Аннотация: We studied the effects of anti-inflammatory substances incorporated in polymeric microparticles made of degradable natural polyhydroxyalkanoate polyesters on experimental skin wounds caused by chemical burns in laboratory animals. Treatment with encapsulated forms of anti-inflammatory substances (applied in gel) accelerated wound healing in comparison with routine therapy (estimated by area of burn wound, wound healing activity, number of acanthotic cells, and number of hair and sebaceous follicles). The results showed the perspectives of usage of developed form of substances (degradable polymeric microparticles) for treatment of skin defects.

Аннотация: The purpose of this study was to develop micro and nano sized drug carriers from poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), and study the cell and skin penetration of these particles. PHBV micro/nanospheres were prepared by o/w emulsion method and were stained with a fluorescent dye, Nile Red. The particles were fractionated by centrifugation to produce different sized populations. Topography was studied by SEM and average particle size and its distribution were determined with particle sizer. Cell viability assay (MTT) was carried out using L929 fibroblastic cell line, and particle penetration into the cells were studied. Transdermal permeation of PHBV micro/nanospheres and tissue reaction were studied using a BALB/c mouse model. Skin response was evaluated histologically and amount of PHBV in skin was determined by gas chromatography-mass spectrometry. The average diameters of the PHBV micro/nanosphere batches were found to be 1.9 mu m, 426 and 166 nm. Polydispersity indices showed that the size distribution of micro sized particles was broader than the smaller ones. In vitro studies showed that the cells had a normal growth trend. MTT showed no signs of particle toxicity. The 426 and 166 nm sized PHBV spheres were seen to penetrate the cell membrane. The histological sections revealed no adverse effects. In view of this data nano and micro sized PHBV particles appeared to have potential to serve as topical and transdermal drug delivery carriers for use on aged or damaged skin or in cases of skin diseases such as psoriasis, and may even be used in gene transfer to cells.

Аннотация: The present study reports construction of wound dressing materials from degradable natural polymers such as hydroxy derivatives of carboxylic acids (PHAs) and 3-hydroxybutyrate/4-hydroxybutyrate [P(3HB/4HB)] as copolymer. The developed polymer films and electrospun membranes were evaluated for its wound healing properties with Grafts—elastic nonwoven membranes carrying fibroblast cells derived from adipose tissue multipotent mesenchymal stem cells. The efficacy of nonwoven membranes of P(3HB/4HB) carrying the culture of allogenic fibroblasts was assessed against model skin defects in Wistar rats. The morphological, histological and molecular studies revealed the presence of fibroblasts on dressing materials which facilitated wound healing, vascularization and regeneration. Further it was also observed that cells secreted extracellular matrix proteins which formed a layer on the surface of membranes and promoted the migration of epidermal cells from the neighboring tissues surrounding the wound. The wounds under the P(3HB/4HB) membrane carrying cells healed 1.4 times faster than the wounds under the cell-free membrane and 3.5 times faster than the wounds healing under the eschar (control).The complete wound healing process was achieved at Day 14. Thus the study highlights the importance of nonwoven membranes developed from degradable P(3HB/4HB) polymers in reducing inflammation, enhancing angiogenic properties of skin and facilitating better wound healing process. © 2016, Springer Science+Business Media New York.

Аннотация: An integrated study has been performed to investigate the process of formation of ultrafine fibers and nonwoven membranes by electrospinning from natural degradable polymers-polyhydroxyalkanoates (PHAs); physical, mechanical, and biological properties of the products have been studied. Then, electrospinning was used to prepare ultrafine fibers from PHAs with different compositions: P(3HB) and its copolymers P(3HB-co-4HB), P(3HB-co-3HV), and P(3HB-co-3HHx). The main process parameters, that influence UF-fiber diameter and properties of fibrous non-woven membranes) (polymer concentration, solution feeding rate, working distance, and applied voltage), were investigated and their effects evaluated. This study was the first to compare biological and physical/mechanical parameters of PHAs with different chemical compositions as dependent upon the fractions of monomers, constituting the polymers and fiber orientation. Electrospun polymer membranes, prepared from the [P(3HB-co-4HB)], were tested as wound dressings. The developed nonwoven membranes can be used as the equivalent of collagen skine dressings in the treatment of burns of degree II. Experiments on laboratory animals with model skin defects showed, that the membranes fitted the wound shape good, protected the wound from external influences, and facilitated wound healing, promoting fast repair. The successful experiments on laboratory animals were followed by pilot clinical trials of nanomembranes, comprised of PHA membranes as wound dressings in the treatment of septic wounds. During the regeneration phase, PHA membranes served as a scaffold for the new tissue on the skin and filled out soft tissue defects. The formation of the uniform and sufficiently vascularized tissue is a prerequisite for quicker wound healing and can serve as a basis for the subsequent skin grafting and spontaneous re-epithelialization of superficial wounds. The wound dressing, tested in this clinical trial, performs important physiological functions of natural skin, provides a barrier against secondary infection, reduces fluid loss, and, at the same time, does not keep the air out. © 2017 Nova Science Publishers, Inc.

Аннотация: Experiments in vivo demonstrated the protective effect of modified nanodiamonds on guinea pig skin after local exposure cobalt ions, but not chrome ions. The observed differences are determined by different adsorption of these ions by nanodiamonds: in vitro experiments showed that nanodiamonds adsorbed cobalt ions, but not chrome ions from water solutions. The perspectives of using modified nanodiamonds as a new adsorbent for prevention of allergic contact dermatitis induced by ions of bivalent metals are discussed. © 2014, Springer Science+Business Media New York.

Аннотация: Hybrid wound dressings have been constructed using two biomaterials: bacterial cellulose (BC) and copolymer of 3-hydroxybutyric and 4-hydroxybutyric acids [P(3HB/4HB)] – a biodegradable polymer of microbial origin. Some of the experimental membranes were loaded with drugs promoting wound healing and epidermal cells differentiated from multipotent adipose-derived mesenchymal stem cells. A study has been carried out to investigate the structure and physical/mechanical properties of the membranes. The in vitro study showed that the most effective scaffolds for growing fibroblasts were composite BC/P(3HB/4HB) films loaded with actovegin. Two types of the experimental biotechnological wound dressings – BC/P(3HB/4HB)/actovegin and BC/P(3HB/4HB)/fibroblasts – were tested in vivo, on laboratory animals with model third-degree skin burns. Wound planimetry, histological examination, and biochemical and molecular methods of detecting factors of angiogenesis, inflammation, type I collagen, and keratin 10 and 14 were used to monitor wound healing. Experimental wound dressings promoted healing more effectively than VoskoPran – a commercial wound dressing. © 2019

Аннотация: Several polymorphisms in the melanocortin-1 receptor gene (MC1R) have been associated with melanoma risk. In particular, rs1805007, rs1805008, and rs1805009 mutations, which result in R151C, R160W, and D294H amino acid substitutions, respectively, and are associated with the phenotype of red-hair mutations, have also been connected with melanoma and nonmelanoma skin cancer risks. This work describes a method of detecting these polymorphisms using primer extension with subsequent dual bioluminescent assay. Model plasmids carrying polymorphic MC1R fragments, as well as several clinical DNA samples, were tested using the proposed technique. The results agreed well with those obtained by Sanger sequencing. © 2015, Pleiades Publishing, Inc.
