Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.

Аннотация: There is considerable data on molecular evolution, but there remains no approach to systematizing them within the framework of the key problems of biology. To search for the most common properties of evolving systems, the heuristic method has been proposed. Artificial networks of formal neurons were chosen as the heuristic model object. The paper examines the divergent component of evolutionary trajectory formation. As a result of the simulation, the dependence of the potential variability parameter on the position of the fitness function landscape was obtained. The simulation results are in agreement with the real data of molecular evolution experiments. © Published under licence by IOP Publishing Ltd.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: A procedure has been developed to fabricate polymer tubes using solution casting inside template tubes. Polyester solution placed inside the vertically fixed template evaporated forming a hollow tube on the inner walls of the template. Silicone tubes used as the templates were permeable to solvent vapors and had relatively low adhesion to materials of the fabricated tubes, enabling their effortless removal. Parameters of the polymer tubes were determined by a type of the polymer, its concentration in the solution, and the inner diameter of the template. In the first method, a clamp was placed on the lower end of the template tube. A thickness of the new tube walls gradually increased from the open to the clamped end of the template tube. In another method viscous polymer solution without clamping was used; in this case, a polymeric bulkhead was formed in the middle of the tube. Two microbial polyesters, poly-3-hydroxybutyrate and poly-3-hydroxybutyrate-co-3-hydroxyvalerate, and also synthetic poly-L-lactide were used as model polymers. (C) 2020 Elsevier B.V. Аннотация: The study addresses 242Pu accumulation by Elodea canadensis, one of the abundant species of submerged plants in the Yenisei River. 242Pu in water samples of the "Elodea - Yenisei River water" model system and in the biomass fractions was determined ?-spectrometrically, following radiochemical recovery of 242Pu using 236Pu - a chemical yield tracer. The experiments on accumulation of 242Pu by Elodea biomass showed that the activity concentration of 242Pu can reach 21 В± 2 Bq/g dry wt, with the concentration factor for 242Pu 13100 В± 2100 L/kg dry wt. Results of chemical fractionation proved that during the first few hours of the experiment 242Pu contained in Elodea was mainly concentrated in the exchangeable and the adsorbed fractions of biomass (about 100%). As Elodea biomass accumulated 242Pu, the absolute amount of 242Pu in the exchangeable and the adsorbed fractions remained almost unchanged, although the portion of 242Pu tightly bound to biomass increased. At the end of the experiment, on day 7, 242Pu tightly bound to biomass (fractions of organics and mineral residue) constituted 43-63% (in different experiments) of the total 242Pu in the biomass. Аннотация: The proposed elementary mathematical model of formation and decomposition of soil organic matter (SOM) is based on using equations of chemical kinetics to describe the multistage process of SOM transformation. The model both describes each step of transformation in accordance with the law of mass action and postulates the trend of increasing stability of the matter towards further transformation, which is common for all steps. Analysis of the model demonstrates that it is extremely difficult to construct a realistic model of SOM dynamics by assembling elementary models of the type presented in this study into the full description of SOM transformation processes. © 2018 Elsevier B.V.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.

Аннотация: The present study describes production of bacterial cellulose composites with silver nanoparticles and antibiotics and compares their properties. Bacterial cellulose (BC) composites synthesized in the culture of the strain of acetic acid bacterium Komagataeibacter xylinus VKPM B-12068 with silver nanoparticles, BC/AgNps, were produced hydrothermally, under different AgNO3 concentrations (0.0001, 0.001, and 0.01 M) in the reaction medium. The presence of silver in the BC/AgNp composites was confirmed by elemental analysis conducted using scanning electron microscopy with a system of X-ray spectral analysis. Analysis showed that the average atomic number of silver particles in composite samples depended on the concentration of AgNO3: as AgNO3 concentration in the reaction solution was increased, silver content in the composites increased from 0.044 to 0.37 mg/cm2. BC composites with amikacin and ceftriaxone were prepared by immersing dry BC films in solutions containing different concentrations of the antibiotics. The surface structure and properties and physicochemical and mechanical characteristics of composites were investigated using SEM, DSC, X-ray analysis, the system for measuring water contact angles, and electromechanical tensile testing machine. The disk-diffusion method and the shake-flask culture method used in this study showed that all experimental composites had pronounced antibacterial activity against E. coli, Ps. eruginosa, K. pneumoniae, and St. aureus, and the BC/antibiotic composites were more active than BC/AgNp ones; S. aureus was the most susceptible to the effect of BC composites. No potential cytotoxicity was detected in any of the BC/AgNp composites in the NIH 3T3 mouse fibroblast cell culture, in contrast to the BC/antibiotic composites. These results suggest that BC composites constructed in the present study hold promise as dressings for managing wounds, including contaminated ones. © 2017 Elsevier Ltd

