Аннотация: The problem of correlation between rheological properties in macro- and micro- scales of media with biopolymers of polypeptide (gelatin) and polysaccharide (starch) nature is investigated. The viscosity of the biopolymer solutions with concentrations 0.5-5 wt% was estimated by standard rotational rheometry technique and with fluorescent molecular rotor at 15-50 A degrees C. Opposite trends were observed for relationship between microviscosity eta (m) and macroviscosity eta for two biopolymers: eta (m) << eta for gelatin and eta (m) >> eta for starch solutions. The temperature dependence of eta (m) followed the monoexponential decay law in all samples over the whole temperature range indicating insensitivity of microviscosity to gel mesh melting under heating. The dissimilarity of macro- and micro-rheological properties of gelatin and starch-containing media is discussed in terms of difference in architecture of the gels.

Аннотация: The physicochemical properties such as the degree of crystallinity and temperature and molecularmass characteristics of a number of polyhydroxyalkanoates of various chemical composition synthesized on a complex carbon substrate by bacteria Cupriavidus eutrophus В10646 have been investigated. Two-, three-, and four-component copolymer samples have different sets and ratios of monomers with various lengths of carbon chains: 3-hydroxybutyrate (3HB), 4-hydroxybutyrate (4HB), 3-hydroxyvalerate (3HV), 3-hydroxyhexanoate (3HH), 3-hydroxy-4-methyl valerate (3H4MV), and diethylene glycol (DEG). It has been shown that weight-average molar mass Мw and polydispersity vary in a wide range with no correlation existing with the composition of copolymer polyhydroxyalkanoates and that thermal stability is preserved in the temperature interval between the melting temperature and the thermal degradation temperature from 100 to 120–140°С. The composition and ratio of monomers most notably affect the degree of crystallinity of polyhydroxyalkanoates. Significant differences between the degrees of crystallinity of three- and four-component polyhydroxyalkanoates have been found for the first time. The degree of crystallinity for copolymers P(3HB/3HV/4HB) is 9–22%, and the degree of crystallinity for copolymers P(3HB/3HV/3HH) and P(3HB/3GV/3H4MV) is 41–63%; this value is close to the degree of crystallinity for diblock copolymers P(3HB)/DEG, which is 56–69%. For the four-component copolymers P(3HB/3GV/4HB/3HH), the degree of crystallinity is 30–41%. The values of Мw for the copolymers P(3HB/DEG) are inhomogeneous and the polymers contain fractions uneven with respect to molecular mass: a high-molecular-mass polymer (Мw from 2700 to 4900 kDa) and a low-molecular-mass polymer (Мw = 46–167 kDa). For the copolymers P(3HB)/DEG and P(3HB/3HV/3H4MV), two peaks are observed in the region of melting with the gap between these peaks being 4–20°С. All of the types of copolymer samples, regardless of the monomer ratio, show an increase in elongation at break against the background of a decrease in tensile stress and Young’s modulus, with these effects being pronounced to different extents. On the whole, the properties of multicomponent polyhydroxyalkanoates differ appreciably. © 2017, Pleiades Publishing, Ltd.

Аннотация: According to the results of seasonal monitoring, in 2007–2013 purple sulfur bacteria morphologically similar to Thiocapsa sp. Shira_1 (AJ633676 in EMBL/GenBank) predominated in the anoxygenic phototrophic community of the water column of the meromictic Lake Shira (Khakassia, Siberia). No pronounced seasonal periodicity in the total cell number in the water column was revealed during the period of observation. In some years cell number during the period when the lake was covered with ice was reliably higher than in summer. The absence of seasonal periodicity was probably due to the low amplitude of seasonal variations in temperature and illumination in the redox zone, resulting from its relatively deep location (12–16 m). The year-to-year dynamics was characterized by a reliable decrease of the total cell number in 2009–2010 and maxima in 2007 and 2011–2012. Canonical correlation analysis revealed that water temperature in the redox zone was the best predictor of the PSB abundance in Lake Shira. Water temperature, in turn, depended on the depth of mixing of the water column. Intense mixing in 2009–2011 was probably responsible for decreased PSB abundance in the lake. On the other hand, the absence of deep winter mixing, resulting in stable conditions in the chemocline, favored the preservation of relatively high PSB biomass. Prediction of circulation depth, which depends mainly on the weather conditions and dynamics of the water level, is required for prediction of PSB abundance in Lake Shira. These results may be useful for paleolimnological reconstructions of the history of the lake based on the remnants of purple sulfur bacteria in bottom sediments. © 2016, Pleiades Publishing, Ltd.

