Аннотация: A top-down nanofabrication approach involving molecular beam epitaxy and electron beam lithography was used to obtain silicon nanowire-based back gate field-effect transistors with Schottky contacts on silicon-on-insulator (SOI) wafers. The resulting device is applied in biomolecular detection based on the changes in the drain-source current (IDS). In this context, we have explained the physical mechanisms of charge carrier transport in the nanowire using energy band diagrams and numerical 2D simulations in TCAD. The results of the experiment and numerical modeling matched well and may be used to develop novel types of nanowire-based biosensors. © 2021 Elsevier B.V.

Аннотация: The present study addresses the seasonal dynamics of productivity and species composition of the meadow and steppe vegetation communities in Khakassia, determined using the ground-based and satellite data of 2017. The MODIS/Terra satellite data were used to analyze the Normalized Difference Vegetation Index (NDVI) and the Land Surface Water Index (LSWI). The NDVI and LSWI were found to be related to the productivity of the meadow and steppe vegetation. The NDVI increased as the portion of the mesophyte grasses in the grass canopy became larger. The LSWI was higher in the steppe communities, which had lower projective coverage, with spots of bare soil, than in the meadow communities, with their abundant vegetation. © Published under licence by IOP Publishing Ltd.

Аннотация: The recently described bioluminescent system from fungi has great potential for developing highly efficient tools for biomedical research. Luciferase enzyme is one of the most crucial components of this system. The luciferase from Neonothopanus nambi fungus belongs to the novel still undescribed protein family. The structure data for this protein is almost absent. A detailed study of the N. nambi luciferase properties is necessary for the improvement of analytical methods based on the fungal bioluminescent system. Here we present the positions of key amino acid residues and their effect on enzyme function described using bioinformatic and experimental approaches. These results are useful for further fungal luciferase structure determination.

Аннотация: The paper presents a brief overview of the results of the implementation of the project "Agropreparations of the new generation: a strategy of construction and realization". The first part contains the analysis of the growth of the wild-type strain Cupriavidus necator B-10646 (formerly eutrophus) and the synthesis of polyhydroxyalkanoates by this strain on various substrates: glycerol, palm oil, Siberian oil seed, sunflower seed oils, and oleic acid. On refined glycerin, a highly productive process is implemented when scaling up, allowing to obtain 128 ± 11 g / L PHA. Evaluation of oils has shown that palm oil is the best carbon substrate. The second part presents the results of the development of environmentally friendly slow-release pesticide formulations. They are a degradable matrix of poly-3-hydroxybutyrate mixed with natural materials (peat, clay, wood flour), into which a pesticide (metribuzin, tribenuron-methyl, fenoxaprop-P-ethyl, azoxystrobin, epoxiconazole, and tebuconazole) has been. The developed preparations showed high activity against pathogenic fungi and weeds and had a much weaker negative effect on the soil microflora. Studies of the degradation of the developed preparations and the release of pesticides into the soil confirm their effectiveness over a long period of time, up to 90 days. © Published under licence by IOP Publishing Ltd.

Аннотация: Mycelia of various Armillaria fungi are bioluminescent while the fruit bodies do not emit light. The presence in fruit bodies of Armillaria species of enzymes involved in the fungal bioluminescence was investigated by treating them with an exogenous analogue of the substrate for the light-emitting reaction. For this, hot extracts from nonluminous fungus Pholiota squarrosa were used. Upon spraying the pristine and transversely cut fruit bodies with the extracts, light emitting regions of different intensity were revealed. This suggests that the fruit bodies of the studied species are nonluminous due to lack of the substrate for light luminescent reaction. The prolonged incubation of the fruit bodies in water elevated the bioluminescence level. A possible mechanism which can explain this phenomenon is discussed. © 2021 Institute of Physics Publishing. Potential consumers of chironomids adults, the orb-weaver spiders Larinoidessuspicax, which inhabited both zones, also differed in fatty acid composition. The main difference in their biochemical composition was a significantly higher level of EPA in spiders from the riparian zone that likely to be explained by consumption of the Glyptotendipes adults enriched in this fatty acid and concentrated only within this zone. The higher level of EPA, which is deficient in terrestrial ecosystems, in orb-weaver spiders from the riparian zone of the saline lake may potentially promote a successful survival of the consumers in the arid landscape. © 2021, Pleiades Publishing, Ltd.