Аннотация: Recombinant apoobelin and recombinant luciferase are separated from bacterial cells of Escherichia coli with the use of detonation nanodiamonds. The application of nanodiamonds has a number of points in its favor, namely, (i) simplifies the procedures for purifying the proteins, (ii) decreases the time of their separation to 30-40 min, (iii) eliminates the necessity of using special chromatographic equipment, and (iv) makes it possible to prepare high-purity apoobelin and luciferase materials with protein yields of 35-45 and 45-60%, respectively. Аннотация: Specimens of 14C-labeled polymer of 3-hydroxybutyric acid, P(3-HB), with different initial molecular weights, were used to prepare microparticles, whose morphology was not influenced by the M w of the polymer. During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: Microspheres have been prepared from the resorbable linear polyester of ?-hydroxybutyric acid (polyhydroxybutyrate, PHB) by the solvent evaporation technique and investigated in vitro and in vivo. Biocompatibility of the microspheres has been proved in tests in the culture of mouse fibroblast cell line NIH 3T3 and in experiments on intramuscular implantation of the microspheres to Wistar rats for 3 months. Tissue response to the implantation of polymeric microspheres has been found to consist in a mild inflammatory reaction, pronounced macrophage infiltration that increases over time, involving mono- and poly-nuclear foreign body giant cells that resorb the polymeric matrix. No fibrous capsules were formed around polymeric microparticles; neither necrosis nor any other adverse morphological changes and tissue transformation in response to the implantation of the PHB microparticles were recorded. The results of the study suggest that polyhydroxybutyrate is a good candidate for fabricating prolonged-action drugs in the form of microparticles intended for intramuscular injection. Аннотация: The paper presents a brief overview of the results of the implementation of the project "Agropreparations of the new generation: a strategy of construction and realization". The first part contains the analysis of the growth of the wild-type strain Cupriavidus necator B-10646 (formerly eutrophus) and the synthesis of polyhydroxyalkanoates by this strain on various substrates: glycerol, palm oil, Siberian oil seed, sunflower seed oils, and oleic acid. On refined glycerin, a highly productive process is implemented when scaling up, allowing to obtain 128 ± 11 g / L PHA. Evaluation of oils has shown that palm oil is the best carbon substrate. The second part presents the results of the development of environmentally friendly slow-release pesticide formulations. They are a degradable matrix of poly-3-hydroxybutyrate mixed with natural materials (peat, clay, wood flour), into which a pesticide (metribuzin, tribenuron-methyl, fenoxaprop-P-ethyl, azoxystrobin, epoxiconazole, and tebuconazole) has been. The developed preparations showed high activity against pathogenic fungi and weeds and had a much weaker negative effect on the soil microflora. Studies of the degradation of the developed preparations and the release of pesticides into the soil confirm their effectiveness over a long period of time, up to 90 days. © Published under licence by IOP Publishing Ltd.

