content of metabolites in differentiating and mature phloem elements, in the cambium zone, and in the xylem cells growing in the radial direction depended on cell specificity, stage of their development, and type of forming wood, early or late, which differ in the cell wall parameters and, hence, requirement of assimilates. Significant differences were described between the content of low molecular weigh carbohydrates, amino acids, organic acids, and phenol compounds using two methods of calculation: per dry weight and per cell. Аннотация: The fraction of incomplete degradation ?1 of the hydrogen bonds net in the matrix of wood substance by the action of dimethyl sulfoxide, N,N-dimethyl formamide, ethylene glycol, methyl cellosolve, tertiary butyl alcohol and propyl alcohol was studied. A relationship between ?1 and a nondimensional parameter which is a combination of the molecular packing coefficient, specific heat, temperature, hydrogen-bonding contribution to the three-dimensional solubility parameter and surface tension for the given solvent was found.

Аннотация: Aluminum, Sc, Th, Hf, and lanthanoids belong to groups of elements that are, slightly, very slightly, and moderately captured in marsh ecosystems. In the genesis of the peatbog the biogeochemical migration of elements is mainly determined by the quality of the humus barriers as well as by the capability to form intermetallic compounds of widely varying compositions. In the surface layer (10 cm) of the peatbog the highest content of Al, Hf, Ce, and Sm over the entire history of marsh development was observed. This fact reflects the modem tendency to forming technogenic streams of the above content.

Аннотация: We conducted a comparative study on the composition of water soluble carbohydrate-protein complexes of primary cell walls and cytoplasm in xylem cells of Siberian larch (Larix sibirica Ldb.). Preparations were obtained in the process of isolating protoplasts from xylem cells at the stage of elongation. Water-soluble high-molecular-weight compounds were fractionated on DEAE cellulose. It was found that the dominant fraction in composition of these compounds consists of a carbohydrate-protein complex. Comparison of the composition of neutral sugars, uronic acids, and amino acids of these complexes showed that water-soluble arabinogalactan proteins were absent in the cytoplasm, but present in the solution after plasmolysis. They were bound with pectins and arabinoxylans, detected in solutions obtained after isolation and purification of protoplasts, and capable of forming a gel at low water content and low temperature. It is hypothesized that arabinogalactan proteins are components of primary cell walls, that they are located for the most part on the surface, and that their solubility in an aqueous medium depends upon binding with pectins of the middle plate and with other cell wall components.

    Lignin deposition during earlywood and latewood formation in Scots pine stems

Аннотация: Lignin deposition at consecutive secondary wall thickening stages of early and late xylem cells during annual ring wood formation in Scots pine (Pinus sylvestris L.) stems was studied. Lignin patterns, isolated by thioglycolic acid method, consisted of alcohol-soluble (LTGA-I) and alkali-soluble (LTGA-II) fractions. The sum of two fractions, being the total lignin content, gradually increased in the course of lignification. However, the increments of lignin amount at each development stage of early and late tracheids were different. The intensity of lignin deposition increased in the course of earlywood tracheid maturation and decreased toward the end of latewood cell differentiation. The deposition of two lignin fractions in each layer of forming wood also occurred oppositely. The increment of LTGA-I descended, whereas that of LTGA-II increased from the beginning to the end of early xylem lignification. In contrast, LTGA-I increment dropped, whereas LTGA-II rose during late xylem lignification. Gel permeation chromatography showed that the lignins, formed at the beginning of lignification, were more homogeneous and had higher molecular weight compared with the lignins at the end of cell differentiation. Besides, the content of cellulose, estimated as the residue after lignin isolation, and of cell wall substances, presented as cell wall cross-section areas, at consecutive maturation stages of early and late xylem cells have been found to be different. The data show that lignin deposition occurred in different conditions and with opposite dynamics during early and late xylem formation. © 2014 Springer-Verlag Berlin Heidelberg.

Аннотация: A genetic library is being currently developed for leafmining insects, attacking native and alien woody plants in Siberia, that may allow a quick and reliable identification of the immature stages of potential pests and invaders. Integration of molecular data with ecological characteristics of leafminers, particularly larval and pupal morphology, and diagnostic characteristics of mines will accelerate the detection and tracking of insect pests.

Аннотация: The objective of this study was to obtain a genetically stable haploid in vitro-derived line from Siberian larch (Larix sibirica Ledeb.) using megagametophyte explants, which then could be used for different molecular genetic studies, including whole genome de novo sequencing. However, cytogenetic analysis and genotyping of 11 microsatellite loci showed high levels of genomic instability and a high frequency of mutation in the obtained megagametophyte-derived callus cultures. All cultures contained new mutations in one or more microsatellite loci.

Аннотация: Europe has one of the best-known Lepidopteran faunas in the world, yet many species are still being discovered, especially in groups of small moths. Here we describe a new gracillariid species from the southeastern Alps, Callisto basistrigella Huemer, Deutsch & Triberti, sp. n. It shows differences from its sister species C. coffeella in morphology, the barcode region of the cytochrome c oxidase I gene and the nuclear gene histone H3. Both C. basistrigella and C. coffeella can co-occur in sympatry without evidence of admixture. Two C. basistrigella specimens show evidence of introgression. We highlight the importance of an integrative approach to delimit species, combining morphological and ecological data with mitochondrial and nuclear sequence data. Furthermore, in connection with this study, Ornix blandella Muller - Rutz, 1920, syn. n. is synonymized with C. coffeella (Zetterstedt, 1839).

Аннотация: While phylogeographic patterns of organisms are often interpreted through past environmental disturbances, mediated by climate changes, and geographic barriers, they may also be strongly influenced by species-specific traits. To investigate the impact of such traits, we focused on two Eurasian spruce bark beetles that share a similar geographic distribution, but differ in their ecology and reproduction. Ips typographus is an aggressive tree-killing species characterized by strong dispersal, whereas Dendroctonus micans is a discrete inbreeding species (sib mating is the rule), parasite of living trees and a poor disperser. We compared genetic variation between the two species over both beetles' entire range in Eurasia with five independent gene fragments, to evaluate whether their intrinsic differences could have an influence over their phylogeographic patterns. We highlighted widely divergent patterns of genetic variation for the two species and argue that the difference is indeed largely compatible with their contrasting dispersal strategies and modes of reproduction. In addition, genetic structure in I.typographus divides European populations in a northern and a southern group, as was previously observed for its host plant, and suggests past allopatric divergence. A long divergence time was estimated between East Asian and other populations of both species, indicating their long-standing presence in Eurasia, prior to the last glacial maximum. Finally, the strong population structure observed in D. micans for the mitochondrial locus provides insights into the recent colonization history of this species, from its native European range to regions where it was recently introduced.
