Аннотация: 1. Aquatic systems are considered to be a main source of essential long-chain n-3 polyunsaturated fatty acids (PUFA), which are preferentially synthesized by microalgae and transferred along food chains to terrestrial consumers. Emerging aquatic insects comprise a significant part of this transfer of the essential PUFA from water to land. Quantitative data on PUFA content and composition are available mainly for rheophilic insects while taxa that are characteristic of wetlands and stagnant water bodies, such as aquatic Heteroptera, remain relatively unstudied. 2. We investigated the role of various waterbug taxa (Heteroptera: Nepomorpha and Gerromorpha) inhabiting different ecoregions in temperate Central Siberia (Russia) as potential sources of PUFA. The ecoregions were steppe, forest-steppe, hemiboreal forest and montane coniferous forest. Although these waterbugs insects are aquatic in both larval and adult stages, they can disperse through the landscape and transfer essential PUFAs from water to land so making them potentially available to terrestrial consumers. 3. Species of Naucoridae, Notonectidae and Corixidae were generally dominant in all ecoregions, attaining maximum biomass in the steppe. We showed that habitat or ecoregion played a major role in the variability of fatty acid composition of Notonectidae and Gerridae but not Corixidae. In contrast, the biochemical composition of the only naucoridae, Ilyocoris cimicoides, was largely affected by life stage. 4. Both the dominant families and species within them differed with respect to their mass-specific contents of essential long-chain PUFA of the n-3 family, namely eicosapentaenoic and docosahexaenoic acids. Corixid species had the highest content of these two essential PUFAs among the waterbug studies, and relative to literature reports for other aquatic insects. Corixids thus appear to be a potentially important vector for transfer of the essential biochemical compounds from water to land, especially in steppe ecoregions with numerous ephemeral water bodies.

Аннотация: Bryophytes are abundant in streams and are a habitat for many invertebrates, but their contribution to the diet of fluvial zoobenthos is still debated. To estimate the amount of bryophyte-derived organic matter assimilated by benthic invertebrates, we used a combination of fatty acid and stable isotope analyses during a four-year monthly study of a littoral site in the Yenisei River (Siberia, Russia). Acetylenic acids, which are highly specific biomarkers of the water moss Fontinalis antipyretica, were found in lipids of all dominant benthic animals: gammarids, ephemeropterans, chironomids and trichopterans. The dominant zoobenthic species, Eulimnogammarus viridis, had maximum levels of the biomarkers in its biomass during winter, and minimum levels in summer. The zoobenthos in the studied site regularly consume and assimilate bryophyte-derived organic matter as a minor supplemental food. Аннотация: Coelenteramide-containing fluorescent proteins are products of bioluminescent reactions of marine coelenterates. They are called ‘discharged photoproteins’. Their light-induced fluorescence spectra are variable, depending considerably on external conditions. Current work studies a dependence of light-induced fluorescence spectra of discharged photoproteins obelin, aequorin, and clytin on excitation energy. It was demonstrated that photoexcitation to the upper electron-excited states (260–300 nm) of the discharged photoproteins initiates a fluorescence peak in the near UV region, in addition to the blue-green emission. To characterize the UV fluorescence, the light-induced fluorescence spectra of coelenteramide (CLM), fluorophore of the discharged photoproteins, were studied in methanol solution. Similar to photoproteins, the CLM spectra depended on photoexcitation energy; the additional peak (330 nm) in the near UV region was observed in CLM fluorescence at higher excitation energy (260–300 nm). Quantum chemical calculations by time depending method with B3LYP/cc-pVDZ showed that the conjugated pyrazine-phenolic fragment and benzene moiety of CLM molecule are responsible for the additional UV fluorescence peak. Quantum yields of CLM fluorescence in methanol were 0.028 ± 0.005 at 270–340 nm photoexcitation. A conclusion was made that the UV emission of CLM might contribute to the UV fluorescence of the discharged photoproteins. The study develops knowledge on internal energy transfer in biological structures – complexes of proteins with low-weight aromatic molecules. © 2016 Elsevier B.V.

