Аннотация: In cold smoked species of genus Coregonus, identified by molecular genetic analysis, contents of fatty acids and heavy metals and arsenic were measured. The highest content of sum of long-chain polyunsaturated fatty acids of omega-3 family (LC-PUFA), namely eicosapentaenoic (EPA) and docosahexaenoic (DHA) fatty acids, 6.53 ± 0.78 mg g?1 wet weight, was characteristic of tugun Coregonus tugun. This is the first quantitative estimation (mg LC-PUFA per g of product) of the nutritive value of smoked fish. Thus, to obtain a daily personal doze of EPA + DHA of 1 g, recommended for prevention of cardiovascular diseases, one needs to consume 153 g of the smoked tugun. Metals contents did not exceed standards for fish meat except Pb in least cisco Coregonus sardinella. Accordingly, values of hazard quotients, which estimate benefit-risk ratio of fish intake, indicate that most of the smoked fish species are safe product for human nutrition, except least cisco regarding Pb content. © 2020 Elsevier Inc.

Аннотация: The harmful potential of bulk bottom sediments of the Yenisei River has been assessed by the Allium test. Sediment samples have been taken in ten sites on a plot of about 100 km in length in the middle reaches of the Yenisei River affected by urban activity of the city of Krasnoyarsk and industrial plants. The samples of sediments differ considerably in the content of potentially toxic substances (heavy metals, artificial radionuclides, and organic pollutants) and nutrients and in physical properties. We use the mean length of onion root as an indicator of general toxicity, the mitotic index as an indicator of cytotoxicity, and the percentage of anatelophase cells in the apical root meristem with abnormal chromosomes as an indicator of genotoxicity. Both the stimulation and inhibition of indicator endpoints are revealed in sediment samples of the Yenisei River relative to the control (artificial sediments). An increased genotoxicity is recorded in the sample of sediments contaminated by a mixture of organic and chemical toxicants. A significant positive correlation between the percentage of abnormal cells and the content of copper and petrochemicals in the samples is revealed. No extremely strong reactions of onion endpoints to the quality of sediment samples are revealed, which is probably related to relatively low contamination of the sediments studied. This is in agreement with the data of chemical analyses. Based on the testing results, we can conclude that the endpoints of the Allium test are sensitive enough to the quality of bottom sediments of the Yenisei River and can be used for their biotesting.

Аннотация: Enzymatic luminescent systems are a promising tool for rapid detection of heavy metals ions for water quality assessment. Nevertheless, their widespread use is limited by the lack of test procedure automation and available sensitive handheld luminometers. Herein we describe integration of disposable microfluidic chips for bioluminescent enzyme-inhibition based assay with a handheld luminometer, which detection system is based on a thermally stabilized silicon photomultiplier (SiPM). Microfluidic chips were made of poly(methyl methacrylate) by micro-milling method and sealed using a solvent bonding technique. The composition of the bioluminescent system in microfluidic chip was optimized to achieve higher luminescence intensity and storage time. Results indicate that developed device provided comparable sensitivity with bench-scale PMT-based commercial luminometers. Limit of detection for copper (II) sulfate reached 2.5 mg/L for developed biosensor. Hereby we proved the concept of handheld enzymatic optical biosensors with disposable chips for bioassay. The proposed biosensor can be used as an early warning field-deployable system for rapid detection of heavy metals salts and other toxic chemicals, which affect bioluminescent signal of enzymatic reaction.

