Аннотация: MOX-fuel production and spent nuclear fuel reprocessing started recently at the Mining-and-Chemical Combine (MCC, Zheleznogorsk, Russia) have caused an increase in controlled releases of plutonium to the Yenisei River. In this study, we analyzed time-dependent trends of plutonium (239,240Pu and 238Pu) in biota and bottom sediments of the Yenisei during 2008-2019, to estimate comparatively the potential of abundant representatives of biota as bio-monitors of contamination of the Yenisei by plutonium. Gamma-emitting radionuclides (40 K; 60Co; 137Cs; 152Eu; 241Am) were measured in environmental samples of the Yenisei as well. Samples of bottom sediments, water moss (Fontinalis antipyretica), shining pondweed (Potamogeton lucens), caddisfly larvae with casings (Apatania crymophila), and amphipods (Eulimnogammarus viridis and Palaseopsis cancelloides) were collected downstream and upstream of the radioactive discharge site. Environmental samples of the Yenisei collected downstream of the radioactive discharge site differed considerably in activity concentrations of plutonium but were similar in time-dependent trends of plutonium, reflecting the trends of annual discharges of plutonium. In 2018, the year of a sharp increase in controlled discharge of plutonium, the concentration of 239,240Pu in water moss (26 Bq kg-1 d.w.) was higher than in sediments (14 Bq kg-1 d.w.). In other years, the highest activity concentration of 2391,240Pu was observed in bottom sediments. In view of the higher magnitude of increase in plutonium concentration, water moss and shining pondweed can be considered as more sensitive indicators of increased fresh releases of plutonium than bottom sediments. Taking into account the food-related mechanism of plutonium uptake by amphipods, this representative of biota can be regarded as a sensitive monitor of bioavailable plutonium in the Yenisei.

Аннотация: The present study investigates, for the first time, the structure of the microbial community of cryogenic soils in the subarctic region of Siberia and the ability of the soil microbial community to metabolize degradable microbial bioplastic - poly-3-hydroxybutyrate [P(3HB)]. When the soil thawed, with the soil temperature between 5-7 and 9-11 degrees C, the total biomass of microorganisms at a 10-20-cm depth was 226-234 mg g(-1) soil and CO2 production was 20-46 mg g(-1)W day(-1). The total abundance of microscopic fungi varied between (7.4 +/- 2.3) x 10(3) and (18.3 +/- 2.2) x 10(3) CFU/g soil depending on temperature; the abundance of bacteria was several orders of magnitude greater: (1.6 +/- 0.1) x 10(6) CFU g(-1) soil. The microbial community in the biofilm formed on the surface of P(3HB) films differed from the background soil in concentrations and composition of microorganisms. The activity of microorganisms caused changes in the surface microstructure of polymer films, a decrease in molecular weight, and an increase in the degree of crystallinity of P(3HB), indicating polymer biodegradation due to metabolic activity of microorganisms. The clear-zone technique e plating of isolates on the mineral agar with polymer as sole carbon source e was used to identify P(3HB)-degrading microorganisms inhabiting cryogenic soil in Evenkia. Analysis of nucleotide sequences of rRNA genes was performed to identify the following P(3HB)degrading species: Bacillus pumilus, Paraburkholderia sp., Pseudomonas sp., Rhodococcus sp., Stenotrophomonas rhizophila, Streptomyces prunicolor, and Variovorax paradoxus bacteria and the Penicillium thomii, P. arenicola, P. lanosum, Aspergillus fumigatus, and A. niger fungi. Аннотация: The efficacy of slow-release formulations of tribenuron-methyl (TBM) embedded in the matrix of degradable poly(3-hydroxybutyrate) blended with birch wood flour [polymer/wood flour/herbicide 50/30/20 wt.%] was compared with the efficacy of TBM as the active ingredient of the Mortira commercial formulation, which was applied as post-emergence spray to treat spring wheat cv. Novosibirskaya 15. The study was conducted in Central Siberia (in the environs of the city of Krasnoyarsk, Russia) from May to August 2020. The biological efficacy of the embedded TBM was 92.3%, which was considerably higher than the biological efficacy of the Mortira formulation used as the post-emergence spray (15.4%). The embedding of TBM into degradable blended matrix enabled long-duration functioning of this unstable herbicide in soil. The sensitivity of weed plants to TBM differed depending on the species. TBM was more effective against A. retroflexus and A. blitoides, which were killed at an earlier stage, than against C. album and G. aparine, whose percentage increased in the earlier stage and which were controlled by the herbicide less effectively and at later stages. On the plot treated with the embedded herbicide, the parameters of the wheat yield structure were the best, and the total yield was the highest: 3360 +/- 40 kg/ha versus 3250 +/- 50 kg/ha in the group of plants sprayed with the Mortira formulation. The grain produced in all groups was of high quality and was classified as Grade 1 food grain. The highest quality parameters (grain hectoliter mass, gluten, and protein contents) were obtained in the group of plants treated with the embedded herbicide. The study of the embedded TBM confirmed the high efficacy of the experimental formulation.

