Аннотация: Colonies grown from vegetative Bacillus subtilis 2335 cells had a standard structure, with bacillar cells occupying the whole colony volume. At the same time, the colonies of this bacterium grown from germinated spores had an abnormal structure characterized by the location of cells in a surface layer 100-200 mum thick at the colony boundary with the air. The glycocalyx of the colonies grown from spores was characterized by a wetting angle theta(e) of 120degrees-160degrees, whereas that of the colonies grown from vegetative cells had an angle theta(c) as low as 5degrees-30degrees. It is suggested that spores and vegetative cells follow different strategies of substrate colonization and that the architectonics of bacterial colonies is determined by the physicochemical properties of the glycocalyx.

Аннотация: A specific index of nucleotide sequence redundancy, the specific restriction length of a finite frequency dictionary, was determined for a complete set of genes in some viral genomes and a genome of a bacterium, Bacillus subtilis. The distribution of the gene number over the specific restriction length was shown to be bimodal for viral genomes and unimodal for the Bac. subtilis genome. These results agree with earlier data.

Аннотация: The natural microflora of Middle Siberian karst cavities, which comprises psychrotolerant bacteria and fungi capable of growing at 3-15 and 3...+28degreesC, respectively, has been studied. Bacteria are ubiquitous in caves, their count varying from 10(3) to 10(7) cells/g ground. The bacteria have been identified as Pseudomonas, Arthrobacter, Bacillus, and coryneform bacteria. Fungi have been found in places exposed to increased anthropogenic impact, their count being as large as 10(6) to 10(7) cells/g ground. Mucor Penicillium, and Chrysosporium were dominant fungal genera.

Аннотация: The role of key environmental factors in adaptation of spore-forming and non-spore-forming transgenic microorganisms (TM) have been studied in model ecosystems. Model TM Escherichia coli Z905 (bearing plasmid genes of bacterial luminescence Ap(r)Lux(+)) has been found to have a higher adaptation potential than TM Bacillus subtilis 2335/105 (bearing genes of human alpha 2-interferon Km(r)lnf(+)), planned for employment as a living vaccine under varying environmental conditions. Effects of abiotic factors on migration of natural and recombinant plasmids between microorganisms under model ecosystem conditions has been estimated. The transgenic microorganisms with low copy number survived better under introduction conditions in the microcosms studied. Аннотация: Adaptation of the Bacillus subtilis strain 2335/105 (Km(r)Inf(+)) containing a recombinant plasmid encoding the extracellular human interferon alpha 2 was studied under various conditions. Stability of the plasmid in the population of B. subtilis 2335/105 was estimated under nonselective conditions. The plasmid-free cells and cells with a low number of plasmid copies were found to accumulate progressively, constituting 80% of the population after 10 culture passages, indicating the poor competitiveness of cells carrying a high number of plasmid copies. The behavior of vegetative cells of the recombinant strain introduced into aquatic microcosms differing in trophic chain length was studied. Within the first 10 days, the lysis of vegetative cells of B. subtilis 2335/105 occurred; the number of viable spores was very low but remained constant for half a year.

Аннотация: Biodegradation of microbial linear polymers of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) by soil microbial communities of different structures has been studied during two field seasons in different weather conditions. This process was shown to be influenced by the polymer chemical composition, temperature, humidity, and the microbial soil component. The PHA degradation was accompanied by a decrease in the polymer molecular weight and an increase in the degree of crystallinity, indicating the preferential destruction of the amorphous phase compared to the crystalline one. The quantity of the true PHA destructors developing at the surface of the polymer samples was lower than the quantity of accompanying bacteria. The dominant PHA degrading microorganisms under the test conditions were identified as bacteria of the genera Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus, and Xanthomonas and as micromycetes from Penicillium, Paecilomyces, Acremonium, Verticillium, and Zygosporium.

Аннотация: Electron microscopy has shown that the colonies obtained as a result of joint growth of bacterial monocultures Flavobacterium sp. 56 and Bacillus cereus 60 consist of three main cell types. Two cell types of the mixed colonies preserve the fine structure specific for bacteria in the monocultures. At the same time, formation of a large amount of the third type of cells is stimulated, which occur as single cells in the monoculture Flavobacterium sp. 56. The cell types are not intermixed in the colony volume and have certain spatial positions.

