Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.

Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, ?-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/ 3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physical-mechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB. Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: A procedure has been developed to fabricate polymer tubes using solution casting inside template tubes. Polyester solution placed inside the vertically fixed template evaporated forming a hollow tube on the inner walls of the template. Silicone tubes used as the templates were permeable to solvent vapors and had relatively low adhesion to materials of the fabricated tubes, enabling their effortless removal. Parameters of the polymer tubes were determined by a type of the polymer, its concentration in the solution, and the inner diameter of the template. In the first method, a clamp was placed on the lower end of the template tube. A thickness of the new tube walls gradually increased from the open to the clamped end of the template tube. In another method viscous polymer solution without clamping was used; in this case, a polymeric bulkhead was formed in the middle of the tube. Two microbial polyesters, poly-3-hydroxybutyrate and poly-3-hydroxybutyrate-co-3-hydroxyvalerate, and also synthetic poly-L-lactide were used as model polymers. (C) 2020 Elsevier B.V. Аннотация: The study addresses the effect of abiotic (medium salinity and copper ions) and biotic (interactions between populations) factors on the formation of structured communities by binary associations consisting of halotolerant bacteria (Alcaligenes sp. 1-1 or Acinetobacter sp. 1-19) and a wild-type B. subtilis 2335 strain or a transgenic strain. The results showed that 250 mg l(-1) of copper ions inhibit formation of biofilms by monocultures of the tested strains. Binary associations of the strains were more resistant to high concentrations (250 mg l(-1)) of copper ions. At the lowest NaCl concentration (0.05% and 2.5%) and in the presence of copper ions, bacilli seemed to help halotolerant bacteria survive. Under increased salinity and in the presence of copper ions, structured communities developed due to halotolerant bacteria. Coexistence under stressful conditions was beneficial for the both groups of bacteria.

Аннотация: A series of films was obtained from blends of poly-3-hydroxybutyrate (PHB) and poly-s-caprolactone (PCL). The structure, physical, mechanical and biological properties of the films were studied, depending on the ratio of the components. The microscopic structure of mixed films was determined by the main component of the blend: the films were porous with a predominance of PHB and relatively smooth with a predominance of PCL. The highest values of elongation at break (229-269%), as well as the local maximum values of tensile strength (14-15 MPa) were noted for blends with 5-25% PHB content. At PHB content of 25% and 75%, local maxima of Young's modulus were observed (507 and 655 MPa, respectively). When the ratio of the components in the blend was equal (50:50 by weight), the films were heterogeneous and had the lowest values of mechanical strength (ultimate strength and elongation at break were 3 MPa and 15.5%, respectively). A study of the biocompatibility of mixed films in a culture of NIH 3T3 mouse fibroblasts based on determination of the number of adhered cells in the MTT test showed the absence of cytotoxic action, as well as high adhesion and cell proliferation, in some cases exceeding the data for films from pure PHB and PCL. This demonstrates the suitability of PHB-PCL blends for the production of cell carriers for tissue engineering and other reconstructive technologies. © 2021 Author(s).

Аннотация: Films from biodegradable poly-3-hydroxybutyrate are treated with chemical reagents to improve their hydrophilicity and biocompatibility. Two approaches are tested: a single treatment with alkali, acids, oxidizing or reducing agents, and a step-by step treatment of the alkali pre-activated surface of polymer films with bromine water and amino-compounds (ammonia or triethylamine). The maximal level of hydrophilicity (the lowest water contact angle and the highest polar component of the surface free energy) is registered after a single treatment with NaOH and after the step-by-step treatment. These samples also showed the best adhesion of mouse fibroblasts of NIH 3T3 line on the film surface. So, the proposed methods can be used to enhance hydropilicity and biocompatibility of biopolymer surface.

Аннотация: Films from biodegradable poly-3-hydroxybutyrate are treated with chemical reagents to improve their hydrophilicity and biocompatibility. Two approaches are tested: a single treatment with alkali, acids, oxidizing or reducing agents, and a step-by step treatment of the alkali pre-activated surface of polymer films with bromine water and amino-compounds (ammonia or triethylamine). The maximal level of hydrophilicity (the lowest water contact angle and the highest polar component of the surface free energy) is registered after a single treatment with NaOH and after the step-by-step treatment. These samples also showed the best adhesion of mouse fibroblasts of NIH 3T3 line on the film surface. So, the proposed methods can be used to enhance hydropilicity and biocompatibility of biopolymer surface. © 2021 Wiley-VCH GmbH

