Аннотация: The effects of medium viscosity on the decay rate of the 4a-hydroperoxyflavin intermediate of the bioluminescent reaction was investigated. It was found that at low concentrations of glycerol or sucrose (viscosity 1.1-1.3 cP) the decay rate rises, whereas a further increase in viscosity to 6.2 cP leads to a decrease in the decay rate following a power function with an exponent of 0.82-0.84. Using molecular dynamics methods, it was shown that the presence of glycerol and sucrose molecules causes a change in the mobility of the amino acid residues in the active center of luciferase, particularly those responsible for binding of flavin. The results obtained are indicative of two opposite effects of viscous media with glycerol and sucrose: (1) destabilization of 4a-hydroperoxyflavin due to a change in the structural and dynamic properties of the protein and (2) stabilization of this intermediate by the decrease in the diffusion rate of its decay products.

Аннотация: The contents of isotopes of plutonium (Pu-238 and(239, 240)Pu) and(137)Cs in samples of bottom sediments and hydrobionts (water moss, amphipods, and caddisfly larvae with casings) taken in the Yenisei River in the vicinity of the radioactive discharge from the Mining and Chemical Combine in 2012 and 2018 were comparatively estimated. It is shown that the content and ratios between the specific activities of plutonium isotopes (238/239 and 240) in samples of BSs and hydrobionts increased after the recommencing of plutonium discharges into the Yenisei due to the beginning of MOX fuel production. The background content and the ratio between plutonium isotopes in BSs of the Yenisei were estimated for the first time. Hydrobionts and BSs were ranged differently according to the content of plutonium and(137)Cs: we recorded the highest content of(137)Cs in BSs and that of plutonium in water moss. The plutonium content in hydrobionts of the Yenisei River varies considerably, which permits specification of representatives of biota (water moss and caddisfly larvae with casings) that, along with BSs, can be used as effective indicators of ecosystem pollution with plutonium.

Аннотация: An anomalous layer enriched with chemical elements indicating the presence of terrigenous matter was discovered in the sediment core of Zapovednoe Lake located 60 km from the epicenter of the Tunguska event (1908) using synchrotron radiation X-ray fluorescence spectroscopy (SR-XRF). Radioisotope measurements indicate that the age of the layer is consistent with the date of the catastrophe. Apparently, the anomalous layer was formed as a result of an intense terrigenous matter inflow from the water catchment area due to massive forest falls and subsequent wildfires caused by the Tunguska event. Thus, it is established that targeted searches for microparticles of extraterrestrial origin can be carried out in the discovered and dated anomalous bottom sediment layer.

Аннотация: The results of experiments on application of a newly developed facility for oxidation of volatile organic compounds on a platinum catalyst are presented. The feasibility of using this method in artificial ecosystems as a whole and in mass exchange of closed biological-technical life support systems in particular is shown. The possibility of deep purification of gas emitted from the reactor of physical-chemical processing of organic wastes is demonstrated. Wheat growing experiment on using the facility for oxidation of volatile organic compounds in a sealed chamber was performed. No adverse effect of probable toxic oxidation products on wheat plants during a 4-day experiment was determined.

Аннотация: Calcium-regulated photoproteins are found in at least five phyla of organisms. The light emitted by those photoproteins can be tuned by mutating the photoprotein and/or by modifying the substrate coelenterazine (CTZ). Thirty years ago, Shimomura observed that the luminescence activity of aequorin was dramatically reduced when the substrate CTZ was replaced by its analogv-CTZ. The latter is formed by adding a phenyl ring to the pi-conjugated moiety of CTZ. The decrease in luminescence activity has not been understood until now. In this paper, through combined quantum mechanics and molecular mechanics calculations as well as molecular dynamics simulations, we discovered the reason for this observation. Modification of the substrate changes the conformation of nearby aromatic residues and enhances the pi-pi stacking interactions between the conjugated moiety ofv-CTZ and the residues, which weakens the charge transfer to form light emitter and leads to a lower luminescence activity. The microenvironments of CTZ in obelin and in aequorin are very similar, so we predicted that the luminescence activity of obelin will also dramatically decrease when CTZ is replaced byv-CTZ. This prediction has received strong evidence from currently theoretical calculations and has been verified by experiments.