Аннотация: For the first time comparative data on localization, structural-functional organization, optimal functioning conditions for two bioluminescent systems of soil enchytraeids Fridericia heliota and Henlea sp. are given. Strong difference of these systems from each other and from all known among Oligochaeta class is shown. The analysis allows stating the discovery of two new systems of bioluminescent systems of enchytraeids, and accounting peroxide-dependent system of Diplocardia longa megascolecid, establishing multiplicity of luminescent mechanisms for Oligochaeta earth-worm.

Аннотация: The trophic transfer efficiency (TTE) is an important indicator of ecosystem functioning. However, TTE data from freshwater food webs are ambiguous due to differences in time scales and methods. We investigated the transfer of essential substances (carbon, nutrients, and polyunsaturated fatty acids) through plankton communities in 30 Polish lakes with different trophic status in the middle of summer. The results of our study revealed that different essential substances were transferred from phytoplankton to zooplankton with varying efficiencies. The average TTE of C, N, P, and the sum of ?-3 PUFA were 6.55%, 9.82%, 15.82%, and 20.90%, respectively. Our results also show a large mismatch between the elemental and biochemical compositions of zooplankton and their food during the peak of the summer stagnation, which may further promote the accumulation of essential substances. There were also large differences in TTEs between trophic conditions, with the highest efficiencies in oligotrophic lakes and the lowest in dystrophic and eutrophic lakes. Therefore, our study indicates that disturbances like eutrophication and dystrophication similarly decrease the TTE of essential substances between phytoplankton and zooplankton in freshwater food webs. © 2021 The Authors. Аннотация: Fullerene is a nanosized carbon structure with potential drug delivery applications. We studied the bioeffects of a water-soluble fullerene derivative, fullerenol, with 10-12 oxygen groups (F10-12); its structure was characterized by IR and XPS spectroscopy. A bioluminescent enzyme system was used to study toxic and antioxidant effects of F10-12 at the enzymatic level. Antioxidant characteristics of F10-12 were revealed in model solutions of organic and inorganic oxidizers. Low-concentration activation of bioluminescence was validated statistically in oxidizer solutions. Toxic and antioxidant characteristics of F10-12 were compared to those of homologous fullerenols with a higher number of oxygen groups:F24-28 and F40-42. No simple dependency was found between the toxic/antioxidant characteristics and the number of oxygen groups on the fullerene’s carbon cage. Lower toxicity and higher antioxidant activity of F24-28 were identified and presumptively attributed to its higher solubility. An active role of reactive oxygen species (ROS) in the bioeffects of F10-12 was demonstrated. Correlations between toxic/antioxidant characteristics of F10-12 and ROS content were evaluated. Toxic and antioxidant effects were related to the decrease in ROS content in the enzyme solutions. Our results reveal a complexity of ROS effects in the enzymatic assay system. © 2021 by the authors. Аннотация: Tissue reaction to implantation of polymeric microparticles from resorbable polymer (polyhydroxybutyrate) is characterized by slight inflammatory reaction and pronounced progressive macrophage infiltration with the presence of mono-and multinuclear foreign body giant cells resorbing the polymeric matrix. No fibrous capsules were formed around the polymeric microparticles; neither necrosis nor other adverse morphological changes and tissue transformation in response to implantation of the PHB microparticles were recorded. The results indicate good prospects of using polyhydroxybutyrate for the construction of long-acting dosage forms as microparticles for intramuscular injection. Аннотация: We investigated time-dependent trends of artificial radionuclides in aquatic moss, zoobenthos (amphipods and caddisfly larvae), and three abundant wild fish species (Northern pike, Arctic grayling, and Siberian dace) inhabiting the Yenisei River in the vicinity of the radioactive discharge site in 2007–2015, in a period before and after the shutdown of the last nuclear reactor plant at the Mining-and-Chemical Combine (MCC), which occurred in 2010. From our research, we learned that concentrations of short-lived radionuclides, whose discharges to the Yenisei either stopped or declined after the shutdown of the reactor plant at the MCC (24Na, 46Sc,51Cr, 54Mn, 58Co, 59Fe, 60Co, 65Zn, 103Ru, 141,144Ce, 152,154Eu, 239Np), decreased in biota samples as well. The ecological half-life (EHL) of 65Zn (0.4–0.7 y) was similar to the physical half-life of this isotope, the EHLs of 60Co (1.2–2.1 y) and 152Eu (1.8 y) were shorter than the physical half-lives of these isotopes. Concentration of 137Cs did not decrease significantly in biota of the Yenisei after the shutdown of the last reactor plant