Аннотация: Ophiostomatoid fungi colonize the conducting tissues of conifer stems, the phloem and the xylem. These pathogenic fungi penetrate into the stem through injuries made by xylophagous insects vectoring these pathogens. In this study the response of the phloem of Scotch pine (Pinus sylvestris L.) to wounding (treatment 1) was compared with the response to wounding combined with application of high-molecular-weight compounds isolated from the mycelium of the ophiostomatoid fungus Ceratocystis laricicola Redfern & Minter (treatment 2). Both treatments induced the appearance of necrosis in the inner bark, the formation of periderm separating living and dead tissues, and formation of the callus alongside the wound perimeter. In addition, the bark accumulated lignin, bound proanthocyanidins, and resins, with a parallel decrease in the content of free proanthocyanidins, low-molecular-weight carbohydrates, and non-lignin components of the cell wall (P > 0.95). The size of necrotic spots, as well as changes in the content of most substances, were significantly higher in the treatment 2 than in the treatment 1 (P > 0.95). The accumulation of lignin in cell walls of phloem sieve cells was delayed in the treatment 2 as compared with that in the treatment 1. This suggested that the mycelial extract temporarily inhibited lignification at the early stage of the wound response. This disturbance of the cell wall protective transformation led to the hypothesis that the fungal suppressors retard the repair of inner bark injured by insects, thereby favoring the invasion of conifer tissues by ophiostomatoid fungi.

Аннотация: The elicitor activity of compounds extracted from the mycelia of six species of phytopathogenic fungi was assessed from the sizes of necrotic lesions on the external surface of the living trunk phloem of five coniferous species inhabiting Siberia: Siberian larch (Larix sibirica L.), Scotch pine (Pinus sylvestris L.), Siberian spruce (Picea obovata Ledeb.), Siberian fir (Abies sibirica L.), and cedar pine (Pinus sibirica (Rupr.) Mayr.). The compounds for inoculation were extracted from the mycelium of ascomycetes imperfect, and basidium fungi; the living mycelia of these fungi were also used. The fungal extract or mycelium was placed into the hollows 7 mm in diameter in the trunk bark. Infection triggered the formation of hypersensitivity necrotic lesions in the inner bark exceeding in size those appeared after control wounding of four tree species (larch, pine, spruce, and cedar); fir was an exclusion. In experiments with tree trunks and conifer calluses, a dependence of immune response parameters (the sizes of necrotic lesions and the content of lignin and bound proanthocyanidins) on the quantity of the fungal preparation was elucidated. The largest necrotic lesions appeared after injection of 500 mu g of the fungal preparation into the hollow in the trunk, and its higher quantities did not increase the indices measured. The size of the necrotic lesion on the trunk bark is supposed to be used as a promising index characterizing the level of tree immunity and tolerance under various ecological conditions.

Аннотация: The contents of ascorbic acid (AA) and its oxidized form, dehydroascorbic acid (DHA), were assessed as related to the tracheid differentiation in the course of early and late wood development in the Siberianlarch (Larix sibirica Ldb.) trees. The samples of the cambium, cell enlargement zone and mature cells were collected at the successive developmental stages by scraping tissues off layer by layer from trunk segments of the 20-year-old trees according to anatomical and histochemical criteria. While cambium initials were rapidly dividing, the AA contents per dry weight and per cell considerably exceeded the corresponding values characteristic of the late xylem development; such difference corresponded to the higher number of early tracheids per annual ring, as compared to the late tracheids. The AA content decreased as cells enlarged. The radial growth of the early wood tracheids, as compared to the late wood tracheids, was accompanied with a threefold increase in the AA and a decline in the DHA contents. The AA/DHA ratio was in line with the early tracheid enlargement. The maximum AA content was observed at the early stage of the secondary cell wall thickening in the tracheids of early and late xylem preceding lignification. During this stage of early wood development, the DHA content exceeded sixfold the corresponding value in the late xylem; as a result, the initial rates of lignification were different in two tissues. The rate of lignification in a newly developing layer of the early xylem increased gradually and was the highest in the completely differentiated tracheids. In the late xylem, the lignification rate was at its highest at the very beginning and then declined in the course of tracheid maturation. The dissimilar patterns of lignification in the early and late xylem were primarily associated with the DHA content, which increased in the early xylem and decreased in the maturing late xylem. Thus, the AA content and its accessibility to oxidation in the growing and mature xylem cells exhibited the diverse developmental patterns in the early and late xylem: two tissues differed in the tracheid number and radial diameter as well as in the rate of lignification.

