Аннотация: Исследованы амфифильные свойства бакт. люциферазы в различных условиях и ее взаимодействие с длинноцепоч. алифатич. альдегидом и соответствующей жирной к-той. Методом малоуглового рассеяния рентген. лучей определены радиусы инерции фермента в р-ре и найдены зависимости их от состояния люциферазы. Исследована реология монослоев, полученных из белка, субстрата, продукта и их комбинаций со строго определенными соотношениями на поверхности различных субфаз. Соответствующие 'пи'-А изотермы показывают, что во всех случаях образуются твердые двумерные пленки, сжимаемость к-рых сильно зависит от соотношения компонент. Монослои перенесены на твердую подложку и получены мультислойные пленки на разных носителях. Рентгеновское исследование позволило определить ближний порядок в толстых пленках, к-рый также зависит от соотношения компонент. Метод позволяет конструировать регулярные неоднородности распределения люциферазы в монослое. Результаты электр. микроскопии чистых и смешанных ионо- и бислоев, перенесенных на твердые подложки, свидетельствуют в пользу микроэмульсионной модели амфифильности люциферазы. Получены ленгмюровские пленки с сохранением активности. Это открывает путь к новому поколению люциферазных твердотельных биосенсоров. Библ. 21

Аннотация: The synthesis of polyhydroxybutyrate by the carboxydobacterium Seliberia carboxydohydrogena under various growth conditions was studied. The bacterium was able to accumulate poly-beta-hydroxybutyric acid and a heteropolymer that was a product of copolymerization of hydroxybutyric and hydroxyvaleric acids. Nitrogen and sulfur exerted the strongest effect on polymer accumulation in carboxydobacteria: The maximum polymer concentrations in cells reached 28%. Specimens of polyhydroxybutyrate films were obtained, and their degradability by soil microflora was shown.

Аннотация: The influence of a monolayer of a palmitic acid on amplitude-frequency characteristics of temperature oscillations caused by thermal spots of a surface film was experimentally determined in a Langmuir tank. An analysis of the power spectra showed that the temperature oscillations were multimodal. In addition, no consistant differences between the spectrum for a pure surface and the spectrum for a surface covered by an acid monolayer were found. A roughened gain-frequency factor was found that is equal to the ratio of the average period between the thermal spots to the dispersion of the time series of temperature oscillations. The factor significantly differed over a wide range of temperatures for a pure surface and for a surface covered with the monolayer. The experimental results do not confirm the existence of a leading frequency in the temperature oscillations of a cold surface film due to an appearance of thermal spots.

Аннотация: Biodegradation of polyhydroxyalkanoates (PHA) - polymers of hydroxylated fatty acids - differing in chemical composition, crystallinity, melting temperature, and molecular weight was studied in soil and water environments under conditions of varying ambient temperature and pH. Under similar conditions, homogeneous films of polyhydroxybutyrate (PHB) with different initial properties were degraded at virtually equal rates. Hydroxybutyrate-hydroxyvalerate copolymers with a lower crystallinity and different microstructure in comparison to PHB were degraded at a 20 to 30% higher rate on average. The ambient temperature significantly influenced the rate of PHA biodegradation.

Аннотация: The surface properties of diamond nanoparticles (40-50 A in diameter) have been investigated by X-ray photoelectron spectroscopy (XPS), UV photoelectron spectroscopy (UPS) and Raman spectroscopy. The diamond nanoparticles have been deposited on flat Si(100) substrates by electrophoresis/dielectrophoresis. The as-deposited films are strongly oxidized and present a 1-2% nitrogen content. After treatment at 850 В°C in H2 plasma for 60 min, the oxygen is removed, and the position of the C 1s core-level peak indicates a n-type electronic comportment of the diamond nanoparticles. Raman spectroscopy of the as-deposited film shows a sp3 contribution at 1321 cm-1 and a sp2 contribution around 1620 cm-1. The 12 cm-1 shift of the sp3 contribution with respect to the bulk diamond peak at 1333 cm-1 is attributed to a phonon confinement effect due to the size of the diamond particles. The H2 plasma treatment induces a size decrease of the nanocrystallites confirmed by Raman and scanning electron microscopy (SEM) measurements. UPS spectroscopy shows a negative electron affinity of -0.2 eV of the hydrogenated nanodiamond film.

