Аннотация: COVID-19 has specific characteristics that distinguish this disease from many other infec-tions. We suggest that the pathogenesis of severe forms of COVID-19 can be associated with acidosis. This review article discusses several mechanisms potentially linking the damaging effects of COVID-19 with acidosis and shows the existence of a vicious cycle between the development of hypoxia and acidosis in COVID-19 patients. At the early stages of the disease, inflammation, difficulty in gas exchange in the lungs and thrombosis collectively contribute to the onset of acidosis. In accordance with the Verigo-Bohr effect, a decrease in blood pH leads to a decrease in oxygen saturation, which contributes to the exacerbation of acidosis and results in a deterioration of the patient’s condition. A decrease in pH can also cause conformational changes in the S-protein of the virus and thus lead to a decrease in the affinity and avidity of protective antibodies. Hypoxia and acidosis lead to dysregu-lation of the immune system and multidirectional pro-and anti-inflammatory reactions, resulting in the development of a “cytokine storm”. In this review, we highlight the potential importance of supporting normal blood pH as an approach to COVID-19 therapy. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: This paper considers an approach to the description of the age-specific and time-dependent dynamics of cancer incidence. This approach is based on a model that describes the age-specific dynamics of cancer as a phase transition of the second order and is widely used in the study of physical systems. The proposed model of cancer development as the second-order phase transitions is quite consistent with the data of medical statistics, is well described using only two free parameters, easily verified by the statistical data, and is interpreted well. The applicability of the model of second-order phase transitions to the description of the processes in the nonphysical systems is likely to be determined by the versatile nature of the processes that occur during the phase transitions. © 2015, Pleiades Publishing, Inc.

Аннотация: In cold smoked species of genus Coregonus, identified by molecular genetic analysis, contents of fatty acids and heavy metals and arsenic were measured. The highest content of sum of long-chain polyunsaturated fatty acids of omega-3 family (LC-PUFA), namely eicosapentaenoic (EPA) and docosahexaenoic (DHA) fatty acids, 6.53 ± 0.78 mg g?1 wet weight, was characteristic of tugun Coregonus tugun. This is the first quantitative estimation (mg LC-PUFA per g of product) of the nutritive value of smoked fish. Thus, to obtain a daily personal doze of EPA + DHA of 1 g, recommended for prevention of cardiovascular diseases, one needs to consume 153 g of the smoked tugun. Metals contents did not exceed standards for fish meat except Pb in least cisco Coregonus sardinella. Accordingly, values of hazard quotients, which estimate benefit-risk ratio of fish intake, indicate that most of the smoked fish species are safe product for human nutrition, except least cisco regarding Pb content. © 2020 Elsevier Inc.

Аннотация: Background: Endosymbionts are microorganisms present in all plant species, and constitute the subject of interest among the scientific community. These symbionts have gained considerable attention in recent years, owing to their emerging biological roles. Global challenges, such as antimicrobial resistance, treatment of infectious diseases such as HIV and tuberculosis, cancer, and many genetic disorders, exist. Endosymbionts can help address these challenges by secreting valueadded bioactive compounds with various activities. Objective: Herein, we describe the importance of plants inhabiting Siberian niches. These plants are considered to be among the least studied organisms in the plant kingdom worldwide. Barcoding these plants can be of interest for exploring bioactive endosymbionts possessing myriad biological properties. Methods: A systematic survey of relevant scientific reports was conducted using the PubMed search engine. The reports were analyzed, and compiled to draft this review. Results: The literature survey on Siberian plants regarding endosymbionts included a few reports, since extremely few exploratory studies have been conducted on the plants in these regions. Studies on the endosymbionts of these plants are highly valuable, as they report potent endosymbionts possessing numerous biological properties. Based on these considerations, this review aims to create awareness among the global scientific community working on related areas. Conclusion: This review could provide the basis for barcoding novel endosymbionts of Siberian plants and their ecological importance, which can be exploited in various sectors. The main purpose of this review is to create awareness of Siberian plants, which are among the least studied organisms in the plant kingdom, with respect to endosymbionts, among the scientific community. © 2018 Higher Education Press and Springer-Verlag GmbH Germany, part of Springer Nature

