Аннотация: Cross sections and macerates were used to study leaf mesostructure in cucumber, sunflower, and tomato plants formed under controlled environmental conditions at an irradiance of 100 W.m-2 in the red and blue regions of the spectrum. White light served as the control. Three types of response to red light were isolated: the first type is a response not associated with harmful action of the light factor (tomato); the second a response associated with reversible changes of mesostructure (sunflower); and the third a response associated with irreversible changes of leaf mesostructure (cucumber). It is concluded that destructive changes during prolonged action of red radiation on the leaf are mainly associated with degradation of palisade parenchyma, whereas spongy parenchyma suffers less and takes upon itself the main functional role in certain species (sunflower). Species specificity was not clarified in the response of leaf mesostructure to radiation in the blue region of the spectrum.

Аннотация: The inhibition of bacterial luminescence has been used in testing industrial enterprises sewage. The toxicity of the sewage is less than the total toxicity of separate components due to neutralization of quinone products of polyphenol oxidation in the reactions with the other phenol components of sewage. Toxicity increase is due to their influence on the cell membrane. Studies of cell ultrastructure confirm this fact. The studied mechanism of the complex effect allowed a more accurate forecast of the ecological situation during the discharge of phenol compounds and metals. It also showed the necessity of taking into account the complex effect of sewage components on contaminant discharge into water reservoirs.

Аннотация: Most scenarios for a manned lunar base include a combination of physical-chemical and bioregenerative life support systems. Especially on the lunar surface, however, there is a series of special environmental factors which seriously affect the organisms suitable for food production and biological regeneration of the habitat atmosphere and water. So, e.g. the lunar day/night period creates difficult problems for higher plant culture. The paper presents the current scientific approaches to bioregenerative life support systems of a lunar base and discusses critically the possibilities of their realization. Moreover, a scientific strategy is developed with the biologist's point of view to implement in a stepwise manner bioregenerative life support modules into a lunar base covering the possibilities of the untilization of chemolytotrophic bacteria, microalgae and higher plants as well as those of animal breeding and protein production in intensive aquaculture systems. Аннотация: Artificial, closed ecosystems are potentially useful tools with which to explore ecosystem interactions, allowing investigations of the biology, physiology and biophysics of complex communities. They can also be used to evaluate the long-term effects of environmental constituents that are usually present at very low concentrations. The introduction of humans into such systems extends their usefulness even further, allowing the study of the influence of the environment on people, and the effects of people on the environment. Because of the great potential of such systems in adding to human knowledge, and their considerable expense, it is appropriate that such activities be international studies. The Institute of Biophysics of the Russian Academy of Sciences (Siberian Branch) has established, The International Research Center for Closed Ecosystems (Biospherics). Аннотация: In the paper an attempt is made to combine three important criteria of LSS comparison: minimum mass, maximum safety and maximum quality of life. Well-known types of BLSS were considered: with higher plant, higher plants and mushrooms, microalgae, and hydrogen-oxidizing bacteria. These BLSSs were compared in terms of "integrated" mass for the case of a vegetarian diet and a "normal" one (with animal proteins and fats). It was shown that the BLSS with higher plants and incineration of wastes becomes the best when the exploitation period is more than 1 yr. The dependence of higher plants' LSS structure on operation time was found. Comparison of BLSSs in terms of integral reliability (this criterion includes mass and quality of life criteria) for a lunar base scenario showed that BLSSs with higher plants are advantageous in reliability and comfort. This comparison was made for achieved level of technology of closing and for perspective one. В© 1997 Elsevier Science Ltd.

Dependence of microorganism catalase activity has been found to depend on residual substrate concentration in proportion to the biomass of microorganisms. Аннотация: Available methods for mineralizing wastes of human activity and inedible biomass of plants used in this country and abroad are divided into two types: dry mineralization at high temperatures up to 1270 degrees K with subsequent partial dissolution of the ash and the other wet oxidation by acids. In this case mineralization is performed at the temperature of 470-460 degrees K and the pressure of 220-270 atmospheres in pure oxygen with the output of mineral solution and dissoluble sediments in the form of scale. The drawback of the former method is formation of dioxins, CO, SO2, NO2 and other toxic compounds. The latter method is too sophisticated and presently is confined to bench testing. The proposed method to mineralize the wastes is in mid-position between the thermal and physical chemical methods. At the temperature of 80-90 degrees C the mixture was exposed to controlled electromagnetic field at normal atmospheric pressure. The method merits simplicity, reliability, produces no dissoluble sediment, emissions noxious for humans and plants. The basic difference from the above said methods is to employ for oxidizer atomic oxygen, its active forms including OH-radicals with hydrogen peroxide as the source. Hydrogen peroxide can be produced with electric power from water inside LSS.