Аннотация: Sediments from eutrophic reservoir Bugach (Siberia, Russia) were tested for possibility to produce biodiesel. We supposed that the sediments could be a promising biodiesel producer. The major reason of high price of biodiesel fuel is cost of a raw material. The use of dredging sediments for biodiesel production reduces production costs, because the dredging sediments are by-products which originated during lake restoration actions, and are free of cost raw materials. Lipid content in sediments was 0.24% of dry weight. To assess the potential of from sediments as a substitute of diesel fuel, the properties of the biodiesel such as cetane number, iodine number and heat of combustion were calculated. All of this parameters complied with limits established by EN 14214 and EN 14213 related to biodiesel quality. В© 2011 Elsevier Ltd.

Аннотация: In vitro experiments proved the usefulness of electron paramagnetic resonance (EPR) spectrometry for detecting detonation nanodiamonds (NDs) in samples of biomaterials (blood and homogenates of organs of mice). A characteristic EPR signal (g = 2.003, Delta H similar or equal to 10 G) was detected in biomaterials containing NDs, and its intensity linearly increased at nanoparticle concentrations of between 1.6 and 200 mu g/ml. In vivo experiments demonstrated that EPR spectrometry was effective for monitoring the inter-organ distribution of NDs intravenously injected to mice. In 2.5 h after the injection of NDs, the nanoparticles mainly accumulated in the lungs and liver of the animals - about 25 and 20%, respectively, of the initially injected NDs. The amounts of NDs accumulated in the heart and kidneys were considerably lower. Also, EPR spectrometry did not detect NDs in the blood, spleen, brain, and femoral muscles of mice. Ten days after injection, EPR spectrometry detected redistribution of NDs in mice. The amounts of nanoparticles decreased approximately by a factor of 3.5 in the lungs and increased almost by a factor of 3 in the liver; NDs were detected in the spleen. This study suggests ways to use EPR spectrometry to study the distribution, accumulation, and elimination of detonation NDs injected into laboratory animals.

Аннотация: Purpose: The aim of the work is to compare the biological activity of carbonic nano-structures of natural and artificial origination, namely, humic substances (HS) and fullerenols. Materials and methods: The representative of the fullerenol group, С60Оy(OH)x where у + x = 20–22, was chosen. Enzyme-based luminescent bioassay was applied to evaluate toxicity and antioxidant properties of HS and fullerenol (F); chemiluminescent luminol method was used to study a content of reactive oxygen species (ROS) in the solutions. Toxicity of the bioactive compounds was evaluated using effective concentrations ЕС50; detoxification coefficients DOxT were applied to study and compare antioxidant activity of the compounds. Antioxidant activity and ranges of active concentrations of the bioactive compounds were determined in model solutions of organic and inorganic oxidizers—1,4-benzoquinone and potassium ferricianide. Results and discussion: Values of ЕС50 revealed higher toxicity of HS than F (0.005 and 0.108 g L?1, respectively); detoxifying concentrations of F were found to be lower. Antioxidant ability of HS was demonstrated to be time-dependent; the 50-min preliminary incubation in oxidizer solutions was suggested as optimal for the detoxification procedure. On the contrary, F’ antioxidant effect demonstrated independency on time. Antioxidant effect of HS did not depend on amphiphilic characteristics of the media (values of DOxT were 1.3 in the solutions of organic and inorganic oxidizers), while this of F was found to depend: it was maximal (DOxT = 2.0) in solutions of organic oxidizer, 1,4-benzoquinone. Conclusions: Both HS and F demonstrated toxicity and low-concentration antioxidant ability; however, quantitative characteristics of their effects were different. The differences were explained with HS polyfunctionality, higher ability to decrease ROS content, non-rigidity, and diffusion restrictions in their solutions. Antioxidant effect of the bioactive compounds was presumably attributed to catalytic redox activity of their ?-fragments. The paper demonstrates a high potential of luminescent enzymatic bioassay to study biological activity of nano-structures of natural and artificial origination. © 2018, Springer-Verlag GmbH Germany, part of Springer Nature.

