Аннотация: The results of the development and utilization of an affine magnetic sorbent with Ni2+ ions immobilized on coal ash microspheres are reported. The applicability of the material in the isolation of Histag proteins is demonstrated by examples of the recombinant green fluorescent protein from Clytia gregaria and the Ca2+ regulated photoprotein obelin from Obelia longissima. The specific sorption capacity of the sorbent was 2-7 mg/cm3 for medium-size proteins (20-30 kDa). The particles are suitable for chromatography with the presence of chaotropic agents and EDTA. They are easy to manipulate as isolation of a target protein takes 30-35 min. On the one hand, the elevated affinity of the sorbent to proteins rich in native histidines may result in a high degree of irreversible sorption; on the other hand, it allows isolation of such proteins without the introduction of artificial polyhistidine fragments. Аннотация: AsLn2, an unusual modified peptide, was isolated from the bioluminescent earthworm Fridericia heliota (Enchytraeidae). Its structure, elucidated by NMR and mass spectrometry, includes residues of tyrosine, CompX (a novel tyrosine modification product, reported in the accompanying paper), and N(omega)-acylated lysine. Chromatography, UV, and 1H NMR data imply a close structural similarity of AsLn2 with F. heliota luciferin. AsLn2 appears to be an intermediate or by-product in F. heliota luciferin biosynthesis. © 2013 Elsevier Ltd. All rights reserved.

Аннотация: The study of the trophic relationships of aquatic animals requires correct estimates of their diets. We compared the quantitative fatty acid signature analysis (QFASA) and the isotope?mixing model IsoError, based on the compound?specific isotope analysis of fatty acids (CSIA?FA), which are potentially effective models for quantitative diet estimations. In a 21?day experiment, Daphnia was fed a mixture of two food items, Chlorella and Cryptomonas, which were supplied in nearly equal proportions. The percentages and isotope values of the FAs of the algal species and Daphnia were measured. The IsoError based on CSIA?FA gave an estimation of algae consumption using only one FA, 18:3n?3. According to this model, the proportion of consumption of Chlorella decreased while the proportion of consumption of Cryptomonas increased during the experiment. The QFASA model was used for two FA subsets—the extended?dietary subset, which included sixteen FAs, and the dietary one, which included nine FAs. According to both subsets, the portion of consumed Chlorella decreased from Day 5 to 10 and then increased at Day 21. The comparison of the two model approaches showed that the QFASA model is a more reliable method to determine the contribution of different food sources to the diet of zooplankton than the CSIA?based mixing model. © 2021 by the authors. Аннотация: Specimens of 14C-labeled polymer of 3-hydroxybutyric acid, P(3-HB), with different initial molecular weights, were used to prepare microparticles, whose morphology was not influenced by the M w of the polymer. During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: Polyhydroxyalkanoates have attracted much attention as biodegradable alternative to petroleum-based synthetic plastics. Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] copolymer is one of the best characterized PHA copolymers because of its high commercial potential. However, commercial use of PHAs has been limited by their high price. One approach to reducing the cost of PHA production is to use inexpensive carbon sources (fatty acids, plant oils, etc.). The aim of this work was to study synthesis of P(3HB-co-3HV) by the Cupriavidus necator B-10646 bacterium grown on oleic acid and different biochemical precursors of 3HV. Bacterial cells were grown for 72 h at 30°C and 200 rpm on an incubator shaker. Salts of propionic or valeric acids were used as precursors of 3HV. The content and the composition of the polymer were determined by gas chromatography of fatty acid methyl esters. Lipids and polymer were extracted from biomass using the method of Folch. The addition of potassium propionate and valerate did not inhibit bacterial growth and polymer synthesis, the cell concentration and polymer content reaching 9.3-9.5 g/L and 80-83%, respectively. The addition of potassium valerate or propionate led to the synthesis of (P(3HB-co-3HV)) copolymer containing 21.2 and 14.3 mol% of 3HV, respectively. The number average molecular weight (Mn) of the polymer synthesized by the bacterium on oleic acid alone was 220 kDa; the polydispersity of the polymer was 3.5. The polymer synthesized in the presence of potassium valerate and propionate was characterized by a lower Mn (156-178 kDa) and a higher polydispersity of the polymer (4.4-4.9). The main fatty acids (FA) of intracellular lipids were oleic (33.26% of the total FA) and palmitic acid (27.48% of the total FA). The addition of potassium propionate or valerate did not cause any significant changes in the composition of the FA of intracellular lipids of the strain studied. This study demonstrates the ability of C. necator B-10646 to synthesize P(3HB-co-3HV) from mixtures of oleic acid and 3HV precursors. The data obtained can be used to develop and implement an economically feasible process of the P(3HB-co-3HV) production. © Siberian Federal University. All rights reserved.