Аннотация: Changes in the intracellular concentration of free ionized calcium ([Ca2+]i) control a host of cellular processes as varied as vision, muscle contraction, neuronal signal transmission, proliferation, apoptosis etc. The disturbance in Ca2+-signaling causes many severe diseases. To understand the mechanisms underlying the control by calcium and how disorder of this regulation relates to pathological conditions, it is necessary to measure [Ca2+]i. The Ca2+-regulated photoproteins which are responsible for bioluminescence of marine coelenterates have been successfully used for this purpose over the years. Here we report the results on comparative characterization of bioluminescence properties of aequorin from Aequorea Victoria, obelin from Obelia longissima, and clytin from Clytia gregaria charged by native coelenterazine and coelenterazine analogues f, i, and hcp. The comparison of specific bioluminescence activity, stability, emission spectra, stopped-flow kinetics, sensitivity to calcium, and effect of physiological concentrations of Mg2+ establishes obelin-hcp as an excellent semisynthetic photoprotein to keep track of fast intracellular Ca2+ transients. The rate of rise of its light signal on a sudden change of [Ca2+] is almost 3- and 11-fold higher than those of obelin and aequorin with native coelenterazine, respectively, and 20 times higher than that of the corresponding aequorin-hcp. In addition, obelin-hcp preserves a high specific bioluminescence activity and displays higher Ca2+-sensitivity as compared to obelin charged by native coelenterazine and sensitivity to Ca2+ comparable with those of aequorin-f and aequorin-hcp. © The Royal Society of Chemistry and Owner Societies 2015.

Аннотация: The authors investigate seasonal dynamics of number of alive and dead zooplankton individuals, their vertical distribution and the rates of sedimetation using special staining. Seasonal dynamics of natural mortality (ageing, parasites, diseases, toxic substances) was calculated. The mortality was maximal in the middle of a summer (the period of zooplankton depression) and minimal - at the beginning and at the end of summer. The method was compared with traditional method of mortality calculation that includes death from predators. It was shown that zooplankton depression in the middle df summer was caused not by press of predators but by other reasons.

Аннотация: The present review critically discusses the latest developments in the field of smart diagnostic systems for cancer biomarkers. A wide coverage of recent biosensing approaches involving aptamers, enzymes, DNA probes, fluorescent probes, interacting proteins and antibodies in vicinity to transducers such as electrochemical, optical and piezoelectric is presented. Recent advanced developments in biosensing approaches for cancer biomarker owes much credit to functionalized nanomaterials due to their unique opto-electronic properties and enhanced surface to volume ratio. Biosensing methods for a plenty of cancer biomarkers has been summarized emphasizing the key principles involved. © 2016 Elsevier B.V.

Аннотация: SARS­CoV­2 virus causes disease that has several distinctive characteristics compared to diseases caused by other viruses. We have put forward a hypothesis that relates COVID­19 pathogenesis with acidosis, which frequently characterizes severe cases of this disease. It has been shown that hypoxia and acidosis affect the progression of severe COVID­19 at various physiological levels such as organs, tissues, and cells. The physiological effects of acidosis and hypoxia range from the level of compensatory capabilities of the whole organism to the functioning of a single hemoglobin molecule. In our work, we consider several mechanisms that link the damaging factors of COVID­19 with acidosis. These mechanisms reveal step­by­step processes with a pronounced positive feedback. In accordance with the well­ known Bohr effect, a decrease in blood pH leads to a drop in blood oxygen saturation. At the same time, this drop in saturation contributes to the further development of acidosis. This indicates a depletion of the body's compensatory capabilities to regulate acidosis and leads to deterioration of the patient's condition. In addition, a decrease in pH can cause conformational changes in the viral S­protein, followed by changes in ability of some antibodies to recognize the virus. This might lead to the decrease in antibodies affinity and avidity, negatively affecting virus clearance. Low levels of pH and hypoxia in blood and tissues can induce a pro­inflammatory innate response even in the absence of antigen stimulation. Therefore, hypoxia and acidosis can lead to a restructuring of the immune system and multidirectional pro­ and anti­inflammatory responses, which often, instead of recovery, lead to the disease aggravation.

