Аннотация: A psychrotolerant, methylotrophic methanogen, strain YSF-03T, was isolated from the saline meromictic Lake Shira in Siberia. Cells of strain YSF-03T were non-motile, irregular cocci and 0.8–1.2?m in diameter. The methanogenic substrates utilized by strain YSF-03T were methanol and trimethylamine. The temperature range of growth for strain YSF-03T was from 0 to 37 °C. The optimum growth conditions were 30–37 °C, pH 7.0–7.4 and 0.17M NaCl. The G+C content of the genome of strain YSF-03T was 41.3 mol%. Phylogenetic analysis revealed that strain YSF-03T was most closely related to Methanolobus profundi MobMT (98.15% similarity in 16S rRNA gene sequence). Genome relatedness between strain YSF-03T and MobMT was computed using the Genome-to-Genome Distance Calculator and average nucleotide identity, which gave values of 23.5 and 79.3 %, respectively. Based on the morphological, phenotypic, phylogenetic and genomic relatedness data presented here, it is evident that strain YSF-03T represents a novel species of the genus Methanolobus, for which the name Methanolobus psychrotolerans sp. nov. is proposed. The type strain is YSF-03T (=BCRC AR10049T=DSM 104044T=NBRC 112514T). © 2018 IUMS.

Аннотация: We studied structure, dynamics and functional (biogeochemical) activity of microbial complexes of cryogenic soils in larch forests in Central Evenkia and polygonal tundra on Samoilovskii Island, Lena Delta. We found that daily flux of methane from soil surface is 3–5 times less in forest soil than in the center of polygon in tundra. Short-term heating to 18.5–22.5 °C of permafrost-affected soil in larch forest caused sweetening of soil solution, shrinkage of eco-trophic groups of microorganisms and microbial biomass, as well as increase in greenhouse gases (CO2 and CH4) emission to the air. Notably the permafrost-affected soil on sandy deposits in tundra had highest microbial diversity of methanogenic archaea including Methanobacteriaceae, Methanomicrobiaceae, Methanosarcinaceae, Methanosaetaceae families. On the other hand only Methanosarcinacea were found in cryosols of larch forest. Both type I and type II methanotrophs were found in the forest soil, while only type II methanotrophs occurred in tundra soil. © 2017, Izdatel’stvo Nauka. All rights reserved.

Аннотация: The results of a study of the specific features of morphological and genetic polymorphism of the population of the anadromous White Sea common whitefish (Coregonus lavaretus L.) from the Keret' River are presented. As assessed by its morphological features, the whitefish population of the Keret' is homogenous. A comparison of the Keret' whitefish with whitefish populations of both Europe and Siberia revealed clear differences. These differences relate first and foremost to the fish locomotion and orientation in space: body depth, length and positions of fins, and interorbital width. The characteristics of skull features (craniological analysis) of the White Sea whitefish are presented for the first time. The results of an analysis of the polymorphism of the ND1 fragment of the mitochondrial DNA confirmed an earlier suggestion about the mixed origin of the Keret' River whitefish: the descendants of three different phylogenetic lineages were revealed in the population.

Аннотация: The seasonal dynamics of the vertical structure of small saline Lake Uchum, located in the steppe arid zone of the south of Siberia (Krasnoyarsk krai), has been studied in detail for the first time. This lake is a meromictic water body. We have revealed a heterogeneous vertical distribution of plankton organisms and a dense population of purple sulfuric bacteria in the redox zone. The taxonomic composition and seasonal dynamics of phyto- and zooplankton are described. Presumably, the meromixis of Lake Uchum is due to the inflow of fresh water to the surface of the saline water body during the rise of its level in the early 20th century, similarly to lakes Shira and Shunet located nearby. The processes of salt displacement into the solution during the formation of ice, as well as the precipitation of salts in the winter, also contribute to the maintenance of permanent stratification. The information on the current state of the lake can be useful for reconstructing the climate by bottom sediments, as well as for creating a model of water quality and investigating the therapeutic properties of lake mud.

Аннотация: We estimated the age and sedimentation rate of bottom sediments in Cheko Lake located in southern Evenkia, in the territory of Tunguska Nature Reserve, near the supposed epicenter of the so-called 1908 Tunguska Event. The vertical distributions of Cs-137 and Pb-210 activity and visually counted varves in the core of lake bottom sediments indicate that Cheko Lake is significantly older than the 1908 Tunguska Event; therefore, the lake basin cannot be a crater or a trace of the explosion as was supposed earlier by some researchers.