Аннотация: Many Cladocera species under unfavourable conditions produce resting eggs that can survive for years in deep diapause. Resting eggs form egg banks at the bottom sediments, which serve as a source of genetic diversity and replenish the population after periods of decline. Despite the obvious importance of resting eggs for the ecosystem functioning, studies assessing the sensitivity of resting eggs to different toxicants are scarce. We reviewed published data on the sensitivity of resting eggs to the effect of heavy metals, organic pollutants and ionizing radiation. Analysis shows that the effects of toxicants of different types on resting eggs will have different environmental consequences. Egg banks may suffer from prolonged contact of dormant eggs with heavy metals. However, the ecological relevance of these effects is low, since the effective concentrations of toxicants must be very high. In addition, the effect of heavy metals on resting eggs is not transmitted to hatchlings from exposed eggs. Taking into account high toxicity of heavy metals to active animals, we assume that the toxic effect of heavy metals is critical for active zooplankters and relatively safe for resting stages. Accumulation of artificial radionuclides in bottom sediments can have a significant impact on aquatic ecosystems through chronic effects both on survival of resting eggs and on the life cycle parameters of animals hatched from irradiated eggs. Resting eggs during reactivation are more sensitive to the effect of ionizing radiation. Pesticides and complex chemical compounds produce similar toxic effects on both resting eggs and life history parameters of hatchlings from exposed resting eggs. The bottom line is that to predict the effect of contamination of bottom sediments by different pollutants on the ecosystem structure and functioning, it is highly important to investigate the viability of the resting eggs under a wide range of concentrations of various toxicants and different durations of direct contact of eggs with toxicants.

Аннотация: Accumulation of essential/beneficial and non-essential chemical elements by submerged (Stuckenia pectinata (L.) Borner) and emergent (Phragmites australis (Cav.) Trin. ex Steud.) rooted macrophytes in three water bodies was studied as dependent on water salinity (specific conductivity) and main environmental factors (temperature, pH of water, growing period, element contents in water and sediments). Inductively coupled plasma optical emission spectrometry (ICP - OES) was used for the element detection in the plants, sediments, and water. Multivariate statistics redundancy analysis (RDA) showed that investigated factors together explained 37.8 - 44.7% and 29.9 - 42.6% of the essential/beneficial and non-essential elements variability, respectively, whereas salinity was one of the main factors determining the accumulation of a number of elements (Mg, S, Na, Li, Sr, Sb) by the macrophytes. A decrease in the essential element contents (K, P, N, Cu, Zn) was observed in both species of the macrophytes during the growing season. Environmental factors (salinity, growing period, and pH) were more significant than the element contents in water and the sediments for the essential/beneficial element accumulation by two species of the macrophytes. The element contents in the sediments to a greater extent determined non-essential element accumulation by P. australis than by S. pectinata. The study showed that the efficiency of using P. australis and S. pectinata for remediation of aquatic ecosystems contaminated by heavy metals should not decrease with a salinity increase. © 2020 Elsevier Masson SAS

Аннотация: Optically enhanced biosensing strategies are prerequisites for developing miniature and highly sensitive multiplexed analytical platforms. Such smart biosensing systems are highly promising for use in the fields of biomedicine and environmental monitoring. Optical signal enhancement during bioassays is attributed to the complex opto-electronic interactions of incoming photonic signals at the nanomaterial interface. Research on the use of metals other than gold and silver for such purposes tends to extend the spectral window to observe luminescence enhancement effects. Such manifold increase in luminescence may be explained by the principles of plasmon coupling, directional emission led high collection efficiency, Rayleigh scattering and related opto-electronic events. The present review begins with a mechanistic description of important phenomena associated with metal-induced luminescence enhancement, particularly focusing on the origin of metal-enhanced luminescence. This review further analyses the hybrid nanostructure capabilities responsible for maintaining unique opto-electronic properties during bio-functionalisation. Current research trends in this area, future scope of this field for designing useful bioassays and concluding remarks are then discussed. (C) 2017 Elsevier B.V. All rights reserved.