Аннотация: The study deals with the vertical distribution and seasonal dynamics of planktonic ciliates in the pelagic and littoral zones of a saline meromictic Lake Shira. Fourteen species of free-living ciliates have been found in the Lake, seven of them inhabiting the pelagic zone. The richness of ciliates is higher both in terms of the number of species and biomass in the littoral zone compared to the pelagic zone. Although the ciliate species diversity is low in the pelagic zone, in certain seasons, the biomass of some of the species may reach considerable values, up to 3.4 g m(-2) in the water column. The biomass of ciliates in Lake Shira is generally comparable to the average values for other mesotrophic lakes. The abundance and composition of ciliate populations in Lake Shira vary considerably with depth. Changes in the Lake ecosystem caused by meromixis breakdown in 2015-2016, which induced considerable variations in many Lake components, affected the vertical distribution of planktonic ciliates. However, the annual average biomass of the species that inhabited the Lake before meromixis breakdown remained unchanged. The total ciliate biomass increased due to the presence of the new species. Аннотация: Most of Earth's bacteria have yet to be cultivated. The metabolic and functional potentials of these uncultivated microorganisms thus remain mysterious, and the metagenome-assembled genome (MAG) approach is the most robust method for uncovering these potentials. However, MAGs discovered by conventional metagenomic assembly and binning are usually highly fragmented genomes with heterogeneous sequence contamination. In this study, we combined Illumina and Nanopore data to develop a new workflow to reconstruct 233 MAGs-six novel bacterial orders, 20 families, 66 genera, and 154 species-from Lake Shunet, a secluded meromictic lake in Siberia. With our workflow, the average N50 of reconstructed MAGs greatly increased 10-40-fold compared to when the conventional Illumina assembly and binning method were used. More importantly, six complete MAGs were recovered from our datasets. The recovery of 154 novel species MAGs from a rarely explored lake greatly expands the current bacterial genome encyclopedia. Chen and colleagues develop a workflow for assembling high quality metagenome-associated genomes for microbial species using long and short reads, in this case from a meromictic lake. A full, detailed workflow is provided in for use by the community.

Аннотация: Meromictic lakes usually harbour certain prevailing anoxygenic phototrophic bacteria in their anoxic zone, such as the purple sulfur bacterium (PSB) Thiocapsa sp. LSW (hereafter LSW) in Lake Shunet, Siberia. PSBs have been suggested to play a vital role in carbon, nitrogen and sulfur cycling at the oxic–anoxic interface of stratified lakes; however, the ecological significance of PSBs in the lake remains poorly understood. In this study, we explored the potential ecological role of LSW using a deep-sequencing analysis of single-cell genomics associated with flow cytometry. An approximately 2.7 Mb draft genome was obtained based on the co-assembly of five single-cell genomes. LSW might grow photolithoautotrophically and could play putative roles not only as a carbon fixer and diazotroph, but also as a sulfate reducer/oxidizer in the lake. This study provides insights into the potential ecological role of Thiocapsa sp. in meromictic lakes. © 2021 The Authors.

Аннотация: The world map of anthropogenic atmospheric nitrogen deposition and its effects on natural ecosystems is not described with equal precision everywhere. In this paper, we report atmospheric nutrient, sulfate and spheroidal carbonaceous particle (SCP) deposition rates, based on snowpack analyses of a formerly unexplored Siberian mountain region. Then, we discuss their potential effects on lake phytoplankton biomass limitation. We estimate that the nutrient depositions observed in the late-season snowpack (40 +/- 16 mgNO(3)-Nm(-2) and 0.58 +/- 0.13 mg TP-Pm-2; TP for total phosphorous) would correspond to yearly depositions lower than 119 +/- 71 mgNO(3)-Nm(-2) yr(-1) and higher than 1.71 +/- 0.91 mg TP-Pm-2 yr(-1). These yearly deposition estimates would approximately fit the predictions of global deposition models and correspond to the very low nutrient deposition range, although they are still higher than world background values. In spite of the fact that such a low atmospheric nitrogen deposition rate would be enough to induce nitrogen limitation in unproductive mountain lakes, phosphorus deposition was also extremely low, and the resulting lake water N: P ratio was unaffected by atmospheric nutrient deposition. In the end, the studied lakes' phytoplankton appeared to be split between phosphorus and nitrogen limitation. We conclude that these pristine lakes are fragile sensitive systems exposed to the predicted climate warming, increased winter precipitation, enhanced forest fires and shifts in anthropogenic nitrogen emissions that could finally couple their water chemistry to that of atmospheric nutrient deposition and unlock temperature-inhibited responses of phytoplankton to nutrient shifts.