Аннотация: The in-situ data on the vertical structure and stability of the vertical stratification of saline Lake Shira over the past decade (2007–2015) are analyzed. Simplified mathematical models have shown that strong wind in the autumn of 2014 together with rather thick ice in the winter of 2015 caused a change in the circulation regime of this water reservoir from meromictic (incomplete mixing) to holomictic (compete mixing). Based on the results obtained, a circulation regime for deep saline lakes located in the continental climate zone, in particular, in the arid zones of Southern Siberia (Khakassia, Transbaikal, and Altai) can be predicted under various climate scenarios of the future. © 2017, Pleiades Publishing, Ltd.

Аннотация: Even though bioluminescent oligochaetes rarely catch people's eyes due to their secretive lifestyle, glowing earthworms sighting reports have come from different areas on all continents except Antarctica. A major breakthrough in the research of earthworm bioluminescence occurred in the 1960s with the studies of the North American Diplocardia longa. Comparative studies conducted on 13 earthworm species belonging to six genera showed that N-isovaleryl-3-aminopropanal (Diplocardia luciferin) is the common substrate for bioluminescence in all examined species, while luciferases appeared to be responsible for the color of bioluminescence. The second momentous change in the situation has occurred with the discovery in Siberia (Russia) of two unknown luminous enchytraeids. The two bioluminescent systems belong to different types, have different spectral characteristics and localization, and different temperature and pH optima. They are unique, and this fact is confirmed by the negative results of all possible cross-reactions. The bioluminescent system of Henlea sp. comprises four essential components: luciferase, luciferin, oxygen and calcium ion. For Friderica heliota, the luminescent reaction requires five components: luciferase, luciferin, ATP, magnesium ion and oxygen. Along with luciferin, more than a dozen analogues were isolated from worm biomass. These novel peptide-like natural compounds represent an unprecedented chemistry found in terrestrial organisms. © 2017 The American Society of Photobiology

Аннотация: Saline Lake Shira (Southern Siberia, Russia) was meromictic through the observation period 2002-2015. During the under-ice periods of 2015 and 2016, complete mixing of the water column was recorded for the first time, and hydrogen sulphide temporarily disappeared from the water column of the lake; i.e. in those years the lake turned to holomixis. In the summer of 2015, a sharp increase in chlorophyll a, organic carbon, zooplankton, and phytoflagellates was observed in the lake, which was probably due to the release of nutrients from the monimolimnion. Purple sulfur bacteria completely disappeared from the lake after the first mixing in 2015, and did not reappear despite the restoration of meromixis in 2017. Thus, it was demonstrated that purple sulfur bacteria are sensitive to the weakening of the stratification of Lake Shira. Based on the data of the seasonal monitoring of temperature and salinity profiles over the period 2002-2017, it was presumed that the main cause of deep mixing in 2015 was the weakening of the salinity gradient due to strong wind impact and early ice retreat in the spring of 2014. In addition, it was shown that in previous years a significant contribution to the maintenance of meromixis was made by an additional influx of fresh water, which caused a rise in the lake level in the period 2002-2007. Thus, we identified a relationship between the stratification regime of the lake and the change in its level, which provides valuable information both for the forecast of Water quality and for reconstruction of the Holocene climate humidity in this region of Southern Siberia from the sediment cores of Lake Shira.

Аннотация: The small synanthropic and peregrine earthworm Microscolex phosphoreus (Duges, 1837) is reported for the first time from Siberia. Morphological and DNA barcode (COI) analyses of this and widely separate samples worldwide demonstrate that, as currently identified, M. phosphoreus is a heterogeneous taxon, with divergent lineages occurring often in the same locality and hardly providing geographically structured genetic signals. The combined morphological and genetic evidence suggests that at least four of the found clades should be reclassified as separate species, both morphologically and genetically distinct from each other. However, as the specimen number was limited and only the COI gene was studied for the genetic work, we hesitate in formally describing new species. There would also be the problem of assigning the available names to specific lineages. Our findings encourage careful external and anatomical examination and using reliable characters such as the interchaetal distances and spermathecal morphology for correct identification and for deeper evaluation of cryptic diversity in this interesting bioluminescent worm.

