Аннотация: The transfer of transuranium element americium-241 (241Am) from food and water to organs and tissues of freshwater fish (Carassius auratus gibelio, crucian carp) has been investigated in laboratory experiments. The fishes were fed with homogenized biomass of submerged macrophytes labeled with 241Am, through catheter. For the first two days after force-feeding, fishes excreted up to 80% of ingested americium, for four days - 98%. 241Am was registered in internal organs and tissues that had no direct contact with americium (liver, bones, muscles) as early as two days after the feeding and after eight days, when the digestive tract was depurated. Among internal organs, the highest activity concentration was recorded in the liver (up to 427 Bq/kg). Activity concentration in bones (19-31 Bq/kg) was several times higher than in muscles, indicating the affinity of americium to bone tissue. Assimilation of 241Am in organs and tissues of crucian carp from water occurred mainly via the digestive tract too. В© 2011 EDP Sciences.

Аннотация: The integral bioluminescent biotest with lyophilized fluorescent bacteria was used for monitoring of LPO processes in tissue extracts and serum of rats exposed to stress. A relationship between the content of MDA (LPO indicator) and fluorescence of bacteria was observed in all biological samples.

Аннотация: Long-chain polyunsaturated n-3 fatty acids (LC-PUFA), including eicosapentaenoic (EPA), and docosahexaenoic (DHA) are an essential part of human diets, but their main source, the wild fish catch, is limited. Terrestrial production animals, such as cattle, pigs, and chicken, can synthesize these compounds from the plant-derived precursor -linolenic acid, and the synthesis occurs mainly in liver. We estimated the putative contribution of liver products to the LC-PUFA supply in human nutrition, as an alternative to fish. We measured EPA and DHA contents in raw and cooked livers of the animals. Calculations of global production of EPA and DHA in the livers of these animals were done. Mean contents of LC-PUFA in raw livers of chicken, pigs, and cattle were comparable with those in some fish. Culinary treatments of the liver did not result in a decrease of EPA and DHA contents. Global production of EPA and DHA in animals' livers was estimated as approximate to 410(6)kg/year, while the global EPA+DHA supply through the wild fish catch is known to be 18010(6)kg/year. Thus, liver of production animals is an additional source of LC-PUFA for human nutrition, rather than an alternative to fish. Practical applications: Consumption of food with a high PUFA content and a low n-6/n-3 ratio is recommended for humans to prevent cardiovascular disease and psychiatric disorders. We found that cooked pork and beef liver is a valuable source of n-3 LC-PUFA in human nutrition. In contrast, cooked chicken liver had too high of a n-6/n-3 ratio and, thereby, had a lower nutritive value concerning n-3 LC-PUFA. An estimate has been made of the putative contribution of liver products of terrestrial animals to the LC-PUFA supply as alternative to fish in human nutrition.

Аннотация: The two paradoxical major health problems namely obesity and anemia are confirmed to affect millions around the world. Hepcidin, a protein synthesized in liver is a negative iron binding regulator. There is an affirmative relation between hepcidin and leptin levels and an inverse co-relation between hepcidin and iron status due to inflammation mediated by obesity in adolescents. So this implicates an alliance between anemia and obesity wherein weight reduction can be a powerful medium to improve iron absorption in obese adolescents. Also the Body Mass Index can serve as a preliminary non-invasive screening tool to identify potential adolescents prone to anemia. © 2018

Аннотация: Hepatocyte proliferation in the liver regenerating after partial hepatectomy ceases when the organ is restored, and the mechanism of this phenomenon is still unclear. In the experiments on fusing hepatocytes from the reoenerated mouse liver (15 days after partial hepatectomy) with NIH 3T3 mouse fibroblasts, we revealed no DNA synthesis in the nuclei of stimulated fibroblasts in heterokaryons (in the presence of hepatocyte nuclei), whereas DNA synthesis in nonfused cells was undisturbed. In this work, our purpose was to find out whether the suppression of DNA synthesis in heterokaryons could be due to the appearance in hepatocytes of some endogenous factors having an inhibitory effect on proliferation. To this end, hepatocytes from the mouse liver regenerated after partial hepatectomy were treated with cycloheximide for 1-4 h and were then fused with stimulated fibroblasts. Such a short-term treatment of hepatocytes with cycloheximide proved to result in the loss of their ability to inhibit DNA synthesis in the nuclei of stimulated or quiescent fibroblasts in heterokaryons, but hepatocytes proper actively proliferated in the medium with a low serum content (0.2%). When the mice with the liver reoenerated after partial hepatectomy were treated with a single sublethal dose of cycloheximide (3 mg/kg), their hepatocytes taken two days after this treatment had no inhibitory effect. Puromycin, another inhibitor of protein synthesis, had the same effect on hepatocytes. These results may be interpreted as evidence that the final stage of liver regeneration after damage is controlled by the factors having a C C negative effect on cell proliferation.