Аннотация: Morphological and cytoembryological studies of unique forms of Siberian pine (Pinus sibirica Du Tour) with the annual cycle of ovulate cone development were carried out. The morphogenesis of female shoots in these anomalous trees with the annual cycle of development was shown to provide for an accelerated development and very rapid growth of megastrobili and accelerated formation of reproductive structures. Cytoembryological studies of ovules in these trees showed significant shortening of the free-nuclear stage of gametophyte development (up to 3 weeks instead of one year) and ultra-early formation of archegonia and egg cells. However, no fertilization of egg cells and the development of embryos occur in the anomalous forms; the egg-cell nucleus divides in the haploid state. Developing seeds are formed without embryo. The author hypothesizes that the acceleration of embryo development in the anomalous pine trees is caused by enhanced hormonal, carbohydrate, and nitrogen metabolism in ovule tissues.

Аннотация: Pine (Pinus sylvestris L.) callus culture was treated with the mycelium extracts from six Fusarium strains. Previously, pine seedlings were infected with a spore suspension in order to test the pathogenicity of the used strains. Callus culture infection resulted in a decrease in the free proanthocyanidin (PA) and an increase in bound PA content. After treating the calli with all strains except F. oxysporum var. orthoceras, the lignin content became lower than the control one. The most considerable changes involved the p-hydroxybenzoic acid (HBA) content, and its greatest change was observed after treating the calli with F. nivale, when the HBA concentration (1229 mug/g, dry wt) exceeded fourfold the control one. There was a positive correlation (R = 0.81) between the HBA content in the callus culture cells treated with a fungal extract and the virulence of Fusarium strains. At the same time, there was an inverse correlation (R = -0.80) between the lignin content in a callus culture and the fungal virulence; the latter did not affect the contents of both free and bound PA.

Аннотация: Twelve-day-old seedlings of Scots pine (Pinus sylvestris L.) were infected with a virulent spore suspension of the fungus Fusarium sporotrichiella var. sporotrichioides Bilai. 72 h after infection, the seedlings were divided into infection-susceptible and infection-resistant ones, and phenolic compounds in the seedling hypocotyls were analyzed. Susceptible seedlings contained 4.3-fold less lignins, 1.4-fold less free and bound proanthocyanidins, 12.7-fold less free hydroxybenzoic acids, 56.6-fold less bound hydroxybenzoic acids, 2-fold less quinic acid, and 5.1-fold less shikimic acid as compared to the resistant seedlings. At the same time, L-phenylalanine content was increased 3-fold and that of L-tyrosine, 1.3-fold. The activity of quinate:NAD(P)(+) oxidoreductase and L-phenylalanine ammonia-lyase in the hypocotyls of susceptible seedlings was 2.3-fold and 1.4-fold lower, respectively, whereas the activity of shikimate:NAD(P)(+) oxidoreductase remained unchanged.

Аннотация: We compared effects of ambient (360 vpm) and elevated (720 vpm) carbon dioxide concentration ([CO(2)]) and high and low nutrient supply rates on stem growth, annual ring structure and tracheid anatomy of Siberian larch (Larix sibirica Ledeb.) seedlings over two growing seasons. Elevated [CO(2)] had no significant effect on either stem height or diameter growth; however, both stem height and diameter growth were enhanced by the high nutrient supply rate, and these increases were stimulated by elevated [CO(2)]. Elevated [CO(2)] tended to increase the width of the annual xylem ring, the number of cells in a radial file spanning the ring, and tracheid lumen diameter, whereas it tended to reduce cell wall thickness, although there were no statistically significant CO(2) effects on tracheid anatomy. Changes in tracheid cell morphology seemed to be dependent on changes in shoot elongation rates.

