Аннотация: The extent of inhibition of the bioluminescence reaction by wheat grain extracts was studied as a function of the scabby kernel content in wheat. The NADH : flavine mononucleotide oxidoreductase-luciferase bienzyme bioluminescence system was found to be the most sensitive to mycotoxins produced by fungi of the genus Fusarium. A biological luciferase test was developed for monitoring wheat grain infection with Fusarium.

Аннотация: Cs-137 concentrations were measured in mushrooms in an area affected by radioactive discharges of the Mining-and-Chemical Combine (MCC) (Rosatom, Russia) in 2002-2017. The sources of radionuclides in the study sites were global fallouts and waterborne and airborne radioactive discharges of the MCC. The mushroom species Suillus granulatus and S. luteus showed the highest Cs-137 concentrations (140-7100 Bq kg(-1)) for this area. Over the entire monitoring period, no significant change in Cs-137 concentration was observed in the Suillus spp. samples collected from the sites with the aerial deposition of radionuclides. In the floodplain site with the radionuclide deposition from water and air, a significant decrease in the average Cs-137 concentration was observed in the period between 2004 and 2017: a three-fold decrease in Suillus spp. and a nine-fold decrease in Lactarius deliciosus. The effective half-lives of Cs-137 in fruiting bodies of the mushrooms L. deliciosus and Suillus spp. in this site were 3.6 +/- 0.6 and 9.2 +/- 2.7 years, respectively.

Аннотация: The development of inexpensive, easy-to-produce, and easy-to-use analytical tools for detection of harmful and toxic substances is a relevant research problem with direct applications in environmental monitoring and protection. In this work, we propose a novel composite material based on alumina nanofibers and detonation nanodiamonds for detection of phenol in aqueous medium. The composite material was obtained by mixing an aqueous suspension of alumina nanofibers with a diameter of 10-15 nm and a length of several microns and a hydrosol of nanodiamonds with an average cluster size of 70 nm. The mechanisms underlying the interaction of these nanomaterials are clarified and the physicochemical properties of the composite are investigated. The SEM and TEM studies show that the obtained composite has a network structure, in which clusters of nanodiamonds (10-20 nm in diameter) are distributed over the surface of nanofibers. Coupling of nanomaterials occurs due to opposite signs of their zeta potentials, which results in electrostatic attraction and subsequent chemical bonding as indicated by the X-ray photoelectron spectroscopy and simultaneous thermal analysis. The bonding apparently occurs between functional groups (mainly carboxyl) on the surface of nanodiamonds and amphoteric hydroxyl groups on the surface of alumina nanofibers. The proposed composite allows an easy-to-perform colorimetric analysis for qualitative and quantitative determination of phenol in aqueous samples with linear response over a wide range of concentrations (0.5-106 mu M). Multiple tests have shown that the composite is reusable and retains its catalytic function for at least 1 year during storage at room temperature.

Аннотация: Upcoming professional sports authorities seek rapid noninvasive biosensing tools for regular monitoring of athletes' physiological states. The analysis of saliva through luminescence-based biosensors has been perceived as a suitable candidate for such purposes. The present study reports a qualitative bioluminescence assay based on a coupled enzyme system that consists of bacterial luciferase (BLuc) and nicotinamide adenine dinucleotide (NADH):flavin mononucleotide (FMN) oxidoreductase (Red), BLuc-Red, for the express diagnostics of athletes' stress levels before and after physical exertion. The volunteers who participated in the study were grouped as freestyle wrestlers and students who adapted to different levels of physical activities. Under physical exertion modelling conditions, the influence of participant saliva on BLuc-Red catalyzed light emission was investigated. Results showed a significant increase in residual luminescence (I-exp, mean maximum bioluminescence intensity of the experimental measurement (I-exp); I-c, luminescence intensity in control; I-exp/I-c, %) values for participants in the wrestler group while a decrease in the student group (P 0.05). Such contrasting residual luminescence values in both groups were found to be dependent on the catalase activity of saliva. The proposed bioluminescence assay can be utilized as a potential nonspecific biosensing tool for determining the physical state of athletes under high loads.