Аннотация: Background Canine parvovirus (CPV) is now recognized as a serious threat to the dog breeding industry worldwide. Currently used CPV vaccines all have their specific drawbacks, prompting a search for alternative safe and effective vaccination strategies such as subunit vaccine. VP2 protein is the major antigen targeted for developing CPV subunit vaccine, however, its production in baculovirus expression system remains challenging due to the insufficient yield. Therefore, our study aims to increase the VP2 protein production by using an improved baculovirus expression system and to evaluate the immunogenicity of the purified VP2 protein in mice. Results The results showed that high-level expression of the full length VP2 protein was achieved using our modified baculovirus expression system. The recombinant virus carrying two copies of VP2 gene showed the highest expression level, with a productivity of 186 mg/L, which is about 1.4-1.6 fold that of the recombinant viruses carrying only one copy. The purified protein reacted with Mouse anti-His tag monoclonal antibody and Rabbit anti-VP2 polyclonal antibody. BALB/c mice were intramuscularly immunized with purified VP2 protein twice at 2 week intervals. After vaccination, VP2 protein could induce the mice produce high level of hemagglutination inhibition antibodies. Conclusions Full length CPV VP2 protein was expressed at high level and purified efficiently. Moreover, it stimulated mice to produce high level of antibodies with hemmaglutination inhibition properties. The VP2 protein expressed in this study could be used as a putative economic and efficient subunit vaccine against CPV infection.

Аннотация: We compared the composition and content of fatty acids (FAs) in the liver, muscles, and subcutaneous fat of Yakut horses inhabiting extreme environment in the Cryolithozone. Essential linoleic and alpha-linolenic acids, supplied to horses with their food, were accumulated in different tissues. Linoleic acid was accumulated in the liver but alpha-linolenic acid was accumulated in muscle and subcutaneous fat. Such a distribution indicates different roles of these fatty acids in the metabolism of horses. Yakut horse meat is a valuable dietary product owing to its fatty acid composition and content.

Аннотация: Ca2+-regulated photoproteins responsible for bioluminescence of a variety of marine organisms are single-chain globular proteins within the inner cavity of which the oxygenated coelenterazine, 2-hydroperoxycoelenterazine, is tightly bound. Alongside with native coelenterazine, photoproteins can also use its synthetic analogues as substrates to produce flash-type bioluminescence. However, information on the effect of modifications of various groups of coelenterazine and amino acid environment of the protein active site on the bioluminescent properties of the corresponding semi-synthetic photoproteins is fragmentary and often controversial. In this paper, we investigated the specific bioluminescence activity, light emission spectra, stopped-flow kinetics and sensitivity to calcium of the semi-synthetic aequorins and obelins activated by novel coelenterazine analogues and the recently reported coelenterazine derivatives. Several semi-synthetic photoproteins activated by the studied coelenterazine analogues displayed sufficient bioluminescence activities accompanied by various changes in the spectral and kinetic properties as well as in calcium sensitivity. The poor activity of certain semi-synthetic photoproteins might be attributed to instability of some coelenterazine analogues in solution and low efficiency of 2-hydroperoxy adduct formation. In most cases, semi-synthetic obelins and aequorins displayed different properties upon being activated by the same coelenterazine analogue. The results indicated that the OH-group at the C-6 phenyl ring of coelenterazine is important for the photoprotein bioluminescence and that the hydrogen-bond network around the substituent in position 6 of the imidazopyrazinone core could be the reason of different bioluminescence activities of aequorin and obelin with certain coelenterazine analogues.

