Аннотация: Environmental conditions causing stress have a significant effect on the generative organs of Scotch pine. The parameters characterizing pine pollen are subject to significant interannual variations and are closely connected with weather and climatic conditions in the period of pollen formation. Airborne pollutants affect the quality of pine pollen, which is especially apparent in the years favorable for the formation of microspores. The ability of Scotch pine pollen to germinate and form pollen tubes in the regions with different technogenic loads proved to be significantly lower than in tree stands of the background areas.

Аннотация: Morphological and cytoembryological studies of unique forms of Siberian pine (Pinus sibirica Du Tour) with the annual cycle of ovulate cone development were carried out. The morphogenesis of female shoots in these anomalous trees with the annual cycle of development was shown to provide for an accelerated development and very rapid growth of megastrobili and accelerated formation of reproductive structures. Cytoembryological studies of ovules in these trees showed significant shortening of the free-nuclear stage of gametophyte development (up to 3 weeks instead of one year) and ultra-early formation of archegonia and egg cells. However, no fertilization of egg cells and the development of embryos occur in the anomalous forms; the egg-cell nucleus divides in the haploid state. Developing seeds are formed without embryo. The author hypothesizes that the acceleration of embryo development in the anomalous pine trees is caused by enhanced hormonal, carbohydrate, and nitrogen metabolism in ovule tissues.

Аннотация: Structure of crown and viability of generative organs of Siberian fir were studied in disturbed forest ecosystems near Baikal Lake. The crown structure changes in damaged trees: a zone of subapical drying appears and apical dominance is lost. Needle chlorosis and necrosis appear and proceed in sprouts of male sexualization. The tree top including the female generative organs does not show degradation symptoms during about 30 years and produces seed cones. Processes of degradation in male generative zone greatly influence on fir tree pollen. Pollen viability of fir trees varied from year to year in different trees but always pollen quality of trees growing in the Khamar-Daban Mts in the air pollution zone was more poor in comparison with the pollen of trees at die Baikal Lake shore. Tree pollen growing in the Khamar-Daban Mts germinates in vitro but pollen tubes are formed short and not viable. Seed quality of fir trees is very low, A lot of seeds are sterile. Empty seeds prevail. The formation of empty seeds was caused by the low pollen germination in ovules and as a whole absence of pollen in ovules. The high sterility of generative organs and especially of Abies sibirica pollen can limit gametic selection and decrease fir diversity in forest ecosystems near Baikal Lake.

Аннотация: The prolongation of the warm season during autumn, which has become typical for Siberia in the past 5-6 years, has caused changes in the terms of reproductive processes in microsporangia of Scotch pine in the Krasnoyarsk region. Meiosis has started to begin in the autumn of the year preceding pollination, and then come to an end the next spring, whereas usually this process has occurred in the spring of the year of pollination. Under such conditions the division of microsporocytes is accompanied by different abnormalities, causing various anomalies and the incomplete development of pollen grains.

Аннотация: The objective of this study was to obtain a genetically stable haploid in vitro-derived line from Siberian larch (Larix sibirica Ledeb.) using megagametophyte explants, which then could be used for different molecular genetic studies, including whole genome de novo sequencing. However, cytogenetic analysis and genotyping of 11 microsatellite loci showed high levels of genomic instability and a high frequency of mutation in the obtained megagametophyte-derived callus cultures. All cultures contained new mutations in one or more microsatellite loci.

Аннотация: Morphogenesis and maturation of somatic embryos, ploidy, and genotyping of cell lines (CL) of embryogenic cultures of Larix sibirica Ledeb. in vitro were investigated during 2–6 years. It was revealed that from 2000 to 11103 globular somatic embryos were formed in proliferating CL. However, the ability of somatic embryos to the maturation and germination decreased. Cytogenetic study of embryonal-suspensor masses (ESM) of Larix sibirica demonstrated that cells of long-term cultivated cultures remained diploid. According to microsatellite analysis, proliferating CL of Siberian larch were characterized by weak allelic variability, and cell line 6 and cloned seedlings of this line were genetically stable and corresponded to the donor tree. Embryogenic cell lines composed the collection bank, which will be successfully used for plantation forest growing. © 2016, Pleiades Publishing, Ltd.

Аннотация: Cytoembryological research of the ovules in experiments with interspecific hybridization of Pinus sibirica (pollination be the pollen of P. koraiensis, P. armandii, P. parviflora, P. strobus, P. hokkaidensis, P. wallichiana, P. monticola, and P. Nembra) revealed that the development of megagametophytes occurred in them by the usual scenario and resulted in the formation of mature archegonia. Pollen successfully germinated on the nucellus of ovules. However, disturbances were observed in the process of male gametophyte development, and pollen tubes on the nucellus were not visible by the period of archegonia maturation. Fertilization was usually absent. The development of embryonic channel is determined by egg cell maturity. The only exception was the variant of the controlled pollination of Pinus sibirica x P. Nembra, in which the embryo has been formed.

