ferrihydrite nanoparticles -- concentration effect -- microorganisms -- Klebsiella oxytoca -- neutrophilic granulocytes -- chemiluminescence -- toxicology
Аннотация: Biogenic ferrihydrite nanoparticles were synthesized as a result of the cultivation of Klebsiella oxytoca microorganisms. The distribution of nanoparticles in the body of laboratory animals and the physical properties of the nanoparticles were studied. The synthesized ferrihydrite nanoparticles are superparamagnetic at room temperature, and the characteristic blocking temperature is 23-25 K. The uncompensated moment of ferrihydrite particles was determined to be approximately 200 Bohr magnetons. In vitro testing of different concentrations of ferrihydrite nanoparticles for the functional activity of neutrophilic granulocytes by the chemiluminescence method showed an increase in the release of primary oxygen radicals by blood phagocytes when exposed to a minimum concentration and a decrease in secondary radicals when exposed to a maximum concentration. In vivo testing of ferrihydrite nanoparticles on Wister rats showed that a suspension of ferrihydrite nanoparticles has chronic toxicity, since it causes morphological changes in organs, mainly in the spleen, which are characterized by the accumulation of hemosiderin nanoparticles (stained blue according to Perls). Ferrihydrite can also directly or indirectly stimulate the proliferation and intracellular regeneration of hepatocytes. The partial detection of Perls-positive cells in the liver and kidneys can be explained by the rapid elimination from organs and the high dispersion of the nanomaterial. Thus, it is necessary to carry out studies of these processes at the systemic level, since the introduction of nanoparticles into the body is characterized by adaptive-proliferative processes, accompanied by the development of cell dystrophy and tension of the phagocytic system.

Аннотация: For analytical purposes bioluminescence can be used in three main ways: 1. luminescence measurement of bioluminescent system components isolated in vitro; 2. determination of luminous organisms' reaction to the in vivo test-action; 3. measurement of bioluminescence in marine ecological systems. The majority of the reports of this Symposium are dealing with the first two topics. The aim of our presentation is to draw attention to the third one. The possibilities of bioluminescent analysis are wider than its traditional scheme of applications in the laboratory, when the emitting system is withdrawn from a native source and is placed in a cuvette of the light measuring device. The reverse scheme is also possible, i.e. the device can be introduced into light emitting system such as a marine biocenosis--the community of the sea inhabitants--where we obtain a highly sensitive and rapid means of gaining the information on the vital activity of marine ecosystems, i.e. their spatial structure, rhythms, man's influence upon them, etc. The present communication will consider the possibilities of this form of bioluminescent analysis.

Аннотация: The influence of nanodiamonds synthesized by detonation on human red and white blood cells in vitro was studied. Nine nanodiamond samples produced by different manufacturers and two samples of modified nanodiamonds with high colloidal stability were used in the study. It was shown that nanoparticles cause the destruction of white cells and the hemolysis of erythrocytes. Possible mechanisms of the desctructive effect of detonation nanodiamonds on blood-forming elements are discussed.

Аннотация: A green flavoprotein (GFP) was isolated and purified to homogeneity from Photobacterium leiognathi, strain 208. GFP is a homodimer of molecular weight 54,000 and contains two molecules of an unusual flavin per molecule of protein. Various biochemical characteristics including isoelectric point, trypsin and chymotrypsin degradation, SDS and temperature influence on subunit dissociation and the dissociation of the flavin chromophore, were investigated. The sequence of 23 N-terminal amino acids was determined and found to be concurrent with the N-terminal amino acid sequence encoded by the lux G(N) gene of P. leiognathi. This fact suggests that GFP is a structural component of the Photobacterium luminescence system.

Аннотация: The examination of four species of luminous bacteria Photobacterium leiognathi, Photobacterium phosphoreum, Vibrio fischeri and Vibrio harveyi has enabled us to reveal some nutrient medium components effecting growth, luminescence intensity and luciferase synthesis. These agents are nucleic components (nucleotides, nucleotides and amine bases), amino acids and vitamins, which are part of hydrolysates from the biomass of various lithotrophic microorganisms, hydrogen-oxidizing, iron-oxidizing and carboxydobacteria. The effect of promoting agents essentially alters the physiological state and ultrastructure of the cells of luminous bacteria and increases luciferase biosynthesis two- to three-fold compared to a control.

Аннотация: The study includes anthropometry of 172 young male, obtained data on the length and body mass, measured the transverse diameters of the shoulders and pelvis, various body types was identified by the J.M. Tanner sexual dimorphism index (andromorphic, mesomorphic, gynecomorphic). The chemiluminescent and bioluminescent study of saliva and blood was conducted in the examined young male. We studied the indicators of the antioxidant defense system under the influence of stress. The antioxidant status of saliva was determined using the H2 O2-luminol-dependent chemiluminescence method. Data on the activity of NAD (P)-dependent dehydrogenases in blood lymphocytes was obtained from a bioluminescent method of research. Young male of andromorphic body type had large overall and transverse body sizes. Indicators of antioxidant protection of saliva and blood in men of adolescence, the body type of the sexual dimorphism index J.M. Tanner was different. The persons of the andromorphic body type differed in terms of chemiluminescence in comparison with the young male of gynecomorphic body type. The results of bioluminescent blood tests suggest a violation of the catabolic and anabolic processes of carbohydrate and fat metabolism in young men of mesomorphic and gynecomorphic body types. Indicators of the system of antioxidant protection of saliva and blood reflect the sexual characteristics of the body of young male and can be used as additional criteria for diagnosing sex inversion and assessing the risk of developing socially attributed diseases. © 2020, Klinichescheskaya Laboratornaya Diagnostika. Аннотация: Repeated plasmapheresis sessions attended by chemiluminescent evaluation of cellular immunity parameters lead to the recovery of the whole blood leucocyte activity in patients with unhealing gastroduodenal ulcers. The kinetic analysis of the cellular chemiluminescent response has shown that, among all the components of the plasmapheresis procedure, hemorrhage is the most important factor increasing the functional activity of phagocytes.