Аннотация: Multiplex simultaneous genotyping technique was developed for four polymorphisms in genes coding for blood coagulation factors and homocysteine metabolism which are considered as thrombophilia related mutations: FV Leiden, FII G20210A, MTHFR C677T, and FVII G10976A. It is based on primer extension reaction with the following bioluminescent solid-phase microassay. At that, two different in bioluminescence obelin mutants were applied to simultaneous detection of two gene allelic variants. The assay is carried out in microtiter plate format and provides fast and reliable genotyping of four single nucleotide polymorphisms in four different genes within 2.5 hours. A large number of clinical samples were analyzed and the obtained results were found to be in complete correlation with those obtained by using conventional RT-PCR techniques. © 2015 Bentham Science Publishers.

Аннотация: This study aimed to analyze the relationship between chlorophyll potential, chlorophyll content, and grain yield of different wheat cultivars by using ground remote sensing and laboratory data. Chlorophyll potential is the difference between the reflectance factor integrals with and without chlorophyll absorption (in the 550–730 nm wavelength range). Ground-truth data were obtained at the experimental fields located in the Krasnoyarsk region, Russia (2002–2012). Experiments were conducted in different seasons under various lighting conditions by controlling plants states and soil types. Spectral measurements were obtained using a double-beam spectroradiometer, which was installed on a mobile work platform at a height of 5–18 m. The photometric area varied from 0.5 to 2 m2. The study showed good correlation (R2 = 0.9) between chlorophyll potential and chlorophyll content for different wheat cultivars. However, the correlation between chlorophyll potential and grain yield was less (R2 = 0.8). The values of chlorophyll potentials depended on the type of wheat during the growing season. The novelty of the approach is that it calculates the chlorophyll potential with additional spectral regions when compared with normalized difference vegetation index. We used a spectroradiometer with high spectral resolution. This increased the accuracy and stability of measurements in rapidly changing conditions. © 2016

Аннотация: The harmful potential of bulk bottom sediments of the Yenisei River has been assessed by the Allium test. Sediment samples have been taken in ten sites on a plot of about 100 km in length in the middle reaches of the Yenisei River affected by urban activity of the city of Krasnoyarsk and industrial plants. The samples of sediments differ considerably in the content of potentially toxic substances (heavy metals, artificial radionuclides, and organic pollutants) and nutrients and in physical properties. We use the mean length of onion root as an indicator of general toxicity, the mitotic index as an indicator of cytotoxicity, and the percentage of anatelophase cells in the apical root meristem with abnormal chromosomes as an indicator of genotoxicity. Both the stimulation and inhibition of indicator endpoints are revealed in sediment samples of the Yenisei River relative to the control (artificial sediments). An increased genotoxicity is recorded in the sample of sediments contaminated by a mixture of organic and chemical toxicants. A significant positive correlation between the percentage of abnormal cells and the content of copper and petrochemicals in the samples is revealed. No extremely strong reactions of onion endpoints to the quality of sediment samples are revealed, which is probably related to relatively low contamination of the sediments studied. This is in agreement with the data of chemical analyses. Based on the testing results, we can conclude that the endpoints of the Allium test are sensitive enough to the quality of bottom sediments of the Yenisei River and can be used for their biotesting.

Аннотация: Vegetation indices derived from remote sensing measurements are commonly used to describe and monitor vegetation. However, the same plant community can have a different NDVI (normalized difference vegetation index) depending on weather conditions, and this complicates classification of plant communities. The present study develops methods of classifying the types of plant communities based on long-term NDVI data (MODIS/Aqua). The number of variables is reduced by introducing two integrated parameters of the NDVI seasonal series, facilitating classification of the meadow, steppe, and forest plant communities in Siberia using linear discriminant analysis. The quality of classification conducted by using the markers characterizing NDVI dynamics during 2003-2017 varies between 94% (forest and steppe) and 68% (meadow and forest). In addition to determining phenological markers, canonical correlations have been calculated between the time series of the proposed markers and the time series of monthly average air temperatures. Based on this, each pixel with a definite plant composition can be characterized by only four values of canonical correlation coefficients over the entire period analyzed. By using canonical correlations between NDVI and weather parameters and employing linear discriminant analysis, one can obtain a highly accurate classification of the study plant communities. © 2019 by the authors.

