Аннотация: We studied fatty acid (FA) composition in samples from bodies and intestinal contents of the littoral amphipod Gammarus lacustris Sars, from the Bugach freshwater reservoir. Simultaneously, samples of seston and bottom sediments were also collected from the reservoir during early August. There were no differences in FA composition of gut contents, seston and sediments of pebbly bottom. Seston was the main food source of Gammarus but some FAs Gammarus got from sediments. The FA composition of G. lacustris and seston from the Bugach freshwater reservoir were compared with those of the animals from the saltwater Lake Shira (Siberia). While FA composition of the two Gammarus populations differed significantly, those of seston were practically similar: the composition of long-chain unsaturated fatty acids, 20:5omega3, 22:6omega3 and 20:4omega6, were significantly higher in animals from saline Shira Lake, whereas 16:1 and 16:0 were higher in the freshwater populations of amphipods from the Bugach freshwater reservoir. Taking into account the relevant literature data, we hypothesise that this difference in C16 acid might be a distinguishing characteristic of FA composition of freshwater and saltwater crustaceans.

Аннотация: Properties of non-canonical GC base pairs and their relations with mechanochemical cleavage of DNA are analyzed. A hypothesis of the involvement of the transient GC wobble base pairs both in the mechanisms of the mechanochemical cleavage of DNA and epigenetic mechanisms involving of 5-methylcytosine, is proposed. The hypothesis explains the increase in the frequency of the breaks of the sugar-phosphate backbone of DNA after cytosines, the asymmetric character of these breaks, and an increase in break frequency in CpG after cytosine methylation. As an alternative hypothesis, probable implication of GC(+)Hoogsteen base pairs is considered, which now exemplify the best-studied non-canonical GC base pairs in the DNA double helix. Аннотация: This study investigates synthesis of poly(3-hydroxybutyrate)/diethylene glycol copolymers (P3HB/DEG) by Cupriavidus eutrophus B-10646 cells as related to DEG concentration in the medium and the time when it is added to the culture of cells synthesizing P3HB. The study determines the limits of physiological effect of DEG on C. eutrophus cells, showing that at DEG concentrations above 30 g/L, it inhibits cell growth, decreasing cell concentration and total P3HB/DEG yield and inducing an increase in the degree of saturation of fatty acids in lipids of cell cytoplasmic membrane. A series of copolymers containing different molar fractions of DEG (between 0.13 and 3.0 mol%) have been synthesized and their physicochemical, physical/mechanical, and biological properties have been investigated as related to the chemical composition and proportions of DEG monomers of the polymers. © 2016 American Institute of Chemical Engineers Biotechnol. Аннотация: The effect of ambient temperature on the composition of intracellular fatty acids and the release of free fatty acids (FFA) into a medium by cyanobacterium Spirulina platensis and eukaryotic microalgae, Chlorella vulgaris and Botryococcus braunii, was studied using their batch cultures. It was found that all the species studied, regardless of their taxonomic status, responded to the temperature regime by similar changes in their intracellular fatty acid composition: the relative content of more unsaturated fatty acids decreased with the elevation of temperature. At the same time, in the prokaryote, this temperature shift blocked, first of all, the elongation of 16:0 to 18:0 and then their further desaturation. In eukaryotes, the change in the desaturation of dienoic to trienoic fatty acids was the most pronounced process. The ratio of dienoic to trienoic fatty acids remained almost unchanged in S. platensis. The relative content of extracellular unsaturated FFA increased in the prokaryotic organism S. platensis at a higher temperature. But no significant changes in the composition of extracellular unsaturated FFA were detected in eukaryotic algae upon temperature elevation.

Аннотация: The synthesis of polyhydroxybutyrate by the carboxydobacterium Seliberia carboxydohydrogena under various growth conditions was studied. The bacterium was able to accumulate poly-beta-hydroxybutyric acid and a heteropolymer that was a product of copolymerization of hydroxybutyric and hydroxyvaleric acids. Nitrogen and sulfur exerted the strongest effect on polymer accumulation in carboxydobacteria: The maximum polymer concentrations in cells reached 28%. Specimens of polyhydroxybutyrate films were obtained, and their degradability by soil microflora was shown.

