Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Results of the study of tissue response to biodegradable polyoxyalkanoate suture threads in the postoperative period are analyzed. These suture threads have been shown to have the required strength and meet a number of other requirements for currently available materials.

Аннотация: Tissue reaction to implantation of polymeric microparticles from resorbable polymer (polyhydroxybutyrate) is characterized by slight inflammatory reaction and pronounced progressive macrophage infiltration with the presence of mono-and multinuclear foreign body giant cells resorbing the polymeric matrix. No fibrous capsules were formed around the polymeric microparticles; neither necrosis nor other adverse morphological changes and tissue transformation in response to implantation of the PHB microparticles were recorded. The results indicate good prospects of using polyhydroxybutyrate for the construction of long-acting dosage forms as microparticles for intramuscular injection. Аннотация: The vertical distribution and abundance of Gammarus lacustris in the pelagic zone of two fishless meromictic lakes, L. Shira and L. Shunet, in Southern Siberia (Russia), was studied with the underwater video recording system and using vertical hauls. In both lakes, during summer stratification, Gammarus was distributed non-homogenously, with a stable peak in the metalimnion. The average depth of Gammarus population in the pelagic zone was significantly correlated with the depth of the thermocline. Gammarus abundances obtained using vertical plankton hauls with net were quite comparable with those obtained from video records. The peak abundance of Gammarus in the pelagic zone of the lakes observed with underwater video amounted up to 400 individuals m-2, while the peak animal densities in the metalimnion reached 50 ind. m-3. The data are compared with previously published abundances of Gammarus in the littoral of Lake Shira. Both littoral and pelagic can be equally important habitats for amphipods in meromictic lakes. The absence of fish in the pelagic zone, high oxygen concentration, low water temperature, increased seston concentration, elevated water density in the metalimnion and the anoxic hypolimnion can be the most probable combination of factors that are responsible for the peak of Gammarus in the metalimnion of these lakes. Аннотация: COVID-19 has specific characteristics that distinguish this disease from many other infec-tions. We suggest that the pathogenesis of severe forms of COVID-19 can be associated with acidosis. This review article discusses several mechanisms potentially linking the damaging effects of COVID-19 with acidosis and shows the existence of a vicious cycle between the development of hypoxia and acidosis in COVID-19 patients. At the early stages of the disease, inflammation, difficulty in gas exchange in the lungs and thrombosis collectively contribute to the onset of acidosis. In accordance with the Verigo-Bohr effect, a decrease in blood pH leads to a decrease in oxygen saturation, which contributes to the exacerbation of acidosis and results in a deterioration of the patient’s condition. A decrease in pH can also cause conformational changes in the S-protein of the virus and thus lead to a decrease in the affinity and avidity of protective antibodies. Hypoxia and acidosis lead to dysregu-lation of the immune system and multidirectional pro-and anti-inflammatory reactions, resulting in the development of a “cytokine storm”. In this review, we highlight the potential importance of supporting normal blood pH as an approach to COVID-19 therapy. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: Chloride salts are the most commonly used deicing materials for winter maintenance of roads. Numerous studies indicate a significant increase in the salinity of aquatic ecosystems associated with the long-term use of deicing materials in countries located in cold climates. The functioning of ecosystems largely depends on salinity, since salinity is one of the key factors determining the species composition, the structure of food webs and the productivity of aquatic communities. Given the growing threat of salinization of groundwater and surface waters, it is extremely important to study the effect of deicing materials on the biota and functioning of aquatic ecosystems. The aim of this research is to determine the threshold concentrations of solutions of the deicing salt mixture "Bionord" containing sodium and calcium chlorides, at which negative effects on the development of animal and plant test objects are observed. In this study, we used the salt-containing mixture "Bionord" as a model deicer. Similarly, with the most commonly used ice melting chemicals, the "Bionord" salt mixture contains a large amount of sodium and calcium chlorides (about 85% of the total weight). To evaluate the