Аннотация: Remote sensing applications have greatly enhanced ability to monitor and manage in the areas of forestry. Accurate measurements of regional and global scale vegetation dynamics (phenology) are required to improve models and understanding of inter-annual variability in terrestrial ecosystem carbon exchange and climate-biosphere interactions. Study of vegetation phenology is required for understanding of variability in ecosystem. In this paper, monitoring of vegetation dynamics using time series of satellite data is presented. Vegetation variability (vegetation rate) in different topoclimatic areas is investigated. Original software using IDL interactive language for processing of satellite long-term data series was developed. To investigate growth dynamics vegetation rate inferred from remote sensing was used. All estimations based on annual time series of Moderate Resolution Imaging Spectroradiometer (MODIS) imagery. Vegetation rate for Enhanced Vegetation Index (EVI) and Normalized Difference Vegetation Index (NDVI) was calculated using MODIS data. The time series covers spring seasons of each of 9 years, from 2000 to 2008. Comparison of EVI and NDVI derived growth rates has shown that NDVI derived rates reveal spatial structure better. Using long-term data of vegetation rates variance was estimated that helps to reveal areas with anomalous growth rate. Such estimation shows sensitivity degree of different areas to different topoclimatic conditions. Woods of heights depend on spatial topoclimatic variability unlike woods of lowlands. Principal components analysis shows vegetation with different rate conditions. Also it reveals vegetation of same type in areas with different conditions. It was demonstrated that using of methods for estimating the dynamic state of vegetation based on remote sensing data enables successful monitoring of vegetation phenology. В© 2011 COSPAR. To preserve the biosphere and to make appropriate use of it, it is imperative to have a deep insight into the long-term dynamics of the primary production on the planet. To investigate the dynamics of chlorophyll concentration based on satellite data, the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) has developed a geoinformation system. It was used to detect the areas in the global ocean that are quasistationary in relation to seasonal dynamics of chlorophyll concentration. Areas such as these, found in the Indian Ocean, are described in this work. These areas form the basis for the analysis of long-term dynamics of chlorophyll concentration. In these quasistationary zones systematic monitoring of phytopigment concentration is conducted by space-borne and marine craft. The work presents long-term satellite-based data on the space distribution of anomalous deviations of chlorophyll concentration in the ocean. An anomaly criterion is proposed and maps of Pacific Ocean areas with a high probability of anomalies are presented. В© 2004 Taylor and Francis Ltd.

Аннотация: The source of radioactive contamination of the Yenisei River floodplain, including contamination with transuranic elements, is the Mining-and-Chemical Combine of the Russian Ministry of Atomic Energy, which has for many years been producing weapons-grade plutonium. Transuranic elements have been detected not only in the soil and sediment of the river but also in the biomass of aquatic plants. This work is an investigation of accumulation and release of 241Am by a submerged macrophyte of the Yenisei River (Elodea canadensis) in laboratory experiments. In 2000-2003, laboratory experiments were carried out with biomass of E. canadensis Mich. and filtered river water. The samples were collected from the Yenisei River upstream of the discharge of the Combine's radioactive effluent. The experiments showed that 241Am is accumulated by Elodea biomass: the activity concentration of 241Am can reach 3280 В± 240 Bq/g, with the concentration factor for 241Am 16 600 В± 2200 l/kg. Results of chemical fractionation have proved that in the course of 241Am accumulation by Elodea biomass, 241Am tightly bound to biomass increases from 11% to 27% of the total 241Am in the biomass. Release of 241Am from the decaying Elodea biomass has been evaluated experimentally. By the end of the experiment (lasting up to 127 days), the Elodea plants had lost up to 65% of their initial 241Am activity and the rate of 241Am release into the water environment reached 23 Bq/day. Аннотация: The study addresses 242Pu accumulation by Elodea canadensis, one of the abundant species of submerged plants in the Yenisei River. 242Pu in water samples of the "Elodea - Yenisei River water" model system and in the biomass fractions was determined ?-spectrometrically, following radiochemical recovery of 242Pu using 236Pu - a chemical yield tracer. The experiments on accumulation of 242Pu by Elodea biomass showed that the activity concentration of 242Pu can reach 21 В± 2 Bq/g dry wt, with the concentration factor for 242Pu 13100 В± 2100 L/kg dry wt. Results of chemical fractionation proved that during the first few hours of the experiment 242Pu contained in Elodea was mainly concentrated in the exchangeable and the adsorbed fractions of biomass (about 100%). As Elodea biomass accumulated 242Pu, the absolute amount of 242Pu in the exchangeable and the adsorbed fractions remained almost unchanged, although the portion of 242Pu tightly bound to biomass increased. At the end of the experiment, on day 7, 242Pu tightly bound to biomass (fractions of organics and mineral residue) constituted 43-63% (in different experiments) of the total 242Pu in the biomass. Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.

Аннотация: The bacterial bioluminescence has high sensitivity to the action of various inhibitors of biological activity. The lyophilized luminous bacteria Photobacterium phosphoreum (Microbiosensor B17 677F) and luminous strain Escherichia coli (Microbiosensor EC) from the Culture Collection IBSO were used to create bioluminescent biotests. They have been applied in ecological monitoring to determine the overall toxicity of the Yenisei and Angara Rivers and some water sources of Altai Territory. As a rule the heaviest pollution of water in studied rivers was registered near cities and settlements. The luminous bacteria biotests are simple and convenient in work, standardized and quantitative, have rapid response to actions of different substances and high sensitivity to environmental pollutants. It takes less than 30 min to do the biotest (the other biotests take 48-96 h). Copyright В© 1999 John Wiley & Sons, Ltd.