Аннотация: Strain AIS(T), an aerobic halophilic, Gram-reaction-negative, heterotrophic bacterium isolated from the water of Shira Lake in Khakasia, southern Siberia, was characterized using a polyphasic approach. Our analysis of the 16S rRNA gene sequences showed that 'Aliidiomarina haloalkalitolerans', 'Allidiomarina sanyensis', Idiomarina maris and AIS(T) formed a distinct lineage. The sequence similarities between AIS(T) and the type strains of species of the genera Idiomarina and Aliidiomarina were 91.6-95.1 % and 94.0-96.9 %, respectively. The major isoprenoid quinone of AIS(T) was ubiquinone 8 (Q-8). Predominant cellular fatty acids were iso-C-17:0, iso-C-15:0 and summed feature 9. The genomic DNA G+C content was 45.8 mol%. It is concluded that AIS(T) represents a novel species of the genus Aliidiomarina, and the name Aliidiomarina shirensis sp. nov. is herein proposed for it. The type strain is AIST (=JCM 17761(T)=BCRC 80327(T)). Based on its fatty acid profile and our phylogenetic analysis, we propose that Idiomarina mans be transferred to the genus Aliidiomarina.

We hypothesised that there was an imbalance between the composition and content of amino acids in the biofilm and its consumers, the zoobenthos, as well as between those in the zoobenthos and fish. Based on monthly sampling from 2007 to 2010, there was seasonal variation in AA profiles in the epilithic biofilms, probably caused by the succession of microalgal and cyanobacterial species. Overall, there was an imbalance in the percentage of the essential amino acids (lysine and histidine) between benthic animals and their food (the epilithic biofilm), which suggests that benthic animals may be limited by food quality. Moreover, the zoobenthos had a significantly higher content of AA, relative to carbon, than the biofilm. Based on sampling in 2012, there was an imbalance between the AA profiles of zoobenthos and that of their main consumer, the Siberian grayling (Thymallus arcticus), particularly in the percentages of two essential amino acids, lysine and leucine. In terms of overall content of essential amino acids, the nutritional value to fish of gammarids, which have recently invaded the river, was significantly lower than that of indigenous taxa, trichopteran and chironomid larvae. Аннотация: We have reviewed foreign and domestic literature devoted to the study of the amino acid (AA) composition of aquatic organisms representing major groups of producers (green microalgae and diatoms, and cyanobacteria) and primary consumers (zooplankton). Based on published data, we estimate the composition of essential and nonessential AAs of microalgae, cyanobacteria, and zooplankton and determine their differences. It is concluded that the AA composition of major groups of plankton is heterogeneous. The role of AAs as a limiting factor for the development of herbivorous zooplankton is discussed. The prospects and the need for further study of AA composition in order to develop a complete theory of functioning of aquatic ecosystems have been demonstrated.

Аннотация: Total dependence of aquaculture on imports of feeds can significantly compromise Russia's food supply security; hence the task of designing new domestic feeds is of primary importance. A major challenge of world aquaculture is providing fish with a balanced diet, in particular, with a well-balanced composition of amino acids. In this review, we analyze international publications on the composition of amino acids in fish feed over the last two years. We summarize the data on the amino acid composition of prospective food sources for fish aquaculture: agricultural crops, insects, annelids, higher crustaceans, animal waste, phytoplankton, zooplankton, macrophytes, and microorganisms. We performed a comparative analysis of the composition of amino acids in promising aquafeed sources and fish flour traditionally used for fish feeding. Based on the results, we conclude that each of the examined food sources can be used as a component of fish nutrition and offers realistic prospects for production of alternative feeds. To ensure high quality of fish aquaculture products, the priority should be given to development of technologies which allow to control amino acid feed composition during the production process with respect to fish species-specific needs. It is hoped that this review will be useful to researchers and practitioners involved in designing and producing new domestic aquafeed for fish. © Siberian Federal University. All rights reserved.