Аннотация: Effects of temperature on bioluminescent patterns of luciferases from luminescent bacteria Vibrio harveyi, Vibrio fischeri, Photobacterium leiognathi, and Photobacterium phosphoreum were studied. The highest luminescence level was observed at 15-25 degrees C for the luciferase from P. phosphoreum, at 20-30 degrees C for the V. fischeri and P. leiognathi enzymes, and at 30-37 degrees C for the enzyme from V. harveyi. All the luciferases were significantly stabilized at increased salt concentrations, at low pH values, or in the presence of dithiothreitol (DTT) and EDTA. The addition of DTT and EDTA affected the reversible stage of enzyme inactivation, while salts reduced the rate of the irreversible stage. A peak corresponding to aggregated protein was detected by gel chromatography of irreversibly inactivated luciferase. Activation energies were calculated for each luciferase in bioluminescent reactions with decanal, dodecanal, tetradecanal, and without aldehydes. The activation energy of the reaction with tetradecanal was much lower than those with the other aldehydes. The temperature dependence of the lifetime of the long-lived reaction intermediate showed that in the 10-30 degrees C interval all the luciferases, except for the enzyme from V. harveyi, have only one active form.

Аннотация: Three canned fish species - Pacific saury (Cololabis saira), Pacific herring (Clupea harengus) and Baltic sprat (Sprattus sprattus) - most common and popular in Russia, were analyzed for fatty acids. Special attention was paid to long-chain essential polyunsaturated fatty acids: eicosapentaenoic acid (20:5?3) and docosahexaenoic acid (22:6?3). Sums of eicosapentaenoic acid and docosahexaenoic acid in saury, herring and sprat were, on average, 2.42, 1.80 and 1.43 g/100 g product, respectively. Contents of these essential acids in all the canned fish species were found to be very high compared with many other fish reported in the available literature. All the canned fish appeared to be highly valuable products for human nutrition concerning the content of eicosapentaenoic and docosahexaenoic acids.

Аннотация: Contents of highly unsaturated long-chain fatty acids of omega-3 family (HUFA); eicosapentaenoic acid (EPA; 20:5n-3); and docosahexaenoic acid (DHA; 22:6n-3) are the principal indicators of the nutritive quality of fish for humans. Effects of environmental factors, first of all water temperature, on EPA and DHA contents in fish tissue are not currently completely understood. To reveal the putative effect of water temperature, fatty acid composition and contents were studied for 6 fish species, inhabiting the cold waters of the Yenisei River (Siberia, Russia) downstream of a dam (hypolimnetic release from reservoir) and its adjacent warm water tributaries (the Mana River and the Kacha River). It was hypothesized that (a) fish species from the cold river would have higher HUFA contents than fish from the warm rivers and (b) temperature would be negatively correlated with HUFA content in fish species. Using gas chromatography-mass spectrometry, distinct species-specific fatty acid profiles were observed, whereas contents of the essential fatty acids, EPA and DHA, in fish species from the cold Yenisei River were in general similar to that from warm tributaries. Thus, in contrast to the first hypothesis, phylogenetic factors overweighed the effect of water temperature and food (benthic invertebrate) composition, on fatty acid composition and HUFA content in fish. For the second hypothesis, for the 2 species inhabiting both cold and warm rivers, only one had higher EPA and DHA content at lower temperatures. Consequently, the response of EPA and DHA content in fish tissue to temperature variations may be species-specific.