Аннотация: The tangible and intangible legacy of the World Universiades is a debatable topic in the science literature of the world. The Universiade-2019 has become a powerful stimulus for the development of sports education, science, mass and elite student sports at Siberian Federal University (SibFU). In the field of physical culture and sports SibFU has implemented new educational programs, held international scientific conferences, carried out major scientific projects, and built a new infrastructure. The number of students involved in physical culture and sports on a regular basis has increased. Since 2012, SibFU students began to win medals at all major international student competitions, such as the European and World Championships, European Universities Games and the Universiade. For example, according to the results of the Universiade-2019, SibFU took second place among all universities in the world in terms of the number of medals won. It is concluded that the Universiadecan contribute not only to the traditional national, cultural and sports goals, but also to the development of university sports in cities hosting mega-events. An effective way to implement this idea may be to include universities in the Universiade organizing committees. The experience of SibFU in the preparation and holding of the Universiade-2019 can be recommended for replication and successful holding of future Universiades or other international student sports events. © Siberian Federal University. Аннотация: Objective was to study single-nucleotide polymorphisms (SNP) in CAT, NCL, HSPA1L, PCDH15, and PON2 genes and their associations with hearing impairment among the people working among noise-exposed workers of the mashine-building plant (JSC «Krasmash», Krasnoyarsk, Eastern Siberia, Russia). Materials and methods. The 443 employees of Krasmash JSC, who have been working under conditions of increased noise for at least 1 year, were surveyed and examined. A hearing study was performed by speech and tonal audiometry. Tonal audiometry was carried out in accord with according to a standard method in the frequency range 125—8000 Hz. People with chronic hearing impairment, survivors of meningitis and family history of hearing impairment were excluded from the study. The allelic composition of the studied genes was determined in the remaining group of 288 workers (study group). Polymorphisms were detected using bioluminescent method, developed by the authors earlier. The study group comprised 122 people with hearing impairment (experimental group) and 166 people without impairment (control group). Results. The genotyping results of on allelic variants rs494024 (CAT), rs7598759 (NCL), rs2227956 (HSPA1L), rs7095441 (PCDH15) and rs7785846 (PON2) showed that their frequencies in the study group did not differ and were comparable with those for the European population. No statistically significant differences were revealed in the distribution of the genotypes of the studied mutations between the experimental and control groups. Also no statistically significant associations we found between hearing impairment and availability of two or several SNPs, or these SNPs and clinical characteristics of the disease (degree of hearing impairment, tinnitus). In the group of workers with an experience of 5 to 16 years, an association was found for hearing impairment and SNP rs494024, as well as when it is combined with rs7598759. Conclusions. The associations between SNP rs7598759, rs2227956, and rs7095441 and hearing impairment were not found. In the group of workers with 5—16 year experience, this association was found for SNP rs494024, as well as when it is combined with rs7598759. Аннотация: The trophic transfer efficiency (TTE) is an important indicator of ecosystem functioning. However, TTE data from freshwater food webs are ambiguous due to differences in time scales and methods. We investigated the transfer of essential substances (carbon, nutrients, and polyunsaturated fatty acids) through plankton communities in 30 Polish lakes with different trophic status in the middle of summer. The results of our study revealed that different essential substances were transferred from phytoplankton to zooplankton with varying efficiencies. The average TTE of C, N, P, and the sum of ?-3 PUFA were 6.55%, 9.82%, 15.82%, and 20.90%, respectively. Our results also show a large mismatch between the elemental and biochemical compositions of zooplankton and their food during the peak of the summer stagnation, which may further promote the accumulation of essential substances. There were also large differences in TTEs between trophic conditions, with the highest efficiencies in oligotrophic lakes and the lowest in dystrophic and eutrophic lakes. Therefore, our study indicates that disturbances like eutrophication and dystrophication similarly decrease the TTE of essential substances between phytoplankton and zooplankton in freshwater food webs. © 2021 The Authors. Аннотация: We present the presence/absence species list (Table 1) of rotifer, cladoceran, and copepod (Calanoida, Harpacticoida, and Cyclopoida) fauna from seven Arctic regions of Russia (the Kola Peninsula, the Pechora River Delta, the Bolshezemelskaya tundra, the Polar Ural, the Putorana Plateau, the Lena River Delta, and the Indigirka River Basin) based on our own and literature data. Our own records were obtained by analyzing samples of zooplankton, meiobenthos, and two cores of bottom sediments (from the Kola Peninsula and the Bolshezemelskaya tundra lakes) that we collected once in July or August in 1992, 1995–2017. To supplement the list, we used relevant literature with periods of research from the 1960s to the 2010s. The list is almost identical to “Dataset 2: Zooplankton and Meiofauna across Arctic Regions of Russia”, which was analyzed but not published in [1]. The detailed analysis of this list revealed the specific composition of the aquatic fauna associated with the climatic and geographical factors [1]. The data provide information on the current state of biodiversity and species richness in Arctic fresh waters and can serve as the basis for monitoring these environments and predicting how they are likely to change in the future. © 2021