Аннотация: There is a poor relationship between nutrient intake and existing nutritional biomarkers due to variety of factors affecting their sensitivity and specificity. To explore the impact of nutrients at molecular level and devising a sensitive biomarker, proteomics is a central technology with sirtuins as one of the most promising nutritional biomarker. Sirtuins (seven mammalian sirtuins reported so far) have been reported to perform protein deacetylases and ADP-ribosyltransferases activity. It is distributed in different cellular compartments thereby controlling several metabolic processes. Sirtuins are oxidized nicotinamide adenine dinucleotide dependent, which implicates a direct effect of the metabolic state of the cell on its activity. Calorie restriction upregulates the mammalian sirtuin protein levels in variety of tissues and organs where it acts upon both histone and nonhistone substrates. Sirtuin senses nutrient availability and impacts gluconeogenesis, glycolysis, and insulin sensitivity. It deacetylates and inhibits the nuclear receptor that activates fat synthesis and adipogenesis in the body, leading to fat loss and bringing favorable cellular and health changes. Sirtuins mediates intracellular response that promotes cell survival, DNA damage repair thereby increasing the cell longitivity. The activation of sirtuins brings a wide spectrum of other health benefits and its activity levels are indicative of nutritional status as well as disease progression in cancer, inflammation, obesity, cardiovascular diseases, and viral infections. There are several foods that activate sirtuin activity and offer significant health benefits by their consumption.

Аннотация: The study includes anthropometry of 172 young male, obtained data on the length and body mass, measured the transverse diameters of the shoulders and pelvis, various body types was identified by the J.M. Tanner sexual dimorphism index (andromorphic, mesomorphic, gynecomorphic). The chemiluminescent and bioluminescent study of saliva and blood was conducted in the examined young male. We studied the indicators of the antioxidant defense system under the influence of stress. The antioxidant status of saliva was determined using the H2 O2-luminol-dependent chemiluminescence method. Data on the activity of NAD (P)-dependent dehydrogenases in blood lymphocytes was obtained from a bioluminescent method of research. Young male of andromorphic body type had large overall and transverse body sizes. Indicators of antioxidant protection of saliva and blood in men of adolescence, the body type of the sexual dimorphism index J.M. Tanner was different. The persons of the andromorphic body type differed in terms of chemiluminescence in comparison with the young male of gynecomorphic body type. The results of bioluminescent blood tests suggest a violation of the catabolic and anabolic processes of carbohydrate and fat metabolism in young men of mesomorphic and gynecomorphic body types. Indicators of the system of antioxidant protection of saliva and blood reflect the sexual characteristics of the body of young male and can be used as additional criteria for diagnosing sex inversion and assessing the risk of developing socially attributed diseases. © 2020, Klinichescheskaya Laboratornaya Diagnostika. Аннотация: The purpose of this study was to develop micro and nano sized drug carriers from poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), and study the cell and skin penetration of these particles. PHBV micro/nanospheres were prepared by o/w emulsion method and were stained with a fluorescent dye, Nile Red. The particles were fractionated by centrifugation to produce different sized populations. Topography was studied by SEM and average particle size and its distribution were determined with particle sizer. Cell viability assay (MTT) was carried out using L929 fibroblastic cell line, and particle penetration into the cells were studied. Transdermal permeation of PHBV micro/nanospheres and tissue reaction were studied using a BALB/c mouse model. Skin response was evaluated histologically and amount of PHBV in skin was determined by gas chromatography-mass spectrometry. The average diameters of the PHBV micro/nanosphere batches were found to be 1.9 mu m, 426 and 166 nm. Polydispersity indices showed that the size distribution of micro sized particles was broader than the smaller ones. In vitro studies showed that the cells had a normal growth trend. MTT showed no signs of particle toxicity. The 426 and 166 nm sized PHBV spheres were seen to penetrate the cell membrane. The histological sections revealed no adverse effects. In view of this data nano and micro sized PHBV particles appeared to have potential to serve as topical and transdermal drug delivery carriers for use on aged or damaged skin or in cases of skin diseases such as psoriasis, and may even be used in gene transfer to cells.