Аннотация: Treatment of patients with mechanical jaundice (MJ) has been one of the topical problems of current medicine. The fraction of patients having MJ of oncologic character that can undergo radical surgery does not exceed 25-30%. A way out of such situation is to use minimally invasive endobiliary interventions, the main type of which is endoprosthesis replacement of bile duct lumens. The aim of the work was to study the biological properties of the experimental models of polymer stents made of PHA for endobiliary prosthetics and to investigate the biological properties of PHA suture material for forming of biliary-enteric anastomoses (cholecystoduodenostomy). Experiments were performed on 20 adult mongrel dogs, weighing 10-12 kg. The animals were divided to three groups: the negative control group (intact animals); the positive control group (the animals with implanted endobiliary silicon stents); and the experimental group (the animals with the PHA stents). The animals were monitored for 100 days. The clinical blood analysis was made before the operation, on day 7, 30, 60 and 100 days. During autopsy the presence of exudate, commissural process in the free abdominal cavity and subhepatic spatium, the appearance of choledoch where the prosthesis was located, as well as the appearance of cholecystoenterostomy, liver and duodenum. We did not found any signs of inflammation, cicatrical changes were estimated in the free abdominal cavity and subhepatic spatium. All implanted PHA stents were at their initial places of implantation. After the end of the experiment inflammatory reaction and anastomositis were absent. Macroscopic changes of liver and duodenum were also not detected. Liver function did not have any pathological deviations. These positive results give grounds to conclude that application of PHA as endobiliary stents in reconstructive surgery of bile passages and as suture material is a promising technology. Аннотация: Serum-deprived (0.2 %) resting and serum-stimulated (10 %) proliferating NIH 3T3 mouse fibroblasts were fused with hepatocytes from intact, regenerating and embryonic mouse livers to elucidate mechanisms of liver cell proliferation, DNA synthesis being investigated in nuclei of heterokaryons and non-fused cells using radioautography. Hepatocytes in heterokaryons were found to have no inhibitory effect on the entry of stimulated fibroblast nuclei into the S-period, but on the contrary they were involved in DNA synthesis. In addition, the nuclei in heterokaryons mutually stimulated each other to enter the S-period. In their turn, the resting fibroblasts did not prevent the proliferating hepatocytes from the regenerating and embryonic livers to enter the S-period. Possible reasons of the absence of inhibitory effect of differentiated cells in heterokaryons are discussed. The data obtained enable us to conclude that the mechanism of proliferative process control in resting immortalized cells differs from that in differentiated cells where proliferation seems to be stopped without affecting the endogenous inhibitor postulated for the resting and ageing fibroblasts.

Аннотация: Hematological parameters and the state of liver cells of rats were examined in vivo after the animals received aflatoxin B(1) (AfB(1)) alone and together with modified nanodiamonds (MND) synthesized by detonation. The rats that had received the MND hydrosol had elevated leukocyte levels, mainly due to higher granulocyte counts and somewhat increased monocyte counts compared to control rats. Hematological parameters of the rats that had received AfB(1) alone differed from those of the control rats in another way: total white blood cell counts were significantly lower due to the decreased lymphocyte counts. In rats that had consumed AfB(1) with the MND hydrosol, changes in hematological parameters were less pronounced than in rats that had consumed either AfB(1) or MND. Electron microscopy showed that hepatocytes of the rats that had received the MND hydrosol or AfB(1) with the MND hydrosol contained elevated levels of lipid inclusions and lysosomes. Hyperplasia of the smooth endoplasmic reticulum (EPR) was revealed in liver specimens of the rats that had received AfB(1). Results of the study suggest the conclusion about mutual mitigation of the effects of nanoparticles and the mycotoxin on rats blood and liver cells after AfB(1) has adsorbed on MND.