Аннотация: Fatty acid (FA) and element contents were studied in 14 brands of canned (in its own juice and with sunflower oil) saury (Cololabis saira), a popular product of Russian market. Canned saury is a valuable source of essential polyunsaturated FA - eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3). To obtain personal daily dose of EPA + DHA of 1 g for prevention of cardiovascular diseases, neural and inflammatory disorders one needs to intake from 26 to 76 g of canned saury, as was calculated for studied brands. ICP-OES analysis of 24 elements showed that Pb concentration in one brand and Cd content in most of studied saury samples exceeded standards for fish meat established by the European Commission. However, values of hazard quotient, HQEFA, which estimate benefit -risk ratio of fish intake, indicate that canned saury is safe product for human nutrition. (C) 2016 Elsevier Ltd. Аннотация: The research aimed to determine critical concentrations of heavy metals at which survival of resting eggs of the cladoceran Moina macrocopa is negatively affected. Resting eggs’ viability was not affected over a 30-days exposure towards copper, cadmium, zinc or nickel at concentrations up to 60–70 g/L. When resting eggs were exposed to sediment contaminated with heavy metals for 8 months, the hatching success was affected at 30 g copper/kg. Thus, resting eggs of Cladocera can tolerate heavy metals at concentrations that far exceed lethal concentrations of heavy metals to active life stage and exceed low or moderate levels of environmental pollution. Follow up investigation of life table parameters of hatchlings from resting eggs exposed to heavy metals demonstrated that neither lifespan nor fecundity of hatchlings differ from control animals. Аннотация: A number of approaches have been proposed and tested to transfer enzymatic reactions into the functional elements of microfluidic chips on the example of the bienzyme bioluminescent reaction involving NAD(P)H:FMN-oxidoreductase and luciferase. Measurement of the catalytic activity of these enzymes (under the influence of pollutants) is the basis of enzymatic bioassay of various liquids. It was found that all of the components of the reaction must be placed in the same cell of the chip to improve the reproducibility of the measurements. The use of starch gel as a carrier for immobilization and gelatin as a scaffold in the reactor of the chip enables the preservation of enzyme activity in the course of sealing the chip at room temperature. It is shown that the components of the reaction should be vigorously stirred in a microfluidic chip reactor to improve the efficiency of the analysis. As a result of the studies, a prototype of microfluidic chip based on the enzymatic bioluminescent reaction is proposed. It is characterized by a detection limit of copper sulfate of 3 mu M that corresponds to the sensitivity of traditional lux-biosensors based on living cells. The analysis time is reduced to 1 min, and the analysis can be performed by individuals without special laboratory skills.

Аннотация: The relevance of the study is determined by the necessity to obtain the data on concentrations of chemical elements in water of the Yenisei River, which is continuously affected by human activities. The aim of the study is to determine concentrations of such chemical elements as Na, K, Ca, Mg, Zn, Cu, Ba, Al, Mn, U, Mo, Cr, Ni, As, Co, Sr, Fe, Pb, Cd, and Bi in water of the Yenisei River between the city of Krasnoyarsk and the outfall of the Angara River. The methods used in the study. Total contents of the elements were determined by inductively coupled plasma mass spectrometry, using an Agilent 7500a instrument. The results. The study showed the changes in average concentrations of 20 chemical elements in water of the Yenisei River between the city of Krasnoyarsk and the outfall of the Angara River (the village Strelka), for 2010-2015. Comparison of the data obtained with the levels of maximum permissible concentrations showed that the examined part of the Yenisei River contained the increased concentrations of Fe, Cu, Mn, Al, and Zn. For the study period, the magnitudes of average concentrations of Fe reached 1...2 maximum permissible concentrations (MPC) and Cu concentrations reached 1...5 MPC The average concentrations of Mn amounted to 1...3 MPC in the parts of the river over 90 km downstream of Krasnoyarsk (at the villages Atamanovo and Strelka). Cu, Fe, and Zn concentrations tend to increase somewhat in the parts of the river over 90 km downstream of Krasnoyarsk, which may be caused by the influence of the industrial complex of the town Zheleznogorsk and the outfall of the Kan River, with the large industrial facilities located along its banks, in Zelenogorsk, in particular.

Аннотация: The bacterial composition of water and sediments in Lake Pyasino and adjacent rivers has been studied by high-throughput sequencing for the first time. No hydrocarbon-degrading bacteria are found in the river sections exposed to the diesel spillage in May 2020. The Daldykan and Ambarnaya rivers, located downstream of the spill site, are dominated by bacteria that oxidize sulfur and heavy metals. At the same time, these bacteria are not found in Lake Pyasino, which is dominated by small cyanobacteria. Thus, the composition of bacteria in the water and sediments of the Norilsk-Pyasino lake-river system corresponds to the technogenic impact to which it is exposed and also demonstrates the buffer properties of Lake Pyasino as water flows into the Kara Sea.

