Аннотация: All of man's former space flights were not real ventures into space in the biological sense, as his life was supported with unregenerated earth supplies. The coming stage of space exploration requires man's long existence in the cosmos and on the other planets. This stage of man's activity outside the earth become possible only by creating small man-made ecosystems, permitting the support of his metabolism by the recycling of substances of the terrestrial biosphere. Creation of such systems is a new scientific and technical task. Man-made ecosystems are a new product of man's activity, which have no complete analogy, either in nature, or in technology. Stochastic mechanisms, which stabilize biogeocenosis, cannot be effective in small ecosystems. A technique of parametric control over biosynthesis made it possible to calculate, and put to practice, an ecosystem for man with a cyclic regeneration of the atmosphere, water and, partially, food. The specific bio-technological properties of small man-made ecosystems are being analysed. The possibility of their application for man's excursions into space and for the settlement of other planets is being considered.

Аннотация: We describe the experimental system having maximal possible closure of material recycling in an ecosystem, including people and plants, which was carried out in a hermetically sealed experimental complex "BIOS-3", 315 m2 in volume. The system included 2 experimentators and 3 phytotrons with plants (total sowing area of 63 m2). Plants were grown with round-the-clock lamp irradiation with 130 Wm-2 PAR intensity. The plants production was food for people. Water exchange of ecosystem, as well as gas exchange, was fully closed excluding liquids and gas samples taken for chemical analysis outside the system. The total closure of material turnover constituted 91%. Health state of the crew was estimated before, during and after the experiment. A 5-months period did not affect their health. The experiments carried out prove that the closed ecosystem of "man-plants" is a prototype of a life-support system for long-term space expeditions. В© 1989.

Аннотация: For analytical purposes bioluminescence can be used in three main ways: 1. luminescence measurement of bioluminescent system components isolated in vitro; 2. determination of luminous organisms' reaction to the in vivo test-action; 3. measurement of bioluminescence in marine ecological systems. The majority of the reports of this Symposium are dealing with the first two topics. The aim of our presentation is to draw attention to the third one. The possibilities of bioluminescent analysis are wider than its traditional scheme of applications in the laboratory, when the emitting system is withdrawn from a native source and is placed in a cuvette of the light measuring device. The reverse scheme is also possible, i.e. the device can be introduced into light emitting system such as a marine biocenosis--the community of the sea inhabitants--where we obtain a highly sensitive and rapid means of gaining the information on the vital activity of marine ecosystems, i.e. their spatial structure, rhythms, man's influence upon them, etc. The present communication will consider the possibilities of this form of bioluminescent analysis.

Аннотация: The notion of ecological biophysics as a scientific discipline investigating physical processes and phenomena caused by functioning of the living super-organism systems is formulated. The three main constituents of the ecological biophysics are defined: elaboration of the monitoring methods of the basis of sensing of the biophysical fields (of bioluminescence and fluorescence), mathematical and physical modeling and investigation of the part of living organisms in the hydrophysical processes of the ecosystem scale. Examples of realization of the ecological biophysical approach in the process of investigations of the World Ocean, Lake Baikal, the reservoirs of Dnieper and Yenisei revers are given.

Аннотация: A concept is formulated on ecological and biophysics as a scientific discipline which studies physical processes and phenomena resulting from the functioning of live supra-organism systems. Three main directions of ecological biophysics are defined: creation of methods of monitoring ecosystems on the basis of probing biophysical fields (fields of bioluminescence and fluorescence); mathematical and physical modelling; and study of the role of live organisms in hydrophysical processes on the ecosystem scale. Examples are given of the realization of the ecologo-biophysical approach in research into the ecosystems of the world ocean, Lake Baikal and the Dnieper and Yenisei reservoirs. В© 1994.