Аннотация: Objective was to study single-nucleotide polymorphisms (SNP) in CAT, NCL, HSPA1L, PCDH15, and PON2 genes and their associations with hearing impairment among the people working among noise-exposed workers of the mashine-building plant (JSC «Krasmash», Krasnoyarsk, Eastern Siberia, Russia). Materials and methods. The 443 employees of Krasmash JSC, who have been working under conditions of increased noise for at least 1 year, were surveyed and examined. A hearing study was performed by speech and tonal audiometry. Tonal audiometry was carried out in accord with according to a standard method in the frequency range 125—8000 Hz. People with chronic hearing impairment, survivors of meningitis and family history of hearing impairment were excluded from the study. The allelic composition of the studied genes was determined in the remaining group of 288 workers (study group). Polymorphisms were detected using bioluminescent method, developed by the authors earlier. The study group comprised 122 people with hearing impairment (experimental group) and 166 people without impairment (control group). Results. The genotyping results of on allelic variants rs494024 (CAT), rs7598759 (NCL), rs2227956 (HSPA1L), rs7095441 (PCDH15) and rs7785846 (PON2) showed that their frequencies in the study group did not differ and were comparable with those for the European population. No statistically significant differences were revealed in the distribution of the genotypes of the studied mutations between the experimental and control groups. Also no statistically significant associations we found between hearing impairment and availability of two or several SNPs, or these SNPs and clinical characteristics of the disease (degree of hearing impairment, tinnitus). In the group of workers with an experience of 5 to 16 years, an association was found for hearing impairment and SNP rs494024, as well as when it is combined with rs7598759. Conclusions. The associations between SNP rs7598759, rs2227956, and rs7095441 and hearing impairment were not found. In the group of workers with 5—16 year experience, this association was found for SNP rs494024, as well as when it is combined with rs7598759. Аннотация: The world map of anthropogenic atmospheric nitrogen deposition and its effects on natural ecosystems is not described with equal precision everywhere. In this paper, we report atmospheric nutrient, sulfate and spheroidal carbonaceous particle (SCP) deposition rates, based on snowpack analyses of a formerly unexplored Siberian mountain region. Then, we discuss their potential effects on lake phytoplankton biomass limitation. We estimate that the nutrient depositions observed in the late-season snowpack (40 +/- 16 mgNO(3)-Nm(-2) and 0.58 +/- 0.13 mg TP-Pm-2; TP for total phosphorous) would correspond to yearly depositions lower than 119 +/- 71 mgNO(3)-Nm(-2) yr(-1) and higher than 1.71 +/- 0.91 mg TP-Pm-2 yr(-1). These yearly deposition estimates would approximately fit the predictions of global deposition models and correspond to the very low nutrient deposition range, although they are still higher than world background values. In spite of the fact that such a low atmospheric nitrogen deposition rate would be enough to induce nitrogen limitation in unproductive mountain lakes, phosphorus deposition was also extremely low, and the resulting lake water N: P ratio was unaffected by atmospheric nutrient deposition. In the end, the studied lakes' phytoplankton appeared to be split between phosphorus and nitrogen limitation. We conclude that these pristine lakes are fragile sensitive systems exposed to the predicted climate warming, increased winter precipitation, enhanced forest fires and shifts in anthropogenic nitrogen emissions that could finally couple their water chemistry to that of atmospheric nutrient deposition and unlock temperature-inhibited responses of phytoplankton to nutrient shifts.

Аннотация: Meromictic lakes usually harbour certain prevailing anoxygenic phototrophic bacteria in their anoxic zone, such as the purple sulfur bacterium (PSB) Thiocapsa sp. LSW (hereafter LSW) in Lake Shunet, Siberia. PSBs have been suggested to play a vital role in carbon, nitrogen and sulfur cycling at the oxic-anoxic interface of stratified lakes; however, the ecological significance of PSBs in the lake remains poorly understood. In this study, we explored the potential ecological role of LSW using a deepsequencing analysis of single- cell genomics associated with flow cytometry. An approximately 2.7 Mb draft genome was obtained based on the co- assembly of five single- cell genomes. LSW might grow photolithoautotrophically and could play putative roles not only as a carbon fixer and diazotroph, but also as a sulfate reducer/oxidizer in the lake. This study provides insights into the potential ecological role of Thiocapsa sp. in meromictic lakes.

Аннотация: During the winter of 2018, total zooplankton samples were taken under ice with a plankton net in Sobachye Lake, one of the large Norilsk lakes, in the western (April 13, May 6) and middle (May 6) parts at 70, 80, and 100 m depths. The previous assumption that Senecella in the large Norilsk lakes is reproduced in winter and is a univoltine species with a long (one-year) generation development time has been confirmed. The assignment of adult individuals to the species Senecella siberica Vyshkvartzeva 1994 has also been confirmed.