Аннотация: Callus cultures of Siberian larch (Lar ix sibirica Ledeb.) and Siberian spruce (Picea obovata Ledeb.) were used to demonstrate the elicitor activity of two ophiostomatoid fungal species, Ceratocystis laricicola and Ceratocystis polonica, as the pioneer settlers on larch and spruce, respectively. The extract from C. laricicola mycelium stimulated the accumulation of lignin in larch cells by 37% and that of hound proanthocyanidins by 25%. In spruce callus cultures, C. laricicola and C. polonica increased the bound PA content by 25 and 46%, respectively. In the callus cultures of larch and spruce, the addition of extract of C. laricicola increased the concentration of p-hydroxybenzoic acid 13-fold and nearly 4-fold, respectively. The metabolic characteristics of accumulation of phenolic compounds in conifer cells are discussed.

Аннотация: We assessed above- and belowground biomass and net primary production (NPP) of a mature Larix gmelinii (Rupr.) Rupr. forest (240-280 years old) established on permafrost soils in central Siberia. Specifically, we investigated annual carbon budgets in roots in relation to root system development and availability of soil resources. Total stand biomass estimated by allometry was about 39 Mg ha(-1). Root biomass (17 Mg ha(-1)) comprised about 43% of total biomass. Coarse root (greater than or equal to 5 mm in diameter) biomass was about twice that of fine roots (< 5 mm). The aboveground biomass/root biomass ratio (T/R) of the larch stand was about unity, which is much less than that of other boreal and subalpine conifer forests. The proportion of fine roots in total root biomass (35%) was relatively high compared with other cold-climate evergreen conifer forests. Total NPP, defined as the sum of annual biomass increment of woody parts and needle biomass, was estimated to be 1.8 Mg ha(-1) year(-1). Allocation of total NPP to needle production was 56%. The proportion of total NPP in belowground production (27%) was less than for evergreen taiga forests. However, belowground NPP was probablyunderestimatedbecauserootmortalitywasexcluded.We conclude that L. gmelinii trees invested annual carbon gains largely into needle production or roots, or both, at the expense of growth of aboveground woody parts. This carbon allocation pattern, which resulted in the construction of exploitative root networks, appeared to be a positive growth response to the nutrient-poor permafrost soil of central Siberia.

Аннотация: Androgenesis in vitro in plants is a phenomenon of developmental switching of male generative cells, microspores, from their normal gametophytic to sporophytic pathway. We obtained androgenic callus and embryoids (embryo-like structures derived from microspores) of the conifer plant, Siberian larch (Larix sibirica Ledeb.) in the in vitro culture. The immune-enzyme analysis of the hormonal balance of larch androgenic cultures showed a substantial increase in the content of all phytohormones, especially cytokinins and ABA, as compared with initial explants (microstrobiles). This was evidently related to active cell divisions and embryoid formation. A comparison of androgenic cultures derived from trees nonifested and infested with larch gall midges (Dasineura rozhkovi Main. et Nik.) revealed a cytokinin content increase (by two times) and an ABA content decrease (by two times), which indicates more intense growth of cultures derived from healthy trees. Phytohormone content in the androgenic callus was compared with their accumulation in the embryos of larch seeds harvested from noninfested trees. We concluded that successful growth of androgenic cultures and embryoid formation demand an additional medium supplement with auxins (not more than 0.5 mg/l).

Аннотация: Changes in the levels of ascorbic acid (AA), its oxidized form, dehydroascorbic acid (DHA), and uronic acids as initial precursors for the AA synthesis were studied as related to the degree of xylem and phloem cell development in the course of early and late wood formation in the trunks of Scots pine (Pinus sylvestris L.). The cells of mature and conducting phloem, cambial zone, differently developed cells in the zones of cell enlargement and maturation were obtained by successive scraping tissue layers from trunk segments of 20-25-year-old trees; tissue identification was checked anatomically and histochemically. The contents of compounds tested were calculated per dry weight and per cell basis. We found great differences in the contents of AA and DHA and also in their ratio in dependence of the wood type developing in the pine trunks during growth period and on the stage of differentiation of xylem and phloem cells. Changes in the AA content during xylem cell differentiation were accompanied by changes in the content of uronic acids. The amounts of AA, DHA, and uronic acids were the highest at the stage of early lignification and reduced with tracheid maturation. The AA to DHA ratio changed differently in the course of early and late xylem lignification. It reduced from the start of lignification to the formation of early mature xylem and, in contrast, increased in mature late wood; this indicates a difference in the level of redox processes in these tissues.