Аннотация: Using experimental microecosystems the kinetics of phenol disappearance in river and reservoir water were investigated. In river water the disappearance kinetics could be described by A first-order equation, the same kinetics took place in reservoir water before and after the period of ''bloom'' of blue-green algae. During the ''bloom'', the phenol seemed to be mineralized by bacteria which grew at the expense of another compound, and the model of best fit was the model of exponential growth and low concentration of the test substrate. In the river, two sections differed according to the difference between the mean values of the specific disappearance rates. In the reservoir these rates were lower than those in the river. In general the specific disappearance rate values did not correlate with the values of initial bacterioplankton density, the concentrations of inorganic nutrients nor the chemical oxygen demand. Conclusions about the integral influence of ecosystems on the disappearance rates and dependence of self-purification kinetics on the type of aquatic ecosystem were made.

Аннотация: Artificial, closed ecosystems are potentially useful tools with which to explore ecosystem interactions, allowing investigations of the biology, physiology and biophysics of complex communities. They can also be used to evaluate the long-term effects of environmental constituents that are usually present at very low concentrations. The introduction of humans into such systems extends their usefulness even further, allowing the study of the influence of the environment on people, and the effects of people on the environment. Because of the great potential of such systems in adding to human knowledge, and their considerable expense, it is appropriate that such activities be international studies. The Institute of Biophysics of the Russian Academy of Sciences (Siberian Branch) has established, The International Research Center for Closed Ecosystems (Biospherics). Аннотация: The experience of human existence in the last few decades shows clearly that developed civilizations come into greater and greater antagonism with nature. Modern technology cannot on its own coexist easily with the biosphere without destroying key biological components. The necessity of comprehending the laws of development of the biosphere as a single whole is becoming more and more obvious and urgent. Because it is so precious and fragile, the biosphere cannot be subjected to any direct study which may harm it. Therefore this science depends on the study of analogs and small models such as artificial ecological systems with differing degrees of complexity and closure. On such model ecosystems we can (and must) study both the particular laws of development of individual elements and components of the ecosystems, and the general principles of turnover of the entire biospheric system. As this new science is being formed, it is necessary to develop the scientific basis of harmonizing the relationship of humanity and nature, to open the path to the next phase, termed the noosphere by Vernadsky. The principal objects of study are closed ecological systems, from simple microsystems to more sophisticated human life-support systems under extreme conditions on the Earth and in space. Biospherics by its very nature knows no political boundaries, and is by necessity an international effort. As such, it is deserving of support both intellectually and economically from all peoples.

Аннотация: To create a life support system based on biological and physical-chemical processes is the optimum solution providing full-valued condidtions for existence and efficient work of people at a lunar base. Long-standing experinece in experimental research or closed ecosystems and their components allows us to suggest a realistic functional structure of the lunar base and to estimate qualitatively its parameters. The original restrictions are as follows: 1) the basic source of energy to support the biological processes has to be the solar radiation; 2) the initial amount of basic biological elelments forming the turnover of substances (C, O, H, P, K, N) has to be delivered from Earth; 3). Moon materials are not to be used in the biological turnover inside the base; 4) the base is to supply the crew fully with atmosphere and water, and with 90% (A scenario) or 40% (B scenario) of food. Experimental data about the plant productivity under the "Moon" rhythm of light and darkness allow us to suggest that the A scenario requires per one human: plant area - 40 m2 irradiated during the lunar day by 250-300 W/m2 PAR producing 1250 g of dry biomass a terrestrial day; a heterotrophic component of "biological incineration" of inedible plant biomass (800 g/day) including the aquaculture of fish to produce animal products and contaminating the environment less than birds and mammals, and the culture of edible mushrooms; a component of physical-chemical correction for the LSS envi ronment including the subsystems of: deep oxidation of organic impurities in the atmosphere and of water, organic wastes of human activity and that biological components (420 g/day) Co2 concentration in "Moon" nights, damping O2 in "Moon" days, etc. The stock of presotred or delivered from Earth substances (food additions, seeds, etc.) to be involved in biological turnover is to be about 50 kg/year per man. Increase of the mass of prestored substances per man up to 220 kg/year would reduce twice the plant area and consumed amount of radiant energy to exclude the components of "biological incineration" and physical-chemical destruction of organic wastes. Аннотация: The chromatographic conditions presented allowed the separation of the nitrophenacyl derivatives of standards of eleven free fatty acids (FFA) using a portable high-performance chromatograph, suitable for use aboard a research vessel. A statistically significant linear correlation between UV absorbance and amount of the analytes injected was obtained. The method was tested on FFA from algae cultural media. The method can be used for the ecological monitoring of natural waters.