Аннотация: Bioluminescence of isolated enzymes is a perspective phenomenon for biosensors development due to simplicity of registration of a physiological parameter - light intensity. Enzyme-based bioluminescent assay is widely used to evaluate a decrease in biochemical toxicities. Also the enzyme-based assay is used for the direct biochemical monitoring of oxidative toxicity. This work considers antioxidant properties of fullerenols, water-soluble polyhydroxylated derivatives of fullerenes and perspective pharmaceutical agents, in solutions of model inorganic and organic toxicants of oxidative type K-3[Fe(CN)(6)] and 1,4-benzoquinone. Two fullerenol preparations were used: C60O2-4(OH)(20-24) and mixture of two types of fullerenols C60O2-4(OH)(20-24)+C70O2-4(OH)(20-24). The enzyme-based assays showed the peculiarities of the detoxification processes: ultralow concentrations of fullerenols were active (ca 10(-17)-10(-5)g/L); no monotonic dependence of detoxification efficiency on fullerenol concentrations was observed, and detoxification of organic oxidizer solutions was more effective than that of the inorganic oxidizer. The antioxidant effects of highly diluted fullerenol solutions were attributed to hormesis phenomenon; the detoxification was concerned with stimulation of adaptive cellular response under low-dose exposures. Аннотация: This work considers antioxidant properties of natural detoxifying agents-humic substances (HS) in solutions of model inorganic and organic compounds of oxidative nature-complex salt K-3[Fe(CN)(6)] and 1,4-benzoquinone. Bioluminescent system of coupled enzymatic reactions catalyzed by NAD(P) H:FMN-oxidoreductase and bacterial luciferase was used as a bioassay in vitro to monitor toxicity of the oxidizer solutions. Toxicities of general and oxidative types were evaluated using bioluminescent kinetic parameters-bioluminescence intensity and induction period, respectively. Antioxidant activity of HS was attributed to their ability to decrease both general and oxidative toxicities; the HS antioxidant efficiency was characterized with detoxification coefficients D-GT and D-OxT, respectively. Dependencies of D-GT and D-OxT on HS concentration and time of preliminary incubation of the oxidizers with HS were demonstrated. The optimal conditions for detoxification of the oxidizers were 20-min incubation time and 0.5x10(-4) to 2x10(-4) M of HS concentration. The present study promotes application of the enzymatic luminescent bioassay to monitor toxicity of pollutants of oxidative nature in environmental and waste waters in remediation procedures.

Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.

Аннотация: A formula was derived for quantification of benefit-risk ratio (hazard quotient) for the intake of a product containing essential polyunsaturated fatty acids vs. heavy metals. The quotient was used in a three year case study of the contents of essential fatty acids and heavy metals in Siberian grayling, the main food fish from the middle section of the Yenisei River. As found, in general the fish intake was potentially very beneficial for human health, except on a few occasions, when the risk overweighed the benefit because of high contents of chromium in the fish muscle tissue. The data demonstrated the necessity for regular monitoring of the hazard quotients for food fish in wild conditions, based at least on monthly sampling frequency. В© 2008 Elsevier Ltd. Аннотация: In vitro experiments proved the usefulness of electron paramagnetic resonance (EPR) spectrometry for detecting detonation nanodiamonds (NDs) in samples of biomaterials (blood and homogenates of organs of mice). A characteristic EPR signal (g = 2.003, Delta H similar or equal to 10 G) was detected in biomaterials containing NDs, and its intensity linearly increased at nanoparticle concentrations of between 1.6 and 200 mu g/ml. In vivo experiments demonstrated that EPR spectrometry was effective for monitoring the inter-organ distribution of NDs intravenously injected to mice. In 2.5 h after the injection of NDs, the nanoparticles mainly accumulated in the lungs and liver of the animals - about 25 and 20%, respectively, of the initially injected NDs. The amounts of NDs accumulated in the heart and kidneys were considerably lower. Also, EPR spectrometry did not detect NDs in the blood, spleen, brain, and femoral muscles of mice. Ten days after injection, EPR spectrometry detected redistribution of NDs in mice. The amounts of nanoparticles decreased approximately by a factor of 3.5 in the lungs and increased almost by a factor of 3 in the liver; NDs were detected in the spleen. This study suggests ways to use EPR spectrometry to study the distribution, accumulation, and elimination of detonation NDs injected into laboratory animals.

Аннотация: The influence of a series of quinones and phenols on bacterial bioluminescence systems was investigated. Three bioluminescence systems used in ecological monitoring were compared: (1) water-soluble; (2) immobilized in starch gel coupled enzyme systems: NADH:FMN-oxidoreductase-luciferase; (3) luminescent bacteria. Bioluminescence inhibition constants of quinones and phenols and bioluminescence induction periods were compared. These kinetic parameters are proportional to quinone concentrations and depend on the quinone redox potential. Different effects of the substances are related to structure and properties of the bioluminescence systems. The set of bioluminescence assays for quinones and phenols monitoring should include two bioluminescence systems: 1 (or 2) and 3. (C) 2002 Elsevier Science (USA).
