Аннотация: The bacterial composition of water and sediments in Lake Pyasino and adjacent rivers has been studied by high-throughput sequencing for the first time. No hydrocarbon-degrading bacteria are found in the river sections exposed to the diesel spillage in May 2020. The Daldykan and Ambarnaya rivers, located downstream of the spill site, are dominated by bacteria that oxidize sulfur and heavy metals. At the same time, these bacteria are not found in Lake Pyasino, which is dominated by small cyanobacteria. Thus, the composition of bacteria in the water and sediments of the Norilsk-Pyasino lake-river system corresponds to the technogenic impact to which it is exposed and also demonstrates the buffer properties of Lake Pyasino as water flows into the Kara Sea.

Заглавие : Comparison of rapid methods used to determine the concentration, size structure and species composition of algae
Место публикации : J. Sib. Fed. Univ. - Biol.: Siberian Federal University, 2021. - Vol. 14, Is. 1. - С. 5-27. - ISSN 19971389 (ISSN), DOI 10.17516/1997-1389-0338
Аннотация: Traditionally, the abundance, cell size distribution and species identification of algae are determined by microscopic counts. In recent years, various rapid methods have been developed for routine algal studies. However, each of these methods has its drawbacks. It is important for aquatic ecologists to understand the advantages, disadvantages, and limitations of these methods. We compared the sensitivity of three rapid methods (multichannel fluorimeter FluoroProbe, imaging flow cytometer FlowCam, and CASY particle counter) to changes in cell abundance of three algae species (Chlorella vulgaris Beyerinck, Arthrospira platensis Gomont, and Nostoc sp.). We also assessed the ability of rapid methods to estimate the cell abundance of different species in the mixed samples. All instruments showed high sensitivity to changes in the cell abundance of different algae species and a mixture of these species. Any one of these methods, once calibrated, can be reliably used to estimate the abundance of a single-species/laboratory culture of microalgae. At the same time, FlowCam, without preliminary calibration, recorded the cell abundance closest to microscopic counts. When analysing a mixture of three microalgae differing in their cell sizes and spectral characteristics, FluoroProbe showed the highest accuracy in assessing the proportions of species in the mixture and FlowCam - in assessing their abundance. To study mixtures of algae and/or natural phytoplankton communities, it is advisable to use jointly a flow cytometer and a multichannel fluorimeter. The images of algae saved by the flow cytometer, if necessary, can be used to identify them, with a certain accuracy, to the species. Information on cells size and spectral characteristics obtained by two methods will be detailed enough to perform such common tasks as studying trophic interactions between phyto- and zooplankton or creating warning systems to inform of unwanted blooms of phytoplankton and their individual groups (for example, cyanobacteria). © Siberian Federal University. Заглавие : Ecological role of cyprideis torosa and heterocypris salina (Crustacea, ostracoda) in saline rivers of the lake elton basin: Abundance, biomass, production, fatty acids
Место публикации : Zool. Stud.: Biodiversity Research Center, Academia Sinica, 2021. - Vol. 60. - С. 60-53. - ISSN 10215506 (ISSN), DOI 10.6620/ZS.2021.60-53
Аннотация: Saline rivers are highly productive ecosystems in arid regions. The meiobenthic community (bottom meiofauna) and its dominant representatives are one of the least studied components of these aquatic ecosystems. Ostracods Cyprideis torosa and Heterocypris salina are major consumers among the species of bottom meiofauna in saline rivers flowing into the hyperhaline Lake Elton (Volgograd Region, Russia). We estimated the abundance, biomass and production of C. torosa, the dominant species at the mouth of the polyhaline Chernavka River (average salinity is ~30 g l-1), and H. salina, the dominant species at the mouth of the mesohaline Bolshaya Samoroda River (~13 g l-1), in spring (May) and summer (August). Additionally, we studied the composition and content of fatty acids of the ostracods and their potential food sources (bottom sediments with bacterial-algal mats). We found that the abundance and biomass (wet weight with shells) of C. torosa in the Chernavka River and H. salina in the Bolshaya Samoroda River reached 3.5 ? 106 ind. m-2 and 117 g m-2, and 1.1 ? 105 ind. m-2 and 12 g m-2, respectively. The first species formed on average about 85% of the total abundance and 96% of the total biomass of the meiobenthos, and the second one, about 13% and 31%, respectively. The daily production of C. torosa and H. salina can reach 249 and 36 mg m-2 ash-free dry weight, respectively. The results indicate that these species may play an important role in the total flow of matter and energy in the studied habitats. Based on the fatty acid (FA) composition of the ostracods and their food sources, it was found that C. torosa mainly consumed diatoms, while H. salina preferred bacteria, cyanobacteria, and green algae. Differences between the species were greater than differences between the bottom sediments from the rivers. It may mean that the ostracods selectively consumed different food items that may be related to the different nutrient requirements of the species. Seasonal changes in the FA compositions of the ostracods were higher than in their food sources (bottom sediments), which also indicates selective feeding of the species. © 2021 Academia Sinica, Taiwan.

