Аннотация: The full-length cDNA genes encoding five new isoforms of Ca2 +-regulated photoprotein mitrocomin from a small tissue sample of the outer bell margin containing photocytes of only one specimen of the luminous jellyfish Mitrocoma cellularia were cloned, sequenced, and characterized after their expression in Escherichia coli and subsequent purification. The analysis of cDNA nucleotide sequences encoding mitrocomin isoforms allowed suggestion that two isoforms might be the products of two allelic genes differing in one amino acid residue (64R/Q) whereas other isotypes appear as a result of transcriptional mutations. In addition, the crystal structure of mitrocomin was determined at 1.30 A resolution which expectedly revealed a high similarity with the structures of other hydromedusan photoproteins. Although mitrocomin isoforms reveal a high degree of identity of amino acid sequences, they vary in specific bioluminescence activities. At that, all isotypes displayed the identical bioluminescence spectra (473–474 nm with no shoulder at 400 nm). Fluorescence spectra of Ca2 +-discharged mitrocomins were almost identical to their light emission spectra similar to the case of Ca2 +-discharged aequorin, but different from Ca2 +-discharged obelins and clytin which fluorescence is red-shifted by 25–30 nm from bioluminescence spectra. The main distinction of mitrocomin from other hydromedusan photoproteins is an additional Tyr at the C-terminus. Using site-directed mutagenesis, we showed that this Tyr is not important for bioluminescence because its deletion even increases specific activity and efficiency of apo-mitrocomin conversion into active photoprotein, in contrast to C-terminal Pro of other photoproteins. Since genes in a population generally exist as different isoforms, it makes us anticipate the cloning of even more isoforms of mitrocomin and other hydromedusan photoproteins with different bioluminescence properties. © 2016 Elsevier B.V.

Аннотация: Effects of variable levels of photosynthetically active radiation (PAR) and NaCl concentrations, typical of closed ecological life support systems, on growth of Salicornia europaea L. plants, CO2 exchange, mineral composition, and the content of malondialdehyde (MDA) and photosynthetic pigments were investigated. The plants were grown for 25 days at different salinities of nutrient Knop solution (171, 342, and 513 mM NaCl) under two PAR levels (690 and 1150 ?mol/(m2 s)). At PAR of 690 ?mol/(m2 s), the plant productivity did not show significant changes at increasing salinities; at 1150 ?mol/(m2 s), the maximal productivity was observed at NaCl concentrations of 171 and 342 mM. The increase in NaCl concentration from 171 to 513 mM in the nutrient solution led to a substantial increase in the relative Na content in aboveground organs at PAR level of 1150 ?mol/(m2 s). The MDA content in aboveground organs by the end of the growth period was independent of PAR intensity. The content of photosynthetic pigments in the assimilatory tissue decreased with the increase in salinity from 342 to 513 mM NaCl at PAR level of 1150 ?mol/(m2 s) but not at the lower irradiance. The combination of 1150 ?mol/(m2 s) PAR intensity with the salinity as high as 342 mM NaCl was found to be the most effective for optimal productivity of S. europaea plants. © 2016, Pleiades Publishing, Ltd.

Аннотация: Currently, the patients with purulent wounds account for about 40% of the patients of surgical profilt. Since the treatment of septic wounds under the bandage remains the main method, as it is the most convenient and economically beneficial, but not effective enough, the development of new advanced coatings possessing simultaneously by multiple properties, is considered to be relevant. In the process of wound healing a physician faces with many problems including the issue of the selection option of a wound dressing, as they differ significantly in their design and properties. A disadvantage of many wound dressings for wound care is the adherence to wound. This results in the regenerating tissue injury and the dressings become sore. Many of the positive properties of dressings reduced as a result of damage caused by daily dressings. In recent years the biocompatible materials for injuries and wounds contribute to more effective healing and regeneration. In this regard, the paper presents the main types of current wound dressings used in the treatment of purulent wounds, their properties, indications and methods of applicatio.

