Аннотация: Several polymorphisms in the melanocortin-1 receptor gene (MC1R) have been associated with melanoma risk. In particular, rs1805007, rs1805008, and rs1805009 mutations, which result in R151C, R160W, and D294H amino acid substitutions, respectively, and are associated with the phenotype of red-hair mutations, have also been connected with melanoma and nonmelanoma skin cancer risks. This work describes a method of detecting these polymorphisms using primer extension with subsequent dual bioluminescent assay. Model plasmids carrying polymorphic MC1R fragments, as well as several clinical DNA samples, were tested using the proposed technique. The results agreed well with those obtained by Sanger sequencing. © 2015, Pleiades Publishing, Inc.

Аннотация: There is considerable data on molecular evolution, but there remains no approach to systematizing them within the framework of the key problems of biology. To search for the most common properties of evolving systems, the heuristic method has been proposed. Artificial networks of formal neurons were chosen as the heuristic model object. The paper examines the divergent component of evolutionary trajectory formation. As a result of the simulation, the dependence of the potential variability parameter on the position of the fitness function landscape was obtained. The simulation results are in agreement with the real data of molecular evolution experiments. © Published under licence by IOP Publishing Ltd.

Аннотация: 137Cs concentrations were measured in mushrooms in an area affected by radioactive discharges of the Mining-and-Chemical Combine (MCC) (Rosatom, Russia) in 2002–2017. The sources of radionuclides in the study sites were global fallouts and waterborne and airborne radioactive discharges of the MCC. The mushroom species Suillus granulatus and S. luteus showed the highest 137Cs concentrations (140–7100 Bq kg?1) for this area. Over the entire monitoring period, no significant change in 137Cs concentration was observed in the Suillus spp. samples collected from the sites with the aerial deposition of radionuclides. In the floodplain site with the radionuclide deposition from water and air, a significant decrease in the average 137Cs concentration was observed in the period between 2004 and 2017: a three-fold decrease in Suillus spp. and a nine-fold decrease in Lactarius deliciosus. The effective half-lives of 137Cs in fruiting bodies of the mushrooms L. deliciosus and Suillus spp. in this site were 3.6 ± 0.6 and 9.2 ± 2.7 years, respectively. © 2020, © 2020 Informa UK Limited, trading as Taylor & Francis Group.

Аннотация: We describe a low-cost three-dimensional underwater particle tracking velocimetry system to directly measure particle settling rate and flux in low-turbulence aquatic environments. The system consists of two waterproof cameras that acquire stereoscopic videos of sinking particles at 48 frames s?1 over a tunable sampling volume of about 45 ? 25 ? 24 cm. A dedicated software package has been developed to allow evaluation of particle velocities, concentration and flux, but also of morphometric parameters such as particle area, sinking angle, shape irregularity, and density. Our method offers several advantages over traditional approaches, like sediment trap or expensive in situ camera systems: (1) it does not require beforehand particle collection and handling; (2) it is not subjected to sediment trap biases from turbulence, horizontal advection, or presence of swimmers, that may alter particulate load and flux; (3) the camera system enables faster data processing and flux computation at higher spatial resolution; (4) apart from the particle settling rates, the particle size distribution, and morphology is determined. We tested the camera system in Lake Stechlin (Germany) in low turbulence and mean flow, and analyzed the morphological properties and settling rates of particles to determine their sinking behavior. The particle flux assessed from conventional sediment trap measurements agreed well with that determined by our system. By this, the low-cost approach demonstrated its reliability in low turbulence environments and a strong potential to provide new insights into particulate carbon transport in aquatic systems. Extension of the method to more turbulent and advective conditions is also discussed. © 2019 The Authors. Аннотация: We used a Lake Shira numerical model to estimate the response of the ecosystem of a saline meromictic lake to variations in weather parameters during the growing season. The sensitivity analysis of the model suggests that compared to other external (nutrient inflows) and internal (spring biomasses of food-web components) factors, weather parameters are among the most influential for both mixolimnetic (phyto- and zooplankton) and monimolimnetic (purple sulfur bacteria, sulfur reducing bacteria and hydrogen sulfide) food-web components. Calculations with different weather scenarios shows how changes in the water temperature and mixing depth affect mixolimnetic and monimolimnetic food-web components and the depth of the oxic-anoxic interface in a meromictic lake. When weather forcing stimulates an increase in the biomass of food-web components in the mixolimnion, it produces cascading effects that lead to three results: 1) a higher content of detritus in the water column; 2) a higher content of hydrogen sulfide in the monimolimnion; 3) raising of the oxic-anoxic interface closer to the water-air surface. This cascading effect is complicated by the negative correlation between two light dependent primary producers located at different depths—phytoplankton in the mixolimnion and purple sulfur bacteria at the oxic-anoxic interface. Thus, weather conditions that stimulate higher phytoplankton biomass are associated with a higher detritus content and lower biomass of purple sulfur bacteria, a higher content of hydrogen sulfide and a shallower oxic-anoxic interface. The same weather conditions (higher wind, lower cloud cover, and lower air temperature) promote a scenario of less stable thermal stratification. Thus, our calculations suggest that weather parameters during the summer season strongly control the mixing depth, water temperature and the mixolimnetic food web. An effect of biogeochemical and physical interactions on the depth of the oxicanoxic interface is also detectable. However, intra- and interannual climate and weather effects will be more important for the control of meromixis stability. © 2018, Chinese Society for Oceanology and Limnology, Science Press and Springer-Verlag GmbH Germany, part of Springer Nature.