Аннотация: The periodic production of large seed crops by trees (masting) and its interaction with stern growth has long been the objective of tree physiology research. However, very little is known about the effects of masting on stern growth and total net primary productivity (NPP) at the stand scale. This study was conducted in an old-growth, mixed deciduous forest dominated by Fagus sylvatica (L.) and covers the period from 2003 to 2007, which comprised wet, dry and regular years as well as two masts of Fagus and one mast of the co-dominant tree species Fraxinus excelsior (L.) and Acer pseudoplatanus (L.). We combined analyses of weather conditions and stem growth at the tree level (inter- and intra-annual) with fruit, stem and leaf production, and estimates of total NPP at the stand level. Finally, we compared the annual demand of carbon for biomass production with net canopy assimilation (NCA), derived from eddy covariance flux measurements, chamber measurements and modelling. Annual stem growth of Fagus was most favoured by warm periods in spring and that of Fraxinus by high precipitation in June. For stem growth of Acer and for fruit production, no significant relationships with mean weather conditions were found. Intra-annual stem growth of all species was strongly reduced when the relative plant-available water in soil dropped below a threshold of about 60% between May and July. The inter-annual variations of NCA, total NPP and leaf NPP at the stand level were low (mean values 1313, 662 and 168 g C m(-2) year(-1), respectively), while wood and fruit production varied more and contrarily (wood: 169-241 g C m(-2) year(-1); fruits: 21-142 g C m(-2) year(-1)). In all years, an annual surplus of newly assimilated carbon was calculated (on average 100 g C m(-2) year(-1)). The results suggest that stem growth is generally not limited by insufficient carbon resources; only in mast years a short-term carbon shortage may occur in spring. In contrast to common assumption, stem growth alone is not a sufficient proxy for total biomass production or the control of carbon sequestration by weather extremes.

    Regeneration patterns in boreal Scots pine glades linked to cold-induced photoinhibition

Аннотация: Regeneration patterns of Pinus sylvestris L. juveniles in central Siberian glades were studied in relation to cold-induced photoinhibition. Spatial distribution of seedlings in different height classes revealed higher seedling densities beneath the canopy than beyond the canopy, and significantly higher densities of seedlings < 50 cm tall on the north side of the trees. These patterns coincided with differences in light conditions. Compared with plants on the north side of canopy trees (north-exposed), photosynthetic photon flux (PPF) received by plants on the south side of canopy trees (south-exposed) was always higher, making south-exposed plants more susceptible to photoinhibition, especially on cool mornings. Chlorophyll fluorescence data revealed lower photochemical efficiency and increased non-photochemical quenching of small (20-50 cm in height), south-exposed seedlings from spring to early autumn, indicating increased excitation pressure on photosynthesis. Maximum rate of oxygen evolution was less in south-exposed plants than in north-exposed plants. Increased pools of xanthophyll cycle pigments and formation of the photoprotective zeaxanthin provided further evidence for the higher susceptibility to photoinhibition of south-exposed seedlings. A linear mixed model analysis explained many of the physiological differences observed in seedlings according to height class and aspect with early morning temperature and PPF as predictors. The link between photoinhibition and differential distribution of seedlings by height class suggests that photoinhibition, together with other environmental stresses, decreases the survival of small, south-exposed P sylvestris seedlings, thereby significantly affecting the regeneration pattern of central Siberian pine glades.