because the discharges of this radionuclide to the Yenisei continued at the same level. On a longer-term scale (since 1973 and since 1991), concentration of 137Cs in fish muscle had significantly decreased, following the decrease in annual discharges of this radionuclide to the Yenisei, and the EHL of 137Cs was estimated as 6.5–12.8 y. Statistically significant correlation with annual discharges of 137Cs was revealed for the concentration of this radionuclide in grayling (whole bodies and muscle); dace (muscle), and amphipods. Despite their ability to accumulate high concentrations of 137Cs, aquatic moss and caddisfly larvae (analyzed together with their stony casings) were not sensitive to interannual fluctuations in the releases of this radionuclide to the Yenisei. Among the analyzed fish species of the Yenisei, the highest activity concentration of 137Cs was revealed in pike (body and muscle), indicating biomagnification of this radionuclide in the top level of the trophic chain. © 2019 Elsevier Ltd

Аннотация: The vertical structure of the ecosystem in the euphotic zone of the western Equatorial Pacific has been studied. The studies were based on a continuous sounding of the bioluminescence field, with simultaneous, vertically aimed sampling made with the aid of a 5l water bottle and plankton nets. The bioluminescence field has a two-maxima structure with a more pronounced and permanent lower maximum found in the oligotrophic regions as deep as 60 to 100 m. In the narrow (10 m) layer of maximum bioluminescence, the concentration of zooplankton, as well as the concentration, activity and production of bacteria and phytoplankton, are several times higher than those in immediately adjacent waters at greater or lesser depth. At the same time, the concentration of nutrient salts in that layer diminishes sharply and approaches zero in the overlying water. Analysis of the data obtained shows that the production processes above the lower maximum layer are dependent not only on the turbulent influx of nutrient salts from the underlying layers, but also on the horizontal supply from the zone of upwelling. A pelagic community, in its different time aspects from the moment of water ascending until its sinking in the convergence zone, is suggested as a common system for modelling. Аннотация: We studied the effects of anti-inflammatory substances incorporated in polymeric microparticles made of degradable natural polyhydroxyalkanoate polyesters on experimental skin wounds caused by chemical burns in laboratory animals. Treatment with encapsulated forms of anti-inflammatory substances (applied in gel) accelerated wound healing in comparison with routine therapy (estimated by area of burn wound, wound healing activity, number of acanthotic cells, and number of hair and sebaceous follicles). The results showed the perspectives of usage of developed form of substances (degradable polymeric microparticles) for treatment of skin defects.

Аннотация: Interactions between phytoplankton and zooplankton affect the overall functioning of lakes. Herbivores are habitually confronted with food of inferior quality, usually a result of low nutrient concentrations in plant material. Large-bodied cladocerans are better competitors for food than small-bodied species but they are more vulnerable to low food quality. Understanding the effects of food quality on zooplankton structure and competition between small - large bodied herbivorous is of considerable interest. We want to find out how differences in C:N:P ratios between phytoplankton and zooplankton communities affect their abundances in a freshwater food web. We want also to assess the role of phytoplankton and zooplankton as sinks of the phosphorus and nitrogen. Therefore, we conducted a 31-day mesocosms experiment with water from a mesotrophic and a eutrophic lake (with natural plankton communities). To simulate changes in the plankton communities large-bodied Daphnia magna and Daphnia pulicaria were added. Samples for zooplankton, phytoplankton and water chemistry were taken every 10 days. Samples for elemental analysis (C:N:P) of seston and zooplankton were collected on the first, and on the final day of the experiment. Our mesocosms experiment showed mismatch in C:P between seston (high) and zooplankton (low), which suggests that most of the phosphorus is incorporated in zooplankton biomass. This evidenced that zooplankton is an effective sink of phosphorus, while nitrogen is accumulated mainly by primary producers. Our results also indicated more stability in stoichiometry with increasing trophic levels of organisms. However, there were significant changes in the zooplankton structure. The increasing dominance of large Daphnia resulted in reduction of C:P ratio in zooplankton. Low food quality (C:P) did not limit the growth of large Daphnia in the experimental conditions, which competed effectively with small planktonic cladocerans and with Rotifera. Over time, inedible algae began to dominate resulting in increase of relative biomass of periphyton grazers, which suggests that plankton community is transformed into littoral system in mesocosms for about 30 days. © 2019

