Remains the most comprehensive collection of methods available for studying natural killer cells in both mice and humans. Examines a wide array of molecular and cellular techniques for manipulating and assaying NK cell functions both in vitro and in vivo . Includes techniques to assay NK cell phenotype, development, signaling, and functional responses to tumor cells, virus infection and malaria infection . Contains a wide array of techniques to assay NK cells by flow cytometry. Provides molecular techniques to assay important genotypes, map functionally important genetic loci in mice, and identify genetic regulatory elements. Supplies several specialized protocols to identify novel ligands for NK cell receptors. Includes specialized techniques for studying NK cells from tonsils, lymph nodes, uterine tissues, and gut mucosa. Includes clinically relevant assays on human killer cell Ig-like receptor (KIR) genotype and surface expression phenotype by flow cytometry, as well as detailed information on KIR nomenclature and ligand specificities. Highlights information on nomenclature, cell lines, available antibodies, and receptor ligands in its valuable appendices. As the research has continued, it has become increasingly clear that natural killer (NK) cells are critical sentinels of the innate immune response, playing important roles in protecting the body from numerous pathogens and cancer in addition to contributing to normal pregnancy and impacting the outcomes of transplantation. While the first edition provided a valuable collection of classical cellular and in vivo techniques to study NK cell functions, the Second Edition of Natural Killer Cell Protocols: Cellular and Molecular Methods brings together more recently developed methods, more refined techniques, and detailed protocols designed to study NK cells within specialized tissue sites in both mice and humans. In this collection of methods, international leaders in the field cover topics ranging from the analysis of the various stages of NK cell development and maturation to specialized techniques for the identification of ligands for NK cell receptors. This volume also includes an appendix, providing a rich resource summarizing available reagents to study NK cells, cross-referencing KIR nomenclature, and detailing the many HLA ligands for various KIR family members. As a volume in the highly successful Methods in Molecular Biology™ series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and thorough notes sections, highlighting tips on troubleshooting and avoiding known pitfalls. Comprehensive and cutting-edge, Natural Killer Cell Protocols: Cellular and Molecular Methods, Second Edition seeks to aid researchers and further advance our understanding of the functions, maturation, and regulation of these fascinating and dynamic cells.

Аннотация: Plant diseases are due mainly to the action of fungi, but some are caused by bacteria, a few by plasmodia, while others in which no particulate living organisms have, so far, been discovered, are believed to be due to virus infection. In plant viruses that have been studied in any detail the invisible agent multiplies with great readiness only in the living host from which the virus has been isolated in the form of a nucleoprotein. Other diseases, again, but of a non-parasitic nature, are attributed to physiological or to deficiency troubles, and probably all are influenced by the factors of the environment.

epizootic disease, polyhedrosis virus, insect population

Аннотация: The polyhedrosis of the alfalfa caterpillar is an important factor in the natural reduction of populations of this insect in California. The results of preliminary field tests indicate that: (1) The virus, applied as a spray, is capable of causing infection in and markedly reducing populations of the alfalfa caterpillar. (2) It is possible initiate an epizootic of the disease in populations of low density (20 to 30 larvae per 10 sweeps), and that even these low population can be substantially reduced by the artificial dissemination of the virus. (3) It is possible to initiate an epizootic of the disease in a population of caterpillars earlier than it would occur naturally, thus curtailing the amount of damage done to the crop by insect

disease -- virus -- infection

Аннотация: The purpose of present paper to recount seven new instances in which a single insect species is subject to infection by two types of virus; in one case both types occurred simultaneously in the same specimen. The number of insect species known to be susceptible to both a nuclear polyhedrosis virus and a granulosis virus is not great. Nevetheless, a perusal of the literature shows as many as seven insect species recorded, at different times, as being attacked by a nuclear polyhedrosis and a granulosis virus. It is to be hoped that eventually a comparative morphological, developmental, and biochemical study will be made of two types of virus as they occurr in a single host species

The results of the diagnoses made, according to the kind of causative microorganism or the cause of death were grouped as follows percentagewise: fungous infections, 34 per cent; bacterial infections, 20 per cent; virus infections, 18 per cent; protozoan infections, 11 per cent; accessions only for the purpose of ascertaining the presence or absence of symbiotes, 6 per cent; nematode infections, 2 per cent; and deaths resulting from nonmicrobial causes, 2 per cent. No diagnoses were rendered in 7 per cent of the accession received

opaque -- stress -- polyhedrosis virus

Аннотация: A cytoplasmic polyhedrosis has been found affecting the mid-gut of the alfalfa caterpillar, Colias philodice eurytheme Boisduval. Externally, the diseased larvae exhibit a yellowish-green color over the mid-gut region. Upon dissection, the mid-gut is seen to have assumed a yellowish opaque appearance. Histological sections show the cytoplasm of the gut cells to contain polyhedra of two general size groups. Electron micrographs of alkaline-dissolved polyhedra reveal the presence of numerous spherical particles presumed to be the virus. The diseased larvae were originally observed during laboratory experiments in specimens subjected to the stress of crowding; but similarly diseased caterpillars have also been collected from alfalfa fields

Аннотация: Miscellaneous observations on polyhedroses in several species of Lepidoptera are reported. The genera of Lepidoptera represented include Peridroma, Prodenia, Colias, and Heliothis. In the case of Peridroma, the data pertaining to a nuclear polyhedrosis were compared with those relating to a granulosis. Incubation periods, strain differences, cross infectivities, symptomatology, and the effects of certain stressors were determined or studied in individual instances. Also reported are new records of virus-diseased insects received as accessions for diagnosis