Аннотация: Humans who eat fish are exposed to mixtures of healthful nutrients and harmful contaminants that are influenced by environmental and ecological factors. Marine fisheries are composed of a multitude of species with varying life histories, and harvested in oceans, coastal waters and estuaries where environmental and ecological conditions determine fish exposure to both nutrients and contaminants. Many of these nutrients and contaminants are thought to influence similar health outcomes (i.e., neurological, cardiovascular, immunological systems). Therefore, our understanding of the risks and benefits of consuming seafood require balanced assessments of contaminants and nutrients found in fish and shellfish. In this paper, we review some of the reported benefits of fish consumption with a focus on the potential hazards of mercury exposure, and compare the environmental variability of fish oils, selenium and mercury in fish. A major scientific gap identified is that fish tissue concentrations are rarely measured for both contaminants and nutrients across a range of species and geographic regions. Interpreting the implications of seafood for human health will require a better understanding of these multiple exposures, particularly as environmental conditions in the oceans change. © Marine Biological Association of the United Kingdom, 2015.

Аннотация: Triploid fish has become an important item of commercial aquaculture, but data on its fatty acid (FA) composition are still controversial, especially regarding essential polyunsaturated fatty acids, eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA). We studied FA composition and content of diploid and triploid pink salmon Oncorhynchus gorbuscha, reared in aquaculture in a bay of the White Sea (Russia). FA composition, measured as percentages of total FA of triploids and immature diploid females significantly differed from that of mature diploid fish. Specifically, mature diploids had higher percentage of EPA and DHA in their muscle tissue (filets) compared to that of triploids and immature diploid females. Nevertheless, the contents of EPA and DHA per mass of the filets in diploid and triploid specimens were similar. Thus, no special efforts are needed to improve EPA and DHA contents in filets of triploids. © 2016 Elsevier Ltd

Аннотация: The study describes preparation and testing of porous 3D implants of natural degradable polymer of 3-hydroxybutyric acid P(3HB) for regeneration of bone tissue defects. The ability of the P(3HB) implants to favor attachment and facilitate proliferation and directed differentiation of mesenchymal stem cells (MSCs) was studied in the culture of MSCs isolated from bone marrow and adipose tissue. Tissue-engineered hybrid systems (grafts) constructed using P(3HB) and P(3HB) in combination with osteoblasts were used in experiments on laboratory animals (n=48) with bone defect model. The defect model (5 mm in diameter) was created in the rat parietal bone, and filling of the defect by the new bone tissue was monitored in the groups of animals with P(3HB) implants, with commercial material, and without implants (negative control). Computed tomography (CT) and histologic examination showed that after 120 days, in the group with the osteoblast-seeded P(3HB) implants, the defect was completely closed; in the group with the cell-free P(3HB) implants, the remaining defect was no more than 10% of the initial one (0.5 mm); in both the negative and positive controls, the size of the defect was about 1.0-1.2 mm. Аннотация: An integrated study has been performed to investigate the process of formation of ultrafine fibers and nonwoven membranes by electrospinning from natural degradable polymers-polyhydroxyalkanoates (PHAs); physical, mechanical, and biological properties of the products have been studied. Then, electrospinning was used to prepare ultrafine fibers from PHAs with different compositions: P(3HB) and its copolymers P(3HB-co-4HB), P(3HB-co-3HV), and P(3HB-co-3HHx). The main process parameters, that influence UF-fiber diameter and properties of fibrous non-woven membranes) (polymer concentration, solution feeding rate, working distance, and applied voltage), were investigated and their effects evaluated. This study was the first to compare biological and physical/mechanical parameters of PHAs with different chemical compositions as dependent upon the fractions of monomers, constituting the polymers and fiber orientation. Electrospun polymer membranes, prepared from the [P(3HB-co-4HB)], were tested as wound dressings. The developed nonwoven membranes can be used as the equivalent of collagen skine dressings in the treatment of burns of degree II. Experiments on laboratory animals with model skin defects showed, that the membranes fitted the wound shape good, protected the wound from external influences, and facilitated wound healing, promoting fast repair. The successful experiments on laboratory animals were followed by pilot clinical trials of nanomembranes, comprised of PHA membranes as wound dressings in the treatment of septic wounds. During the regeneration phase, PHA membranes served as a scaffold for the new tissue on the skin and filled out soft tissue defects. The formation of the uniform and sufficiently vascularized tissue is a prerequisite for quicker wound healing and can serve as a basis for the subsequent skin grafting and spontaneous re-epithelialization of superficial wounds. The wound dressing, tested in this clinical trial, performs important physiological functions of natural skin, provides a barrier against secondary infection, reduces fluid loss, and, at the same time, does not keep the air out. © 2017 Nova Science Publishers, Inc.