toxicity of the deicer solutions, we used acute and chronic toxicity tests with cladoceran Moina macrocopa (Straus, 1820) (Cladocera: Moinidae) and standard onion-based test with Allium cepa L. (Liliopsida: Amaryllidaceae) (Allium-test). In acute and chronic toxicity tests with Cladocera, the females on the first day of their life (body size 0.5-0.6 mm) were placed individually in jars with aged (not less than for 72 h) tap water with a volume of 20 ml with the addition of a deicer at a certain concentration. A group of animals that was placed in the medium without the deicer was used as a control. In the acute toxicity tests, we used the following concentration of the deicer: 1.3; 2.5; 4.0, 5.0; 6.0; 8.0; 10.0 g/l. The mortality of animals was recorded 24 and 48 hours after the start of the experiment. The concentration of the deicer (LC50) at which 50% of animals was observed to die, compared to the control, was determined in the acute toxicity test. In a chronic toxicity test, animals were tested in the following range of concentrations of the deicer: 0.3; 0.6; 1.3; 2.5; 5.0; 6.0 and 8.0 g/l. The chronic toxicity test was conducted until the death of all test animals. Based on the data obtained in the chronic toxicity test, the specific growth rate of juvenile females, average fecundity, and average life span of M. macrocopa were calculated for each concentration of the deicer. Bulbs of onions of the Stuttgartenrisen variety with a diameter of 1.8 +/- 0.1 cm and a weight of 2.27 +/- 0.17 g were used in the onion test. Bulbs with their bottoms were placed in test tubes containing 20 ml of a solute of the deicer or tap water for 48 hours. Three bulbs were tested for each concentration and for the control. The following concentrations of the deicer were used in the onion test: 1.0; 2.5; 5.0; 7.0; 10.0; 15.0; 20.0; 50.0 g/l. The general toxic and cytotoxic effects were evaluated in the onion test. The average root length and the total root length on each bulb were used as indicators of the total toxicity of the solutions of deicer. To evaluate proliferative activity, we calculated the mitotic index as the fraction of dividing cells in the apical root meristem to the total number of cells. Based on the results of the experiments, we determined median effective mixture concentrations (EC50) at which there is a 50% decrease, compared to the control, in the values of root growth indicators: average root length, sum of root lengths on each bulb and mitotic index. Median lethal concentration (LC50) of the deicing salt determined in the 48-hour acute toxicity test with females of Al. macrocopa was equal to 5.1 g/l. In the chronic test, we showed that the exposure to the solutions of the deicing salt in the range of concentrations from 0.3 to 5.0 g/l does not affect the life span, specific growth rate of juveniles and fecundity of females of M. macrocopa. The median effective concentration (EC50) of the deicing salt determined in the Allium-tests were 6.3, 5.2 and 10.4 g/l for the sum of root lengths, average root length on each bulb and proliferative activity at the tips of roots (mitotic index), respectively (See Table 1 and 2). Complete inhibition of onion root growth was observed at the concentration of the decider equal to 20 g/l, while the death of all test animals in the acute toxicity test occurred at the concentration of the deicer equal to 8,0 g/l (See Fig. 1). Thus, we demonstrated that similar concentrations of the deicer induced 50% inhibition of the growth of onion roots and 50% mortality of cladocerans. These values, in general, corresponded to a critical salinity of 5-8 %o above which qualitative changes occur both in the external and internal condition of aquatic animals. The electrical conductivity of the deicer solutions, which had a negative effect on the selected test species, coincides with the previously obtained values of the electrical conductivity of sodium chloride solutions harmful to cladocerans. We can assume that the main mechanism of the effect of the deicing material that we study is associated with the biological effect of its chlorine and sodium salts Taking this into account, the value of electrical conductivity measured for solutions of deicing salt can be used to assess its negative potential effects. We estimated that in the absence of timely cleaning, regulated by the rules for using the material, the runoff from each square meter of the treated surface can lead to the pollution of 8-13 liters of fresh water. Thus, the basic requirement for the use of deicing salts on roads is the need to comply with the cleaning regime of the treated surfaces. Otherwise, the gradual accumulation of sodium and calcium chlorides in water bodies can cause an increase in salinity which will affect the survival of freshwater aquatic organisms and lead to serious disturbances in the functioning of aquatic ecosystems.

