Аннотация: The present study describes production of bacterial cellulose composites with silver nanoparticles and antibiotics and compares their properties. Bacterial cellulose (BC) composites synthesized in the culture of the strain of acetic acid bacterium Komagataeibacter xylinus VKPM B-12068 with silver nanoparticles, BC/AgNps, were produced hydrothermally, under different AgNO3 concentrations (0.0001, 0.001, and 0.01 M) in the reaction medium. The presence of silver in the BC/AgNp composites was confirmed by elemental analysis conducted using scanning electron microscopy with a system of X-ray spectral analysis. Analysis showed that the average atomic number of silver particles in composite samples depended on the concentration of AgNO3: as AgNO3 concentration in the reaction solution was increased, silver content in the composites increased from 0.044 to 0.37 mg/cm2. BC composites with amikacin and ceftriaxone were prepared by immersing dry BC films in solutions containing different concentrations of the antibiotics. The surface structure and properties and physicochemical and mechanical characteristics of composites were investigated using SEM, DSC, X-ray analysis, the system for measuring water contact angles, and electromechanical tensile testing machine. The disk-diffusion method and the shake-flask culture method used in this study showed that all experimental composites had pronounced antibacterial activity against E. coli, Ps. eruginosa, K. pneumoniae, and St. aureus, and the BC/antibiotic composites were more active than BC/AgNp ones; S. aureus was the most susceptible to the effect of BC composites. No potential cytotoxicity was detected in any of the BC/AgNp composites in the NIH 3T3 mouse fibroblast cell culture, in contrast to the BC/antibiotic composites. These results suggest that BC composites constructed in the present study hold promise as dressings for managing wounds, including contaminated ones. © 2017 Elsevier Ltd

Аннотация: The possibility of creating new screening methods with a cell-free translation system has been demonstrated with a quantitative determination of diphtheria toxin and some antibiotics (puromycin, kanamycin and tetracycline) as examples. The approach proposed follows from the ability of various substances to inhibit protein synthesis. We used a wheat-germ cell-free translation system stabilized by freeze-drying in the presence of trehalose with the mRNA of the Ca2+-activated photoprotein obelin as a reporter template. This freeze-dried cell-free translation system allows prolonged storage of the detecting system before it is required, increases the reproducibility of the results and simplifies the application procedure. The obelin mRNA extends the sensitivity of the method owing to the high sensitivity of detection of the synthesized protein.

Аннотация: An experimental dosage form of rubomycin is developed: the drug is incorporated in absorbable polymeric (polyhydroxybutyrate) matrix in the form of microparticles. Antitumor efficiency of this rubomycin dosage form was studied in laboratory mice with transplanted Ehrlich ascitic carcinoma. Rubomycin deposited in polymeric microparticles exhibited pronounced antitumor activity, inhibited the proliferative activity of Ehrlich ascitic carcinoma, and improved survival of mice with tumors. Аннотация: From 1996 to 1999 heterotrophic bacteria of the brackish-water Lake Shira (Republic of Khakasia, Russia) were studied to understand the seasonal dynamics of their antibiotic resistance. During the winter, these bacteria were represented primarily by forms that could not be cultured and were psychrotolerant. In the summer period, heterotrophic, mesophilic bacteria increased in number. The percentages of isolates with multiple, antibiotic resistance isolated from the lake region near the resort area of the lake were 2-3 times higher than those from the central part of the lake. A decline in the bacterial numbers with multiple antibiotic resistance was observed during the cold period (February-March). Various mechanisms of multiple, antibiotic resistance of heterotrophic bacteria isolated from Shira lake are discussed.