Аннотация: We used a Lake Shira numerical model to estimate the response of the ecosystem of a saline meromictic lake to variations in weather parameters during the growing season. The sensitivity analysis of the model suggests that compared to other external (nutrient inflows) and internal (spring biomasses of food-web components) factors, weather parameters are among the most influential for both mixolimnetic (phyto- and zooplankton) and monimolimnetic (purple sulfur bacteria, sulfur reducing bacteria and hydrogen sulfide) food-web components. Calculations with different weather scenarios shows how changes in the water temperature and mixing depth affect mixolimnetic and monimolimnetic food-web components and the depth of the oxic-anoxic interface in a meromictic lake. When weather forcing stimulates an increase in the biomass of food-web components in the mixolimnion, it produces cascading effects that lead to three results: 1) a higher content of detritus in the water column; 2) a higher content of hydrogen sulfide in the monimolimnion; 3) raising of the oxic-anoxic interface closer to the water-air surface. This cascading effect is complicated by the negative correlation between two light dependent primary producers located at different depths—phytoplankton in the mixolimnion and purple sulfur bacteria at the oxic-anoxic interface. Thus, weather conditions that stimulate higher phytoplankton biomass are associated with a higher detritus content and lower biomass of purple sulfur bacteria, a higher content of hydrogen sulfide and a shallower oxic-anoxic interface. The same weather conditions (higher wind, lower cloud cover, and lower air temperature) promote a scenario of less stable thermal stratification. Thus, our calculations suggest that weather parameters during the summer season strongly control the mixing depth, water temperature and the mixolimnetic food web. An effect of biogeochemical and physical interactions on the depth of the oxicanoxic interface is also detectable. Аннотация: The variability of the state of forest vegetation was studied basing on the analysis of NDVI time series (2003–2016) of coniferous and deciduous stands and climate in Krasnoyarsk Territory (Stolby Nature Reserve). The initial data were 8-day Modis satellite information (MOD09Q1 product) and meteorological information from terrestrial weather stations. It was revealed that the trends of the averaged NDVI of forest vegetation for the period May – September and the maximum NDVI were negative. An analysis of the relationship between the dynamics of NDVI forest vegetation and the hydrothermal factor on the territory of the reserve in the 14-year cycle revealed an insignificant correlation between these variables. The negative NDVI trends of coniferous and deciduous stands indicate degradation processes. Deterioration of the state of forest vegetation in the study area detected by satellite data is explained by a combination of factors: climate change, anthropogenic impact of Krasnoyarsk and presence of old-aged forest. © 2018 Space Research Institute of the Russian Academy of Sciences. Аннотация: A series of two-component polyhydroxyalkanoates consisting of hydroxybutyrate and hydroxyvalerate monomer at different ratios were synthesized using the bacterium Ralstonia eutropha B5786. The properties of polyhydroxyalkanoates were compared with those of the homopolymer of hydroxybutyric acid by X-ray structure analysis, IR spectroscopy, differential scanning calorimetry, and viscosimetry. With an increase in the molar fraction of hydroxyvalerate, an equalization of the ratio of the crystalline and amorphous phases in the copolymer was observed. The degree of crystallinity of the polymer decreased from 70-80 to 45-50%; in the range of an increase in the hydroxyvalerate molar fraction from several to 25-30 mol %, the dependence was linear. The temperature characteristics, the melting temperature (T-m), and the degradation temperature (T-d) were lower in polyhydroxyalkanoates than in polyhydroxybutyrate, for which T-m and T-d were 168-170 and 260-265degreesC, respectively. In the copolymer, as the molar fraction of hydroxyvalerate grew, both parameters decreased. In the range of variation of monomer ratio studied, they decreased to 150-160 and 200-220degreesC, respectively. No distinct correlation between the composition of the polymer and its molecular mass was found.