Аннотация: The behavior of Escherichia coil Z905, carrying a recombinant plasmid pPHL7 with genes determining ampicillin resistance and bacterial luminescence, and the efficiency of expression of cloned genes were studied after introduction of the strain into model aqueous ecosystems with different trophic chain lengths. The E. coli Z905 variants isolated from ecosystems after different periods of time were found to vary in their resistance to ampicillin (from 50 to 0.05 mu g/ml) and in the intensity of bioluminescence. An increase in the concentration of the selective factor (ampicillin) or in the extent of the aqueous microcosm blooming restored the expression of the recombinant plasmid genes in some clones.

Аннотация: Dynamics of catalase activity has been shown to reflect the growth curve of microorganisms in batch cultivation (celluloselythic bacteria Bacillus acidocaldarius and bacteria of the associated microflora Chlorella vulgaris). Gas and substrate closure of the three component ecosystems with spatially separated components "producer-consumer-reducer" (Chl. vulgaris-Paramecium caudatum-B. acidocaldarius, two bacterial strains isolated from the associated microflora Chl. vulgaris) demonstrated that the functioning of the reducer component can be estimated by the catalase activity of microorganisms of this component. Аннотация: The dynamics of biomass and species composition of microalgae and of composition of extracellular free fatty acids was studied for 98 days in laboratory experimental ecosystems inoculated with water from a forest pond. The microecosystems were kept at two temperature levels: 14-17 and 22-25В°C. During the experiment, a community of planktonic microalgae with the predominance of diatoms was succeeded by a community consisting mostly of green, filamentous algae. The succession of species composition of the algal community was accompanied by changes in the composition of extracellular free fatty acids. Variations of temperature did not markedly affect either the species composition of the microalgal community or the ratio of unsaturated to saturated free fatty acids. Аннотация: Using experimental microecosystems the kinetics of phenol disappearance in small forest pond waters (Siberia, Russia) in the summer of 1995-96 were investigated. Despite of high variability of components of the ecosystem (plankton biomass and species composition) and two pronounced 'blooms' of green algae Volvox aureus the same kinetics of the disappearance took place over the investigated period. Half-lives of the pollutant depended on water temperature only. A comparison of the self-purification of the pond with that of the Krasnoyarsk reservoir, 'blooming' with blue-greens was carried out. Half-lives in the pond were significantly lower than that in the reservoir. During the periods of 'blooms' of the green algae in the pond the concentrations of inorganic nutrients were comparatively high and the phenol-degrading bacteria likely were not limited by these nutrients, in contrast to the periods of 'bloom' of the blue-green algae in the reservoir.

Аннотация: We studied preservation of recombinant Escherichia coli strain Z905 (AprLux+) in liquid microecosystems (LME) after the introduction. E. coli cells were shown to remain viable and preserve the ability to express the cloned lux genes for a long time (more than a year) in LME. The majority of the clones have reduced efficiency of the expression due to either changed regulation of the lux operon or decreased number of copies of the plasmid. These mechanisms could be realized either independently or simultaneously depending on LME conditions. We have exposed the major factors affecting the metabolic activity of the E. coli strain Z905 (AprLux+) introduced into model ecosystems and the level of expression of the cloned genes.