Аннотация: The flow of long-chain polyunsaturated fatty acids (PUFAs) of the omega-3 family, namely, eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA), exported by amphibian metamorphs from water to terrestrial ecosystems in the Medveditsa River floodplain, was quantified for the first time. The total biomass export by three amphibian species (Pelobates fuscus, Bombina bombina, and Pelophylax ridibundus) per unit area of the lake surface was 0.594 g/m(2) per year (as a mean for 2 years). The biomass flow per unit area of land was 0.726 g/ha per year (0.302 g/ha per year for organic carbon) in 2015-2016. The average annual total removal of EPA + DHA by amphibians from the floodplain lake was 1.47 mg/m(2) of water surface area. Due to the high content of EPA and DHA in biomass, amphibians are potentially a valuable food for terrestrial predators having no access to other sources of essential PUFAs.

Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: The study of the composition of fatty acid markers of tadpoles of cohabiting amphibian species for the first time revealed differences in their diets: the moor frog Rana arvalis prefers bacteria not associated with plant detritus, whereas the diet of the common spadefoot Pelobates fuscus is based on cyanobacteria, green algae, diatoms, and possibly higher plants. Major differences in the fatty acid composition are determined by the difference in the percentage of eicosapentaenoic and myristic acids.

Аннотация: The study of the trophic relationships of aquatic animals requires correct estimates of their diets. We compared the quantitative fatty acid signature analysis (QFASA) and the isotope?mixing model IsoError, based on the compound?specific isotope analysis of fatty acids (CSIA?FA), which are potentially effective models for quantitative diet estimations. In a 21?day experiment, Daphnia was fed a mixture of two food items, Chlorella and Cryptomonas, which were supplied in nearly equal proportions. The percentages and isotope values of the FAs of the algal species and Daphnia were measured. The IsoError based on CSIA?FA gave an estimation of algae consumption using only one FA, 18:3n?3. According to this model, the proportion of consumption of Chlorella decreased while the proportion of consumption of Cryptomonas increased during the experiment. The QFASA model was used for two FA subsets—the extended?dietary subset, which included sixteen FAs, and the dietary one, which included nine FAs. According to both subsets, the portion of consumed Chlorella decreased from Day 5 to 10 and then increased at Day 21. The comparison of the two model approaches showed that the QFASA model is a more reliable method to determine the contribution of different food sources to the diet of zooplankton than the CSIA?based mixing model. © 2021 by the authors. Аннотация: 1. We studied variation in the composition of fatty acids in the seston of a small freshwater reservoir with changes in phytoplankton composition during four growth seasons. We focused on the dynamics of the ?3 fatty acids because of their potential importance for zooplankton nutrition. 2. Total diatoms were related to the 20:5?3 fatty acid (eicosapentaenoic, EPA) content in seston. Among two dominant diatom genera, Cyclotella was not associated with EPA content. In contrast, there was a significant correlation between Stephanodiscus and the percentage contribution and content of EPA throughout the study. Hence, freshwater diatoms can differ strongly in content of the essential EPA. 3. We considered abundant cyanobacteria as a potential source of 18:3?3 fatty acid (linolenic, ALA) to aquatic food webs. Among four dominant cyanobacteria species, two (Anabaena flos-aquae and Planktothrix agardhii) showed significant correlation with the ALA content of the seston, while the other two (Aphanizomenon flos-aquae and Microcystis aeruginosa) did not. 4. Dinophyta had a relatively high level of 22:6?3 (docosahexaenoic, DHA) for freshwater species and can be also a source of EPA to aquatic food webs. 5. Our results show that various species of diatoms as well as cyanobacteria can be of contrasting nutritional value for zooplankton because of their different content of the essential PUFAs. Diatoms, which are low in EPA, could not be considered as a valuable food, while some field populations of cyanobacteria might be valuable sources of essential ALA.