Аннотация: The time-resolved and steady-state fluorescence techniques,were employed to elucidate possible interactions of four aromatic compounds (anthracene, POPOP, MSB and 1,4-naphthalendiol) with bacterial luciferase. Fluorescence spectra and fluorescence anisotropy decays of these compounds were studied in ethanol, water-ethanol solutions and in the presence of bacterial luciferase. Shifts of fluorescent spectra and differences in rotational correlation times are interpreted in terms of weak (hydrophobic) interactions of the molecules with the enzyme. These interactions suggest the feasibility of intermolecular energy transfer by an exchange resonance mechanism with a collision-interaction radius as a way of excitation of these compounds in the reaction catalysed by bacterial luciferase. Аннотация: 1. We studied the composition of fatty acids (FAs) in the seston from two small freshwater reservoirs (Bugach and Lesnoi) with distinct periodicity of domination by cyanobacteria and eukaryotic algae during the growth season. 2. The diatoms in the both reservoirs were characterised by a high content of 14:0 and C16 unsaturated acids, whereas that of the essential FA 20:5omega3 [eicosapentanoic acid (EPA)] was low. The correlation between this polyunsaturated FA (PUFA) and diatom biomass was not significant in either reservoir. The percentage of 20:5omega3 in seston significantly correlated with the biomass of euglenophyta in Bugach and dinophyta in Lesnoi. Hence the diatoms, usually referred as a valuable food for zooplankton, were not an important source of the essential PUFA in these systems. 3. The dominant cyanobacteria in Bugach, and the green algae in Lesnoi, both contained the same marker acids: 18:3omega3 and 18:2omega6. Hence, a discrimination between these two phytoplanktonic groups on the basis of FA biomarkers may be difficult in some cases. 4. We found no significant correlation between the content of 20:5omega3 in seston and the biomass of the dominant daphniids in either reservoir. This is contrary to expectations, based on the literature, that EPA is generally important. Rather, the biomass of the two dominant Daphnia species in Bugach correlated strongly with the content of 18:3omega3 in the seston. The cyanobacteria were a probable source of this omega3 FA for Daphnia . We conclude that EPA is not always important for Daphnia populations although, in such cases, some other PUFA (e.g. 18:3omega3) might be related to their growth.

Аннотация: An optical remote assay for biological production of crops in the field during the vegetation period is proposed. Our calculations demonstrate a good correlation between the S(t) value and crop yield (in the range from 0.85 to 0.90); the higher is S(t) for the vegetation period, the higher is the crop yield. S(t) value can be used to calculate the increase in dry and wet biomass for various crops during the whole vegetation period with an error of less than 10-12%. The absolute error of crop yield for 110 cultivars of wheat, oats, and barley during the experimental period was +/-3-3.5 quintal/ha with the mean crop yield of 14-38 quintal/ha.

Аннотация: The bioluminescent bacterial enzyme system NAD(P)H:FMN-oxidoreductase-luciferase has been used as a test system for ecological monitoring. One of the modes to quench bioluminescence is the interaction of xenobiotics with the enzymes, which inhibit their activity. The use of endogenous flavin fluorescence for investigation of the interactions of non-fluorescent compounds with the bacterial luciferase from Photobacterium leiognathi and NAD(P)H:FMN-oxidoreductase from Vibrio fischeri has been proposed. Fluorescence spectroscopy methods have been used to study characteristics of endogenous flavin fluorescence (fluorophore lifetime, the rotational correlation time). The fluorescence anisotropy behaviour of FMN has been analysed and compared to that of the enzyme-bound flavin. The fluorescence characteristics of endogenous flavin of luciferase and NAD(P)H:FMN-oxidoreductase have been shown to be applicable in studying enzymes' interactions with non-fluorescent compounds. Аннотация: Multiplex simultaneous genotyping technique was developed for four polymorphisms in genes coding for blood coagulation factors and homocysteine metabolism which are considered as thrombophilia related mutations: FV Leiden, FII G20210A, MTHFR C677T, and FVII G10976A. It is based on primer extension reaction with the following bioluminescent solid-phase microassay. At that, two different in bioluminescence obelin mutants were applied to simultaneous detection of two gene allelic variants. The assay is carried out in microtiter plate format and provides fast and reliable genotyping of four single nucleotide polymorphisms in four different genes within 2.5 hours. A large number of clinical samples were analyzed and the obtained results were found to be in complete correlation with those obtained by using conventional RT-PCR techniques.