Аннотация: Bioluminescent proteins are widely used as reporter molecules in various in vitro and in vivo assays. The smallest isoform of Metridia luciferase (MLuc7) is a highly active, naturally secreted enzyme which, along with other luciferase isoforms, is responsible for the bright bioluminescence of marine copepodMetridia longa. In this study, we report the construction of two variants of a hybrid protein consisting of MLuc7 and 14D5a single-chain antibody to the surface glycoprotein E of tick-borne encephalitis virus as a model fusion partner. We demonstrate that, whereas fusion of a single-chain antibody to either N- or C-terminus of MLuc7 does not affect its bioluminescence properties, the binding site on the single-chain antibody influences its binding capacity. The affinity of 14D5a-MLuc7 hybrid protein (K-D= 36.2 nM) where the C-terminus of the single-chain antibody was fused to the N-terminus of MLuc7, appeared to be 2.5-fold higher than that of the reverse, MLuc7-14D5a (K-D= 87.6 nM). The detection limit of 14D5a-MLuc7 hybrid protein was estimated to be 45 pg of the recombinant glycoprotein E. Although the smallest isoform ofM. longaluciferase was tested as a fusion partner only with a single-chain antibody, it is reasonable to suppose that MLuc7 can also be successfully used as a partner for genetic fusion with other proteins.

Аннотация: The present study addresses the herbicidal activity and biological effects of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides used to control various weed species (Amaranthus retroflexus, Sinapis arvensis,andLeucanthemum maximum). The effects of the free herbicides and the herbicides embedded in granules of degradable polymer poly-3-hydroxybutyrate [P(3HB)] blended with birch wood flour were compared. Metribuzin, regardless of the form, caused 100% mortality of the three weeds by day 21. The herbicidal activity of tribenuron-methyl was lower than that of metribuzin, but the embedded TBM was superior to the free herbicide in the length and strength of its action on the weeds. Both metribuzin forms dramatically decreased the main parameters of fluorescence: maximum quantum yield of photosystem-II [Y(II)(max)], maximum quantum yield of non-photochemical quenching [Y(NPQ)(max)], and maximum rate of non-cyclic electron transport [ETRmax] and concentrations of chlorophyllaandb. The effect of the embedded TBM on the photosynthetic activity of the weeds was lower in the first two weeks of the growth of herbicide-treated plants but lasted longer than the effect of the free TBM and increased over time. Embedding of metribuzin in the matrix of degradable blend did not decrease its herbicidal activity.

The inhibitory effect of the straw subjected to physicochemical oxidation increased with a decrease in the degree to which the straw had been oxidized. Only supplementation with the straw mineralized to the highest possible degree did not inhibit plant growth and development, and the crop yield in that treatment was higher than in the other treatments.

Аннотация: Many Cladocera species under unfavourable conditions produce resting eggs that can survive for years in deep diapause. Resting eggs form egg banks at the bottom sediments, which serve as a source of genetic diversity and replenish the population after periods of decline. Despite the obvious importance of resting eggs for the ecosystem functioning, studies assessing the sensitivity of resting eggs to different toxicants are scarce. We reviewed published data on the sensitivity of resting eggs to the effect of heavy metals, organic pollutants and ionizing radiation. Analysis shows that the effects of toxicants of different types on resting eggs will have different environmental consequences. Egg banks may suffer from prolonged contact of dormant eggs with heavy metals. However, the ecological relevance of these effects is low, since the effective concentrations of toxicants must be very high. In addition, the effect of heavy metals on resting eggs is not transmitted to hatchlings from exposed eggs. Taking into account high toxicity of heavy metals to active animals, we assume that the toxic effect of heavy metals is critical for active zooplankters and relatively safe for resting stages. Accumulation of artificial radionuclides in bottom sediments can have a significant impact on aquatic ecosystems through chronic effects both on survival of resting eggs and on the life cycle parameters of animals hatched from irradiated eggs. Resting eggs during reactivation are more sensitive to the effect of ionizing radiation. Pesticides and complex chemical compounds produce similar toxic effects on both resting eggs and life history parameters of hatchlings from exposed resting eggs. The bottom line is that to predict the effect of contamination of bottom sediments by different pollutants on the ecosystem structure and functioning, it is highly important to investigate the viability of the resting eggs under a wide range of concentrations of various toxicants and different durations of direct contact of eggs with toxicants.

