Аннотация: The study addresses the effect of humic substances on marine luminous bacteria Photobacterium phosphoreum exposed to Am-241 (3,000 Bq L -1, water solution). Luminescent intensity of the bacteria was applied as a marker of their physiological activity. Humic substances have been found to reduce the effect of Am-241 on luminescence, decrease damage to cells, and change distribution of Am-241 between bacterial cells and intercellular media. It was shown that water-soluble humic substances, being products of natural transformation of organic substances in soil and bottom sediments, can serve as protecting agents for water microorganisms exposed to alpha radionuclides. Аннотация: The study addresses the ability of luminous bacteria of different taxa (Photobacterium leiognathi, Photobacterium phosphoreum, Vibrio harveyi, Vibrio fischeri) to synthesize polyesters of hydrocarbon acids (polyhydroxyalkanoates, PHAs) as storage macromolecules. The screened strains widely varied in their PHA productivity. Conditions for attaining high polymer yields (including two- and three-component polymers) in batch culture have been determined. The attained polymer yields reached 40-70% of dry cell biomass. The results suggest a conclusion that luminous microorganisms can be considered as producers of multi-component PHAs. Аннотация: A set of bioluminescent tests was developed to monitor water quality in natural and laboratory ecosystems. It consisted of four bioluminescent systems: luminous bacteria, coupled enzyme system NADH:FMN-oxidoreductase-luciferase and triplet enzyme systems with alcohol dehydrogenase and trypsin. The set of biotests was applied for a small forest pond (Siberia, Russia), laboratory microecosystems polluted with benzoquinone and a batch culture of blue-green algae. Thereby effects of natural water compared to those of models of heavy pollution and "bloom" of blue-greens on the bioluminescent tests were revealed. The set of biotests was not affected by a natural seasonal variability of water quality in the unpolluted pond, but responded to the heavy pollution and the "bloom" of blue-greens. The set of biotests could be recommended as the alarm test to control the acute toxicity of natural water bodies. В© 2001 Elsevier Science Ltd.

Аннотация: New results obtained in the detection of rhythms in the low frequency range in the light signal of a small number of photo bacteria are reported. The luminescence of a small number of bacteria was recorded with an apparatus the sensitivity of which was 12 quanta/pulse. To check the possible rhythms in the luminescence of the bacteria, correlation analysis was made of the interpulse intervals of the detection of the signal with a computer. This revealed a definite rhythm in the bacterial luminescence the fundamental frequency of which lies in the region 8 c/s. Profound modulation of a bioluminescent signal at this and multiple frequencies was demonstrated. The harmonic components found in the spectrum of the signal refute the existing view of the continuous character of bacterial luminescence.

Аннотация: 1. (1) The increase in the intensity of the luminescence of bacteria observed on addition of glucose follows a curve with a well-marked maximum after which attenuation sets in. 2. (2) For the relations studied by us between the amount of glucose and the amount of bacterial suspension (1 ml 0В·1 m solution of glucose per ml suspension) the time of attaining the maximum intensity of luminescence and the ratio of the maximum intensity of emission to its level before addition of glucose do not depend on the concentration of bacteria in the suspension. 3. (3) The highest uptake of glucose by bacteria occurs in the period from addition of glucose to the moment of maximum rise in luminescence. Change in the intensity of luminescence after addition of glucose follows a curve similar to that of glucose uptake. 4. (4) We have determined the radiant flux and all the radiant energy released by bacteria in the period from addition of glucose to the moment of attaining the maximum intensity. One bacterial cell on average emits 100 quanta/sec. 5. (5) Bioluminescence accounts for 10-6 parts of the entire energy released on oxidation of glucose. В© 1966.

Аннотация: A concept is formulated on ecological and biophysics as a scientific discipline which studies physical processes and phenomena resulting from the functioning of live supra-organism systems. Three main directions of ecological biophysics are defined: creation of methods of monitoring ecosystems on the basis of probing biophysical fields (fields of bioluminescence and fluorescence); mathematical and physical modelling; and study of the role of live organisms in hydrophysical processes on the ecosystem scale. Examples are given of the realization of the ecologo-biophysical approach in research into the ecosystems of the world ocean, Lake Baikal and the Dnieper and Yenisei reservoirs. В© 1994.

Аннотация: Characteristics of the diurnal rhythm of the bioluminescence field are analyzed on the basis of experimental data obtained in the region around Kamchatka peninsula (50В°3 1'N; 163В°50'E). The feasibility of reconstructing the nocturnal structure of the bioluminescence field by measurements made at other times of the day has been demonstrated. Diurnal rhythm parameters for the bioluminescent field of several other studied areas of the ocean have been defined. The temporal parameters feature linear latitudinal dependence. At the equator the boundaries of morning and evening transition periods are 05:06 to 07:30 and 17:48 to 19:54 hrs, respectively. In the Barents Sea in spring these boundaries are 02:42 to 04:06 and 19:48 to 21:12 hrs. Boundary times of diurnal rhythm do not deviate from the linear regression by more than 10%.

Аннотация: The luminescence of obelin is initiated by NaOCl in a reaction mixture containing no calcium. The addition of Mn2+ enhances the light emission 300-fold. Sodium azide and histidine, as singlet oxygen quenchers, inhibit NaOCl-activated obelin luminescence in the presence or absence of Mn2+. This suggests that the addition of NaOCl to the mixture causes singlet oxygen formation (stimulated by Mn2+ ions), and singlet oxygen initiates the light-emitting reaction.