Аннотация: During the study of cyanobacteria and algae from littoral of the meromictic Lake Shira (Republic of Khakassia, Russia) forty-eight taxa were identified: Cyanobacteria - 7, Chlorophyta - 5, and Bacillariophyta - 36 species. Cyanobacteria were represented by cosmopolitan taxa Leptolyngbya voronichiniana, cf. Trichocoleus hospitus and widely distributed in the freshwater ecosystems species Phormidium cf. paulsenianum, Pseudophormidium cf. golenkinianum, P. pauciramosum, Leptolyngbya perforans, and L. subtilissima. Among green algae typical freshwater taxa Chloroidium saccharophilum, Desmodesmus abundans, Oocystis lacustris, Chlorella sp., Stigeoclonium sp. were detected. Among Bacillariophyta the most frequent species were Amphora ovalis, Cymbella affinis, Encyonema silesiacum, Eunotia fallax, Gomphonella olivacea, G. parvulum, Kobayasiella subtilissima, Navicula minima, N. radiosa, N. veneta, Nitzchia fonticola, N. palea, Grunowia tabellaria, Surirella undulata. Several rare species cf. Fragilaria tenera, Grunowia tabellaria and cf. Ulnaria capitata were found. In relation to mineralization indifferents was the largest group (73%). In relation to pH, a unique feature of the investigated lake was the predominance of alkaliphilic species of diatoms (56%), such as Amphora ovalis, Cymbella affinis, Gomphonema truncatum, Hantzschia amphioxys, Navicula minima, N. veneta, Nitzschia cf. linearis, N. palea, Planothidium lanceolatum and other. In the geographic structure cosmopolitan group included 25 species (69%), boreal group - 4 species (11%), arcto-apline group - 2 species (6%). To obtain more accurate information on the biodiversity of cyanobacteria and algae of Lake Shira further studies using molecular-genetic and electron microscopic methods are needed.

METABARCODING APPROACH
RAPID ASSESSMENT
Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.

Ключевые слова (''Своб.индексиров.''): essential polyunsaturated fatty acids--fish--food quality--salinity--temperature
Аннотация: Aquatic invertebrates are valuable dietary sources of essential polyunsaturated fatty acids, eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), for fish. Phylogeny, diet, and various ecological factors affect the fatty acid composition of aquatic invertebrates. We focused our study on the effect of ecological factors to a cosmopolitan species inhabiting lakes that differed in salinity, temperature, and presence/absence of predators (fish). To avoid the effect of phylogeny, which strongly influences the fatty acid composition of animals, we studied several populations of one cosmopolitan benthic species, Gammarus lacustris Sars. We found that differences in fatty acid percentages of G. lacustris were mainly affected by differences in their diets. Some populations preferred dinoflagellates, cryptophytes, green algae/cyanobacteria, and bacteria; other populations selected diatoms; and still other populations consumed zooplankton or allochthonous (terrestrial) organic matter. The salinity and presence/absence of fish affected the contents of EPA and DHA in G. lacustris. Populations from saline and fishless lakes had significantly higher contents of EPA and DHA. Thus, stocking of fishless lakes dominated by G. lacustris with fish could lead to a decrease in EPA and DHA contents in the gammarids. We propose that some saline and fishless lakes could be used as a source of gammarids for aquaculture fish feeding. © 2018, © 2018 International Society of Limnology (SIL).

Аннотация: The study of the composition of fatty acid markers of tadpoles of cohabiting amphibian species for the first time revealed differences in their diets: the moor frog Rana arvalis prefers bacteria not associated with plant detritus, whereas the diet of the common spadefoot Pelobates fuscus is based on cyanobacteria, green algae, diatoms, and possibly higher plants. Major differences in the fatty acid composition are determined by the difference in the percentage of eicosapentaenoic and myristic acids.