Аннотация: Five permanent sample plots (SPs; 200-250 trees per plot) were established in middle-aged high-grade suburban pine stands near the industrial city of Krasnoyarsk, Siberia, Russia. Needle damage, inventory parameters of the stands, and the defense response of the stem phloem were evaluated annually for the years 2002-2019 and attributed to acute or chronic toxic exposures (creeping fire or industrial pollutants, respectively). The results form a basis for using trees as bioindicators. A newly elaborated stem lesion test was formed from a hypothesis on the upward sugar transport for the regeneration of an injured crown, based on Eschrich's model of bidirectional sugar transport in the phloem. The formation of a phloem lesion was induced by inoculation of the stem with a mycelial extract of the ophiostomatoid fungus Ceratocystis laricicola. The lesion length and its shift relative to the inoculation hole were measured. An increase in the length of needles at early stages of stand weakening by pollutants was found to correspond to the hormesis model (Selye's adaptation syndrome). A possibility of assessing the chronology of pollutant toxicity and the duration of the recovery period after creeping fire was shown.

Аннотация: In cold smoked species of genus Coregonus, identified by molecular genetic analysis, contents of fatty acids and heavy metals and arsenic were measured. The highest content of sum of long-chain polyunsaturated fatty acids of omega-3 family (LC-PUFA), namely eicosapentaenoic (EPA) and docosahexaenoic (DHA) fatty acids, 6.53 +/- 0.78 mg g(-1) wet weight, was characteristic of tugun Coregonus tugun. This is the first quantitative estimation (mg LC-PUFA per g of product) of the nutritive value of smoked fish. Thus, to obtain a daily personal doze of EPA + DHA of 1 g, recommended for prevention of cardiovascular diseases, one needs to consume 153 g of the smoked tugun. Metals contents did not exceed standards for fish meat except Pb in least cisco Coregonus sardinella. Accordingly, values of hazard quotients, which estimate benefit-risk ratio of fish intake, indicate that most of the smoked fish species are safe product for human nutrition, except least cisco regarding Pb content.

Аннотация: The relations between the spectral surface characteristics of the elements of the soil cover structure and soil properties in the Krasnoyarsk forest-steppe of Central Siberia were investigated. It was revealed that the most informative parameters for field spectrometry are the content of humus, carbonate carbon dioxide and the prevailing particle-size fractions. A statistically significant relationship between the elements of the soil cover structure and the reflectivity of soils has been confirmed by means of multidimensional statistics. The wave lengths with the greatest coupling force are highlighted. Regression equations for remote study of soil cover structure have been obtained, which can be used if additional point studies are carried out in a wider range of test parameters. Аннотация: This study compares the field data of the seasonal dynamics of halophytic plant productivity as dependent upon soil salinity level and soil type. The field study was carried out in the coastal area of Lake Kurinka (the Republic of Khakasia, south of Middle Siberia) between May and September of 2014 and 2016. Two plant communities with different soil salinity levels were studied. Results of the field investigation show that there is a correlation between plant growth and the soil salinity level. With high-salinity (3.72%) soils, the productivity of halophyte plants is lower than productivity with low-salinity (0.175%) soils.

Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.

Аннотация: A halotolerant, psychrotolerant and methylotrophic methanogen, strain SY-01(T), was isolated from the saline Lake Tus in Siberia. Cells of strain SY-01(T) were non-motile, cocci and 0.8-1.0 mu m in diameter. The only methanogenic substrate utilized by strain SY-01(T) was methanol. The temperature range of growth for strain SY-01(T) was from 4 to 40 degrees C and the optimal temperature for growth was 30 degrees C. The pH range of growth was from pH 7.2 to 9.0, with optimal growth at pH 8.0. The NaCl range of growth was 0-1.55 M with optimal growth at 0.51 M NaCl. The G+C content of the genome of strain SY-01(T) was 43.6 mol% as determined by genome sequencing. Phylogenetic analysis revealed that strain SY-01(T) was most closely related to Methanolobus zinderi SD1(T) (97.3% 16S rRNA gene sequence similarity), and had 95.5-97.2 % similarities to other Methanolobus species with valid names. Genome relatedness between strain SY-01(T) and DSM 21339(T) was computed using average nucleotide identity and digital DNA-DNA-hybridization, which yielded values of 79.7 and 21.7 %, respectively. Based on morphological, phenotypic, phylogenetic and genomic relatedness data presented here, it is evident that strain SY-01(T) represents a novel species of the genus Methanolobus, and the name Methanolobus halotolerans sp. nov. is proposed. The type strain is SY-01(T) (=BCRC AR10051(T)=NBRC 113166(T)=DSM 107642(T)).