Аннотация: The results of a hydrochemical analysis of the consequences of an accidental fuel release in the Norilsk-Pyasino water system are presented. The pollution of watercourses in the catchment of Lake Pyasino (a nameless (Nadezhdinsky) creek, the Daldykan River, and the Ambarnaya River) with oil products, phenols, easily oxidizable and hard to oxidize organic matter (COD, PO, and BOD5), suspended solids, inorganic salts, and heavy metals at concentrations exceeding the background levels and MPC for fishery water bodies, as well as a temperature rise in waters of the nameless creek near Norilsk Heat and Power Plant 3 (CHPP-3), have been revealed. The contamination of the surface water decreases downstream in ascending order: nameless creek-Daldykan River-Ambarnaya River. The occurrence of oil products, phenols, and organic substances in the surface waters 2 months after the fuel spill is obviously due to their diffusion from the river bottom sediments, which accumulated a considerable quantity of heavy fractions of diesel fuel after the accident. Increased concentrations of Ca, Cu, Zn, Mn, Co, and Ni in the waters of the studied tributaries of Lake Pyasino are not directly related to the accident; they result from the general technogenic pollution of the territory and the increased geochemical background for these elements. Water contamination with oil products and phenols in the studied areas of Lake Pyasino (its central and northern parts) and the Pyasino River has not been detected. However, Pb concentrations exceed the MPC and Cd is recorded in the water, which is probably due to pollutants that accumulated in previous years.

Аннотация: The presence of potentially toxic xenobiotics in complex matrices has become rather the rule than the exception. Therefore, there is a need for highly sensitive inexpensive techniques for analyzing environmental and food matrices for toxicants. Enzymes are selectively sensitive to various toxic compounds, and, thus, they can be used as the basis for detection of contaminants in complex matrices. There are, however, a number of difficulties associated with the analysis of complex matrices using enzyme assays, including the necessity to take into account properties and effects of the natural components of the test media for accurate interpretation of results. The present study describes the six-stage procedure for designing new enzyme sensors intended for assessing the quality of complex matrices. This procedure should be followed both to achieve the highest possible sensitivity of the biosensor to potentially toxic substances and to minimize the effect of the uncontaminated components of complex mixtures on the activity of the biosensor. The proposed strategy has been tested in designing a bioluminescent biosensor for integrated rapid assessment of the safety of fruits and vegetables. The biosensor is based on the coupled enzyme system NAD(P)H:FMN-oxidoreductase and luciferase as the biorecognition element. The study describes methods and techniques for attaining the desired result in each stage. The proposed six-stage procedure for designing bioluminescent enzyme biosensors can be used to design the enzymatic biosensors based on other enzymes.

Аннотация: The trophic position of fish is one of the most important factors controlling the accumulation of potentially toxic elements and compounds in fish tissues, primarily via the spectrum of fish nutrition. In this study, the content of potentially toxic transition metals (Cu, Zn, Mn, Pb, and Cd) and radiocesium (Cs-137) in the edible tissues (muscles and liver) of two representatives of the fish-eating ichthyofauna of the Yenisei River, northern pike (Esox lucius) and burbot (Lota lota), have been comparatively studied relatively to the size of the fish. A significant decrease in the content of radiocesium and zinc in the muscles of pike and zinc in the liver of pike with an increase in body size has been recorded in juvenile pikes with a body weight (W) of less than 0.35 kg. For larger sexually mature pikes, no significant size dependences of the accumulation of metals in tissues are found. A positive correlation is found between the content of Cs-137, Cu, Zn, and Mn in muscles and the size of burbot in the W range from 0.42 to 1.62 kg. The tissues of burbots and pikes of the same size significantly (p < 0.05) differ in the concentration of metals in their tissues: concentrations of Zn and Mn are 1.6-2.2 times higher in the muscle of burbot; the concentration of Pb is 1.8 times higher in muscle of pike; Cu is twice as high in the liver of burbot; and Zn and Mn are 4.7 and 1.6 times higher in the liver of pike, respectively. These differences may be due to the different food spectra of pike and burbot. Concentrations of Cu, Zn, and Mn in the liver of pike are 3-7 times higher than in muscle; the concentration of Cu in the liver of burbot is 5 times higher than in muscle. Pb and Cd tend to be higher in liver than muscle for both fish species. Despite the revealed size dependences, the concentrations of potentially toxic metals and radiocesium in the muscles and liver of fish are below the permissible concentrations for food. These results can be used to assess environmental risks for the population consuming fish, as well as to plan for the long-term environmental monitoring of rivers using representatives of piscivorous fish.