Аннотация: The problem of how a gradual development of ecological and morphological adaptations combines with large genome rearrangements, which have been found to occur in the phylogeny of many groups of organisms, is a matter of discussion in the literature. The objective of this work was to study the problem with the example of salmonids, whose evolution included at least six events of multiple chromosome fusions. Large karyotype rearrangements are associated with a decrease in ecological and morphological diversity in salmonids. In the above example, genome rearrangements seem to distort the function of the genetic systems that are responsible for the occurrence of certain ecological forms in salmonids. © 2017 by the authors; Licensee MDPI, Basel, Switzerland.

Аннотация: An essentially new method is proposed for studying the statistical properties of gene sequences (g.s.) based on construction of a frequency-correlation dictionary (f.c.d.) for the g.s. being investigated (the f.c.d. is the set of all subsequences (words) with a length from 1 to N together with the frequencies of their encounter in the g.s. studied). Аннотация: The application of nanobiotechnology is an emerging area of nanoscience and nanotechnology. Nanodi-amond has been a potent antibacterial, antifungal, antioxidant, and antiplatelet agent. In the present study, nanodiamonds were reduced by green synthesis and characterization was done through SEM, TEM, FTIR, and XRD. Further they were tested for their biological applications. The antimicrobial activity was investigated/studied/examined? through well diffusion method. The best activity was observed against Trichoderma reesei (16 mm) at 140 ?g/ml. The antioxidant activity was investigated through DPPH and FRAPS method. It was observed that the biologically reduced nanodiamonds reduce the Fe3+ ions to Fe2+ ions at 600 mM/l/g concentration. In DPPH assay, inhibitory concentration was found to be 4.58 ?g/ml. Further the antiplatelet activity was investigated by prothrombin time (PT) and activated partial throm-boplastin time (APTT) assay, and it was observed that biologically reduced nanodiamonds have potent an-tiplatelet activity. © 2021 Sumy State University

Аннотация: A procedure has been developed to fabricate polymer tubes using solution casting inside template tubes. Polyester solution placed inside the vertically fixed template evaporated forming a hollow tube on the inner walls of the template. Silicone tubes used as the templates were permeable to solvent vapors and had relatively low adhesion to materials of the fabricated tubes, enabling their effortless removal. Parameters of the polymer tubes were determined by a type of the polymer, its concentration in the solution, and the inner diameter of the template. In the first method, a clamp was placed on the lower end of the template tube. A thickness of the new tube walls gradually increased from the open to the clamped end of the template tube. In another method viscous polymer solution without clamping was used; in this case, a polymeric bulkhead was formed in the middle of the tube. Two microbial polyesters, poly-3-hydroxybutyrate and poly-3-hydroxybutyrate-co-3-hydroxyvalerate, and also synthetic poly-L-lactide were used as model polymers. © 2020 Elsevier B.V.

Аннотация: The present study addresses the seasonal dynamics of productivity and species composition of the meadow and steppe vegetation communities in Khakassia, determined using the ground-based and satellite data of 2017. The MODIS/Terra satellite data were used to analyze the Normalized Difference Vegetation Index (NDVI) and the Land Surface Water Index (LSWI). The NDVI and LSWI were found to be related to the productivity of the meadow and steppe vegetation. The NDVI increased as the portion of the mesophyte grasses in the grass canopy became larger. The LSWI was higher in the steppe communities, which had lower projective coverage, with spots of bare soil, than in the meadow communities, with their abundant vegetation. © Published under licence by IOP Publishing Ltd.