Аннотация: Dynamics of catalase activity has been shown to reflect the growth curve of microorganisms in batch cultivation (celluloselythic bacteria Bacillus acidocaldarius and bacteria of the associated microflora Chlorella vulgaris). Gas and substrate closure of the three component ecosystems with spatially separated components "producer-consumer-reducer" (Chl, vulgaris-Paramecium caudatum-B. acidocaldarius, two bacterial strains isolated from the associated microflora Chl. vulgaris) demonstrated that the functioning of the reducer component can be estimated by the catalase activity of microorganisms of this component. Аннотация: The study deals with the effects of herbicides (metribuzin, tribenuron-methyl, fenoxaprop-P-ethyl) and fungicides (tebuconazole, epoxiconazole, azoxystrobin) applied to soil as free pesticides or as slow release formulations embedded in a biodegradable composite matrix on the structure of the soil microbial community. The matrix consisted of a natural biopolymer poly-3-hydroxybutyrate [P(3HB)] and a filler-one of the natural materials (peat, clay, and wood flour). The soil microbial community was characterized, including the major eco-trophic groups of bacteria, dominant taxa of bacteria and fungi, and primary P(3HB)-degrading microorganisms, such asPseudomonas,Bacillus,Pseudarthrobacter,Streptomyces,Penicillium, andTalaromyces. The addition of free pesticides adversely affected the abundance of soil microorganisms; the decrease varied from 1.4 to 56.0 times for different types of pesticides. The slow release pesticide formulations, in contrast to the free pesticides, exerted a much weaker effect on soil microorganisms, no significant inhibition in the abundance of saprotrophic bacteria was observed, partly due to the positive effects of the composite matrix (polymer/natural material), which was a supplementary substrate for microorganisms. The slow release fungicide formulations, like the free fungicides, reduced the total abundance of fungi and inhibited the development of the phytopathogensFusariumandAlternaria. Thus, slow release formulations of pesticides preserve the bioremediation potential of soil microorganisms, which are the main factor of removing xenobiotics from the biosphere.

Аннотация: The present study investigates, for the first time, the structure of the microbial community of cryogenic soils in the subarctic region of Siberia and the ability of the soil microbial community to metabolize degradable microbial bioplastic - poly-3-hydroxybutyrate [P(3HB)]. When the soil thawed, with the soil temperature between 5-7 and 9-11 degrees C, the total biomass of microorganisms at a 10-20-cm depth was 226-234 mg g(-1) soil and CO2 production was 20-46 mg g(-1)W day(-1). The total abundance of microscopic fungi varied between (7.4 +/- 2.3) x 10(3) and (18.3 +/- 2.2) x 10(3) CFU/g soil depending on temperature; the abundance of bacteria was several orders of magnitude greater: (1.6 +/- 0.1) x 10(6) CFU g(-1) soil. The microbial community in the biofilm formed on the surface of P(3HB) films differed from the background soil in concentrations and composition of microorganisms. The activity of microorganisms caused changes in the surface microstructure of polymer films, a decrease in molecular weight, and an increase in the degree of crystallinity of P(3HB), indicating polymer biodegradation due to metabolic activity of microorganisms. The clear-zone technique e plating of isolates on the mineral agar with polymer as sole carbon source e was used to identify P(3HB)-degrading microorganisms inhabiting cryogenic soil in Evenkia. Analysis of nucleotide sequences of rRNA genes was performed to identify the following P(3HB)degrading species: Bacillus pumilus, Paraburkholderia sp., Pseudomonas sp., Rhodococcus sp., Stenotrophomonas rhizophila, Streptomyces prunicolor, and Variovorax paradoxus bacteria and the Penicillium thomii, P. arenicola, P. lanosum, Aspergillus fumigatus, and A. niger fungi. Аннотация: The paper dwells upon the use of autochthonous microbial cinosis as a source of antagonistic strains for bioprotection of wheat against Fusarium in biotechnical life support systems (BT LSS). Six bacterial strains antagonistic to 9 phytopathogenic fungi Fusarium responsible for seedling blight of wheat in BT LSS were isolated from hydroponic solutions and artificial soils and subject to the genetic typing. Five strains represent bacteria g. Bacillus, one strain - Chryseobacterium. The 16S rRNA sequencing showed 94 to 100 % genetic affinity with typical cultures. All spore-forming strains-antagonists were capable of germinating and going through the whole development cycle In the presence of swelling wheat seeds. The isolates did not demonstrate antagonism to each other and can grow in a mixed culture. Spectra and levels of the antiobiotic activity of the antagonists, along with the Fusarium sensitivity to their antibiotic action, differed statistically (p 0.001). None of the antagonists was capable of suppressing the entire Fusarium spectrum detected in BT LSS individually. At the same time, treatment of seeds with a mixture of antagonists suppressed totally fungal development in wheat artificially inoculated by Fusarium conidia from a BT LSS roll culture, and had a statistical stimulating effect (p

Аннотация: The effect of high temperatures (35 and 45 degreesC) on microflora of the root zone of radish plants grown in phytotron was evaluated by the response of microorganisms from 9 indicator groups. Phytotron air temperature elevated to 35 degreesC for 20 hours caused no significant changes in qualitative and quantitative composition of the root microflora in experimental plants. By the end of the experiment, the species diversity of microflora had changed. The amount of phytopathogenic microorganisms decreased which can be interpreted as more stable co-existence of microflora with plants. The numbers of microbes from other indicator groups was in dynamic equilibrium. The plants' condition did not deteriorate either. Exposure to the temperature of 45 degreesC for 7 hours have been found to change the numbers and species diversity in the radish root zone microflora. The microorganisms were observed to increase their total numbers at the expense of certain indicator groups. Bacteria increased spore forms at the stage of spores. Colon bacillus bacteria of increased their numbers by the end,of experiment by an order. By the end of experiment the roots of experiment plants had microscopic fungi from. Mucor, Aspergillus, Trichoderma, Cladosporium genera. The observed changes in the microbial complex seem to be associated with the changes of root emissions and general deterioration of the plants' condition. It is suggested that the response of the microorganisms can be indicative of the condition of plants under investigation. (C) 2002 Published by Elsevier Science Ltd on behalf of COSPAR.