Аннотация: Bioluminescent proteins are widely used as reporter molecules in various in vitro and in vivo assays. The smallest isoform of Metridia luciferase (MLuc7) is a highly active, naturally secreted enzyme which, along with other luciferase isoforms, is responsible for the bright bioluminescence of marine copepod Metridia longa. In this study, we report the construction of two variants of a hybrid protein consisting of MLuc7 and 14D5a single-chain antibody to the surface glycoprotein E of tick-borne encephalitis virus as a model fusion partner. We demonstrate that, whereas fusion of a single-chain antibody to either N-or C-terminus of MLuc7 does not affect its bioluminescence properties, the binding site on the single-chain antibody influences its binding capacity. The affinity of 14D5a-MLuc7 hybrid protein (KD = 36.2 nM) where the C-terminus of the single-chain antibody was fused to the N-terminus of MLuc7, appeared to be 2.5-fold higher than that of the reverse, MLuc7-14D5a (KD = 87.6 nM). The detection limit of 14D5a-MLuc7 hybrid protein was estimated to be 45 pg of the recombinant glycoprotein E. Although the smallest isoform of M. longa luciferase was tested as a fusion partner only with a single-chain antibody, it is reasonable to suppose that MLuc7 can also be successfully used as a partner for genetic fusion with other proteins. © 2020 by the authors. Аннотация: COVID-19 has specific characteristics that distinguish this disease from many other infec-tions. We suggest that the pathogenesis of severe forms of COVID-19 can be associated with acidosis. This review article discusses several mechanisms potentially linking the damaging effects of COVID-19 with acidosis and shows the existence of a vicious cycle between the development of hypoxia and acidosis in COVID-19 patients. At the early stages of the disease, inflammation, difficulty in gas exchange in the lungs and thrombosis collectively contribute to the onset of acidosis. In accordance with the Verigo-Bohr effect, a decrease in blood pH leads to a decrease in oxygen saturation, which contributes to the exacerbation of acidosis and results in a deterioration of the patient’s condition. A decrease in pH can also cause conformational changes in the S-protein of the virus and thus lead to a decrease in the affinity and avidity of protective antibodies. Hypoxia and acidosis lead to dysregu-lation of the immune system and multidirectional pro-and anti-inflammatory reactions, resulting in the development of a “cytokine storm”. In this review, we highlight the potential importance of supporting normal blood pH as an approach to COVID-19 therapy. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: Simple Summary Recently, several studies have shown that acidosis, which is increased acidity in the blood and other body tissues, is often associated with severe COVID-19. In this article, we look at the mechanisms and consequences of acidosis that can lead to an exacerbation of COVID-19. We want to draw the attention of readers to the threshold values of such disease characteristics as hypoxia and acidosis, which are associated with a sharp deterioration in the patient's condition. Hypoxia and acidosis mutually reinforce each other according to the principle of a vicious cycle (that is, they are involved in a system of positive feedbacks). Elevated blood lactate (lactic acid) levels are associated with poor clinical outcomes in COVID patients. As a practical recommendation, we propose to pay more attention to the prevention of acidosis, including in the early stages of the disease, when the adjustment of homeostasis requires less effort and is less risky. Аннотация: A new method to compare two (or several) symbol sequences is developed. The method is based on the comparison of the frequencies of the small fragments of the compared sequences; it requires neither string editing, nor other transformations of the compared objects. The comparison is executed through a calculation of the specific entropy of a frequency dictionary against the special dictionary called the hybrid one; this latter is the statistical ancestor of the group of sequences under comparison. Responses of root and shoot length growth were studied 6 and 10 days after irradiation. Results: Our results showed inhibition of the root and shoot length growth 6 days after exposure at all doses, including the low dose ? 0.1 Gy. At a later point in time (day 10), root and shoot inhibition was only observed after irradiation at high doses (above 5 Gy), and that suggested the occurrence of cell repair after irradiation at low doses. The results indicated that the length of seedling roots was more sensitive to gamma-irradiation than the shoot length. Conclusion: The results of the study suggested that short-term gamma-irradiation of onion seedlings (absorbed doses of 0.1?10 Gy) caused inhibition of plant growth 6 and 10 days after irradiation. The dose dependence of the onion root length was linear.