Аннотация: The study reports results of using a CO2-laser in continuous wave (3 W; 2 m/s) and quasi-pulsed (13.5 W; 1 m/s) modes to treat films prepared by solvent casting technique from four types of polyhydroxyalkanoates (PHAs), namely poly-3-hydroxybutyrate and three copolymers of 3-hydroxybutyrate: with 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate (each second monomer constituting about 30 mol.%). The PHAs differed in their thermal and molecular weight properties and degree of crystallinity. Pristine films differed in porosity, hydrophilicity, and roughness parameters. The two modes of laser treatment altered these parameters and biocompatibility in diverse ways. Films of P(3HB) had water contact angle and surface energy of 92? and 30.8 mN/m, respectively, and average roughness of 144 nm. The water contact angle of copolymer films decreased to 80–56? and surface energy and roughness increased to 41–57 mN/m and 172–290 nm, respectively. Treatment in either mode resulted in different modifications of the films, depending on their composition and irradiation mode. Laser-treated P(3HB) films exhibited a decrease in water contact angle, which was more considerable after the treatment in the quasi-pulsed mode. Roughness parameters were changed by the treatment in both modes. Continuous wave line-by-line irradiation caused formation of sintered grooves on the film surface, which exhibited some change in water contact angle (76–80? ) and reduced roughness parameters (to 40–45 mN/m) for most films. Treatment in the quasi-pulsed raster mode resulted in the formation of pits with no pronounced sintered regions on the film surface, a more considerably decreased water contact angle (to 67–76? ), and increased roughness of most specimens. Colorimetric assay for assessing cell metabolic activity (MTT) in NIH 3T3 mouse fibroblast culture showed that the number of fibroblasts on the films treated in the continuous wave mode was somewhat lower; treatment in quasi-pulsed radiation mode caused an increase in the number of viable cells by a factor of 1.26 to 1.76, depending on PHA composition. This is an important result, offering an opportunity of targeted surface modification of PHA products aimed at preventing or facilitating cell attachment. © 2021 by the authors. Аннотация: The trophic transfer efficiency (TTE) is an important indicator of ecosystem functioning. However, TTE data from freshwater food webs are ambiguous due to differences in time scales and methods. We investigated the transfer of essential substances (carbon, nutrients, and polyunsaturated fatty acids) through plankton communities in 30 Polish lakes with different trophic status in the middle of summer. The results of our study revealed that different essential substances were transferred from phytoplankton to zooplankton with varying efficiencies. The average TTE of C, N, P, and the sum of omega-3 PUFA were 6.55%, 9.82%, 15.82%, and 20.90%, respectively. Our results also show a large mismatch between the elemental and biochemical compositions of zooplankton and their food during the peak of the summer stagnation, which may further promote the accumulation of essential substances. There were also large differences in TTEs between trophic conditions, with the highest efficiencies in oligotrophic lakes and the lowest in dystrophic and eutrophic lakes. Therefore, our study indicates that disturbances like eutrophication and dystrophication similarly decrease the TTE of essential substances between phytoplankton and zooplankton in freshwater food webs.

