Аннотация: Specimens of 14C-labeled polymer of 3-hydroxybutyric acid, P(3-HB), with different initial molecular weights, were used to prepare microparticles, whose morphology was not influenced by the M w of the polymer. During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: We studied the possibility of evaluation of the intensity of pathological oxidative processes in rat liver using an integral bioluminescent test with controlled perfusion of the isolated organ. The test revealed a significant correlation between the level of TBA-reactive products and bioluminescence intensity.

Аннотация: Hybrid wound dressings have been constructed using two biomaterials: bacterial cellulose (BC) and copolymer of 3-hydroxybutyric and 4-hydroxybutyric acids [P(3HB/4HB)] – a biodegradable polymer of microbial origin. Some of the experimental membranes were loaded with drugs promoting wound healing and epidermal cells differentiated from multipotent adipose-derived mesenchymal stem cells. A study has been carried out to investigate the structure and physical/mechanical properties of the membranes. The in vitro study showed that the most effective scaffolds for growing fibroblasts were composite BC/P(3HB/4HB) films loaded with actovegin. Two types of the experimental biotechnological wound dressings – BC/P(3HB/4HB)/actovegin and BC/P(3HB/4HB)/fibroblasts – were tested in vivo, on laboratory animals with model third-degree skin burns. Wound planimetry, histological examination, and biochemical and molecular methods of detecting factors of angiogenesis, inflammation, type I collagen, and keratin 10 and 14 were used to monitor wound healing. Experimental wound dressings promoted healing more effectively than VoskoPran – a commercial wound dressing. © 2019

Аннотация: Abstract: Fatty acid composition and content of 2 forms of sockeye salmon Oncorhynchus nerka from lakes in Kamchatka Peninsula (Russia) were compared. One form of sockeye salmon was anadromous ("marine"), that is, adult fish migrated in ocean to feed and grow and than return in the lake to breed. Fish of another form, kokanee, never migrate in the ocean. Per cent levels of the main indicators of nutritive value, eicosapentaenoic acid (EPA, 20:5n-3), and docosahexaenoic acid (DHA, 22:6n-3), were significantly higher in the landlocked O. nerka. However, concentrations of EPA and DHA per wet weight of filets were higher in the marine form, because of the relatively higher content of sum of fatty acids in their muscle tissue. As concluded, fish fed in marine environment had higher contents of long-chain n-3 fatty acids per wet weight than fish of the same species, fed in fresh waters. In general, both the anadromous sockeye salmon and the landlocked kokanee salmon can be recommended for human diet as a valuable product concerning contents of EPA and DHA. Аннотация: Seasonal dynamics of contents of essential polyunsaturated fatty acids (PUFA) in dominant groups of benthic invertebrates: gammarids (Gammaridae, Amphipoda), oligochaetes (Oligochaeta), chironomid larvae (Chironomidae, Diptera) and caddisfly larvae (Trichoptera), and dominant benthivorous fish, Siberian grayling Thymallus arcticus, have been studied in ecosystem of the large Siberian river. During the year of the study most benthic invertebrate taxa showed significant variations in the contents of both C-18 and long chain C-20-22 PUFAs. In contrast, the fish, which consumed the zoobenthos, had no significant seasonal variations in long chain PUFAs' contents. Thereby, the fish, as organisms of relatively higher evolution level than invertebrates, was supposed to have more strict metabolic control of long chain PUFAs' contents. Evidence was obtained that the studied fish species, grayling, may be capable to convert dietary EPA into DHA. В© 2006 Elsevier Inc. All rights reserved.

Аннотация: Microparticles obtained by using 14C-labeled resorbable hydroxyaminobutyric acid polymer were injected into the caudal vein of laboratory animals without negative aftereffects for their growth and development and without changes in the macro- and microstructure of organs and tissues. The distribution of microparticles in the viscera and the dynamics of accumulation of carbon-containing polymer degradation products in the viscera were studied. The main targets for the particles are liver tissues, as well as renal and splenic tissues. The polymeric matrix of the microparticles is most actively destroyed in the spleen and liver. The presence of high-molecular- weight polymeric matrix in organs indicates the integrity of microparticles and the possibility of long-term (up to 12 weeks) functioning of polymeric particles in vivo. Аннотация: Various chronic inflammatory and necrotic processes in the liver parenchyma are accompanied by pathological morphofunctional changes, which are associated with hepatocyte death and hyperplasia of the connective tissue. Regeneration of the liver parenchyma should include not only prevention of fibrosis, but also stimulation of hepatocyte prolifearation. The adrenoceptor agonist dobutamine stimulated proliferative activity of cultured hepatocytes and prevented the development of postintoxication liver cirrhosis in mice produced by chronic poisoning with CCl4.

Аннотация: Populations of brown trout in the Mzymta River and its tributaries include anadromous (mainly female) and resident (mainly males) fish. Some resident males in the basin of the Mzymty River attain sexual maturity at the age 1+, and resident females mature at the age 2+ or 3+. The maximum age of resident fish is 4+ in the samples studied. Migrations of anadromous brown trout to the sea occur at the ages 1+, 2+, or 3+. Future spawners spend from 1 to 4 years at feeding grounds in the sea. Smolts of the population are characterized by performing not only spring but also autumn migrations to the sea. One smolt specimen has been detected upstream from the dam in the river where spawners of anadromous brown trout do not migrate; this means that the capability for sea migrations persists long in the population represented only by resident specimens of brown trout. The diversity of life cycles and ecological forms in populations of brown trout is not lower than in populations of brown trout in Northern and Western Europe. The comparison of the data obtained with published data makes it possible to come to the conclusion about the high plasticity of ontogenesis of Black Sea brown trout.