Аннотация: The present study reports construction of wound dressing materials from degradable natural polymers such as hydroxy derivatives of carboxylic acids (PHAs) and 3-hydroxybutyrate/4-hydroxybutyrate [P(3HB/4HB)] as copolymer. The developed polymer films and electrospun membranes were evaluated for its wound healing properties with Grafts—elastic nonwoven membranes carrying fibroblast cells derived from adipose tissue multipotent mesenchymal stem cells. The efficacy of nonwoven membranes of P(3HB/4HB) carrying the culture of allogenic fibroblasts was assessed against model skin defects in Wistar rats. The morphological, histological and molecular studies revealed the presence of fibroblasts on dressing materials which facilitated wound healing, vascularization and regeneration. Further it was also observed that cells secreted extracellular matrix proteins which formed a layer on the surface of membranes and promoted the migration of epidermal cells from the neighboring tissues surrounding the wound. The wounds under the P(3HB/4HB) membrane carrying cells healed 1.4 times faster than the wounds under the cell-free membrane and 3.5 times faster than the wounds healing under the eschar (control).The complete wound healing process was achieved at Day 14. Thus the study highlights the importance of nonwoven membranes developed from degradable P(3HB/4HB) polymers in reducing inflammation, enhancing angiogenic properties of skin and facilitating better wound healing process. © 2016, Springer Science+Business Media New York.

Аннотация: The present study describes changes in the mycelium of the fungus growing on the luminescent wood collected on Borneo Island in early December 2013 that occurred during 31 months of storage. The study shows that wood samples retain their ability to emit light, forming two types of luminescent mycelium: surface mycelium and aerial mycelium. The hyphae of the surface mycelium form on the surface of the wood sample and then spread over the surface of the polyethylene bag or over the surface of the bottom of tissue culture flasks containing the samples. The aerial mycelium develops later and only in tissue culture flasks, forming biomass composed of local interlaced hyphae, growing upward. The surface mycelium is characterized by non-uniform "flickering" luminescence along the hyphae. There is no diurnal periodicity in the luminescence of this fungus, but luminescence is increased by mechanical disturbance or exposure to ultraviolet radiation. The local impact of these factors causes an increase in luminescence of the mycelium regions that have not been directly affected. It has been assumed that the variable level of luminescence is an individual trait of this fungus species. The results obtained in this study suggest that luminescent wood found on Borneo Island contains mycelium of a fungus species, whose luminescent properties are essentially different from those of the fungi described in the scientific literature.

Аннотация: Long-chain polyunsaturated n-3 fatty acids (LC-PUFA), including eicosapentaenoic (EPA), and docosahexaenoic (DHA) are an essential part of human diets, but their main source, the wild fish catch, is limited. Terrestrial production animals, such as cattle, pigs, and chicken, can synthesize these compounds from the plant-derived precursor -linolenic acid, and the synthesis occurs mainly in liver. We estimated the putative contribution of liver products to the LC-PUFA supply in human nutrition, as an alternative to fish. We measured EPA and DHA contents in raw and cooked livers of the animals. Calculations of global production of EPA and DHA in the livers of these animals were done. Mean contents of LC-PUFA in raw livers of chicken, pigs, and cattle were comparable with those in some fish. Culinary treatments of the liver did not result in a decrease of EPA and DHA contents. Global production of EPA and DHA in animals' livers was estimated as approximate to 410(6)kg/year, while the global EPA+DHA supply through the wild fish catch is known to be 18010(6)kg/year. Thus, liver of production animals is an additional source of LC-PUFA for human nutrition, rather than an alternative to fish. Practical applications: Consumption of food with a high PUFA content and a low n-6/n-3 ratio is recommended for humans to prevent cardiovascular disease and psychiatric disorders. We found that cooked pork and beef liver is a valuable source of n-3 LC-PUFA in human nutrition. In contrast, cooked chicken liver had too high of a n-6/n-3 ratio and, thereby, had a lower nutritive value concerning n-3 LC-PUFA. An estimate has been made of the putative contribution of liver products of terrestrial animals to the LC-PUFA supply as alternative to fish in human nutrition.