Аннотация: Meromictic lakes located in landlocked steppes of central Asia (similar to 2500 km inland) have unique geophysiochemical characteristics compared to other meromictic lakes. To characterize their bacteria and elucidate relationships between those bacteria and surrounding environments, water samples were collected from three saline meromictic lakes (Lakes Shira, Shunet and Oigon) in the border between Siberia and the West Mongolia, near the center of Asia. Based on in-depth tag pyrosequencing, bacterial communities were highly variable and dissimilar among lakes and between oxic and anoxic layers within individual lakes. Proteobacteria, Bacteroidetes, Cyanobacteria, Actinobacteria and Firmicutes were the most abundant phyla, whereas three genera of purple sulfur bacteria (a novel genus, Thiocapsa and Halochromatium) were predominant bacterial components in the anoxic layer of Lake Shira (similar to 20.6% of relative abundance), Lake Shunet (similar to 27.1%) and Lake Oigon (similar to 9.25%), respectively. However, few known green sulfur bacteria were detected. Notably, 3.94% of all sequencing reads were classified into 19 candidate divisions, which was especially high (23.12%) in the anoxic layer of Lake Shunet. Furthermore, several hydro-parameters (temperature, pH, dissolved oxygen, H2S and salinity) were associated (P0.05) with variations in dominant bacterial groups. In conclusion, based on highly variable bacterial composition in water layers or lakes, we inferred that the meromictic ecosystem was characterized by high diversity and heterogenous niches.

Аннотация: We studied biogeochemical characteristics, including organic carbon and nitrogen contents, fatty acid (FA) composition, stable isotope ratios, and primary production in conjunction with species composition of bacterioplankton, using next generation sequencing, in the Yenisei River along a distance similar to 1800km. Basing on FA composition of particulate organic matter (POM) and on other indicators of sources of POM, the river was subdivided into four sections. The upper section 1, situated in mountain region, was the net source of high-quality autochthonous organic matter, produced primarily by diatoms and partly consumed by specialized bacteria species. Section 2 in plain taiga was net sink of high quality allochthonous and autochthonous organic matter, produced by cyanobacteria and green algae. Section 3 was net sink of organic matter, primarily allochthonous, consumed by the specialized species of bacteria. The lowest section 4, situated in tundra, was primarily the conduit of recalcitrant terrestrial organic matter, but also the net source of autochthonous organic matter, produced by diatoms. Biogeochemical traits of sections of the Yenisei River evidently shaped dominant species composition of bacterioplankton of these sections. Regarding the biogeochemical and microbiological data, we concluded that the Yenisei River ecosystem complexly combines features of river mosaic, river continuum, and "neutral pipe."

Аннотация: Understanding the factors that regulate the abundance, size structure and community structure of cladocerans is an important goal of aquatic ecologists. While both top-down and bottom-up factors help to structure cladoceran communities, there may be interactions between these factors. We conducted a mesocosm study to determine how alien large-bodied Daphnia, zebra mussels and fish affected cladoceran community and size structure. We found that large-bodied Daphnia reduced algal resources and the fecundity of smaller bodied cladocerans. Fish removed the large-bodied Daphnia magna from the mesocosms and shifted the cladoceran community to a smaller body size. Fish also appeared to promote increases in cladoceran diversity through the coexistence of several smaller bodied taxa. In contrast, zebra mussels increased cyanobacteria and helped to promote the success of the alien Daphnia, but reduced the biomass of small-bodied cladocerans. Zebra mussels reduced the carbon (C):phosphorus (P) ratio of the phytoplankton in the mesocosms which may have favored the relatively P-limited Daphnia. Combined, our results highlight the complex interactions of multiple factors that help to regulate cladoceran community and size structure.

Ключевые слова (''Своб.индексиров.''): bacterial community--diversity--16s rrna gene--yenisei river
Аннотация: The biodiversity of bacterial communities along the Yenisei River at section c. 1800 km was studied using next-generation sequencing of 16S rRNA genes and common biodiversity indices. Overall, 3022 unique operational taxonomic units were identified. Actinobacteria and Proteobacteria were the dominant phyla at all sampling sites. The highest alpha-diversity values were found in the middle section of the studied river. The beta-diversity of bacterial assemblages in the river was related to the surrounding landscape (biome): three distinctly different bacterial assemblages occurred in sections of the river, situated in mountain taiga, plain taiga and in a region of permafrost, covered by forest-tundra and tundra. Tributaries arising from these different landscapes likely contributed substantially to the variations of Yenisei bacterial communities. In contrast to a prediction of the river continuum concept, the proportion of photoautotrophic Cyanobacteria in bacterial assemblages did not increase downstream, but peaked at the middle section.

