Аннотация: Of all NMR observable isotopes (19)F is the one perhaps most convenient for studies on biodegradation of environmental pollutants. The reasons underlying this potential of (19)F NMR are discussed and illustrated on the basis of a study on the biodegradation of fluorophenols by four Rhodococcus strains. The results indicate marked differences between the biodegradation pathways of fluorophenols among the various Rhodococcus species. This holds not only for the level and nature of the fluorinated biodegradation pathway intermediates that accumulate, but also for the regioselectivity of the initial hydroxylation step. Several of the Rhodococcus species contain a phenol hydroxylase that catalyses the oxidative defluorination of ortho-fluorinated di- and trifluorophenols. Furthermore, it is illustrated how the (19)F NMR technique can be used as a tool in the process of identification of an accumulated unknown metabolite, in this case most likely 5-fluoromaleylacetate. Altogether, the (19)F NMR technique proved valid to obtain detailed information on the microbial biodegradation pathways of fluorinated organics, but also to provide information on the specificity of enzymes generally considered unstable and, for this reason, not much studied so far.

Аннотация: This closing technology developed for experimental BIOS-3 facility seems to be highly expedient for providing normal life conditions in high-latitude settlements and first of all for life-support of Antarctic stations. Obvious advantages of ecological life-support system based on photosynthesis, in comparison with used now in Antarctic Continent, are: 1) highly effective cascade usage of energy for performance of all functions of life-support; 2) minimization of environmental pollution due to processing human excretions and polluted water; 3) providing crew with fresh vegetable food all-the-year-round; 4) contact with alive plants is powerful source of positive emotions and effective means of psychological relaxation; 5) significant reduction of the transport charges on delivery of foodstuff to Antarctic Continent. First of all, the usage of closed ecosystem can be recommended for high-mountainous station "Concordia". Similar ecosystems can be used in northern high-latitude settlements - Russian North, Canadian Northern territories, Alaska. Finally, the experience of closed ecological life-support system operation in Antarctic Continent can serve as the best range for modeling some aspects of Martian mission - its ecological, biotechnological and psychophysiological problems - since there are no natural conditions on the Earth closer to Mars with respect to severity of a place.

Аннотация: MOX-fuel production and spent nuclear fuel reprocessing started recently at the Mining-and-Chemical Combine (MCC, Zheleznogorsk, Russia) have caused an increase in controlled releases of plutonium to the Yenisei River. In this study, we analyzed time-dependent trends of plutonium (239,240Pu and 238Pu) in biota and bottom sediments of the Yenisei during 2008-2019, to estimate comparatively the potential of abundant representatives of biota as bio-monitors of contamination of the Yenisei by plutonium. Gamma-emitting radionuclides (40 K; 60Co; 137Cs; 152Eu; 241Am) were measured in environmental samples of the Yenisei as well. Samples of bottom sediments, water moss (Fontinalis antipyretica), shining pondweed (Potamogeton lucens), caddisfly larvae with casings (Apatania crymophila), and amphipods (Eulimnogammarus viridis and Palaseopsis cancelloides) were collected downstream and upstream of the radioactive discharge site. Environmental samples of the Yenisei collected downstream of the radioactive discharge site differed considerably in activity concentrations of plutonium but were similar in time-dependent trends of plutonium, reflecting the trends of annual discharges of plutonium. In 2018, the year of a sharp increase in controlled discharge of plutonium, the concentration of 239,240Pu in water moss (26 Bq kg-1 d.w.) was higher than in sediments (14 Bq kg-1 d.w.). In other years, the highest activity concentration of 2391,240Pu was observed in bottom sediments. In view of the higher magnitude of increase in plutonium concentration, water moss and shining pondweed can be considered as more sensitive indicators of increased fresh releases of plutonium than bottom sediments. Taking into account the food-related mechanism of plutonium uptake by amphipods, this representative of biota can be regarded as a sensitive monitor of bioavailable plutonium in the Yenisei.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: Cs-137 concentrations were measured in mushrooms in an area affected by radioactive discharges of the Mining-and-Chemical Combine (MCC) (Rosatom, Russia) in 2002-2017. The sources of radionuclides in the study sites were global fallouts and waterborne and airborne radioactive discharges of the MCC. The mushroom species Suillus granulatus and S. luteus showed the highest Cs-137 concentrations (140-7100 Bq kg(-1)) for this area. Over the entire monitoring period, no significant change in Cs-137 concentration was observed in the Suillus spp. samples collected from the sites with the aerial deposition of radionuclides. In the floodplain site with the radionuclide deposition from water and air, a significant decrease in the average Cs-137 concentration was observed in the period between 2004 and 2017: a three-fold decrease in Suillus spp. and a nine-fold decrease in Lactarius deliciosus. The effective half-lives of Cs-137 in fruiting bodies of the mushrooms L. deliciosus and Suillus spp. in this site were 3.6 +/- 0.6 and 9.2 +/- 2.7 years, respectively.