A relationship between the content of MDA (LPO indicator) and fluorescence of bacteria was observed in all biological samples.

Аннотация: Various chronic inflammatory and necrotic processes in the liver parenchyma are accompanied by pathological morphofunctional changes, which are associated with hepatocyte death and hyperplasia of the connective tissue. Regeneration of the liver parenchyma should include not only prevention of fibrosis, but also stimulation of hepatocyte prolifearation. The adrenoceptor agonist dobutamine stimulated proliferative activity of cultured hepatocytes and prevented the development of postintoxication liver cirrhosis in mice produced by chronic poisoning with CCl4.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.

Аннотация: We studied the possibility of evaluation of the intensity of pathological oxidative processes in rat liver using an integral bioluminescent test with controlled perfusion of the isolated organ. The test revealed a significant correlation between the level of TBA-reactive products and bioluminescence intensity.

Аннотация: Additional integral and kinetic parameters of induction curves have been introduced on the basis of analysis of currently employed parameters of slow induction of fluorescence (SIF) for a more complete quantitative description of SIF changes during ontogenesis of higher plant leaves. High sensitivity of the additional parameters to the age state of leaf tissue as compared with previously employed parameters is demonstrated on the example of cucumber leaves (Cucumis sativus L.). Analysis of the relationship between the photosynthetic rate and SIF parameters of separate cucumber leaf disks revealed the presence of a high positive correlation between the photosynthetic rate and the greatest possible average decline in fluorescence intensity during the inductive period for mature regions of the leaf and regions starting to senesce (the correlation coeffcient attained values of 0.95), which indicates the possibility of using the given SIF parameter in further elaboration of methods for estimation of chloroplast photosynthetic activity at the level of the higher plant leaf.

Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types-polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV)-featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 mum thick, which became as thin as 40-60 mum after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner. Аннотация: Various chronic inflammatory and necrotic processes in the liver parenchyma are accompanied by pathological morphofunctional changes, which are associated with hepatocyte death and, hyperplasia of the connective tissue. Regeneration of the liver parenchyma should include not only prevention of fibrosis, but also stimulation of hepatocyte proliferation. The adrenoceptor agonist dobutamine stimulated proliferative activity of cultured hepatocytes and prevented the development of postintoxication liver cirrhosis in mice produced by chronic poisoning with CCl4.

Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 mum thick during weeks 4-8, which in 4-6 months was reduced to 40-60 mum in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Throughout the period of monitoring no differences in the tissue response to the implantation of the polymer filaments of two PHA types were recorded. (C) 2003 Elsevier Science B.V. Аннотация: Composite films and nonwoven mats of the poly-3-hydroxybutyrate and chitosan natural polymers were prepared and investigated. DSC and FTIR techniques were used to confirm that P(3HB) blending with chitosan resulted in a decrease in P(3HB) crystallinity to 47% and 62% in the films and nonwoven mats, respectively. Scanning electron microscopy showed that addition of chitosan induced changes in the surface morphology of the composite films and a reduction in the diameter of ultrafine fibers in the nonwoven mats from 800 nm to 460 nm. The values of water contact angle for films (53°) and nonwoven mats (50.6°) suggested that chitosan enhanced hydrophilic properties and moisture absorption capacity of the composite materials. On the other hand, P(3HB) showed its reinforcing ability and improved the physical/mechanical properties of chitosan. The work included studies of in vitro biodegradation of the composite specimens and their ability to maintain cell growth and attachment in NIH 3T3 fibroblast culture. © 2020