Аннотация: The study addressed the ability of the transgenic strain (TM) B. subtilis 2335/pBMB 105 (Km(r)Inf(+)) to form biofilms on the surface of liquid media of various compositions, inoculated with vegetative cells and spores. The morphological features of these biofilms do not differ from those of the films formed by the recipient strain (WT) B. subtilis 2335 (Km(s)). However, the TM and the natural one differ in the dynamics of biofilm formation and the cellular composition of the films. Biofilms of the TM are formed earlier, develop at a higher rate, but decompose later than the films of the WT. When the medium is inoculated with vegetative cells, sporulation in the biofilms of both strains undergoes glucose repression; no such effect is observed when the medium is inoculated with spores. The TM does not form films when the medium is inoculated with spores and supplemented with glycerin and kanamycin.

Аннотация: The study addresses degradation of polyhydroxyalkanoates (PHA) with different chemical compositions-the polymer of 3-hydroxybutyric acid [P(3HB)] and copolymers of P(3HB) with 3-hydroxyvalerate [P(3HB/3HV)], 4-hydroxybutyrate [P(3HB/4HB)], and 3-hydroxyhexanoate [P(3HB/3HHx)] (10-12 mol%)-in the agro-transformed field soil of the temperate zone. Based on their degradation rates at 21 and 28 A degrees C, polymers can be ranked as follows: P(3HB/4HB) P(3HB/3HHx) P(3HB/3HV) P(3HB). The microbial community on the surface of the polymers differs from the microbial community of the soil with PHA specimens in the composition and percentages of species. Thirty-five isolates of bacteria of 16 genera were identified as PHA degraders by the clear zone technique, and each of the PHA had both specific and common degraders. P(3HB) was degraded by bacteria of the genera Mitsuaria, Chitinophaga, and Acidovorax, which were not among the degraders of the three other PHA types. Roseateles depolymerans, Streptomyces gardneri, and Cupriavidus sp. were specific degraders of P(3HB/4HB). Roseomonas massiliae and Delftia acidovorans degraded P(3HB/3HV), and Pseudoxanthomonas sp., Pseudomonas fluorescens, Ensifer adhaerens, and Bacillus pumilus were specific P(3HB/3HHx) degraders. All four PHA types were degraded by Streptomyces.

Аннотация: The study addresses the effect of abiotic (medium salinity and copper ions) and biotic (interactions between populations) factors on the formation of structured communities by binary associations consisting of halotolerant bacteria (Alcaligenes sp. 1-1 or Acinetobacter sp. 1-19) and a wild-type B. subtilis 2335 strain or a transgenic strain. The results showed that 250 mg l(-1) of copper ions inhibit formation of biofilms by monocultures of the tested strains. Binary associations of the strains were more resistant to high concentrations (250 mg l(-1)) of copper ions. At the lowest NaCl concentration (0.05% and 2.5%) and in the presence of copper ions, bacilli seemed to help halotolerant bacteria survive. Under increased salinity and in the presence of copper ions, structured communities developed due to halotolerant bacteria. Coexistence under stressful conditions was beneficial for the both groups of bacteria.

Аннотация: The copy number of R plasmids weakly depends on the selective pressure of the respective antibiotic but does depend on the physiology of the host species and the type of plasmids and cloned genes, whose expression leads to a further load on the biosynthetic apparatus of cells. The last factor is critical in the maintenance of recombinant plasmids in transgenic microorganisms.

Аннотация: The soil-like substrate (SLS), which is a potential candidate for employment in closed ecological systems, has been tested. CO2 exchange, composition and numbers of soil microflora have been examined in .wheat-SLS. system. The results produced have been compared to analogous characteristics of .wheat-neutral substrate. system. A hydroponic method was used under wheat growing on the neutral substrate (expanded clay aggregate). Plants for both studies were grown in closed environment from seed to physiological maturity. In the .wheat-SLS. system, the net photosynthetic rate of canopy was positive in the course of 6.55 days after planting. The net photosynthetic rate of canopy in .wheat-neutral substrate. system was positive in the entire course of vegetation. According to calculations in the course of vegetation, photosynthesis has withdrawn 3.28 kg m-2 CO2 from the .wheat-SLS. system and 3.40 kg m-2 CO2 from the .wheat-neutral substrate. system. On the SLS dominant among bacteria were the spore-forming bacteria from Bacillus genus, among fungi . from Trichoderma genus. In the hydroponic cultivation on neutral substrate dominant were bacteria from Pseudomonas genus, most commonly found fungi were species from Fusarium and Botrytis genera.