Аннотация: 1. We studied variation in the composition of fatty acids in the seston of a small freshwater reservoir with changes in phytoplankton composition during four growth seasons. We focused on the dynamics of the omega3 fatty acids because of their potential importance for zooplankton nutrition. 2. Total diatoms were related to the 20:5omega3 fatty acid (eicosapentaenoic, EPA) content in seston. Among two dominant diatom genera, Cyclotella was not associated with EPA content. In contrast, there was a significant correlation between Stephanodiscus and the percentage contribution and content of EPA throughout the study. Hence, freshwater diatoms can differ strongly in content of the essential EPA. 3. We considered abundant cyanobacteria as a potential source of 18:3omega3 fatty acid (linolenic, ALA) to aquatic food webs. Among four dominant cyanobacteria species, two (Anabaena flos-aquae and Planktothrix agardhii) showed significant correlation with the ALA content of the seston, while the other two (Aphanizomenon flos-aquae and Microcystis aeruginosa) did not. 4. Dinophyta had a relatively high level of 22:6omega3 (docosahexaenoic, DHA) for freshwater species and can be also a source of EPA to aquatic food webs. 5. Our results show that various species of diatoms as well as cyanobacteria can be of contrasting nutritional value for zooplankton because of their different content of the essential PUFAs. Diatoms, which are low in EPA, could not be considered as a valuable food, while some field populations of cyanobacteria might be valuable sources of essential ALA.

Аннотация: The possibility of symbiotic, mutualistic (++) interrelations between epibiontic microalgae (Characidiopsis ellipsoidea, Colacium vesiculosum) and cyclopoids (Cyclops vicinus, Mesocyclops leukarti) in the Bugach recreational reservoir with a bloom of cyanobacteria is demonstrated. Overgrown cyclopoids, compared to those free from microalgae, have advantages in oxygen supply, which may be insufficient in the period of intensive algal bloom. Calculations have shown that oxygen requirements of cyclopoids may be fully satisfied due to C. vesiculosum photosynthetic activity in the dysphotic zone if the density of overgrowth exceeds 1000 algal cells per individual. There is no significant correlation between the natural mortality of crustaceans and the indices of their overgrowth by microalgae.

(Alaska), Canada, Greenland, Iceland, the Faroe Islands, Norway, Sweden, Finland, and Russia. The main objectives of this study were: (1) to assess spatial patterns of plankton diversity focusing on pelagic communities; (2) to assess dominant component of ? diversity (turnover or nestedness); (3) to identify which environmental factors best explain diversity; and (4) to provide recommendations for future monitoring and assessment of freshwater plankton communities across the Arctic region. Phytoplankton and crustacean zooplankton diversity varied substantially across the Arctic and was positively related to summer air temperature. However, for zooplankton, the positive correlation between summer temperature and species numbers decreased with increasing latitude. Taxonomic richness was lower in the high Arctic compared to the sub- and low Arctic for zooplankton but this pattern was less clear for phytoplankton. Fennoscandia and inland regions of Russia represented hotspots for, respectively, phytoplankton and zooplankton diversity, whereas isolated regions had lower taxonomic richness. Ecoregions with high ? diversity generally also had high ? diversity, and turnover was the most important component of ? diversity in all ecoregions. For both phytoplankton and zooplankton, climatic variables were the most important environmental factors influencing diversity patterns, consistent with previous studies that examined shorter temperature gradients. However, barriers to dispersal may have also played a role in limiting diversity on islands. A better understanding of how diversity patterns are determined by colonisation history, environmental variables, and biotic interactions requires more monitoring data with locations dispersed evenly across the circumpolar Arctic. Furthermore, the importance of turnover in regional diversity patterns indicates that more extensive sampling is required to fully characterise the species pool of Arctic lakes. © 2021 The Authors.