Аннотация: Chloride salts are the most commonly used deicing materials for winter maintenance of roads. Numerous studies indicate a significant increase in the salinity of aquatic ecosystems associated with the long-term use of deicing materials in countries located in cold climates. The functioning of ecosystems largely depends on salinity, since salinity is one of the key factors determining the species composition, the structure of food webs and the productivity of aquatic communities. Given the growing threat of salinization of groundwater and surface waters, it is extremely important to study the effect of deicing materials on the biota and functioning of aquatic ecosystems. The aim of this research is to determine the threshold concentrations of solutions of the deicing salt mixture “Bionord” containing sodium and calcium chlorides, at which negative effects on the development of animal and plant test objects are observed. In this study, we used the salt-containing mixture “Bionord” as a model deicer. Similarly, with the most commonly used ice melting chemicals, the «Bionord» salt mixture contains a large amount of sodium and calcium chlorides (about 85% of the total weight). To evaluate the toxicity of the deicer solutions, we used acute and chronic toxicity tests with cladoceran Moina macrocopa (Straus, 1820) (Cladocera: Moinidae) and standard onion-based test with Allium cepa L. (Liliopsida: Amaryllidaceae) (Allium-test). In acute and chronic toxicity tests with Cladocera, the females on the first day of their life (body size 0.5-0.6 mm) were placed individually in jars with aged (not less than for 72 h) tap water with a volume of 20 ml with the addition of a deicer at a certain concentration. A group of animals that was placed in the medium without the deicer was used as a control. In the acute toxicity tests, we used the following concentration of the deicer: 1.3; 2.5; 4.0, 5.0; 6.0; 8.0; 10.0 g/l. The mortality of animals was recorded 24 and 48 hours after the start of the experiment. The concentration of the deicer (LC50) at which 50% of animals was observed to die, compared to the control, was determined in the acute toxicity test. In a chronic toxicity test, animals were tested in the following range of concentrations of the deicer: 0.3; 0.6; 1.3; 2.5; 5.0; 6.0 and 8.0 g/l. The chronic toxicity test was conducted until the death of all test animals. Based on the data obtained in the chronic toxicity test, the specific growth rate of juvenile females, average fecundity, and average life span of M. macrocopa were calculated for each concentration of the deicer. Bulbs of onions of the Stuttgartenrisen variety with a diameter of 1.8 ± 0.1 cm and a weight of 2.27 ± 0.17 g were used in the onion test. Bulbs with their bottoms were placed in test tubes containing 20 ml of a solute of the deicer or tap water for 48 hours. Three bulbs were tested for each concentration and for the control. The following concentrations of the deicer were used in the onion test: 1.0; 2.5; 5.0; 7.0; 10.0; 15.0; 20.0; 50.0 g/l. The general toxic and cytotoxic effects were evaluated in the onion test. The average root length and the total root length on each bulb were used as indicators of the total toxicity of the solutions of deicer. To evaluate proliferative activity, we calculated the mitotic index as the fraction of dividing cells in the apical root meristem to the total number of cells. Based on the results of the experiments, we determined median effective mixture concentrations (EC50) at which there is a 50% decrease, compared to the control, in the values of root growth indicators: average root length, sum of root lengths on each bulb and mitotic index. Median lethal concentration (LC50) of the deicing salt determined in the 48-hour acute toxicity test with females of M. macrocopa was equal to 5.1 g/l. In the chronic test, we showed that the exposure to the solutions of the deicing salt in the range of concentrations from 0.3 to 5.0 g/l does not affect the life span, specific growth rate of juveniles and fecundity of females of M. macrocopa. The median effective concentration (EC50) of the deicing salt determined in the Allium-tests were 6.3, 5.2 and 10.4 g/l for the sum of root lengths, average root length on each bulb and proliferative activity at the tips of roots (mitotic index), respectively (See Table 1 and 2). Complete inhibition of onion root growth was observed at the concentration of the decider equal to 20 g/l, while the death of all test animals in the acute toxicity test occurred at the concentration of the deicer equal to 8,0 g/l (See Fig. 1). Thus, we demonstrated that similar concentrations of the deicer induced 50% inhibition of the growth of onion roots and 50% mortality of cladocerans. These values, in general, corresponded to a critical salinity of 5-8 % above which qualitative changes occur both in the external and internal condition of aquatic animals. The electrical conductivity of the deicer solutions, which had a negative effect on the selected test species, coincides with the previously obtained values of the