Аннотация: The development of inexpensive, easy-to-produce, and easy-to-use analytical tools for detection of harmful and toxic substances is a relevant research problem with direct applications in environmental monitoring and protection. In this work, we propose a novel composite material based on alumina nanofibers and detonation nanodiamonds for detection of phenol in aqueous medium. The composite material was obtained by mixing an aqueous suspension of alumina nanofibers with a diameter of 10-15 nm and a length of several microns and a hydrosol of nanodiamonds with an average cluster size of 70 nm. The mechanisms underlying the interaction of these nanomaterials are clarified and the physicochemical properties of the composite are investigated. The SEM and TEM studies show that the obtained composite has a network structure, in which clusters of nanodiamonds (10-20 nm in diameter) are distributed over the surface of nanofibers. Coupling of nanomaterials occurs due to opposite signs of their zeta potentials, which results in electrostatic attraction and subsequent chemical bonding as indicated by the X-ray photoelectron spectroscopy and simultaneous thermal analysis. The bonding apparently occurs between functional groups (mainly carboxyl) on the surface of nanodiamonds and amphoteric hydroxyl groups on the surface of alumina nanofibers. The proposed composite allows an easy-to-perform colorimetric analysis for qualitative and quantitative determination of phenol in aqueous samples with linear response over a wide range of concentrations (0.5-106 mu M). Multiple tests have shown that the composite is reusable and retains its catalytic function for at least 1 year during storage at room temperature.

Аннотация: Dynamics of active sludge microorganism activity in aerotanks under chemostat conditions has been studied. Dependence of microorganism catalase activity has been found to depend on residual substrate concentration in proportion to the biomass of microorganisms. Experimental data and field observations has formed the basis to develop a technique to evaluate in relative units the amount of the substrate consumed by biocenosis of the active sludge in the air tanks of purification facilities. Аннотация: The present study addresses the seasonal dynamics of productivity and species composition of the meadow and steppe vegetation communities in Khakassia, determined using the ground-based and satellite data of 2017. The MODIS/Terra satellite data were used to analyze the Normalized Difference Vegetation Index (NDVI) and the Land Surface Water Index (LSWI). The NDVI and LSWI were found to be related to the productivity of the meadow and steppe vegetation. The NDVI increased as the portion of the mesophyte grasses in the grass canopy became larger. The LSWI was higher in the steppe communities, which had lower projective coverage, with spots of bare soil, than in the meadow communities, with their abundant vegetation. © Published under licence by IOP Publishing Ltd.

Аннотация: Adaptation of the Bacillus subtilis strain 2335/105 (Km(r)Inf(+)) containing a recombinant plasmid encoding the extracellular human interferon alpha 2 was studied under various conditions. Stability of the plasmid in the population of B. subtilis 2335/105 was estimated under nonselective conditions. The plasmid-free cells and cells with a low number of plasmid copies were found to accumulate progressively, constituting 80% of the population after 10 culture passages, indicating the poor competitiveness of cells carrying a high number of plasmid copies. The behavior of vegetative cells of the recombinant strain introduced into aquatic microcosms differing in trophic chain length was studied. Within the first 10 days, the lysis of vegetative cells of B. subtilis 2335/105 occurred; the number of viable spores was very low but remained constant for half a year.

Аннотация: The tick-borne encephalitis virus (TBEV) is the causative agent of one of the most severe human neuroinfections. The infection transmitted by ixodid ticks is spread throughout the forest and forest-steppe zones of the temperate climatic belt of the Eurasian continent, including the Siberian region of the Russian Federation. Despite the availability of commercial analytical systems for the detection of TBEV, the task of developing approaches to a quick and reliable analysis that can be performed routinely, particularly in environmental studies, remains topical. A solid-phase bioluminescent immunoassay for determining the tick-borne encephalitis virus (TBEV) in ticks was developed. The assay is based on the hybrid protein consisting of a modified thermostable version of Renilla muelleri luciferase and a single-chain mini-antibody to protein E. This unique protein had been obtained and investigated by the authors earlier. The current study describes the expression of the hybrid protein in two different strains of recombinant E. coli cells. The optimal conditions for obtaining a highly purified protein were found. The bioluminescent reaction of the luciferase domain was triggered with the help of the stable natural form of the substrate, a Ca-dependent coelenterazine-binding protein, the recombinant variant of which was obtained by the authors. The conditions for production and storage of the immunoassay components (the hybrid protein, the stable form of the luciferase substrate, and activated microplates) were determined. Using the developed test system, more than 900 tick samples were analyzed for TBEV. In terms of sensitivity (89.5%) and specificity (98.9%), the proposed method is not inferior to colorimetric detection and is much simpler and faster than the latter. © Siberian Federal University. Аннотация: The source of radioactive contamination of the Yenisei River floodplain, including contamination with transuranic elements, is the Mining-and-Chemical Combine of the Russian Ministry of Atomic Energy, which has for many years been producing weapons-grade plutonium. Transuranic elements have been detected not only in the soil and sediment of the river but also in the biomass of aquatic plants. This work is an investigation of accumulation and release of 241Am by a submerged macrophyte of the Yenisei River (Elodea canadensis) in laboratory experiments. In 2000-2003, laboratory experiments were carried out with biomass of E. canadensis Mich. and filtered river water. The samples were collected from the Yenisei River upstream of the discharge of the Combine's radioactive effluent. The experiments showed that 241Am is accumulated by Elodea biomass: the activity concentration of 241Am can reach 3280 В± 240 Bq/g, with the concentration factor for 241Am 16 600 В± 2200 l/kg. Results of chemical fractionation have proved that in the course of 241Am accumulation by Elodea biomass, 241Am tightly bound to biomass increases from 11% to 27% of the total 241Am in the biomass. Release of 241Am from the decaying Elodea biomass has been evaluated experimentally. By the end of the experiment (lasting up to 127 days), the Elodea plants had lost up to 65% of their initial 241Am activity and the rate of 241Am release into the water environment reached 23 Bq/day. Аннотация: In this work we experimentally estimated the capacities of the key components of the Yenisei River (Russia): particulate suspended matter (seston), diatom microalgae, and submerged macrophytes for accumulating Am-241 from water. In our experiments large particles of seston (>8 mu m), comparable in size with diatoms, took up most of americium from water. The accumulation of americium by isolated diatom algae (Asterionella formosa and Diatoma vulgare) was lower than by total seston. The concentration factors (CFs) of Am-241 for seston of the Yenisei River in our experiments were (2.8-6.9).10(5); for diatoms -(1.5-4.2)-10(4). The Us for aquatic plant Elodea canadensis were within the same order of magnitude as those for diatoms. Activity concentration and CFs of Am-241 were nearly the same in experiments under dark and light conditions. This is indicative of an energy independent mechanism of americium uptake from the water by diatoms and submerged macrophytes. (C) 2009 Elsevier Ltd. All rights reserved.