Аннотация: The previously undescribed dynamics of the heat shock protein HSP70 and subsequent lipid peroxidation products have been assessed alongside lactate dehydrogenase activity for Gammarus lacustris Sars, an amphipod species from the saltwater Lake Shira (Republic of Khakassia). Individuals were exposed to a gradual temperature increase of 1 degrees C/hour (total exposure duration of 26 hours) starting from the mean annual temperature of their habitat (7 degrees C) up to 33 degrees C. A complex of biochemical reactions occurred when saltwater G. lactustris was exposed to the gradual changes in temperature. This was characterized by a decrease in lactate dehydrogenase activity and the launching of lipid peroxidation. The HSP70 level did not change significantly during the entire experiment. In agreement with the concept of oxygen-limited thermal tolerance, an accumulation of the most toxic lipid peroxides (triene conjugates and Schiff bases) in phospholipids occurred at the same time and temperature as the accumulation of lactate. The main criterion overriding the temperature threshold was, therefore, the transition to anaerobiosis, confirmed by the elevated lactate levels as observed in our previous associated study, and by the development of cellular stress, which was expressed by an accumulation of lipid peroxidation products. An earlier hypothesis, based on freshwater individuals of the same species, has been confirmed whereby the increased thermotolerance of G. lacustris from the saltwater lake was caused by differences in energy metabolism and energy supply of nonspecific cellular stress-response mechanisms. With the development of global climate change, these reactions could be advantageous for saltwater G. lacustris. The studied biochemical reactions can be used as biomarkers for the stress status of aquatic organisms when their habitat temperature changes.

Аннотация: The present study proves the presence of seasonal variability of LWRs of Arctic grayling and Siberian dace inhabiting the middle reaches of the Yenisei River. LWRs were estimated using total length (cm) and total body weight (g). The literature data on LWRs of dace and grayling from different regions were compared using log a over b plot. The comparison revealed that seasonal variability of LWRs was wider than the geographic variability for these species. The position of points on the plot can be used to predict the season of sample collection and vice versa.

Аннотация: The large, deep, freshwater Norilsk lakes (Lake Lama, Lake Sobachye, Lake Glubokoe and Lake Keta) are located beyond the Arctic Circle, at the northwestern edge of the Putorana Plateau. They underwent a Pleistocene boreal marine transgression. A relict of that transgression, Limnocalanus macrurus Sars 1863 (Copepoda, Calanoida), presently dominates the crustacean zooplankton of these lakes. Sympatric with the native relict species L. macrurus, Senecella siberica Vyshkvartzeva 1994, was found in our zooplankton samples taken from the deep parts of lakes Lama and Sobachye in the summer-autumn of 2014-2016. Sampling was performed using a Juday net hauled vertically from a depth of 15, 20(25/30) and/or 50 m to the surface. All Senecella specimens were older copepodites, mainly CIV stage in Lama and Sobachye lakes in August and CV in Sobachye Lake in September; their abundance was low (25-2000 ind. m(-2)). They inhabited the central deeper regions of the lakes, contributing up to 7-14% to the total wet biomass of net zooplankton in 0-20 and 0-50 m strata. Previous sporadic and rather superficial samplings in these lakes missed Senecella due to its deep locations, low abundance levels and the absence of both the oldest copepodites and adults in summer.

Аннотация: Microorganisms are critical to maintaining stratified biogeochemical characteristics in meromictic lakes; however, their community composition and potential roles in nutrient cycling are not thoroughly described. Both metagenomics and metaviromics were used to determine the composition and capacity of archaea, bacteria, and viruses along the water column in the landlocked meromictic Lake Shunet in Siberia. Deep sequencing of 265 Gb and high-quality assembly revealed a near-complete genome corresponding to Nonlabens sp. sh3vir. in a viral sample and 38 bacterial bins (0.2-5.3 Mb each). The mixolimnion (3.0 m) had the most diverse archaeal, bacterial, and viral communities, followed by the monimolimnion (5.5 m) and chemocline (5.0 m). The bacterial and archaeal communities were dominated by Thiocapsa and Methanococcoides, respectively, whereas the viral community was dominated by Siphoviridae. The archaeal and bacterial assemblages and the associated energy metabolism were significantly related to the various depths, in accordance with the stratification of physicochemical parameters. Reconstructed elemental nutrient cycles of the three layers were interconnected, including co-occurrence of denitrification and nitrogen fixation in each layer and involved unique processes due to specific biogeochemical properties at the respective depths. According to the gene annotation, several pre-dominant yet unknown and uncultured bacteria also play potentially important roles in nutrient cycling. Reciprocal BLAST analysis revealed that the viruses were specific to the host archaea and bacteria in the mixolimnion. This study provides insights into the bacterial, archaeal, and viral assemblages and the corresponding capacity potentials in Lake Shunet, one of the three meromictic lakes in central Asia. Lake Shunet was determined to harbor specific and diverse viral, bacterial, and archaeal communities that intimately interacted, revealing patterns shaped by indigenous physicochemical parameters.