Аннотация: To create a life support system based on biological and physical-chemical processes is the optimum solution providing full-valued condidtions for existence and efficient work of people at a lunar base. Long-standing experinece in experimental research or closed ecosystems and their components allows us to suggest a realistic functional structure of the lunar base and to estimate qualitatively its parameters. The original restrictions are as follows: 1) the basic source of energy to support the biological processes has to be the solar radiation; 2) the initial amount of basic biological elelments forming the turnover of substances (C, O, H, P, K, N) has to be delivered from Earth; 3). Moon materials are not to be used in the biological turnover inside the base; 4) the base is to supply the crew fully with atmosphere and water, and with 90% (A scenario) or 40% (B scenario) of food. Experimental data about the plant productivity under the "Moon" rhythm of light and darkness allow us to suggest that the A scenario requires per one human: plant area - 40 m2 irradiated during the lunar day by 250-300 W/m2 PAR producing 1250 g of dry biomass a terrestrial day; a heterotrophic component of "biological incineration" of inedible plant biomass (800 g/day) including the aquaculture of fish to produce animal products and contaminating the environment less than birds and mammals, and the culture of edible mushrooms; a component of physical-chemical correction for the LSS envi ronment including the subsystems of: deep oxidation of organic impurities in the atmosphere and of water, organic wastes of human activity and that biological components (420 g/day) Co2 concentration in "Moon" nights, damping O2 in "Moon" days, etc. The stock of presotred or delivered from Earth substances (food additions, seeds, etc.) to be involved in biological turnover is to be about 50 kg/year per man. Increase of the mass of prestored substances per man up to 220 kg/year would reduce twice the plant area and consumed amount of radiant energy to exclude the components of "biological incineration" and physical-chemical destruction of organic wastes. Аннотация: Slow-release fungicide formulations (azoxystrobin, epoxiconazole, and tebuconazole) shaped as pellets and granules in a matrix of biodegradable poly(3-hydroxybutyrate) and natural fillers (clay, wood flour, and peat) were constructed. Infrared spectroscopy showed no formation of chemical bonds between components in the experimental formulations. The formulations of pesticides had antifungal activity against Fusarium verticillioides in vitro. A study of biodegradation of the experimental fungicide formulations in the soil showed that the degradation process was mainly influenced by the type of formulation without significant influence of the type of filler. More active destruction of the granules led to a more rapid accumulation of fungicides in the soil. The content of fungicides present in the soil as a result of degradation of the formulations and fungicide release was determined by their solubility. Thus, all formulations are able to function in the soil for a long time, ensuring gradual and sustained delivery of fungicides.

Аннотация: BACKGROUND The purpose of the present study was to develop ecofriendly herbicide formulations. Its main aim was to develop and investigate slow-release formulations of herbicides (metribuzin, tribenuron-methyl, and fenoxaprop-P-ethyl) of different structure, solubility, and specificity, which were loaded into a degradable matrix of poly-3-hydroxybutyrate (P(3HB)) blended with available natural materials (peat, clay, and wood flour). RESULTS Differences in the structure and physicochemical properties of the formulations were studied depending on the type of the matrix. Herbicide release and accumulation in soil were associated with the solubility of the herbicide. Fourier-transform infrared spectroscopy showed that no chemical bonds were formed between the components in the experimental formulations. Degradation of the formulations in agro-transformed soil in laboratory conditions was chiefly influenced by the shape of the specimens (granules or pellets) while the effect of the type of filler (peat, clay, or wood flour) was insignificant. The use of granules enabled more rapid accumulation of the herbicides in soil: their peak concentrations were reached after 3 weeks of incubation while the concentrations of the herbicides released from the pellets were the highest after 5-7 weeks. Loading of the herbicides into the polymer matrix composed of the slowly degraded P(3HB) and natural materials enabled both sustained function of the formulations in soil (lasting between 1.5 and >= 3 months) and stable activity of the otherwise rapidly inactivated herbicides such as tribenuron-methyl and fenoxaprop-P-ethyl. CONCLUSION The experimental herbicide formulations enabled slow release of the active ingredients to soil. (c) 2019 Society of Chemical Industry