Аннотация: We studied amino acid (AA) composition of epilithic biofilms and zoobenthos near the shore at a middle section of the Yenisei River (Siberia, Russia). We hypothesised that there was an imbalance between the composition and content of amino acids in the biofilm and its consumers, the zoobenthos, as well as between those in the zoobenthos and fish. Based on monthly sampling from 2007 to 2010, there was seasonal variation in AA profiles in the epilithic biofilms, probably caused by the succession of microalgal and cyanobacterial species. Overall, there was an imbalance in the percentage of the essential amino acids (lysine and histidine) between benthic animals and their food (the epilithic biofilm), which suggests that benthic animals may be limited by food quality. Moreover, the zoobenthos had a significantly higher content of AA, relative to carbon, than the biofilm. Based on sampling in 2012, there was an imbalance between the AA profiles of zoobenthos and that of their main consumer, the Siberian grayling (Thymallus arcticus), particularly in the percentages of two essential amino acids, lysine and leucine. In terms of overall content of essential amino acids, the nutritional value to fish of gammarids, which have recently invaded the river, was significantly lower than that of indigenous taxa, trichopteran and chironomid larvae. Аннотация: Recruitment of pelagic populations of cyanobacteria from littoral bottom sediments is assumed to be one of the key factors for the development of harmful blooms. Therefore, it is necessary to obtain more information on the possible factors influencing the spatial distribution of the resting stages in littoral bottom sediments. To test the hypothesis that akinete abundances in littoral bottom sediments are associated with silt and the presence of higher water plants, we sampled spatial and vertical distribution and seasonal dynamics of akinetes of cyanobacterium Anabaena flos-aquae in a small Siberian reservoir. We found that akinetes accumulate in silt at stations located in macrophyte beds. Two experiments were carried out to test if there is relationship between size of inoculum and size of the pelagic population of A. flos-aquae. The proportion of A. flos-aquae in total phytoplankton biomass depended on the initial abundance of akinetes in bottom sediments. The model based on laboratory experiments gave good qualitative description of the field data. These findings can be useful to predict the location of the main akinete accumulation zones in a water body and plan the removal of sediment to prevent the cyanobacterial blooms. В© CSIRO 2011.

Аннотация: During two vegetation seasons (2004-2005), we compared feeding spectra of Arctodiaptomus salinus (Calanoida, Copepoda) populations inhabiting two neighboring salt lakes, Shira and Shunet, Khakasia, Russia, using fatty acid (FA) trophic markers. Sestonic FA composition in two lakes moderately differed, whereas levels of diatom FA markers were higher in Lake Shunet and of Cyanobacteria and green algae markers in Lake Shira. In general, markers in storage lipids-triacylglycerols (TAG) of A. salinus-reflected the differences in sestonic composition of the two lakes. Nevertheless, TAG fraction was also enriched by FA trophic markers of the minor components of seston, which were selectively ingested by the animals. In Lake Shira, A. salinus had significantly higher concentrations of bacterial FA markers in TAG. In Lake Shunet, TAG of A. salinus contained significantly higher relative amounts of 18:4?3, 18:5?3 and C22 polyunsaturated fatty acids (PUFA), which indicated marked contribution of cryptophytes or (and) flagellates into the diet. Laboratory experiments showed feeding on Cryptomonas and sulfur purple bacteria in Lake Shunet and ciliates and colonial picoplankton in both lakes, and generally confirmed the differences in FA trophic markers in A. salinus between the lakes. The two populations of A. salinus markedly differed in levels of essential long-chain PUFA, eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, although the levels of these FA in seston were quite similar between the two lakes. The higher levels of the essential ?3 PUFA in A. salinus in Lake Shunet may be an adaptive response of the animals to a vertical stratification of physico-chemical conditions and significantly higher salinity levels at the boundary of adjacent bottom layer in this lake. Ключевые слова (''Своб.индексиров.''): fatty acids--riverine microalgae--taxa composition--temperature adaptations--algae--bacillariophyta--chlorophyta--cyanobacteria
Аннотация: We studied fatty acid (FA) composition of littoral microalgae in the fast-flowing oligotrophic river, the Yenisei, Siberia, monthly for 3 years. Seasonal dynamics of species composition had similar patterns in all the studied years. In springs, a pronounced dominance of filamentous green algae occurred, in summer and autumn diatoms were abundant, and in late autumn and winter epilithic biofilms consisted primarily of cyanobacteria and detritus. In general, FA composition of the algal periphytic community was dominated by 16:0, 16:1?7, 20:5?3, 14:0, and 18:3?3 throughout the studied period. Several groups of FAs, which had peculiar seasonal dynamics, were differentiated by statistical analysis based on a method of correlation graphs. The seasonal changes in FA composition could be partly explained by the seasonal succession of species composition of the community. Besides, we found that populations of both diatom and green algae grown in summer at a higher water temperature were lower in polyunsaturated fatty acids than those in spring, at a lower temperature. Hence, we suppose that the regular seasonal dynamics of FA composition of the studied littoral microalgae was driven both by changes in species composition and by temperature adaptations of the algal populations. The highest content of essential polyunsaturated FAs, eicosapentaenoic and docosahexaenoic acids, in the spring "psychrophilic" populations of diatoms could make them of the higher nutritive value for zoobenthic primary consumers.
