Аннотация: All of man's former space flights were not real ventures into space in the biological sense, as his life was supported with unregenerated earth supplies. The coming stage of space exploration requires man's long existence in the cosmos and on the other planets. This stage of man's activity outside the earth become possible only by creating small man-made ecosystems, permitting the support of his metabolism by the recycling of substances of the terrestrial biosphere. Creation of such systems is a new scientific and technical task. Man-made ecosystems are a new product of man's activity, which have no complete analogy, either in nature, or in technology. Stochastic mechanisms, which stabilize biogeocenosis, cannot be effective in small ecosystems. A technique of parametric control over biosynthesis made it possible to calculate, and put to practice, an ecosystem for man with a cyclic regeneration of the atmosphere, water and, partially, food. The specific bio-technological properties of small man-made ecosystems are being analysed. The possibility of their application for man's excursions into space and for the settlement of other planets is being considered.

Аннотация: The examination of four species of luminous bacteria Photobacterium leiognathi, Photobacterium phosphoreum, Vibrio fischeri and Vibrio harveyi has enabled us to reveal some nutrient medium components effecting growth, luminescence intensity and luciferase synthesis. These agents are nucleic components (nucleotides, nucleotides and amine bases), amino acids and vitamins, which are part of hydrolysates from the biomass of various lithotrophic microorganisms, hydrogen-oxidizing, iron-oxidizing and carboxydobacteria. The effect of promoting agents essentially alters the physiological state and ultrastructure of the cells of luminous bacteria and increases luciferase biosynthesis two- to three-fold compared to a control.

Аннотация: Two chemolithoautotrophs - the hydrogen-oxidizing bacterium Alcaligenes eutrophus and the carboxydobacterium Seliberia carboxydohydrogena - were investigated with respect to their ability to produce heteropolymeric polyhydroxyalkanoates (PHA) during mixotrophic growth on CO2 and medium-chain-length (C6 to C9) alkanoic acids. Hydroxy derivatives of the alkanoic acids were incorporated into PHA synthesized by the bacteria studied in proportions dependent on the time of cultivation. Irrespective of the nature of the acid added to the medium, PHAs were found to contain hydroxybutyrate as the main constituent and also hydroxyvalerate and hydroxyhexanoate, whose maximal incorporation was observed 4 to 6 h after the addition of an alkanoic acid to the medium. During further cultivation, the fractions of hydroxyvalerate and hydroxyhexanoate in PHA decreased, and the fraction of hydroxybutyrate and the total intracellular content of the polymer increased. Three-component polyhydroxyalkanoates with different proportions of hydroxybutyrate, hydroxyvalerate, and hydroxyhexanoate were obtained, and some of their properties were analyzed. Аннотация: Translation of apoobelin mRNA in a cell-free wheat germ translation system in the presence of coelenterazine and molecular oxygen results in cotranslational formation of active photoprotein. Active obelin formation is recorded by its luminescence, either direct in the translation mixture in the presence of coelenterazine and calcium ions or in aliquots from the translation mixture. In the second case translation is carried out with coelenterazine and EGTA. Registration of the translation course by luminescence of the synthesized product in both cases allows use of apoobelin mRNA at very low concentrations as an internal marker for immediate measure of protein biosynthesis activity of in vitro translation systems. It is shown that the simultaneous translation of any other mRNA does not affect translation of photoprotein mRNAs under standard conditions. Continuous registration of luminescence in a cuvette of a liquid scintillation counter in photon-counting mode varies the time of signal accumulation in a wide temporal range, thus increasing the numerical values of the recorded signals. Registration of photoprotein luminescence during translation can be used to obtain additional information about the translation process, for example codon reading speed, about protein folding, and about the formation of active proteins on ribosomes. Аннотация: Scytalone dehydratase is a molecular target of inhibitor design efforts aimed at preventing the fungal disease caused by Magnaporthe grisea. A method for cocrystallization of enzyme with inhibitors at neutral pH has produced several crystal structures of enzyme-inhibitor complexes at resolutions ranging from 1.5 to 2.2 Angstrom Four high resolution structures of different enzyme-inhibitor complexes are described. In contrast to the original X-ray structure of the enzyme, the four new structures have well-defined electron density for the loop region comprising residues 115-119 and a different conformation between residues 154 and 160. The structure of the enzyme complex with an aminoquinazoline inhibitor showed that the inhibitor is in a position to form a hydrogen bond with the amide of the Asn131 side chain and with two water molecules in a fashion similar to the salicylamide inhibitor in the original structure, thus confirming design principles. The aminoquinazoline structure also allows for a more confident assignment of donors and accepters in the hydrogen bonding network, The structures of the enzyme complexes with two dichlorocyclopropane carboxamide inhibitors showed the two chlorine atoms nearly in plane with the amide side chain of Asn131. The positions of Phe53 and Phe158 are significantly altered in the new structures in comparison to the two structures obtained from crystals grown at acidic pH, The multiple structures help define the mobility of active site amino acids critical for catalysis and inhibitor binding. Аннотация: Modeling of transgenic microorganism introduction into small man-made ecosystems can help forecast changes in expression of cloned genes under different conditions of existence. Introduction of the E. coli Z905/pPHL7 strain containing a plasmid with luminescent system genes of luminous bacteria led to changes in cell and colony morphology, reduction in metabolic activity of cells, and, as a result, a lower level of expression of cloned gene. A low concentration of nutrients has been shown to favor greatly the phenotypic change of cells of the recombinant strain. Expression of cloned genes changed due to: a lower concentration of plasmid DNA, a change in regulation of cloned genes, and a change in cells of biosynthesis of substrates needed for expression of luminescent genes. The conducted investigations can provide a basis for the use of marker transgenic microorganisms in closed ecosystems of different types. (C) 2001 COSPAR. Published by Elsevier Science Ltd. Аннотация: The effects of elevated temperature (35 and 45В°C) on photosynthesis, respiration, and both the qualitative and quantitative compositions of volatile emissions (VE) of wheat (Triticum aestuvum L. cultivar 232) cenoses at light intensities of 70, 150, or 240 W/m2 of photosynthetically available radiation (PAR) were studied. At a PAR of 240 W/m2, the thermal stabilities of photosynthesis and respiration increased at 35В°C and decreased at 45В°C. Elevated temperatures nonuniformly changed the rates and direction of VE syntheses. In this process, the highest increase in VE evolution was observed at 70 W/m2 and 35В°C; the lowest, at 240 W/m2. In addition, the concentrations and composition of VE during the repair period differed from the initial values.