Аннотация: Cupriavidus necator B10646 bacterial cells were cultivated in the mode of synthesis of the reserve polyhydroxyalkanoates (PHAs) in the growth medium that contained, in addition to glucose as the main substrate, precursor substrates of the monomers of various monocarboxylic acids - salts of valeric and hexanoic acids, propionate, and ?-butyrolactone. PHA terpolymers and quaterpolymers with different compositions and proportions of monomers were synthesized, and their physicochemical properties were studied. The terpolymers were composed of monomers of 3-hydroxybutyrate (3HB), 3-hydroxyvalerate (3HV), 4-hydroxybutyrate (4HB), or 3-hydroxyhexanoate (3HHx) and had the following compositions: P(3HB/3HB/4HHx) and P(3HB/3HV/4HHx). The quaterpolymers had the following composition: P(3HB/3HV/4HB/3HHx). All copolymer samples, regardless of the composition and proportions of monomers, had lower molecular weights and higher polydispersity values compared to the highly crystalline 3-hydroxybutyrate homopolymer, but retained thermal stability properties, with a difference between the melting point and thermal degradation of at least 100-110 °C. The inclusion of 3HV, 4HB, and 3HHx monomers in the C-chain of 3HB caused changes in the crystalline to amorphous phase ratio and a significant decrease in the degree of crystallinity (Cx), which depended on the type of monomers and their contents in the copolymer. The maximum decrease in Cx (9-17 %) was detected in the P(3HB/3HV/4HB) terpolymer and the P(3HB/3HV/4HB/3HHx) quaterpolymer (30-36 %). The study confirms that there is the possibility of synthesizing polymers with various compositions, including new ones, which differ significantly in their basic properties. © Siberian Federal University. Аннотация: A key component of the recently described bioluminescent system of higher fungi is luciferase, a new class of proteins. The properties of fungal luciferase and their relationship with its structure are interesting both for improving autoluminescent systems already created on its basis and for creating new ones. Therefore, it is extremely important to understand the spatial structure of this protein. We have performed heterologous expression and purification of Neonothopanus nambi luciferase, obtained a protein suitable for subsequent crystallization, and also determined some biochemical properties of the recombinant luciferase.