Аннотация: The thermal conductivity, k, of diamond/carbon composites with different ratios of sp(2)/sp(3)-bonded carbon is measured by the laser flash technique. The thermal conductivity of nanocomposites containing 6 nm diamond particles falls within the range of k = 0.003-0.017 W/cmK; at room temperature. The thermal conductivity increases while nanopores are gradually filled with pyrolytic carbon (pyrocarbon/diamond mass ratio variation of 0.0-0.5). Transmission electron microscopy data reveal a fairly uniform mixture of two carbon phases, the diamond and matrix having similar grain sizes. Estimates show that the phonon free path is limited by dimensions of carbon matrix layer. Thermal data for coarse-grain (1-2 mu m) composites are also given for comparison. Scanning tunneling-field emission microscopy was used to study the mechanisms of low-field electron emission from the composites, and a model based on quantum well effect at the diamond/graphite interface was proposed and discussed.

Аннотация: Properties of the field electron emission for diamond/pyrocarbon nanocomposites produced from diamond particles surrounded by an sp(2)-bonded pyrocarbon matrix are considered as functions of a size of diamond particles selected in the range of 5 nm - 5 mum, and of an average thickness of the pyrocarbon shell controlled by the pyrocarbon/diamond mass ratio varied from 0 to 0.5. The low-threshold emission at fields of greater than or equal to1 V/mum with ''no activation/no hysteresis'' I-V behavior was observed for these materials using tungsten tip microprobes as well as a fluorescent screen. A specially designed scanning tunneling-field emission microscope was used for simultaneous mapping of field emission intensity, topography, work function, and electrical resistivity to study the mechanisms of the emission from the composites and well-emitting chemical vapor deposition diamond films. It was found that for both of the materials emission centers are associated with interfaces between diamond and sp2-bonded carbon phases. Possible mechanisms of the low-field electron emission for the diamond/graphite composites including local field enhancement are analyzed. A model of the low-field emission based on quantum well effect at the diamond/graphite interface is proposed and discussed. (C) 2001 American Vacuum Society.

Аннотация: This work describes avenues to use satellite information to analyse dynamic processes in aquatic ecosystems. Information for this analysis, was retrieved from AVHRR satellite sensor data. This information consisteds of time series of images of radiation temperature and turbidity. We expect this information will be of great value in analysing inland water bodies. Methods to process satellite information using original software and data processing techniques are proposed. For the investigation of the process and analyses of satellite information Shira Lake (Khakasia, Siberia) was used as a case study. To study the variability of the surface temperature and turbidity of the Lake in summer, the satellite and ground-truth data of the lake was applied. This study represents the first evaluation of the dynamic processes for Lake Shira based on satellite, ground-truth and modelling data. We developed algorithms and software to process satellite images to enable the reconstruction of time dependence of temperature and spectral reflectance of water bodies in the visible range, and to make computer-animated films visualising the spatial and temporal dynamics of the study parameters. The analyses of morphometric, meteorological and hydrological characteristics of Lake Shira have provided a realistic opportunity for processing the satellite information and to develop numerical models of variability of the hydrological regime of the lake. The results obtained demonstrate the feasibility of systematically retrieving the spatial information from the satellite data on the dynamics of the surface water temperature and of the suspended matter in the lake.

Аннотация: The study addressed the ability of the transgenic strain (TM) B. subtilis 2335/pBMB 105 (Km(r)Inf(+)) to form biofilms on the surface of liquid media of various compositions, inoculated with vegetative cells and spores. The morphological features of these biofilms do not differ from those of the films formed by the recipient strain (WT) B. subtilis 2335 (Km(s)). However, the TM and the natural one differ in the dynamics of biofilm formation and the cellular composition of the films. Biofilms of the TM are formed earlier, develop at a higher rate, but decompose later than the films of the WT. When the medium is inoculated with vegetative cells, sporulation in the biofilms of both strains undergoes glucose repression; no such effect is observed when the medium is inoculated with spores. The TM does not form films when the medium is inoculated with spores and supplemented with glycerin and kanamycin.

Аннотация: Samples of olyhydroxyalkanoates (PHAs), polyhydroxybutyrate (PHB) and copolymers poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) with 4 and 18 mol% hydroxyvalerate, synthesized by the bacteria Ralstonia eutropha B5786, were investigated. PHA films in contact with blood did not activate the hemostasis system at the level of cell response, but they did activate the coagulation system and the complement reaction. To detect biologically-active components in the PHAs, a detailed analysis of the composition of the polymers was conducted. Gas chromatography-mass spectrometry revealed long-chain fatty acids (FAs) in the tested PHAs. Their total concentration in the polymer ranged from tenths of mol% to 2-3 mol%, depending on the purification method. C-16:0 constituted the largest proportion, up to 70%. Of the long-chain hydroxy acids, only beta-OH-C-14:0 was detected and it did not exceed 0.06 mol%. The analysis of the hemocompatibility properties of the PHAs purified by a specialized procedure, including the quantitative and morphological estimation of platelets adherent to the surface of polymer films, the plasma recalcification time and complement activation studies, indicated that PHB and PHBV can be used in contact with blood. It has been found out that the lipopolysaccharides of bacteria producing PHAs, which contain mostly long-chain hydroxy acids, can be the factor activating the hemostasis systems. Thus, the technology of PHA purification must satisfy rather stringent specific requirements.