Аннотация: Genes encoding luminescence of Photobacterium leiognathi have been cloned in Escherichia coli. The luminescent clones were readily apparent. Among them, a clone containing a recombinant plasmid with a 13.5-kb insertion was identified. This DNA fragment contained all of the luminescence-encoding genes. The luciferase-encoding genes (lux) in this DNA fragment were localized. We have sequenced a part of the cloned lux region and identified the luxA, luxB and luxG genes encoding the ? and ? subunits of luciferase and a ? protein with an Mr of 26 180, respectively. The analysis of deduced amino acid sequences and comparison with known luciferase sequences from Vibrio harveyi, indicate the common origin of these proteins. В© 1990.

Аннотация: It has been demonstrated that the transgenic microorganism Escherichia coli Z905/pPHL7 (Ap'Lux+) can exist for a long time at an elevated concentration of mineral salts. The microorganism was introduced into microcosms with sterile brackish water (salinity variable from 21 to 22 g 1-1) taken from Lake Shira (Khakasia, Russia). The survivof the microorganism was estimated both by measuring the growth of the colonies on solid nutrient media and by the bioluminescence exhibited by the transgenic strain in samples from the microcosms and in the enrichment culture with the added selective factor - ampicillin (50 ?g/ml). In the enrichment culture, the bioluminescent signal was registered through the 160-day experiment. It has been shown that in the closed microcosms with brackish water the E. coli strain becomes heterogeneous in its ampicillin resistance. The populations of the transgenic strain were mainly represented by isolates able to persist in the medium containing 50 ?g/ml, but there were also the cells (about 10%) with the threshold of ampicillin resistance not more than 0.05 ?g/ml. Thus, it was shown that in the microcosms with brackish water and in the absence of the selective factor the transgenic strain survives and retains the recombinant plasmid. В© 2003 COSPAR. Аннотация: Описан способ выделения и очистки Ca-зависимого фотопротеина - обелина из гидроидных полипов Obelia longissima, включающий: разрушение материала в гипотонич. буфере, фракционирование ПЭГ 6000, фракционирование (NH[4])[2]SO[4] в диапазоне 40-75% насыщения, хроматографию на ДЭАЭ-целлюлозе DE-52, хроматографию на фенил-сефарозе 4В, гель-фильтрацию на сефадексе G-75, гель-фильтрацию на колонке Superose 12 HR 10/30 при помощи системы FPLC. С помощью предложенного метода получен практически чистый обелин с выходом 30-40%. Мол. м. полученного белка, определенная гель-фильтрацией в неденатурирующих условиях, составляет 39,6 кДа, тогда как электрофорез в полиакриламидном геле с додецилсульфатом натрия показывал наличие двух белков с мол. м. 27 и 15,6 кДа соответственно. Аннотация: Computational coefficients for estimating the effectiveness of bioluminescence expression in natural luminescent bacteria Photobacterium leiognathi 54 and transgenic strain E. coli Z905/pPHL7 bearing lux-operon in a multicopy plasmid are suggested and their use at molecular, cell, and population levels was considered. It was shown that at the population level, all transgenic variants have an advantage over natural variants of P. leiognathi 54 irrespective of the type of lux-operon regulation. At the cell level, the effectiveness of bioluminescence expression in the bright and dim variants of the transgenic strain increased by several orders. At the level of one lux-operon, the effectiveness of expression in the bright variant of the transgenic strain is substantially higher than in the natural bright variant; in dim variants, the efficiency values are similar; the effectiveness of bioluminescence expression in the dark variant of E. coli Z905-2/pPHL7 is by two orders of magnitude lower than in the dark variant of P. leiognathi 54. Аннотация: Manifestation of pleiotropic effects in the isogenic variants of the luminescent bacterium Photobacterium leiognathi 54 was investigated. The decrease or increase of the expression level of bioluminescence was caused by changes in lux operon regulation. The dynamics of the bioluminescence of dark and dim variants did not differ from the dynamics of the initial luminescent variant, but dependence of the level of luminescence intensity on the exogenous autoinducer of the lux operon was revealed. The investigated variants of P. leiognathi 54 inherited fairly stable morphological characteristics, colony architectonics, level of luminescence, and activity of some enzymes; variants with reduced bioluminescence formed colonies of the S type. Stable bright variants with S-and R-type colonies appeared both in the initial strain population and in the dark variant population, but with smaller frequency. Populations of the bright variant with R-type colonies were most heterogeneous; this can be determined by the lack of glucose repression of the bioluminescence in contrast to other investigated inherited variants of P. leiognathi. Аннотация: The database "BiolumBase" is designed for the selection and systematization of available world information on microorganisms containing bioluminescent systems; it includes two sections: "natural" and "transgenic" luminous microorganisms. At present, logic schemes of divisions, classification of the objects, presentation of characteristics, and the inputs of relative information, as well as the necessary program modules including links to the database, are developed. The database is constructed on the basis of published data and our own experimental results; the subsequent linkage of the database to the Internet is envisaged. Users will be able to obtain not only the catalogues of strains but also information concerning the properties and functions of the known species of luminous bacteria, the structure, regulatory mechanisms, and application of bioluminescent systems and genetically engineered constructions with live genes, as well as to find references and to search strains by using any set of attributes. The database will provide information that is of interest for the development of microbial ecology and biotechnology, in particular, for the prediction of biological hazard from the application of transgenic strains.