Аннотация: The aim of the paper is to investigate accumulation of artificial radionuclides by the Yenisei river aquatic plants collected in the area affected by the activity of the Mining-and-Chemical Combine (Zheleznogorsk) from 1997 to 2000. The samples of aquatic plants were of four species: Potamogeton lucens, Fontinalis antipyretica, Elodea canadensis, and Ceratophyllum demersum. The gamma-spectrometric analysis of the samples of aquatic plants for artificial radionuclides revealed a wide spectrum of radionuclides. Radionuclides of activation origin were found in the aquatic plants taken both near the Combine and 200 km down of it. The radiochemical analysis of aquatic plants revealed strontium and plutonium isotopes. Among the aquatic plants, the highest concentration factors for the principal radionuclides were recorded in Fontinalis antipyretica (water moss).

Multivariate statistics redundancy analysis (RDA) showed that investigated factors together explained 37.8 - 44.7% and 29.9 - 42.6% of the essential/beneficial and non-essential elements variability, respectively, whereas salinity was one of the main factors determining the accumulation of a number of elements (Mg, S, Na, Li, Sr, Sb) by the macrophytes. A decrease in the essential element contents (K, P, N, Cu, Zn) was observed in both species of the macrophytes during the growing season. Environmental factors (salinity, growing period, and pH) were more significant than the element contents in water and the sediments for the essential/beneficial element accumulation by two species of the macrophytes. The element contents in the sediments to a greater extent determined non-essential element accumulation by P. australis than by S. pectinata. The study showed that the efficiency of using P. australis and S. pectinata for remediation of aquatic ecosystems contaminated by heavy metals should not decrease with a salinity increase. © 2020 Elsevier Masson SAS

Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.

Аннотация: Since almost any waste recycling requires non-zero resources, almost any technology exerts non-zero impact on the biosphere. Therefore, there is an objective of finding technological configuration providing a minimal harmful effect on the biosphere. Correct assessment of the harmful effect requires as broad a framework of consideration as possible. A comparison of ecological impacts of gasoline and electric vehicles was conducted as an example of applying this approach. The contribution of technological processes involved into production, usage, and disposal of vehicles was taken into account. Comparison of the total environmental impact of the vehicles has led to a result that differs radically from widely distributed points of view about the environmental benefits of electric vehicles. It indicates that the use of the proposed approach can significantly change perceptions about the environmental and economic efficiency of various industrial products and processes. Аннотация: The bacterial bioluminescence has high sensitivity to the action of various inhibitors of biological activity. The lyophilized luminous bacteria Photobacterium phosphoreum (Microbiosensor B17 677F) and luminous strain Escherichia coli (Microbiosensor EC) from the Culture Collection IBSO were used to create bioluminescent biotests. They have been applied in ecological monitoring to determine the overall toxicity of the Yenisei and Angara Rivers and some water sources of Altai Territory. As a rule the heaviest pollution of water in studied rivers was registered near cities and settlements. The luminous bacteria biotests are simple and convenient in work, standardized and quantitative, have rapid response to actions of different substances and high sensitivity to environmental pollutants. It takes less than 30 min to do the biotest (the other biotests take 48-96 h). Copyright В© 1999 John Wiley & Sons, Ltd.