Аннотация: Effect of tannin and non-volatile components of lesion resin on the growth of blue-stain fungi - Ceratocystis laricicola, C. polonica, Ophiostoma minus, Leptographium sp. and Ophiostoma sp. - the primary invaders of conifers in Siberia (Russia), has been studied. The fungi under study exhibited tolerance to plant's defensive substances in bioassays. The cultures of Leptographium sp., C. polonica, and Ophiostoma sp. proved to be the most tolerant to tannin. In the range between 0.05 and 0.15% tannin concentration their growth tended to stabilize. Less tolerant to tannin were C. laricicola and O. minus. With an increase in tannin concentration their biomass gradually decreased. The fungi more tolerant to tannin regulated the medium pH about 5.0 and higher, white the species which were less tolerant preferred lower pH values. Lesion resins of conifers differed in their inhibitory effect on fungal growth. Fir resin had the greatest inhibitory effect of all the conifer resins tested, causing reduction to 50-60% in fungal growth. When fungi were grown on the resin of any other conifer, the reduction in the linear growth rate did not exceed 20 to 30%. The assumption that fungi are more tolerant to the resin of a host plant than to that of a non-host plant has not been confirmed.

Аннотация: Effect of defoliation on resistance response of Siberian fir, Abies sibirica Ledeb., to fungal inoculation was investigated within three even-aged fir stands damaged by Siberian moth, Dendrolimus superans sibiricus Tschetw., and located in the Krasnoyarsk Territory (Central Siberia). Histological changes in tissues, accompanying the resistance response were examined in 30 fir trees (90-130 years old) of different defoliation degree (50, 75, 100 %) six weeks after single inoculation with Leptographium sp, vectored by Monochamus urussovi Fisch. (Coleoptera: Cerambycidae). It was recorded that the rate and intensity of defence response of phloem and xylem tissues to inoculation as well as percent of trees infested by the insect-vector differed significantly in dependence of defoliation degree. Trees of 50 percent defoliation significantly differed from trees of more severe defoliation in all parameters of defence response and percentage of colonisation by the beetle. The trees defoliated to 75 and 100% did not differ significantly in defence response activity and the number of the trees attacked by M. urussovi Strong difference in resistance to inoculation and colonisation by the beetle was recorded between trees defoliated in the current year and in the previous year. All infested by M, urussovi trees were characterised by the epidemic level of frequency of blue-stain fungi associated with this insect. The tendency was recorded to a decreased frequency of the pathogenic Ophiostoma sp. and a more frequent occurrence of the saprophyte O. curvicollis in trees defoliated in previous year which lost ability to defend against wounding and fungal infection.

Аннотация: Studies of microsporogenesis in the Siberian larch growing in Krasnoyarsk and its suburbs have shown that meiosis starts in October. Microsporocytes winter at prophase I (leptoneme, diploneme). Reduction divisions in male generative buds are resumed and terminated in spring, in March. However, in the case of thaws during the autumn-winter period, meiotic division proceeds in the larch buds and this leads to the formation of degrading tetrads and pollen. Hence, the organic quiescence is absent in the larch in winter. It was shown that in the larch growing in the city, meiosis proceed more asynchronously than in the background tree stands. An increase of chromosomal aberrations during the reduction division was noted under the conditions of technogenic pollution.

Аннотация: The development of male generative organs of the Siberian stone pine clones of different origin was analyzed in the Krasnoyarsk forest-steppe for many years. Geographical and individual variability was described as concerns the number of microstrobils and pollen variability. The beginning of "flowering", abundance of microstrobils, and their regular formation were shown to markedly vary and, all other conditions being equal, be determined by the presence of clones, their hereditary features, and environmental conditions, as well by the influence of stock.

    Specific features of the development of Siberian stone pine megagametophytes and embryos in vitro

Аннотация: Seedlings were grown in vitro from fertilized eggs and immature embryos of the Siberian stone pine. Cultivation of megagametophytes on a hormone-containing Murashige-Skoog medium from the egg formation until the globular embryo stage made it possible to manipulate fertilization and embryogenesis. Immature embryos are the most promising for in vitro cultivation. Their maturation and germination proceed within seven days of cultivation. When zygotic embryos were cultivated, adventitious buds were formed from cells at the cotyledon base and tips. When adventitious buds were subcultivated on a medium containing benzylaminopurine and naphthylacetic acid, organogenic callus and shoots were formed. Thus, cultivation of megagametophytes and embryos of the Siberian stone pine led to the completion of embryogenesis and formation of viable of seedlings.