Аннотация: We carried out immunoassay of plant hormones [indoleacetic acid (IAA), cytokinins and abscisic acid (ABA)] in the extracts from explants (megagametophytes and embryos) and from four long-term proliferating embryogenic cell lines of Larix sibirica. Immunohistochemical detection of hormones has also been performed at the stage of initiation of somatic embryogenesis and in embryogenic cultures during the process of somatic embryo morphogenesis. During induction of somatic embryogenesis in tissue culture, the first stage of cell development is manifested in their elongation and polarization. Immunohistochemical analysis of hormones showed localization of IAA at one pole of elongated cells of L. sibirica. The polarity of embryogenic structure development persisted later on. The difference has been shown in the content of hormones between the explants and embryogenic cultures. High level of hormones was detected in the cell line (Cl4), distinguished by great number of globular embryos and high capacity for their maturation. Hybrid line Cl5 (L. sibirica ? L. sukaszewii) formed very short suspensor, accumulated higher amount of ABA in the head of somatic embryos (SEs) than did other lines, while SEs of this cell line did not maturate. Cl6 was characterized by low plant hormones content in proliferating embryogenic cultures and low yield of mature somatic embryos. © 2018, Springer Nature B.V.

Аннотация: In vitro cultivation of embryogenic cultures of Siberian larch on AI medium supplemented with different concentrations of antimicrobial peptides of two Trichoderma species, T.citrinoviride (strain TYVI 4/11) and T. viride (strain 346), has been performed to achieve a direct antimicrobial effect and initiate the mechanisms of induced resistance (regulatory function of embryogenic cultures) as well as to study the morphogenesis and growth activity of regenerant plants. The experiment was arranged using four cell lines, CL4, CL5, CL6, and CL12, differing in their embryogenic activity. The effect of treatment of cell cultures with Trichoderma peptides was manifested via significant growth stimulation of proliferating embryogenic cell lines, root growth stimulation, and formation of callus excrescences on regenerant plants. No difference between the treated and untreated variants was observed in seedlings. Conceivably, such peptide treatment may provide an increased immunity of soil-grown seedlings in forest nurseries. Study of the effect of biocontrol strains' use in a plantation reforestation to stimulate the growth and development of calluses and embryos and to obtain disease-resistant regenerant plants of conifer species will provide the development of a clonal silviculture, a new trend that recently appeared abroad (Park 2002, 2014).

Аннотация: The effect of plant antimicrobial peptides on the initiation of callus and embryonal suspensor masses, formation of somatic embryos, and germination of regenerants of Siberian larch has been studied. Protein/peptide extracts isolated fromAmarantus retroflexus(seeds),Nigella sativa(seeds), andElytrigia elongata(spikelets) have been used as objects of plant origin. Peptides have been introduced into the nutrient media at the stage of initiation of embryogenic cultures and somatic embryo germination. The stimulating effect of peptides on the formation of embryogenic cultures of Siberian larch has been found. No other differences in the dynamics of growth in the control and experimental regenerants have been observed. This study is supposed to contribute to enhancing the immunity of the clonal planting stock of Siberian larch.

Аннотация: It was found that glycerides were dominant for both types of cell lines. It was found that triglycerides (TG) and 1,2-diglycerides (1,2-DG) accumulated 1.5-3 times more actively in embryogenic cell lines, which was probably connected with the necessity of the involvement of TG and 1,2-DG in the processes of tissue differentiation. A high relative content of free fatty acids (FFA) was also characteristic of the embryogenic lines. The found differences in the ratio of sterols to their ethers (the content of sterol esters was 1.5-3 times lower in embryogenic cell lines) can indicate that sterols get activated from their inert form to contribute to the embryogenesis processes at a stage of embryo formation.

Аннотация: The contents of auxin (indole-3-acetic acid, IAA), cytokinins, and abscisic acid (ABA) were determined in explants (megagametophyte and embryo), proliferating embryonal-suspensor mass (ESM), and non-embryogenic callus of Pinus sibirica and Larix sibirica. Furthermore, the localization of hormones in the cells was studied with immunohistochemical analysis. It was shown that the formation of embryogenic cultures in both species is associated with the content of phytohormones and their localization in cells. In L. sibirica the content of IAA in ESM was about 100 times higher than in non-embryogenic calli. At the same time, a low ABA content was characteristic of embryogenic cultures. Non-embryogenic callus contains an increased content of cytokinins and ABA. The transition of somatic cells to the path of embryogenic development is characterized by elongation of cells, their asymmetric division, and localization of IAA at one of the ends of elongated cells. Nonembryogenic callus consisted of isodiametric, actively dividing cells. © 2021 The Japan Mendel Society.

Аннотация: The article presents the results of the analysis of available data on the reproduction (sexual reproduction and asexual reproduction or apomixis) of plants of the Pinopsida class. The realization of apomixis in conifers was considered. This method of asexual reproduction is manifested in cases of seed formation's disruption. A specific type of reproduction in conifers is somatic embryogenesis in in vitro culture. It is shown that the transition of somatic cells to the totipotent and embryogenic state in conifers occurs at the morphogenetic, physiological-biochemical, and molecular genetics levels. The commonality of morphogenetic processes underlying somatic and zygotic embryogenesis is revealed. The position of somatic embryogenesis in the reproduction system of conifers is considered.