Аннотация: Streptavidin is a homotetrameric protein produced by Streptomyces avidinii, each subunit of which binds biotin (vitamin H), forming a stable complex (Kd = 10-15 M). Streptavidin-biotin coreaction is widely used in analytical systems, for targeted delivery of compounds, for affinity purification, etc. The aim of this study was to develop a rational technique to produce functionally active recombinant streptavidin. Recombinant Escherichia coli strains producing minimal core and full-sized streptavidin variants were obtained. The E. coli BL21 Codon Plus (DE3) RIPL, as host cells, and the pET19b plasmid carrying gene of minimally-sized core (miniSAV) or full-sized (SAV) streptavidin were used. Synthesis of miniSAV results in its localization as insoluble inclusion bodies. Denatured miniSAV yield was 130 mg per liter of E. coli c ulture. T he r enaturation g ives o nly 10- 15 % of the functionally active protein. Full-sized streptavidin localizes in the cytoplasm in a soluble state, but its toxicity causes low yield of the protein (10-13 mg per liter of the culture). The induction of SAV synthesis at the end of the logarithmic stage of cell growth was found to increase the yield of SAV approximately 2-fold. The yield of functionally active protein was 30 mg per liter culture. SAV was produced practically in individual state after affine chromatography on 2-iminobiotin agarose. One molecule of full-sized streptavidin bound 3.9 biotin molecules as was shown by colorimetric analysis using HABA (4-hydroxyazobenzene-2-carboxylic acid). Both streptavidins form sandwichtype complexes with biotinylated molecules in solid-phase microassay conditions. E. coli BL21 Codon Plus (DE3) RIPL/pET19bSAV strain was stable during storage with 20 % glycerol at -70 °C, which was shown by repeated two-year reseeding. The streptavidin producing strain (E. coli BL21 Codon Plus (DE3) RIPL/pET19bSAV) is deposited in the Collection for extremophile microorganisms and type cultures (Institute of Chemical Biology and Fundamental Medicine SB RAS, Novosibirsk), No. 3505. The method for producing functionally active recombinant streptavidin developed in this study ensures its availability for biotechnological research. © Siberian Federal University. With the use of ion chromatography, it was found that processes related to the absorption of SO 42- and Cl- anions by a solid phase with the release of NO 3- , PO 43- , and F- to a liquid phase competed in the test systems as the concentration of water-soluble organic carbon (WSOC) was increased. Only the test anions were released in the systems without the introduction of an additional amount of WSOC as humic acid. With the use of ?-ray spectrometry, it was found that the release of 60Co, 152Eu, and 241Am radionuclides to the liquid phase in the systems with added humic acid began much earlier than in the system without the addition of humic acid. In this case, the amount of released radionuclides was greater than the amount of radioisotopes released in the system without the addition of humic acid: ?25% 241Am, ?3% 152Eu, and ?0.8% 60Co in the system with added humic acid or 0.8% 152Eu and <0.1% 60Co in the system without the addition of humic acid. The 241Am radionuclide was not determined in the system without the addition of humic acid. An increase in the concentration of WSOC in the experimental system of bottom sediments-Yenisei river water initiated the release of 60Co, 152Eu, and 241Am anthropogenic radionuclides from bottom sediments because of the formation of soluble complexes capable of migration. An increase in the concentration of WSOC had almost no effect on the release of K-40 and Cs-137 radionuclides.

Аннотация: In 11 lakes from cold and warm regions we studied the content of highly unsaturated fatty acids (HUFA) in seston (<130?m) and crustacean zooplankton using gas chromatography-mass spectrometry. An increase of temperature correlated with a decrease of HUFA content in zooplankton. A multivariate canonical correlation analyses revealed, that the decrease of HUFA content was related with a decrease of per cent of copepods in zooplankton communities, which are known to have higher HUFA levels in their biomass, than cladocerans. This means that temperature primarily affected the HUFA levels indirectly, via changing of taxonomic structure of zooplankton community, while the homeoviscous adaptation of zooplankton individuals had comparatively lower importance. As found, water temperature was better predictor of HUFA contents of zooplankton, than the fatty acid composition of seston. Thus, it can be predicted, that a probable climate warming will decrease the content of the essential HUFA in freshwater zooplankton with possible negative consequences for animals of higher trophic levels. Аннотация: The extracellular enzyme with oxidase function was extracted from the Neonothopanus nambi luminescent fungus by using mild processing of mycelium with beta-glucosidase and then isolated by gel-filtration chromatography. The extracted enzyme is found to be a FAD-containing protein, catalyzing phenol co-oxidation with 4-aminoantipyrine without addition of H2O2, which distinguishes it from peroxidases. This fact allowed us to assume that this enzyme may be a mixed-function oxidase. According to gel-filtration chromatography and SDS-PAGE, the oxidase has molecular weight of 60 kDa. The enzyme exhibits maximum activity at 55-70 degrees C and pH 5.0. Kinetic parameters K-m and V-max of the oxidase for phenol were 0.21 mM and 0.40 mu M min(-1). We suggest that the extracted enzyme can be useful to develop a simplified biosensor for colorimetric detection of phenol in aqueous media, which does not require using hydrogen peroxide.