Аннотация: Adaptation of the Bacillus subtilis strain 2335/105 (Km(r)Inf(+)) containing a recombinant plasmid encoding the extracellular human interferon alpha 2 was studied under various conditions. Stability of the plasmid in the population of B. subtilis 2335/105 was estimated under nonselective conditions. The plasmid-free cells and cells with a low number of plasmid copies were found to accumulate progressively, constituting 80% of the population after 10 culture passages, indicating the poor competitiveness of cells carrying a high number of plasmid copies. The behavior of vegetative cells of the recombinant strain introduced into aquatic microcosms differing in trophic chain length was studied. Within the first 10 days, the lysis of vegetative cells of B. subtilis 2335/105 occurred; the number of viable spores was very low but remained constant for half a year.

Аннотация: Measurements of trace elements incoming and exiting ''Bios-3'' biological-engineering life support system evidence presence of a source of these elements inside the system. This source is, most probably, structural units (steel walls, hose rubber, plastic coatings), catalysts of the thermal catalytic incinerator, expanded clay aggregate mechanically fixing plants and so on. The trace elements are received by the nutrient solutions and build up in the plant biomass. As a consequence the crew may receive elevated doses of such trace elements as aluminum, lead, nickel, chromium. To reduce income of trace elements to LSS requires thorough selection and preliminary treatment of structural materials with reduced desorption capacities and running-in of the system in idle mode prior to exploitation. This mode involves operation of the engineering part of the system (without plants and humans, but with nutrient solutions) for several months.

Аннотация: The culture collection maintained at the Institute of Biophysics (IB), Siberian Division, Russian Academy of Sciences, is the only deposit in Russia and CIS which contains about 700 strains of five species of marine luminous bacteria sampled in various sites of the World Ocean. The strains collected are used for identification of luminous bacteria; studying their physiology, biochemistry, and cytology; and developing concepts of organization and metabolism of luminous bacteria under natural and laboratory conditions. Methods of their storage and practical use are developed. The results of these studies are summarized in a data bank, which will be incorporated into the general network of data banks on microbial cultures kept in Russia and into the international network of data banks on microorganisms (MSDN). Systematization of information on luminous bacteria possessing a unique visual marker allows their extensive use in basic research and genetic engineering, for environmental and medical applications, and for biotechnological purposes.

Аннотация: Expression of the lux-genes cloned on the recombinant plasmid pPHL7 (Ap rLux +) in Escherichia coli Z905 cells was studied in various environments, including model aquatic ecosystems. Аннотация: The influence of several suites of pollutants (metallic salts, quinones, and phenols) on bacterial bioluminescence in vivo and in vitro (five test systems) was investigated. The sensitivity of bioluminescence to the different pollutants was evaluated, and inhibition constants were measured. The data obtained were shown to correlate with the physical and chemical characteristics of the substances and the structure of the bioluminescent systems. It has been found that three bioluminescent tests (water-soluble enzyme systems, immobilized enzyme systems, and bioluminescent bacteria) show higher sensitivity to pollutants and cover all types of widespread contamination. These tests were chosen as a set of bioluminescent assays for the detection of pollutants. (C) 1998 John Wiley & Sons, Inc.

Аннотация: Of all NMR observable isotopes (19)F is the one perhaps most convenient for studies on biodegradation of environmental pollutants. The reasons underlying this potential of (19)F NMR are discussed and illustrated on the basis of a study on the biodegradation of fluorophenols by four Rhodococcus strains. The results indicate marked differences between the biodegradation pathways of fluorophenols among the various Rhodococcus species. This holds not only for the level and nature of the fluorinated biodegradation pathway intermediates that accumulate, but also for the regioselectivity of the initial hydroxylation step. Several of the Rhodococcus species contain a phenol hydroxylase that catalyses the oxidative defluorination of ortho-fluorinated di- and trifluorophenols. Furthermore, it is illustrated how the (19)F NMR technique can be used as a tool in the process of identification of an accumulated unknown metabolite, in this case most likely 5-fluoromaleylacetate. Altogether, the (19)F NMR technique proved valid to obtain detailed information on the microbial biodegradation pathways of fluorinated organics, but also to provide information on the specificity of enzymes generally considered unstable and, for this reason, not much studied so far.