Аннотация: Series of cell carriers made of resorbable polyester Bioplastotan, which is a copolymer of 3- And 4-hydroxybutyric acid, are characterized in this work. With use of various technologies membranes, 3D scaffolds, and nanomatrixes formed by ultrafine fibers produced with electrostatic spinning were constructed. All types of scaffolds provide adhesion of multipotent mesenchymal stromal cells and are suitable for the cultivation and differentiation of cells in the osteoblastes, that confirmed by detection of extracellular calcium precipitates, alkaline phosphatase activity and osteopontin production in cell culture. Аннотация: Produced from В«BioplastotanВ» resorbing polyesters (linear polyesters of hydroxyl derivatives alkanoic acids) scaffolds for cell culturing such as films, pressed 3-D forms and nonwoven fabric from ultrathin fibers are characterized. Two types of polymers - a homopolymer of the 3-hydroxybutyric acid and a copolymer formed by monomers of the 3-hydroxybutyric and 3-hydroxyvalerianic acids are studied. Surface properties of developed polymer scaffolds, sterilized with autoclaving and H 2O 2-plasma processing are compared. It is shown that plasma has beneficial effects resulting in decrease of the watering contact angle and increase of surface hydrophilic properties. Positive effects of H 2O 2-plasma processing of scaffold surface on culturing cell adhesion and viability compared with autoclaving sterilization is demonstrated on NIH 3T3 line fibroblast culturing.

Аннотация: CO2 laser radiation was used to process poly(3-hydroxybutyrate) constructs - films and 3D pressed plates. Laser processing increased the biocompatibility of unperforated films treated with moderate uniform radiation, as estimated by the number and degree of adhesion of NIH 3T3 mouse fibroblast cells. The biocompatibility of perforated films modified in the pulsed mode did not change significantly. At the same time, pulsed laser processing of the 3D plates produced perforated scaffolds with improved mechanical properties and high biocompatibility with bone marrow-derived multipotent, mesenchymal stem cells, which show great promise for bone regeneration. © 2015 Taylor & Francis.

Аннотация: Poly-3-hydroxybutyrate (PHB) films were plasma-treated using pure NH3, pure Ar or mixtures of the two different proportions (20%, 30%, 40%, 50%, 70% NH3 in Ar). Surface chemistry and surface topography changes of PHB films were observed after plasma processing in all plasma regimes. The XPS results confirmed the absence of chemical modification in the case of pure Ar plasma treatment. Nitrogen-containing groups (e.g., N–C[dbnd]O) were detected on the surfaces of P3HB films treated with NH3-containing plasma. The surfaces of the untreated P3HB films were hydrophobic, and plasma treatment turned the surfaces hydrophilic, irrespective of the treatment. A significant decrease in the contact angle and an increase in the free surface energy were observed. An insignificant surface ageing effect was observed when P3HB samples were exposed to air for 10 days. In NIH 3T3 mice fibroblast cells, cell adhesion was significantly improved after plasma treatment in an Ar atmosphere, which is likely related to the fact that there was a surface ? potential of 88.6 mV at neutral pH, causing a cleavage of the polymer chains and an increase in surface roughness. © 2018 Elsevier Ltd

Аннотация: Microparticles made from degradable polyhydroxyalkanoates of different chemical compositions a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric and 3-hydroxyvaleric acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoic acids (P3HB/3HHx) were prepared using the solvent evaporation technique, from double emulsions. The study addresses the influence of the chemical compositions on the size and ?-potential of microparticles. P3HB microparticles loaded with doxorubicin have been prepared and investigated. Their average diameter and ?-potential have been found to be dependent upon the level of loading (1, 5, and 10 % of the polymer mass). Investigation of the in vitro drug release behavior showed that the total drug released from the microparticle into the medium increased with mass concentration of the drug. In this study mouse fibroblast NIH 3T3 cells were cultivated on PHA microparticles, and results of using fluorescent DAPI DNA stain, and MTT assay showed that microparticles prepared from PHAs of different chemical compositions did not exhibit cytotoxicity to cells cultured on them and proved to be highly biocompatible. Cell attachment and proliferation on PHA microparticles were similar to those on polystyrene. The cytostatic drug encapsulated in P3HB/3HV microparticles has been proven to be effective against HeLa tumor cells. В© 2013 Springer Science+Business Media New York.