Аннотация: Using the original technique of treating biomass with beta-glucosidase, a pool of extracellular fungal enzymes was obtained for the first time from the mycelium of basidiomycete Neonothopanus nambi. Two protein fractions containing enzymes with oxidase activity were isolated from the extract by gel-filtration chromatography and conventionally called F1 and F2. Enzyme F1 has a native molecular weight of 80-85 kDa and does not contain chromophore components; however, it catalyzes the oxidation of veratryl alcohol with K-m = 0.52 mM. Probably, this enzyme is an alcohol oxidase. Enzyme F2 with a native molecular weight of approximately 60 kDa is a FAD-containing protein. It catalyzes the cooxidation of phenol with 4-aminoantipyrine without the addition of exogenous hydrogen peroxide, which distinguishes it from the known peroxidases. It was assumed that this enzyme may be a mixed-function oxidase. F2 oxidase has K-m value 0.27 mM for phenol. The temperature optimums for oxidases F1 and F2 are 22-35 and 55-70 degrees C, and pH optimums are 6 and 5, respectively.

Аннотация: The purpose of this study was to develop micro and nano sized drug carriers from poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), and study the cell and skin penetration of these particles. PHBV micro/nanospheres were prepared by o/w emulsion method and were stained with a fluorescent dye, Nile Red. The particles were fractionated by centrifugation to produce different sized populations. Topography was studied by SEM and average particle size and its distribution were determined with particle sizer. Cell viability assay (MTT) was carried out using L929 fibroblastic cell line, and particle penetration into the cells were studied. Transdermal permeation of PHBV micro/nanospheres and tissue reaction were studied using a BALB/c mouse model. Skin response was evaluated histologically and amount of PHBV in skin was determined by gas chromatography-mass spectrometry. The average diameters of the PHBV micro/nanosphere batches were found to be 1.9 mu m, 426 and 166 nm. Polydispersity indices showed that the size distribution of micro sized particles was broader than the smaller ones. In vitro studies showed that the cells had a normal growth trend. MTT showed no signs of particle toxicity. The 426 and 166 nm sized PHBV spheres were seen to penetrate the cell membrane. The histological sections revealed no adverse effects. In view of this data nano and micro sized PHBV particles appeared to have potential to serve as topical and transdermal drug delivery carriers for use on aged or damaged skin or in cases of skin diseases such as psoriasis, and may even be used in gene transfer to cells.

Аннотация: The analysis of the literature on the methods of soluble organosolv lignins isolation, their physical-chemical study and on the method of their processing to porous aerogels and liquid hydrocarbons was carried out. A review of the literature allowed us to choice of the most important areas of research. For isolation from wood the soluble lignins free from sulfur the methods of catalytic peroxide delignification at mild conditions (temperature <= 100 degrees C, atmospheric pressure) and methods of lignin extraction by supercritical organic solvents were used. Molecular mass and molecular-mass distribution of ethanol-lignin samples isolated from aspenwood and abies-wood were studied by gel-permeation chromatography. Weighted molecular mass of ethanol-lignin from abies wood is 478 Da and that from aspen wood ethanol-lignin - 750 Da. Thus, the studied samples of ethanol-lignin have rather low molecular mass, what should facilitate their further processing to liquid hydrocarbons and aerogels. For the depolymerization of organosolv lignins to liquid hydrocarbons the processes of their catalytic conversion in supercritical alcohols have good prospects for the use. In the processes of lignin thermal conversion alcohols can to extract the products of lignin depolymerization and to alkylate these products, preventing their repolymerization to high molecular mass substances. To obtain a new class of nanoporous materials based on lignin the methods of organic aerogels synthesis from mixtures of lignin with other natural polymers and crosslinking agents were applied. It was found that the structure and properties of porous materials of aerogel type depend not only from the reaction mixture composition but from the method of drying. Drying in subcritical conditions leads to the formation of xerogels, in supercritical conditions - to the formation aerogels and the freezdrying - of cryogels. Obtained porous materials can have very low density (around 0.2 g/cm(3)), high specific surface area (to 500 m(2)/g) and the pore volume near 5 cm(3)/g.