electrical conductivity of sodium chloride solutions harmful to cladocerans. We can assume that the main mechanism of the effect of the deicing material that we study is associated with the biological effect of its chlorine and sodium salts. Taking this into account, the value of electrical conductivity measured for solutions of deicing salt can be used to assess its negative potential effects. We estimated that in the absence of timely cleaning, regulated by the rules for using the material, the runoff from each square meter of the treated surface can lead to the pollution of 8-13 liters of fresh water. Thus, the basic requirement for the use of deicing salts on roads is the need to comply with the cleaning regime of the treated surfaces. Аннотация: A mathematical model was used to investigate the effect of cannibalism intensity on the net primary production and the dynamics of trophic links in an aquatic ecosystem characterized by cannibalism at the upper trophic level. A mathematical model of an aquatic ecosystem has been constructed, with the following principal trophic links: limiting nutrient concentration, producers (phytoplankton), nonpredatory and predatory zooplankton. The model takes into account the age structure of the predator and includes two age groups (the young and adults). The adult predators are cannibals feeding on both nonpredatory zooplankton and their own young, which consume phytoplankton. It has been found that when cannibalism intensity increases above a certain level, the concentrations of both adults and the young of the predators decrease. At the same time, the concentrations of the nonpredatory zooplankton and of nutrients increase, while the biomass of producers decreases. When the cannibalism intensity is low, the net primary production of the system increases to a certain level correlated with the increase in cannibalism intensity and drops sharply when the level of consumption of young is high. Аннотация: Background: Canine parvovirus (CPV) is now recognized as a serious threat to the dog breeding industry worldwide. Currently used CPV vaccines all have their specific drawbacks, prompting a search for alternative safe and effective vaccination strategies such as subunit vaccine. VP2 protein is the major antigen targeted for developing CPV subunit vaccine, however, its production in baculovirus expression system remains challenging due to the insufficient yield. Therefore, our study aims to increase the VP2 protein production by using an improved baculovirus expression system and to evaluate the immunogenicity of the purified VP2 protein in mice. Results: The results showed that high-level expression of the full length VP2 protein was achieved using our modified baculovirus expression system. The recombinant virus carrying two copies of VP2 gene showed the highest expression level, with a productivity of 186 mg/L, which is about 1.4-1.6 fold that of the recombinant viruses carrying only one copy. The purified protein reacted with Mouse anti-His tag monoclonal antibody and Rabbit anti-VP2 polyclonal antibody. BALB/c mice were intramuscularly immunized with purified VP2 protein twice at 2 week intervals. After vaccination, VP2 protein could induce the mice produce high level of hemagglutination inhibition antibodies. Conclusions: Full length CPV VP2 protein was expressed at high level and purified efficiently. Moreover, it stimulated mice to produce high level of antibodies with hemmaglutination inhibition properties. The VP2 protein expressed in this study could be used as a putative economic and efficient subunit vaccine against CPV infection. © 2020 The Author(s).

Аннотация: Antitumor efficiency of an experimental form of an experimental form of anthracyclin antibiotic (doxorubicin), resorbable microparticles from Bioplastotaneв„ў, was studied on laboratory mice with transplanted Ehrlich's solid carcinoma. Use of the experimental form of the cytostatic in polymeric microparticles from resorbable Bioplastotaneв„ў in animals with solid tumor led to inhibition of the cancerous process, comparable to that in response to intravenous free doxorubicin, but without negative effects on the blood system. Аннотация: Chronic experiments on mature Wistar rats studied the biological properties of suture threads from two types of polyoxyalkanoates (POA), such as polyoxybutyrate (POB) and the copolymer of POB and polyoxyvalerate (POV) (POB/POV) as compared to the conventional surgical material (silk and catgut). The physiological and biochemical parameters that reflect the status of the animals, including their behavior, height and development, the weight of the body and viscera, the morphology of peripheral blood, the biochemical parameters of blood, the activity of some enzymes, as well as histological findings of the viscera, and lymphoid tissue responses were assessed. POA was found to produce no negative effect on the physiological, biochemical, and functional characteristics of the animals irrespective of the duration of contact with the internal medium of the body and of the chemical composition of a material.