Аннотация: There is a need for rapid simple and informative environmental assessment methods. The present investigation is aimed at assessing the possibility of using the combined enzyme system of luminescent bacteria: NAD(P)H:FMN-oxidoreductase + luciferase (Red + Luc) for predicting the potential toxicity of industrial urbostratozems sampled in the city of Krasnoyarsk. Three groups of urbostratozems polluted with fluorine, arsenic and lead, were tested by the methods of chemical analysis and enzymatic bioassay. Only the assessment of the arsenic-contaminated soil samples showed the dependence between the reduced activity of the enzyme system and the arsenic concentration variations. The results reveal that the sensitivity of the Red + Luc enzyme system to the soil pollutants depends on the properties of the studied soil samples. Moreover, the solubility of lead in the soil samples affects the accuracy of the enzymatic bioassays for soil toxicity testing. The results of the enzymatic bioassay of the fluoride-contaminated soil samples are ambiguous. The obtained data show the relevance of the sample preparation during integral bioassays. In addition, soil properties should be taken into account as well. The current study emphasizes the importance of conducting chemical and biological testing as a combined set to obtain comprehensive information about the anthropogenic load. (C) 2021 Elsevier B.V. Аннотация: The review summarizes studies on the detoxification effects of water-soluble humic substances (HS), which are products of the natural transformation of organic substances in soils and bottom sediments that serve as natural detoxifying agents in water solutions. The detoxifying effects of HS on microorganisms are quite complex: HS neutralize free pollutants (indirect bioeffects) and also stimulate the protective response of organisms (direct bioeffects). Prospects and potential problems of bioluminescent bacteria-based assay to monitor toxicity of solutions in the presence of HS are discussed. The main criterion for the bioassay application is versatility and ease of use. The detoxification efficiency of HS in different pollutant solutions was evaluated, and the detoxification mechanisms are discussed. Particular attention was paid to the direct and complex direct + indirect effects of HS. The review focuses on the protective function of HS in solutions of radionuclides and salts of stable metals, with special consideration of the antioxidant properties of HS.

Аннотация: Long-term trends of artificial radionuclides, from 2003 to 2018, in two abundant species of macrophytes, shining pondweed, Potamogeton lucens, and water moss, Fontinalis antipyretica, have been analyzed to estimate the indicative reliability of these two species as biomonitors of radioactive contamination in a river system and to quantify the decrease in the content of artificial radionuclides in the Yenisei River. Time-dependent trends of artificial radionuclides in the biomass of these species were similar, resulting in estimates of effective half-lives for Mn-54, Co-58, Co-60, Zn-65, Cs-137, and Eu-152 similar for both species. Concentrations of artificial radionuclides in biomass of shining pondweed and water moss correlated with annual discharges of the radionuclides to the Yenisei at different levels of significance, and the strongest (R-2 > 0.7) positive correlation (p < 0.05) was obtained for Co-60, Zn-65, and Eu-152. Concentrations of Co-60, Cs-137, and Eu-152 in water moss were 2-7.5 times higher than in shining pondweed, and considerable percentages of those isotopes were recorded in extracellular particulate matter, which was largely represented by epiphytic diatoms. Higher concentrations of artificial radionuclides in the biomass of water moss can be considered as an advantage of water moss as a monitor of radioactive contamination of the Yenisei, while shining pondweed is more useful for estimation of annual deposits of radionuclides in vegetation of the Yenisei and spatial transfer of radionuclides downstream of the discharge site. Despite differences in concentrations of artificial radionuclides, both species can be considered as reliable indicators of radioactive contamination of the river on a long-term scale.