Аннотация: Production of resting eggs in zooplankton is controlled by multiple stimuli. In this chapter, we briefly discussed published data that confirm the effect of infochemicals produced by conspecifics, competitors, predators or preys on the production of resting eggs in zooplankton. We found that the effect of conspecific chemicals on the production of resting eggs is the most convincing. Both experimental data and theoretical research demonstrated that this density-dependent reaction often results in a competitive advantage of individuals in the population that follows such a strategy. The data on the effect of chemicals exuded by competitors or predators are controversial. Data on the effect of chemical interaction on the production of resting eggs in natural habitats are almost absent. Most of the studies of chemical interactions are performed with individuals in laboratory experiments with crowded water. Crowded water is water that contains chemicals exuded by the population. Even though this method has the number of drawbacks, it is still widely used in similar studies. There are several studies focused on the identification of the chemical nature of cues responsible for the production of resting eggs in zooplankton. Most probably, chemicals involved are short proteins. However, the exact identification of the chemicals responsible for the production of resting eggs in zooplankton remains an open task. In order to place chemical interactions into a framework of multiple diapause control theory, it is necessary to determine the nature of chemicals involved and to demonstrate population- and ecosystem-level consequences of this phenomenon in natural habitats.

Аннотация: In the present investigation, the role of dietary intervention in male Wistar rats (n = 8, 3 groups) was studied to observe absolute sirtuin 1 (SIRT1) levels (expressed as ng mg(-1) total protein) in serum, stomach, liver, and kidney. Dietary buckwheat at 30% (w/w) level of incorporation in the standard diet (Buckwheat Enriched Diet, BED) improved SIRT1 with values 0.933 +/- 0.05, 210 +/- 7, 63.26 +/- 4, and 69.89 +/- 3 in serum, stomach, liver, and kidney respectively when compared to the respective control values of 0.536 +/- 0.03, 156 +/- 23.3, 31.07 +/- 2 and 47.11 +/- 4. Moreover, BED though isocaloric to CR diet, led to weight gain (g) by 63.11 +/- 3.8, ca. 10%, and 40% higher than control (56.27 +/- 5.6) and CR (45.05 +/- 4.1) diet groups. A marked rise in Feed Efficiency Ratio (FER) by ca. 37% while a 30% decrease in Feed Conversion Ratio (FCR) was observed for the BED group which supports unexpected weight gain in rats post-dietary intervention. The results justify the superior nutritional profile of buckwheat laden with essential nutrients, essential proteins, and bioactives. In contrast, Calorie Restriction (CR) resulted in a decline of the total protein content in circulation by 19%, while reduction of total protein in stomach, liver, and kidney was estimated to be 95%, 35.2%, and 27% respectively though SIRT1 values were comparatively the highest in all the samples studied. A 30-fold enhancement of SIRT1 in stomach post CR is presumed to counter enhanced stress in gastric tissues. Therefore, mild to moderate expression of SIRT1 may confer beneficial effects such as delayed aging and stress resistance but exceedingly high SIRT1 may evoke increased oxidative stress.

Аннотация: Using the original technique of treating biomass with beta-glucosidase, a pool of extracellular fungal enzymes was obtained for the first time from the mycelium of basidiomycete Neonothopanus nambi. Two protein fractions containing enzymes with oxidase activity were isolated from the extract by gel-filtration chromatography and conventionally called F1 and F2. Enzyme F1 has a native molecular weight of 80-85 kDa and does not contain chromophore components; however, it catalyzes the oxidation of veratryl alcohol with K-m = 0.52 mM. Probably, this enzyme is an alcohol oxidase. Enzyme F2 with a native molecular weight of approximately 60 kDa is a FAD-containing protein. It catalyzes the cooxidation of phenol with 4-aminoantipyrine without the addition of exogenous hydrogen peroxide, which distinguishes it from the known peroxidases. It was assumed that this enzyme may be a mixed-function oxidase. F2 oxidase has K-m value 0.27 mM for phenol. The temperature optimums for oxidases F1 and F2 are 22-35 and 55-70 degrees C, and pH optimums are 6 and 5, respectively.