Аннотация: This paper presents the main results of RP-HPLC-MS and microscale NMR analysis performed on Accompanying similar to Luciferin (AsLn(x)), compounds present in extracts of the bioluminescent earthworm Fridericia heliota that display similarities with Fridericia's luciferin, the substrate of the bioluminescent reaction. Three isomers of AsLn were discovered, AsLn(1), AsLn(2) and AsLn(3), all of which present a molecular weight of 529 Da. Their UV-Vis absorption spectra show maxima at 235 nm for AsLn(1), 238 and 295 nm for AsLn(2) and 241 and 295 nm for AsLn(3). MS n fragmentation patterns suggest the existence of carboxylic acid and hydroxyl moieties, and possibly chemical groups found in other luciferins like pterin or benzothiazole. The major isomer, AsLn(2), presents an aromatic ring and alkene and alkyl moieties. These luciferin-like compounds can be used as models that could give further insights into the structure of this newly discovered luciferin. В© 2010 Elsevier Inc. All rights reserved.

Аннотация: Fish are recognized as the main source of physiologically important omega-3 long-chain polyunsaturated fatty acids, namely, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), for human nutrition. However, muscle tissue contents of these fatty acids in diverse fish species, i.e., their nutritive value for humans, varied within two orders of magnitude. We reviewed contents of EPA and DHA, measured by similar methods using an internal standard during chromatography as mg per g of wet mass in 172 fish species belonging to 16 orders, to evaluate probable variations in phylogenetic and ecological drivers. EPA + DHA content varied from 25.6 mg g(-1) of wet mass (Sardinops sagax) to 0.12 mg g(-1) (Gymnura spp.). Multidimensional redundancy analysis revealed that among phylogenetic, ecomorphological and abiotic environmental factors, the highest proportion of variation contribution belonged to the shared contribution of sets of phylogenetic and ecomorphological factors. Specifically, the highest values of EPA + DHA content were characteristic of fish belonging to the orders Clupeiformes or Salmoniformes, were pelagic fast swimmers, ate zooplankton and inhabited marine waters or migrated from fresh to marine waters (anadromous migrations). High EPA and DHA content in muscle tissues of the above species appeared to be a metabolic adaptation for fast continuous swimming. In contrast to common beliefs, our meta-analysis did not support the significant influence of higher trophic levels (piscivory) and cold environments (homeoviscous adaptation) on EPA and DHA content in fish. However, many causes of high and low levels of physiologically important fatty acids in certain fish species remained unexplained and require evaluation in future studies.

Аннотация: The photoprotein obelin was isolated and purified to homogeneity (as indicated by sodium dodecyl sulfate polyacrylamide gel electrophoresis) from hydroids of Obelia longissima by gel filtration on Sephadex G-75 fine, ion exchange chromatography on Polysil CA-300 (10 mum), hydrophobic chromatography on Phenyl-Sepharose CL-4B, gel filtration on Sephacryl S-200 superfine, ion exchange chromatography on a Mono Q column at pH 7.0, chromatofocusing on a Mono P column (pH gradient 6.0-4.0), and ion exchange chromatography on a Mono Q column at pH 5.5, 8.8, and 7.0. The molecular weight of the native protein was 30 kD, and that measured in the presence of SDS was 19.8 kD. The specific activity of obelin is 4.9.10(15) quanta/mg protein, pseudo-first-order constant of bioluminescence decay 4 sec-1, and quantum yield 0.16 The range of measurable Ca2+ concentrations is 10(-7) to 10(-5) M. The luminescence spectrum of obelin peaks at 469 nm, and the fluorescence emission maximum of the discharged protein is at 455 nm. The optimum pH for luminescence is between 9.0 and 10.5. The molecular ionization constants are pK1 6.8 and pK2 12.2, and the ionization constants for the active site are pK1 9.1 and pK2 10.2