Аннотация: Samples of olyhydroxyalkanoates (PHAs), polyhydroxybutyrate (PHB) and copolymers poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) with 4 and 18 mol% hydroxyvalerate, synthesized by the bacteria Ralstonia eutropha B5786, were investigated. PHA films in contact with blood did not activate the hemostasis system at the level of cell response, but they did activate the coagulation system and the complement reaction. To detect biologically-active components in the PHAs, a detailed analysis of the composition of the polymers was conducted. Gas chromatography-mass spectrometry revealed long-chain fatty acids (FAs) in the tested PHAs. Their total concentration in the polymer ranged from tenths of mol% to 2-3 mol%, depending on the purification method. C16:0 constituted the largest proportion, up to 70%. Of the long-chain hydroxy acids, only ?-OH-C14:0 was detected and it did not exceed 0.06 mol%. The analysis of the hemocompatibility properties of the PHAs purified by a specialized procedure, including the quantitative and morphological estimation of platelets adherent to the surface of polymer films, the plasma recalcification time and complement activation studies, indicated that PHB and PHBV can be used in contact with blood. It has been found out that the lipopolysaccharides of bacteria producing PHAs, which contain mostly long-chain hydroxy acids, can be the factor activating the hemostasis systems. Thus, the technology of PHA purification must satisfy rather stringent specific requirements.

Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.

Аннотация: The study addresses the effect of abiotic (medium salinity and copper ions) and biotic (interactions between populations) factors on the formation of structured communities by binary associations consisting of halotolerant bacteria (Alcaligenes sp. 1-1 or Acinetobacter sp. 1-19) and a wild-type B. subtilis 2335 strain or a transgenic strain. The results showed that 250 mg l(-1) of copper ions inhibit formation of biofilms by monocultures of the tested strains. Binary associations of the strains were more resistant to high concentrations (250 mg l(-1)) of copper ions. At the lowest NaCl concentration (0.05% and 2.5%) and in the presence of copper ions, bacilli seemed to help halotolerant bacteria survive. Under increased salinity and in the presence of copper ions, structured communities developed due to halotolerant bacteria. Coexistence under stressful conditions was beneficial for the both groups of bacteria.

Аннотация: This work is a review of the results of biomedical studies of polymer devices (films, fibers, microparticles, 30 implants) made from resorbable PHAs synthesized by the bacterium Wautersia (Ralstonia) eutropha 65786, using the technology developed at the Institute of Biophysics of the Siberian Branch of the Russian Academy of Sciences. Two types of PHAs - polyhydroxybutyrate (PHB) and a hydroxybutyrate/hydroxyvalerate copolymer (PHB/PHV) - have been proven to be biocompatible in vitro in cultures of fibroblasts, endothelial cells, hepatocytes, and osteoblasts, and in short- and long-duration experiments on animals. Polymer films and membranes have been found to be usable as scaffolds for functioning cells and monofilament suture fibers - for stitching muscular-fascial wounds and in abdominal surgery. Ectopic bone formation assay and experiments with the model of segmental osteotomy showed that 30 PHB and PHB/HA implants can be used for reparative osteogenesis. Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: The effect of laser irradiation on the properties of the surface of films prepared from a bioresorbable polymer poly(hydroxybuturate) has been studied. To determine the spectral region of the polymer optimal for the effective action of radiation on electron molecular bonds, theoretical investigations have been performed, which have shown that, for modifying the surface of PHB scaffolds, it is expedient to use a vacuum laser at a wavelength of 160 nm. Using laser irradiation at a power from 3 to 30 W, a series of films with modified surface, from roughnesses to perforations, have been obtained. The microstructure and properties of the film surface depending on the mode of irradiation have been examined, and conditions have been found under which the contact marginal angles of film wetting with water can be decreased to 50В° (compared with 76-80В° in starting products). Thus, conditions of laser treatment of PHB scaffolds have been theoretically substantiated and experimentally realized that provide a beneficial effect on the properties of the surface without destroying the structure of the material.