Аннотация: The lipid composition of the green alga Botryococcus was studied at three different cultivation temperatures: suboptimal, optimal, and supraoptimal. Cultivation at the supraoptimal temperature was found to considerably inhibit the synthesis of nearly all intracellular lipids, except for triacylglycerides, and to influence their fatty acid composition. In particular, the content of trienoic fatty acids was significantly lower at the supraoptimal than at the optimal cultivation temperature. At the same time, the fatty acid composition of the extracellular lipids of the alga virtually did not depend on cultivation temperature.

Аннотация: Kinetic parameters of a culture of the hydrogen-oxidizing bacterium Ralstonia eutropha grown on a gas substrate under conditions favoring autotrophic biosynthesis of polyhydroxybutyrate were studied. The following parameters, making it possible to control and optimize the process in industrial situations, were determined: specific rate of substrate consumption, physical properties of the culture medium, and coefficients of heat emission and mass transfer.

Аннотация: MELiSSA is a microorganism based artificial ecosystem conceived as a tool for understanding the behavior of ecosystems and developing the technology for future Manned Space Missions. MELiSSA is composed of four compartments colonized by the microorganisms required by the function of this ecosystem : breakdown of waste produced by men, regeneration of atmosphere and biosynthesis of edible biomass. This paper reports the mass balance description of a Biological Life Support System composed of the MELiSSA loop and of a Higher Plant Compartment working in parallel with the photosynthetic Spirulina compartment producing edible biomass. Аннотация: The paper considers problems of biosynthesis of higher plants' biomass and "bioloaical incineration" of plant wastes in a working physical model of biological LSS. The plant wastes are "biologically incinerated" in a special heterotrophic block involving Californian worms, mushrooms and straw. The block processes plant wastes (straw, haulms) to produce soil-like substrate (SLS) on which plants (wheat, radish) are grown. Gas exchange in such a system consists of respiratory gas exchange of SLS and photosynthesis and respiration of plants. Specifics of gas exchange dynamics of high plants - SLS complex has been considered. Relationship between such a gas exchange and PAR irradiance and age of plants has been established. Nitrogen and iron were found to the first to limit plants' growth on SLS when process conditions are deranged. The SLS microflora has been found to have different kinds of ammonifying and denitrifying bacteria which is indicative of intensive transformation of nitrogen-containing compounds. The number of physiological groups of microorganisms in SLS was, on the whole, steady. As a result, organic substances - products of exchange of plants and microorganisms were not accumulated in the medium, but mineralized and assimilated by the biocenosis. Experiments showed that the developed model of a manmade ecosystem realized complete utilization of plant wastes and involved them into the intrasystem turnover. Аннотация: Hepatocyte proliferation in the liver regenerating after partial hepatectomy ceases when the organ is restored, and the mechanism of this phenomenon is still unclear. In the experiments on fusing hepatocytes from the reoenerated mouse liver (15 days after partial hepatectomy) with NIH 3T3 mouse fibroblasts, we revealed no DNA synthesis in the nuclei of stimulated fibroblasts in heterokaryons (in the presence of hepatocyte nuclei), whereas DNA synthesis in nonfused cells was undisturbed. In this work, our purpose was to find out whether the suppression of DNA synthesis in heterokaryons could be due to the appearance in hepatocytes of some endogenous factors having an inhibitory effect on proliferation. To this end, hepatocytes from the mouse liver regenerated after partial hepatectomy were treated with cycloheximide for 1-4 h and were then fused with stimulated fibroblasts. Such a short-term treatment of hepatocytes with cycloheximide proved to result in the loss of their ability to inhibit DNA synthesis in the nuclei of stimulated or quiescent fibroblasts in heterokaryons, but hepatocytes proper actively proliferated in the medium with a low serum content (0.2%). When the mice with the liver reoenerated after partial hepatectomy were treated with a single sublethal dose of cycloheximide (3 mg/kg), their hepatocytes taken two days after this treatment had no inhibitory effect. Puromycin, another inhibitor of protein synthesis, had the same effect on hepatocytes. These results may be interpreted as evidence that the final stage of liver regeneration after damage is controlled by the factors having a C C negative effect on cell proliferation.

Аннотация: MELiSSA is a microorganism based artificial ecosystem conceived as a tool for understanding the behavior of ecosystems and developing the technology for future Manned Space Missions. MELiSSA is composed of four compartments colonized by the microorganisms required by the function of this ecosystem : breakdown of waste produced by men, regeneration of atmosphere and biosynthesis of edible biomass. This paper reports the mass balance description of a Biological Life Support System composed of the MELiSSA loop and of a Higher Plant Compartment working in parallel with the photosynthetic Spirulina compartment producing edible biomass. The recycling efficiencies of the system are determined and compared for various working conditions of the MELiSSA loop with or without the HPC.