Аннотация: Traditionally, the abundance, cell size distribution and species identification of algae are determined by microscopic counts. In recent years, various rapid methods have been developed for routine algal studies. However, each of these methods has its drawbacks. It is important for aquatic ecologists to understand the advantages, disadvantages, and limitations of these methods. We compared the sensitivity of three rapid methods (multichannel fluorimeter FluoroProbe, imaging flow cytometer FlowCam, and CASY particle counter) to changes in cell abundance of three algae species (Chlorella vulgaris Beyerinck, Arthrospira platensis Gomont, and Nostoc sp.). We also assessed the ability of rapid methods to estimate the cell abundance of different species in the mixed samples. All instruments showed high sensitivity to changes in the cell abundance of different algae species and a mixture of these species. Any one of these methods, once calibrated, can be reliably used to estimate the abundance of a single-species/laboratory culture of microalgae. At the same time, FlowCam, without preliminary calibration, recorded the cell abundance closest to microscopic counts. When analysing a mixture of three microalgae differing in their cell sizes and spectral characteristics, FluoroProbe showed the highest accuracy in assessing the proportions of species in the mixture and FlowCam - in assessing their abundance. To study mixtures of algae and/or natural phytoplankton communities, it is advisable to use jointly a flow cytometer and a multichannel fluorimeter. The images of algae saved by the flow cytometer, if necessary, can be used to identify them, with a certain accuracy, to the species. Information on cells size and spectral characteristics obtained by two methods will be detailed enough to perform such common tasks as studying trophic interactions between phyto- and zooplankton or creating warning systems to inform of unwanted blooms of phytoplankton and their individual groups (for example, cyanobacteria). © Siberian Federal University. Аннотация: Purpose: To synthesize and investigate polyhydroxyalkanoates (PHAs) with different monomer composition and percentages and polymer films prepared from them. Results: Various PHAs: homopolymer poly-3-hydroxybutyrate P(3HB) and 2-, 3-, and 4-component copolymers comprising various combinations of 3-hydroxybutyrate (3HB), 3-hydroxyvalerate (3HV), 4-hydroxybutyrate (4HB), and 3-hydroxyhexanoate (3HHx) monomers were synthesized under specialized conditions. Relationships were found between the monomer composition of PHAs and their molecular-weight and thermal properties and degree of crystallinity. All copolymers had decreased weight average molecular weights, Mw (to 390–600 kDa), and increased values of polydispersity (3.2–4.6) compared to the P(3HB). PHA copolymers showed different thermal behavior: an insignificant decrease in Tmelt and the presence of the second peak in the melting region and changes in parameters of crystallization and glass transition. At the same time, they retained thermostability, and the difference between Tmelt and Tdegr was at least 100–120 °C. Incorporation of 4HB, 3HV, and 3HHx monomer units into the 3-hydroxybutyrate chain caused changes in the amorphous to crystalline ratio and decreased the degree of crystallinity (Cx) to 20–40%. According to the degree to which the monomers reduced crystallinity, they were ranked as follows: 4HB – 3HHx – 3HV. A unique set of films was produced; their surface properties and physical/mechanical properties were studied as dependent on PHA composition; monomers other than 3-hydroxybutyrate were found to enhance hydrophilicity, surface development, and elasticity of polymer films. Conclusion: An innovative set of PHA copolymers was synthesized and solution-cast films were prepared from them; the copolymers and films were investigated as dependent on polymer chemical composition. Results obtained in the present study contribute to the solution of a critical issue of producing degradable polymer materials. © 2021 Elsevier B.V.

Аннотация: Abstract: The recently described bioluminescent system from fungi has great potential for developing highly efficient tools for biomedical research. Luciferase enzyme is one of the most crucial components of this system. The luciferase from Neonothopanus nambi fungus belongs to the novel still undescribed protein family. The structure data for this protein is almost absent. A detailed study of the N. nambi luciferase properties is necessary for the improvement of analytical methods based on the fungal bioluminescent system. Here we present the positions of key amino acid residues and their effect on enzyme function described using bioinformatic and experimental approaches. Аннотация: The paper considers a new approach to modeling the relationship between the increase in woody phytomass in the pine forest and satellite-derived Normalized Difference Vegetation Index (NDVI) and Land Surface Temperature (LST) (MODIS/AQUA) data. The developed model combines the phenological and forest growth processes. For the analysis, NDVI and LST (MODIS) satellite data were used together with the measurements of tree-ring widths (TRW). NDVI data contain features of each growing season. The models include parameters of parabolic approximation of NDVI and LST time series transformed using principal component analysis. The study shows that the current rate of TRW is determined by the total values of principal components of the satellite indices over the season and the rate of tree increment in the preceding year. © 2020 by the authors.