Аннотация: SNP genotyping based on the reaction of specific primer extension with the following bioluminescent detection of its products was shown to be potentially applicable for biomedical exploration. The paper describes its elaboration and first application in extensive biomedical research concerning MC1R gene variants’ frequency and associations with clinical characteristics in melanoma patients of Eastern Siberia (Krasnoyarsk region, Russia). Polymorphisms rs 1805007 (R151C), rs 1805008 (R160W), and rs 1805009 (D294H) were detected in 174 DNA samples from patients with histologically proved diagnosis of cutaneous melanoma and in 200 samples from healthy individuals. All the results on bioluminescent SNP genotyping were confirmed by Sanger sequencing. Some features characteristic of the population were found, i.e. melanoma is mostly associated with R160W or R151C while variant D294H is extremely rare; simultaneous carriage of any two investigated variants is also strongly associated with melanoma; R151C is associated with ulceration and consequently the disease course is more aggressive, etc. The design of the technique allows fast evaluation of any known diagnostically important SNP frequencies and associations across population. © 2018 Elsevier B.V.

Аннотация: This paper elucidates the features of colonization by whitefish (Coregonus lavaretus L.) of waterbodies of the European part of Russia in relation to Pleistocene glaciation. Based on the data on the frequencies and phylogenetic relationships of mitochondrial DNA haplotypes, a significant role of migrants from the western periglacial refugia is shown. The first large refugium was located at the North Atlantic; the second was at the South-Eastern Baltic. Until the last glaciation, with the maximum at about 20000 years ago, migrations from Siberia took place as well. However, at present the descendants of these migrants in the waterbodies of Northwestern Russia are not the dominants. The size of the refugium where the population lived prior to expansion may be an important condition for the successful spreading of common whitefish.

Аннотация: Contents of highly unsaturated long-chain fatty acids of omega-3 family (HUFA); eicosapentaenoic acid (EPA; 20:5n-3); and docosahexaenoic acid (DHA; 22:6n-3) are the principal indicators of the nutritive quality of fish for humans. Effects of environmental factors, first of all water temperature, on EPA and DHA contents in fish tissue are not currently completely understood. To reveal the putative effect of water temperature, fatty acid composition and contents were studied for 6 fish species, inhabiting the cold waters of the Yenisei River (Siberia, Russia) downstream of a dam (hypolimnetic release from reservoir) and its adjacent warm water tributaries (the Mana River and the Kacha River). It was hypothesized that (a) fish species from the cold river would have higher HUFA contents than fish from the warm rivers and (b) temperature would be negatively correlated with HUFA content in fish species. Using gas chromatography-mass spectrometry, distinct species-specific fatty acid profiles were observed, whereas contents of the essential fatty acids, EPA and DHA, in fish species from the cold Yenisei River were in general similar to that from warm tributaries. Thus, in contrast to the first hypothesis, phylogenetic factors overweighed the effect of water temperature and food (benthic invertebrate) composition, on fatty acid composition and HUFA content in fish. For the second hypothesis, for the 2 species inhabiting both cold and warm rivers, only one had higher EPA and DHA content at lower temperatures. Consequently, the response of EPA and DHA content in fish tissue to temperature variations may be species-specific.

Аннотация: The model of forest stand growth proposed in this study is based on R. Solow's model of economic growth. The variables introduced into the model are the "capital" (the phytomass of the non-synthesizing tree components in the stand-the stem, roots, and branches) and the "labor" (the phytomass of the photosynthesizing tree components in the stand-leaves or needles). Root phytomass is calculated with a special independent model. The process of energy production by the trees is described with the Cobb-Douglas equation. The proposed approach is used to describe growth processes in the forest stands comprising various species in Siberia and the age dynamics of net primary production. The model can explain a number of effects (such as death of the forest stand after the needles have been consumed by defoliating insects) that cannot be explained by standard logistic models. © 2018 by the authors.

Аннотация: The purpose of this study is to find a relationship between changes in spectral characteristics (MODIS/Terra) of mountain forests and the altitude at which they grow. In the study area, which is located in the West Sayan Mountains (in South Siberia), the types of forest ecosystems change markedly with altitude. The study uses the data of the MODIS-NPP model intended for the evaluation of global net production. Results of the study show that the best approach to dividing mountain forests into different types is to use 8-day composites of satellite data collected at the beginning of the growing season (April-May). This is the time when the most significant differences are recorded between vegetation indices, including Normalized Difference Vegetation Index (NDVI), LAI, and EVI of the mountain forests growing in different altitudinal zones. © Springer International Publishing Switzerland 2015.

Аннотация: The toxicity, cytotoxicity, and genotoxicity of bulk sediments from the Yenisei River (Siberia, Russia) were estimated in laboratory bioassays based on several endpoints in the aquatic plant Elodea canadensis. The bottom sediment samples were collected in the Yenisei River upstream and downstream of the sources of chemical and radioactive contamination.