Аннотация: Long-chain omega-3 polyunsaturated fatty acids (LC-PUFA) essential for human nutrition are mostly obtained from wild-caught fish. To sustain the LC-PUFA supply from natural populations, one needs to know how environmental and intrinsic factors affect fish fatty acid (FA) profiles and contents. We studied seven Salmoniformes species from two arctic lakes. We aimed to estimate differences in the FA composition of total lipids and two major lipid classes, polar lipids (PL) and triacylglycerols (TAG), among the species and to evaluate LC-PUFA contents corresponding to PL and TAG in muscles. Fatty acid profiles of PL and TAG in all species were characterized by the prevalence of omega-3 LC-PUFA and C16-C18 monoenoic FA, respectively. Fish with similar feeding spectra were identified similarly in multivariate analyses of total lipids, TAG and PL, due to differences in levels of mostly the same FA. Thus, the suitability of both TAG and total lipids for the identification of the feeding spectra of fish was confirmed. All species had similar content of LC-PUFA esterified as PL, 1.9-3.5 mg g(-1), while the content of the TAG form strongly varied, from 0.9 to 9.8 mg g(-1). The LC-PUFA-rich fish species accumulated these valuable compounds predominately in the TAG form.

Аннотация: This article presents investigation data on the seasonal dynamics of productivity, status, and species composition of vegetation on fallow lands in the Krasnoyarsk forest steppe (Middle Siberia) obtained from terrain and satellite materials from 2017. The results of the study of grass plant communities on the basis of geobotanical descriptions and terrain spectrometry were have been used for a more accurate interpretation of cosmic photographs of moderate and low resolution. For studying vegetation on fallow lands, we analyze the seasonal dynamics of various vegetative indices (NDVI, EVI, LSWI, and LAI) and parameters (NPP, FPAR, and LST (land surface temperature)) obtained from MODIS satellite images. Our analysis of satellite data shows the absence of evidences of plowing and mowing in the studied area. A positive correlation is revealed between vegetation indices of biomass (NDVI, EVI, LAI, and NPP) and parameters of hydrothermal conditions (LSWI, FPAR, and LST).

Аннотация: Autoluminescent plants engineered to express a bacterial bioluminescence gene cluster in plastids have not been widely adopted because of low light output. We engineered tobacco plants with a fungal bioluminescence system that converts caffeic acid (present in all plants) into luciferin and report self-sustained luminescence that is visible to the naked eye. Our findings could underpin development of a suite of imaging tools for plants.

Аннотация: Light-sensitive Ca2+-regulated photoprotein berovin is responsible for the bioluminescence of the ctenophore Beroe abyssicola. It shares many properties of hydromedusan photoproteins although the degree of identity of its amino acid sequence with those of photoproteins is low. There is a hydrogen bond between C-terminal Pro and Arg situated in the N-terminal alpha-helix of hydromedusan photoproteins that supports a closed conformation of the internal cavity of the photoprotein molecule with bound 2-hydroperoxycoelenterazine. The C- and N-terminal hydrogen bond network is necessary to properly isolate the photoprotein active site from the solvent and consequently to provide a high quantum yield of the bioluminescence reaction. In order to find out which berovin residues perform the same function we modified the N- and C-termini of the protein by replacing or deleting various amino acid residues. The studies on berovin mutants showed that the interaction between C-terminal Tyr208 and Tyr13 localized in the first alpha-helix of the photoprotein is important for the stabilization and proper orientation of the oxygenated coelenterazine adduct within the internal cavity as well as for supporting the closed photoprotein conformation. We also suggest that the interplay between Tyr residues in ctenophore photoproteins occurs rather through the pi-pi interaction of their phenyl rings than through hydrogen bonds as in hydromedusan photoproteins.