Аннотация: Objective: The main objective of this work was to formulate a nanodispersion containing grape seed extract and analyzed its release profile, antioxidant potential of the prepared formulations. Methods: The grape seed extract (GSE) containing proanthocyanidins (PC’s) has been dispersed in polymer matrix soluplus (SOLU) by the freeze-drying method. The morphological analysis was carried out using atomic force microscopy (AFM), scanning electron microscopy (SEM) and Transmission electron microscopy (TEM). The in-vitro release of the nanodispersion formulations was evaluated by simulated intestinal fluid (SIF). The antioxidant activity of GSE and the formulation were evaluated by employing various in-vitro assays such as 2, 2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), 2, 2-diphenyl-1- picrylhydrazyl (DPPH), Ferric reducing antioxidant power (FRAP) and peroxidation inhibiting activity. Results: The formulation FIII (1:5) resulted in a stable formulation with a higher loading efficiency of 95.36%, a particle size of 69.90 nm, a polydispersity index of 0.154 and a zeta potential value of ?82.10 mV. The antioxidant efficiency of GSE-SOLU evaluated by DPPH was found to be 96.7%. The ABTS and FRAP model exhibited a dose-dependent scavenging activity. Linoleic model of FIII formulation and GSE exhibited a 66.14 and 86.58% inhibition respectively at 200 µg/l. Conclusions: The main reason for excellent scavenging activity of the formulations can be attributed to the presence of monomeric, dimeric, oligomeric procyanidins and the phenolic group. The present work denotes that GSE constitutes a good source of PC’s and will be useful in the prevention and treatment of free radical related diseases. © 2020, © 2020 Informa UK Limited, trading as Taylor & Francis Group.

Аннотация: The possibility of using a bioluminescence method for measuring endotoxicosis during therapy has been investigated. Results of our experiments shown that a bioluminescence method can be used as a reliable criterion to monitor the course of disease for patients with bronchitis, ulcerous disease or chronic colecystitis. Assays using both soluble and immobilized reagents are possible. This method does not reveal differences in patients with sepsis, hepatocirrhosis or oncology. The methods are highly sensitive, rapid and simple and allow quatitative determination of the degree of seriousness of illness and estimation of the severity of a patient's condition. Аннотация: Solid-phase bioluminescent immunoassay of thyroid hormones, human thyrotropin (hTSH) and two forms of thyroxine (T4), whose determinations are vitally important for diagnostics of thyroid diseases and the efficiency of treatment, is described. The recombinant obelin, a Ca2+-regulated photoprotein originally derived from the luminous marine hydroid Obelia longissima, is employed as a bioluminescent label. To produce obelin conjugates with anti-hTSH, anti-T4 immunoglobulins (IgG), and T4, additional SH groups are introduced into the obelin molecule using Traut's reagent (2-iminothiolane) and then obelin possessing extra SH groups is conjugated with succinimidyl 4-(N-maleimidomethyl)-cyclohexane-1-carboxylate-activated IgGs or T4. The total yield of obelin conjugates determined by luminescent activity is 60-65% after all chemical and purification procedures. The obtained conjugates are stable to lyophilization and in solution for at least 9 months at 4 degreesC, with loss of activity not exceeding 10%. The application of obelin conjugates for determination of the hTSH, total T4, and free T4 in standard, control, and patient sera displays high sensitivity and reproducibility of results. The results of bioluminescent immunoassays are closely comparable to those obtained by the radioimmunoassay method (R = 0.95-0.99). (C) 2003 Elsevier Inc. All rights reserved.

Аннотация: The paper dwells upon the use of autochthonous microbial cinosis as a source of antagonistic strains for bioprotection of wheat against Fusarium in biotechnical life support systems (BT LSS). Six bacterial strains antagonistic to 9 phytopathogenic fungi Fusarium responsible for seedling blight of wheat in BT LSS were isolated from hydroponic solutions and artificial soils and subject to the genetic typing. Five strains represent bacteria g. Bacillus, one strain - Chryseobacterium. The 16S rRNA sequencing showed 94 to 100 % genetic affinity with typical cultures. All spore-forming strains-antagonists were capable of germinating and going through the whole development cycle In the presence of swelling wheat seeds. The isolates did not demonstrate antagonism to each other and can grow in a mixed culture. Spectra and levels of the antiobiotic activity of the antagonists, along with the Fusarium sensitivity to their antibiotic action, differed statistically (p < 0.001). None of the antagonists was capable of suppressing the entire Fusarium spectrum detected in BT LSS individually. At the same time, treatment of seeds with a mixture of antagonists suppressed totally fungal development in wheat artificially inoculated by Fusarium conidia from a BT LSS roll culture, and had a statistical stimulating effect (p = 0.01) on seedlings. © 2020 Slovo Ltd.