Аннотация: The oxygen and ammonia radio-frequency (RF) plasma treatment of poly(3-hydroxybutyrate) P3HB films was performed. We revealed significant changes in the topography, a decrease in the surface zeta potential from -63 to -75 mV after the oxygen-plasma treatment and an increase after ammonia plasma treatment from -63 to -45 mV at a pH of 7.4. Investigations into the NIH 3T3 fibroblast adhesion and growth demonstrated the best cell vitality and a higher cell number for the ammonia plasma treatment at 150W. (C) 2015 Elsevier B.V. Аннотация: Porous films have been prepared from degradable polymers-poly-3-hydroxybutyrate (PHB), poly-epsilon-caprolactone (PCL) and a blend of these polymers (1:3)-by adding porogen (camphor) to the polymer solution at 10%, 30% or 50% of the total mass of the polymer and porogen, and leaching it out afterwards. After the rinse, camphor content in films decreased to about 0.025%. The structure, physical/mechanical and biological properties of the films were investigated as dependent on their composition and porosity, which varied depending on the amount of camphor added. The surface of PHB films was porous, the PCL films were relatively smooth, and the PHB/PCL films had an intermediate structure. The addition of camphor increased the thickness (from 35 to 45 mu m, from 40 to 80 mu m and from 20 to 65 mu m for PHB, PCL and PHB/PCL, respectively) and porosity (from 4.2(+/- 3.6)% to 50.0(+/- 12.8)%, from 6.4(+/- 5.5)% to 54.5(+/- 6.0)% and from 4.9(+/- 4.8)% to 51.5(+/- 5.8)%, respectively) of the films. The introduction (and removal) of 10% camphor into the PHB and PHB/PCL films led to an approximately twofold increase in the polar component of the free surface energy (from 5.4 +/- 0.38 to 11.8 +/- 1.33 and from 2.7 +/- 0.13 to 5.2 +/- 0.09 mN/m, respectively) but in other cases, on the contrary, a decrease in this indicator was registered. The increase of camphor addition from 0% to 50% gradually impaired mechanical properties of the films: so, Young's modulus decreased from 3.6 to 1.8 GPa, from 0.30 to 0.12 GPa and from 0.50 to 0.20 GPa for PHB, PCL and PHB/PCL, respectively. At the same time, the water vapor transmission rate considerably increased from 197.37 +/- 23.62 to 934.03 +/- 114.34 g/m(2)/d for PHB films; from 1027.99 +/- 154.10 to 7014.62 +/- 280.81 g/m(2)/d for PCL films; and from 715.47 +/- 50.08 to 4239.09 +/- 275.54 g/m(2)/d for PHB/PCL films. Results of biocompatibility testing in the culture of NIH 3T3 mouse fibroblast cells showed that for the most of experimental samples cell adhesion and proliferation were comparable or superior to the corresponding parameters on the initial nonporous films. The best results were obtained for PHB films where at Day 3 of the experiment the registered cell density for experimental samples arrived at 2.66(+/- 0.26) x 10(5) cells/cm(2) versus 1.29(+/- 0.33) x 10(5) cells/cm(2) in the control. So, the proposed method can be used to construct highly porous cell scaffolds for cellular engineering.

At the same time, the water vapor transmission rate considerably increased from 197.37 ± 23.62 to 934.03 ± 114.34 g/m2 /d for PHB films; from 1027.99 ± 154.10 to 7014.62 ± 280.81 g/m2 /d for PCL films; and from 715.47 ± 50.08 to 4239.09 ± 275.54 g/m2 /d for PHB/PCL films. Results of biocompatibility testing in the culture of NIH 3T3 mouse fibroblast cells showed that for the most of experimental samples cell adhesion and proliferation were comparable or superior to the corresponding parameters on the initial nonporous films. The best results were obtained for PHB films where at Day 3 of the experiment the registered cell density for experimental samples arrived at 2.66(±0.26) ? 105 cells/cm2 versus 1.29(±0.33) ? 105 cells/cm2 in the control. So, the proposed method can be used to construct highly porous cell scaffolds for cellular engineering. © 2020 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: Samples of olyhydroxyalkanoates (PHAs), polyhydroxybutyrate (PHB) and copolymers poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) with 4 and 18 mol% hydroxyvalerate, synthesized by the bacteria Ralstonia eutropha B5786, were investigated. PHA films in contact with blood did not activate the hemostasis system at the level of cell response, but they did activate the coagulation system and the complement reaction. To detect biologically-active components in the PHAs, a detailed analysis of the composition of the polymers was conducted. Gas chromatography-mass spectrometry revealed long-chain fatty acids (FAs) in the tested PHAs. Their total concentration in the polymer ranged from tenths of mol% to 2-3 mol%, depending on the purification method. C16:0 constituted the largest proportion, up to 70%. Of the long-chain hydroxy acids, only ?-OH-C14:0 was detected and it did not exceed 0.06 mol%. The analysis of the hemocompatibility properties of the PHAs purified by a specialized procedure, including the quantitative and morphological estimation of platelets adherent to the surface of polymer films, the plasma recalcification time and complement activation studies, indicated that PHB and PHBV can be used in contact with blood. It has been found out that the lipopolysaccharides of bacteria producing PHAs, which contain mostly long-chain hydroxy acids, can be the factor activating the hemostasis systems. Thus, the technology of PHA purification must satisfy rather stringent specific requirements.

Аннотация: The surface properties of poly-3-hydroxybutyrate (P3HB) membranes were modified using oxygen and an ammonia radio-frequency (RF, 13.56 MHz) plasma. The plasma treatment procedures used in the study only affected the surface properties, including surface topography, without inducing any significant changes in the crystalline structure of the polymer, with the exception being a power level of 250 W. The wettability of the modified P3HB surfaces was significantly increased after the plasma treatment, irrespective of the treatment procedure used. It was revealed that both surface chemistry and surface roughness changes caused by the plasma treatment affected surface wettability. A treatment-induced surface aging effect was observed and resulted in an increase in the water contact angle and a decrease in the surface free energy. However, the difference in the water contact angle between the polymers that had been treated for 4 weeks and the untreated polymer surfaces was still significant. A dependence between cell adhesion and proliferation and the polar component of the surface energy was revealed. The increase in the polar component after the ammonia plasma modification significantly increased cell adhesion and proliferation on biodegradable polymer surfaces compared to the untreated P3HB and the P3HB modified using an oxygen plasma. © 2016 Elsevier B.V.
