Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.

Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, c-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physicalmechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers – polyhydroxyalkanoates (PHAs) – of two types – poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 ± 0.06–0.75 ± 0.11 ?m, while the average diameter of copolymer particles varied between 0.43 ± 0.12 and 0.55 ± 0.05 ?m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant. © 2021 Wiley Periodicals LLC.

Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

The investigated properties of the polymer (molecular weight, crystallinity, temperature characteristics) do not differ from those of the polymer synthesised on electrolytic hydrogen.

Аннотация: The paper presents a brief overview of the results of the implementation of the project "Agropreparations of the new generation: a strategy of construction and realization". The first part contains the analysis of the growth of the wild-type strain Cupriavidus necator B-10646 (formerly eutrophus) and the synthesis of polyhydroxyalkanoates by this strain on various substrates: glycerol, palm oil, Siberian oil seed, sunflower seed oils, and oleic acid. On refined glycerin, a highly productive process is implemented when scaling up, allowing to obtain 128 ± 11 g / L PHA. Evaluation of oils has shown that palm oil is the best carbon substrate. The second part presents the results of the development of environmentally friendly slow-release pesticide formulations. They are a degradable matrix of poly-3-hydroxybutyrate mixed with natural materials (peat, clay, wood flour), into which a pesticide (metribuzin, tribenuron-methyl, fenoxaprop-P-ethyl, azoxystrobin, epoxiconazole, and tebuconazole) has been. The developed preparations showed high activity against pathogenic fungi and weeds and had a much weaker negative effect on the soil microflora. Studies of the degradation of the developed preparations and the release of pesticides into the soil confirm their effectiveness over a long period of time, up to 90 days. © Published under licence by IOP Publishing Ltd.

Аннотация: Biodegradation of film specimens from polyhydroxyalkanoates (PHAs) of two types - poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-3- hydroxyvalerate (PHBV) - was analysed in different environments: tropical sea waters of the South China Sea (Nha Trang, Vietnam) and soils in the environs of Hanoi (Vietnam), Nha Trang (Vietnam) and Krasnoyarsk (Siberia, Russia). In seawater, the mass loss of the specimens of both types was almost equal. However, in tropical soils, PHB degraded quicker than PHBV. In the Siberian soil, the degradation rate of the PHBV was generally higher than that of PHBV. Analysis of molecular mass of PHA specimens showed its decreasing during biodegradation. In the tropical sea conditions, PHA degrading microorganisms were represented by bacteria of Enterobacter, Bacillus and Gracilibacillus genera. Among PHA degrading bacteria, Burkholderia, Alcaligenes, Bacillus, Mycobacterium and Streptomyces genera were identified in Vietnamese soils, and Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus and Xanthomonas genera in Siberian soils. Micromycetes of Gongronella, Paecilomyces, Penicillium and Trichoderma genera exhibited PHA degrading activity in Vietnamese soils, and Paecilomyces, Penicillium, Acremonium, Verticillium and Zygosporium genera - in Siberian soils. Аннотация: The biodegradation patterns of two types of PHA, a 3-hydroxybutyrate (3-PHB) polymer and a 3-hydroxybutyrate and 3-hydroxyvalerate (3-PHB/3-PHV) copolymer, were studied in tropical marine environments (Dam Bay, South China Sea, Nha Trang, Vietnam). No reliable differences in the degradation of 3-PHB and 3-PHB/3-PHV were revealed. It was shown that the degradation process depended mainly on the shape of a polymer product and its production method: the degradation of polymer films was found to be more active than that of molded solids. A decrease in the molecular mass of both types of PHA was detected in the course of the degradation of PHA samples. However, the degree of PHA crystallinity did not change; that is, the levels of degradation of both the amorphous and crystalline phases of PHA were almost the same. Among microbial PHA degraders, three bacterial strains, Bacillus sp. IBP-V002, Enterobacter cloacae sp. IBP-V001, and Gracilibacillus sp. IBP-V003, were identified based on the results of morphological, biochemical, and molecular phylogenetic analyses. The ability of the representatives of the genera Gracilibacillus and Enterobacter to degrade PHA was revealed for the first time. В© 2011 Pleiades Publishing, Ltd.

Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: Biodegradation of microbial linear polymers of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) by soil microbial communities of different structures has been studied during two field seasons in different weather conditions. This process was shown to be influenced by the polymer chemical composition, temperature, humidity, and the microbial soil component. The PHA degradation was accompanied by a decrease in the polymer molecular weight and an increase in the degree of crystallinity, indicating the preferential destruction of the amorphous phase compared to the crystalline one. The quantity of the true PHA destructors developing at the surface of the polymer samples was lower than the quantity of accompanying bacteria. The dominant PHA degrading microorganisms under the test conditions were identified as bacteria of the genera Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus, and Xanthomonas and as micromycetes from Penicillium, Paecilomyces, Acremonium, Verticillium, and Zygosporium.

Аннотация: This work is a review of the results of biomedical studies of polymer devices (films, fibers, microparticles, 30 implants) made from resorbable PHAs synthesized by the bacterium Wautersia (Ralstonia) eutropha 65786, using the technology developed at the Institute of Biophysics of the Siberian Branch of the Russian Academy of Sciences. Two types of PHAs - polyhydroxybutyrate (PHB) and a hydroxybutyrate/hydroxyvalerate copolymer (PHB/PHV) - have been proven to be biocompatible in vitro in cultures of fibroblasts, endothelial cells, hepatocytes, and osteoblasts, and in short- and long-duration experiments on animals. Polymer films and membranes have been found to be usable as scaffolds for functioning cells and monofilament suture fibers - for stitching muscular-fascial wounds and in abdominal surgery. Ectopic bone formation assay and experiments with the model of segmental osteotomy showed that 30 PHB and PHB/HA implants can be used for reparative osteogenesis. Аннотация: BACKGROUND: Polyhydroxyalkanoates (PHA) containing 3-hydroxy-4-methylvalerate (3H4MV) have better processing properties than other PHA types and were not prone to ageing. The main challenge in the production of these polymers is to simultaneously achieve high cell biomass, high polymer content, and high molar fraction of 3H4MV. The aim of this study was to investigate the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-3-hydroxy-4-methylvalerate) [P(3HB-co-3 HV-co-3H4MV)] by Cupriavidus eutrophus B-10646, synthesize polymers with different contents of 3H4MV and investigate their properties. RESULTS: It was shown that 4-methylvalerate (4MV, the precursor substrate) concentration in the culture medium must not be higher than 1 g L?1, and that 4MV must be carefully dosed, with at least 10–12 h intervals between supplementations. A series of polymers with molar fractions of 3H4MV between 2.7 and 11.3 mol% was synthesized. Degrees of crystallinity, molecular weight characteristics, and thermal properties of the polymers have been investigated as dependent on proportions of monomers. CONCLUSION: The content of 3H4MV depends on the concentration of 4MV and conditions of carbon nutrition. The molar fraction of 3H4MV had a strong effect on the crystalline to amorphous region ratio. Аннотация: Two chemolithoautotrophs - the hydrogen-oxidizing bacterium Alcaligenes eutrophus and the carboxydobacterium Seliberia carboxydohydrogena - were investigated with respect to their ability to produce heteropolymeric polyhydroxyalkanoates (PHA) during mixotrophic growth on CO2 and medium-chain-length (C6 to C9) alkanoic acids. Hydroxy derivatives of the alkanoic acids were incorporated into PHA synthesized by the bacteria studied in proportions dependent on the time of cultivation. Irrespective of the nature of the acid added to the medium, PHAs were found to contain hydroxybutyrate as the main constituent and also hydroxyvalerate and hydroxyhexanoate, whose maximal incorporation was observed 4 to 6 h after the addition of an alkanoic acid to the medium. During further cultivation, the fractions of hydroxyvalerate and hydroxyhexanoate in PHA decreased, and the fraction of hydroxybutyrate and the total intracellular content of the polymer increased. Three-component polyhydroxyalkanoates with different proportions of hydroxybutyrate, hydroxyvalerate, and hydroxyhexanoate were obtained, and some of their properties were analyzed. Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 85786), it has been first found that under mixotrophic growth conditions -CO2+ co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chain-length monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). A family of short- and medium-chain-length four- and five-component PHAs were synthesized and their physicochemical properties examined.