Аннотация: Long-chain polyunsaturated fatty acids of n-3 family (n-3 LC PUFAs) are physiologically essential compounds required for normal growth and development of animals, including humans. The ability of fish species to synthesize n-3 LC PUFAs varies significantly across different trophic levels. We have studied fatty acid (FA) content (mg/g of wet weight) and level (% of total FAs) in the brain, liver, heart, intestine, female and male gonads, muscle, and adipose tissues of commercially important wild freshwater Baikal grayling. Additionally, FA content and level of Baikal grayling juveniles have been studied. In all tissues of Baikal grayling, some LC PUFAs, namely, 24:5n-3 and 24:6n-3 (C24 PUFAs), have been found. These FAs are the intermediate products in the synthesis of docosahexaenoic acid (DHA, 22:6n-3) by the Sprecher pathway. The levels of C24 PUFAs in tissues differed significantly: the highest levels of C24 PUFAs were found in adipose tissue and the lowest values in the gonads of females, liver, brain, and head of juveniles. According to the dynamics of DHA and C24 PUFAs, the maximum rate of DHA synthesis is achieved in brain of Baikal grayling, while the lowest rate of DHA synthesis probably occurs in adipose tissue. Although all studied tissues had differences in the number of FAs and their levels, 16:0, 18:1n-9, 16:1n-7, 20:5n-3, and DHA dominated. Male gonads contained an extremely high level of furan FAs - presumably beneficial substances for human health. Additionally, the nutritional value of the tissues of Baikal grayling as a source of n-3 LC PUFAs for humans has been estimated.

Аннотация: We investigated the effects of ?-radiation on the survival of resting eggs of the cladoceran Moina macrocopa, on the parameters of the life cycle of neonates hatched from the irradiated eggs and on the performance of the population initiated from irradiated eggs. The study showed that ?-radiation in a range of doses from the background level to 100 Gy had no effect on survival of irradiated eggs. The absorbed dose of 200 Gy was lethal to resting eggs of M. macrocopa. The number of clutches and net reproductive rate (R0) of hatchlings from eggs exposed to radiation were the strongly affected parameters in experiments with individual females. The number of clutches per female was drastically reduced for females hatched from egg exposed to 80–100 Gy. The most sensitive parameter was the R0. The estimated ED50 for the R0 (effective dose that induces 50% R0 reduction) was 50 Gy. Population performance was also affected by the irradiation of the resting stage of animals that initiated population. Populations that was initiated from hatchlings from resting eggs exposed to 100 Gy was of smaller size and with fewer juvenile and parthenogenetic females in comparison with control populations. Thus, we determined the dose-response relationship for the effect of gamma radiation on survival of resting eggs and individual and population responses of hatchlings from irradiated resting eggs. We conclude that for highly polluted areas contamination of bottom sediments with radioactive materials could affect zooplankton communities through adverse chronic effects on resting eggs, which will be transmitted to hatchlings at individual or population levels. © 2017 Elsevier Ltd

Аннотация: Chronic experiments on mature Wistar rats studied the biological properties of suture threads from two types of polyoxyalkanoates (POA), such as polyoxybutyrate (POB) and the copolymer of POB and polyoxyvalerate (POV) (POB/POV) as compared to the conventional surgical material (silk and catgut). The physiological and biochemical parameters that reflect the status of the animals, including their behavior, height and development, the weight of the body and viscera, the morphology of peripheral blood, the biochemical parameters of blood, the activity of some enzymes, as well as histological findings of the viscera, and lymphoid tissue responses were assessed. POA was found to produce no negative effect on the physiological, biochemical, and functional characteristics of the animals irrespective of the duration of contact with the internal medium of the body and of the chemical composition of a material.

Аннотация: Background: Canine parvovirus (CPV) is now recognized as a serious threat to the dog breeding industry worldwide. Currently used CPV vaccines all have their specific drawbacks, prompting a search for alternative safe and effective vaccination strategies such as subunit vaccine. VP2 protein is the major antigen targeted for developing CPV subunit vaccine, however, its production in baculovirus expression system remains challenging due to the insufficient yield. Therefore, our study aims to increase the VP2 protein production by using an improved baculovirus expression system and to evaluate the immunogenicity of the purified VP2 protein in mice. Results: The results showed that high-level expression of the full length VP2 protein was achieved using our modified baculovirus expression system. The recombinant virus carrying two copies of VP2 gene showed the highest expression level, with a productivity of 186 mg/L, which is about 1.4-1.6 fold that of the recombinant viruses carrying only one copy. The purified protein reacted with Mouse anti-His tag monoclonal antibody and Rabbit anti-VP2 polyclonal antibody. BALB/c mice were intramuscularly immunized with purified VP2 protein twice at 2 week intervals. After vaccination, VP2 protein could induce the mice produce high level of hemagglutination inhibition antibodies. Conclusions: Full length CPV VP2 protein was expressed at high level and purified efficiently. Moreover, it stimulated mice to produce high level of antibodies with hemmaglutination inhibition properties. The VP2 protein expressed in this study could be used as a putative economic and efficient subunit vaccine against CPV infection. © 2020 The Author(s).