The population dissociated into R and S variants differing in the size, appearance, and architectonics of colonies, cell ultrastructure, growth rate, enzyme activity, and chemical composition. Аннотация: Procedure for microencapsulation providing stable formation of high quality microspheres was designed. Conditions for deposition of the anthracycline antibiotic daunorubicin to polymer matrix were developed and microsheres loaded with various quantities of the drug were prepared. The kinetics of the in vitro and in vivo release of daunorubicin from the microsheres was studied. The rate of the rubomycin release to the medium in vitro (balanced phosphate buffer at 37.8В°C) in a 300-hour experiment directly depended on the quantity of the incorporated drug and averaged 0.81 В· 10 -4 to 2.3 В· 10 -4 mcg/mlВ·h. The experiment on laboratory animals with intraperitoneal administration of the rubomycin microsperes showed that the drug remained in the blood and abdominal liquid for a long time (up to 10 days). Possible control of the quantity of the rubomycin encapsulation to the polymer matrix, no sharp efflux of the drug at the early stages of the observation and low rate of the drug release to the medium allowed to conclude that the use of the biodegradable polymer microsperes as carriers of the high toxic antibiotic providing its prolonged action was prospective.

Аннотация: Resistance to Ampicillin and Kanamycin displayed by heterotrophic bacteria isolated in Summer and in Spring from the littoral and the central parts of Lake Shira (a therapeutic lake in the Khakasia Republic, Russia) has been investigated. It has been found that in Summer, human and animal microflora featuring multiple antibiotic resistance (to Ampicillin and Kanamycin) predominates in all the studied stations of the littoral zone of the lake. In Spring, concentrations of bacteria featuring multiple antibiotic resistance decrease significantly and bacteria sensitive to antibiotics predominate in the lake. Emergence of multiple antibiotic resistance in bacteria of Lake Shira is caused by the input of allochthonous bacteria into the lake; this feature of heterotrophic bacteria of Lake Shira can be used to monitor the impact on the ecosystem made by health resorts. Аннотация: The application of detonation nanodiamonds (DNDs) in delivery systems has been demonstrated many times for a variety of substances, including anti-inflammatory drugs, antibiotics, antineoplastic agents, vitamins etc. The applicability of detonation nanodiamonds as a carrier for delivery of corticosteroids is investigated in this study. Commercial DNDs were modified to obtain particles with high colloidal stability in aqueous suspensions. Physical adsorption was then used to conjugate the DNDs with methylprednisolone sodium succinate and dexamethasone sodium phosphate. The experimental adsorption isotherms were fit with Langmuir and Freundlich adsorption models. The results of in vitro release study demonstrated an explicit prolonged effect of DNDs, due to which a sustained release of the adsorbed drugs was provided.

Аннотация: Major advances in the development and application of the bioluminescent analysis to detect certain biologically active substances are discussed. The main merit of the method lies in its high sensitivity and specificity along with its simplicity and rapid performance. The available methodologies allow for detection of substances of varying nature: Ca2+, ATP, FMN, NAD(P), long-chain aldehydes, ATP- and NAD(P)-dependent enzymes and their substrates, many xenobiotics and antibiotics, and mutagens. The bioluminescence methodologies may be widely applied in clinical laboratory diagnosis.

Аннотация: A procedure for preparation of microspheres from biodegradable linear polyether of microbiological origin (polyhydroxybutirate, PHB) with using the technology of solvent evaporation was developed considering a specific example of two- and three-component emulsions. The procedure provided permanent preparation of the microspheres of high quality. The influence of the procedure (emulsion type, dispersion process and medium temperature) on the yield of the microspheres, their structure and size was shown. The temperature had a significant impact on incorporation of the antitumor anthracycline antibiotic daunorubicin (rubomycin) to the polymer matrix. The microspheres with various levels of the drug load (29 and 90% of the initial content in the emulsion) were prepared and the kinetics of the in vitro rubomycin release was studied. The dynamics of the highly toxic rubomycin release from the microspheres was on the whole even with the curve profile reaching the plateau in 20-22 hours of the observation period. The rate of the rubomycin release to the medium depended on the value of the antibiotic incorporation and was maximum within the first two hours (3.3 and 13.0 mcg/mlВ·h) that corresponded to the release of 0.97 and 3.89 of the incorporated antibiotic. The average rate of rubomycin release during 300 hours was 0.81В·10-4 and 2.3В·10-4 mcg/mlВ·h. The release constituted respectively 3.9 and 13.11% of the antibiotic incorporated to the microspheres.