Аннотация: This study investigates synthesis of poly(3-hydroxybutyrate)/diethylene glycol copolymers (P3HB/DEG) by Cupriavidus eutrophus B-10646 cells as related to DEG concentration in the medium and the time when it is added to the culture of cells synthesizing P3HB. The study determines the limits of physiological effect of DEG on C. eutrophus cells, showing that at DEG concentrations above 30 g/L, it inhibits cell growth, decreasing cell concentration and total P3HB/DEG yield and inducing an increase in the degree of saturation of fatty acids in lipids of cell cytoplasmic membrane. A series of copolymers containing different molar fractions of DEG (between 0.13 and 3.0 mol%) have been synthesized and their physicochemical, physical/mechanical, and biological properties have been investigated as related to the chemical composition and proportions of DEG monomers of the polymers. © 2016 American Institute of Chemical Engineers Biotechnol. Аннотация: The patterns of the phase portraits of vegetation (agrophytocenosis, woody vegetation) constructed using two-dimensional space radiation temperature values and Normalized Difference Vegetation Index were studied. An analysis of the phenological variability of vegetation in the south of the Krasnoyarsk Territory and the Republic of Khakassia during the growing seasons of 2003 and 2006 was carried out. Distinctive features of the phase portraits of agrophytocenosis and woody vegetation were revealed. The possibility of determining the boundaries of phenological states in the phytocenosis, and the transition range from one state into another was shown. Based on the complex analysis of the reflexive and radiative properties of the plant samples, an algorithm for calculating the start and end of the growing season was developed. © 2017, Pleiades Publishing, Inc.

Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers – polyhydroxyalkanoates (PHAs) – of two types – poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 ± 0.06–0.75 ± 0.11 ?m, while the average diameter of copolymer particles varied between 0.43 ± 0.12 and 0.55 ± 0.05 ?m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant. © 2021 Wiley Periodicals LLC.

Аннотация: Variability of chlorophyll concentration in the ocean is one of the most important components of the primary production process on the planet. To preserve the biosphere and to make appropriate use of it, it is imperative to have a deep insight into the long-term dynamics of the primary production on the planet. To investigate the dynamics of chlorophyll concentration based on satellite data, the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) has developed a geoinformation system. It was used to detect the areas in the global ocean that are quasistationary in relation to seasonal dynamics of chlorophyll concentration. Areas such as these, found in the Indian Ocean, are described in this work. These areas form the basis for the analysis of long-term dynamics of chlorophyll concentration. In these quasistationary zones systematic monitoring of phytopigment concentration is conducted by space-borne and marine craft. The work presents long-term satellite-based data on the space distribution of anomalous deviations of chlorophyll concentration in the ocean. An anomaly criterion is proposed and maps of Pacific Ocean areas with a high probability of anomalies are presented.

Аннотация: The tick-borne encephalitis virus (TBEV) is the causative agent of one of the most severe human neuroinfections. The infection transmitted by ixodid ticks is spread throughout the forest and forest-steppe zones of the temperate climatic belt of the Eurasian continent, including the Siberian region of the Russian Federation. Despite the availability of commercial analytical systems for the detection of TBEV, the task of developing approaches to a quick and reliable analysis that can be performed routinely, particularly in environmental studies, remains topical. A solid-phase bioluminescent immunoassay for determining the tick-borne encephalitis virus (TBEV) in ticks was developed. The assay is based on the hybrid protein consisting of a modified thermostable version of Renilla muelleri luciferase and a single-chain mini-antibody to protein E. This unique protein had been obtained and investigated by the authors earlier. The current study describes the expression of the hybrid protein in two different strains of recombinant E. coli cells. The optimal conditions for obtaining a highly purified protein were found. The bioluminescent reaction of the luciferase domain was triggered with the help of the stable natural form of the substrate, a Ca-dependent coelenterazine-binding protein, the recombinant variant of which was obtained by the authors. The conditions for production and storage of the immunoassay components (the hybrid protein, the stable form of the luciferase substrate, and activated microplates) were determined. Using the developed test system, more than 900 tick samples were analyzed for TBEV. In terms of sensitivity (89.5%) and specificity (98.9%), the proposed method is not inferior to colorimetric detection and is much simpler and faster than the latter. © Siberian Federal University. Аннотация: Accumulation of essential/beneficial and non-essential chemical elements by submerged (Stuckenia pectinata (L.) Borner) and emergent (Phragmites australis (Cav.) Trin. ex Steud.) rooted macrophytes in three water bodies was studied as dependent on water salinity (specific conductivity) and main environmental factors (temperature, pH of water, growing period, element contents in water and sediments). Inductively coupled plasma optical emission spectrometry (ICP - OES) was used for the element detection in the plants, sediments, and water. Multivariate statistics redundancy analysis (RDA) showed that investigated factors together explained 37.8 - 44.7% and 29.9 - 42.6% of the essential/beneficial and non-essential elements variability, respectively, whereas salinity was one of the main factors determining the accumulation of a number of elements (Mg, S, Na, Li, Sr, Sb) by the macrophytes. A decrease in the essential element contents (K, P, N, Cu, Zn) was observed in both species of the macrophytes during the growing season. Environmental factors (salinity, growing period, and pH) were more significant than the element contents in water and the sediments for the essential/beneficial element accumulation by two species of the macrophytes. The element contents in the sediments to a greater extent determined non-essential element accumulation by P. australis than by S. pectinata. The study showed that the efficiency of using P. australis and S. pectinata for remediation of aquatic ecosystems contaminated by heavy metals should not decrease with a salinity increase. © 2020 Elsevier Masson SAS