Аннотация: For the first time, seasonal changes in the content of total lipids (TLs) and phospholipids (PLs) were studied in fodder plants growing in Central Yakutia—a perennial cereal, smooth brome (Bromopsis inermis L.), and an annual cereal, common oat (Avena sativa L.). Both species have concentrated TLs and PLs in autumn under cold hardening. In addition, a significant increase in the content of fatty acids (FAs) of B. inermis was observed during the autumn decrease in temperature. The Yakutian horses, which fed on cereals enriched with nutrients preserved by natural cold (green cryo-fodder), accumulated significant amounts of 18:2n-6 and 18:3n-3, the total content of which in cereals was 75% of the total FA content. We found differences in the distribution of these two FAs in different tissues of the horses. Thus, liver was rich in 18:2n-6, while muscle and adipose tissues accumulated mainly 18:3n-3. Such a distribution may indicate different roles of these FAs in the metabolism of the horses. Аннотация: Long-chain polyunsaturated fatty acids of n-3 family (n-3 LC PUFAs) are physiologically essential compounds required for normal growth and development of animals, including humans. The ability of fish species to synthesize n-3 LC PUFAs varies significantly across different trophic levels. We have studied fatty acid (FA) content (mg/g of wet weight) and level (% of total FAs) in the brain, liver, heart, intestine, female and male gonads, muscle, and adipose tissues of commercially important wild freshwater Baikal grayling. Additionally, FA content and level of Baikal grayling juveniles have been studied. In all tissues of Baikal grayling, some LC PUFAs, namely, 24:5n-3 and 24:6n-3 (C24 PUFAs), have been found. These FAs are the intermediate products in the synthesis of docosahexaenoic acid (DHA, 22:6n-3) by the Sprecher pathway. The levels of C24 PUFAs in tissues differed significantly: the highest levels of C24 PUFAs were found in adipose tissue and the lowest values in the gonads of females, liver, brain, and head of juveniles. According to the dynamics of DHA and C24 PUFAs, the maximum rate of DHA synthesis is achieved in brain of Baikal grayling, while the lowest rate of DHA synthesis probably occurs in adipose tissue. Although all studied tissues had differences in the number of FAs and their levels, 16:0, 18:1n-9, 16:1n-7, 20:5n-3, and DHA dominated. Male gonads contained an extremely high level of furan FAs — presumably beneficial substances for human health. Аннотация: Long-chain polyunsaturated fatty acids of n-3 family (n-3 LC PUFAs) are physiologically essential compounds required for normal growth and development of animals, including humans. The ability of fish species to synthesize n-3 LC PUFAs varies significantly across different trophic levels. We have studied fatty acid (FA) content (mg/g of wet weight) and level (% of total FAs) in the brain, liver, heart, intestine, female and male gonads, muscle, and adipose tissues of commercially important wild freshwater Baikal grayling. Additionally, FA content and level of Baikal grayling juveniles have been studied. In all tissues of Baikal grayling, some LC PUFAs, namely, 24:5n-3 and 24:6n-3 (C24 PUFAs), have been found. These FAs are the intermediate products in the synthesis of docosahexaenoic acid (DHA, 22:6n-3) by the Sprecher pathway. The levels of C24 PUFAs in tissues differed significantly: the highest levels of C24 PUFAs were found in adipose tissue and the lowest values in the gonads of females, liver, brain, and head of juveniles. According to the dynamics of DHA and C24 PUFAs, the maximum rate of DHA synthesis is achieved in brain of Baikal grayling, while the lowest rate of DHA synthesis probably occurs in adipose tissue. Although all studied tissues had differences in the number of FAs and their levels, 16:0, 18:1n-9, 16:1n-7, 20:5n-3, and DHA dominated. Male gonads contained an extremely high level of furan FAs - presumably beneficial substances for human health. Additionally, the nutritional value of the tissues of Baikal grayling as a source of n-3 LC PUFAs for humans has been estimated.

Аннотация: Abstract: We compared the composition and content of fatty acids (FAs) in the liver, muscles, and subcutaneous fat of Yakut horses inhabiting extreme environment in the Cryolithozone. Essential linoleic and alpha-linolenic acids, supplied to horses with their food, were accumulated in different tissues. Linoleic acid was accumulated in the liver but alpha-linolenic acid was accumulated in muscle and subcutaneous fat. Such a distribution indicates different roles of these fatty acids in the metabolism of horses. Yakut horse meat is a valuable dietary product owing to its fatty acid composition and content. © 2020, Pleiades Publishing, Ltd.
