Аннотация: A hypothesis is considered that the establishment and maintenance of mass exchange processes in the Paramecium bursaria symbiotic complex (infusorian-alga) depends on the presence of the third component-bacteria-whose role is underestimated but essential for completing the exchange cycle. The role of this component in the symbiotic cycle of Paramecium bursaria is possible to reveal using special bactericidal preparations specifically inhibiting the bacteria involved in the metabolic cycle but having no effect on the functions of the host cell and the zoochlorella population. Experiments with various preparations of this type were performed, and the response to treatment was estimated from growth rates of the symbiotic complex and its components. This allowed identification of an antibiotic (rifampicin) that had no adverse influence on free-living zoochlorella and chlorella-free paramecia (the basic components of the symbiotic cycle) but drastically inhibited the growth of the P. bursaria symbiotic complex, which could be interpreted as evidence in favor of the hypothesis concerning the existence of the third symbiotic component.

Аннотация: All of man's former space flights were not real ventures into space in the biological sense, as his life was supported with unregenerated earth supplies. The coming stage of space exploration requires man's long existence in the cosmos and on the other planets. This stage of man's activity outside the earth become possible only by creating small man-made ecosystems, permitting the support of his metabolism by the recycling of substances of the terrestrial biosphere. Creation of such systems is a new scientific and technical task. Man-made ecosystems are a new product of man's activity, which have no complete analogy, either in nature, or in technology. Stochastic mechanisms, which stabilize biogeocenosis, cannot be effective in small ecosystems. A technique of parametric control over biosynthesis made it possible to calculate, and put to practice, an ecosystem for man with a cyclic regeneration of the atmosphere, water and, partially, food. The specific bio-technological properties of small man-made ecosystems are being analysed. The possibility of their application for man's excursions into space and for the settlement of other planets is being considered.

Аннотация: Time-resolved fluorescence was used to directly measure the energy transfer rate constant in the protein-protein complex involved in the yellow bioluminescence of Vibrio fischeri, strain Y1. In this reaction the putative donor is the fluorescent transient intermediate, luciferase hydroxyflavin, which exhibits a major fluorescence lifetime of the bound flavin of 10 ns. On addition of the acceptor, the V. fischeri yellow fluorescence protein containing either FMN or riboflavin as ligand, a rapid decay time, 0.25 ns, becomes predominant. The same results are observed using rec-luciferase from Photobacterium leiognathi to produce the donor. Because of favorable spectral separation in this system, this rapid decay rate of 4 ns(-1), can be directly equated to the energy transfer rate. This rate is ten times higher than the rate previously observed in the Photobacterium luciferase hydroxyflavin-lumazine protein, donor-acceptor system, derived from emission anisotropy measurements. This ten-times ratio is close to the ratio of spectral overlaps of the donor fluorescence with the acceptor absorption, between these two systems, so it is concluded that the topology of the protein complexes in both cases, must be very similar. Energy transfer is also monitored by the loss of steady-state fluorescence intensity at 460 nm of the donor, on addition of the acceptor protein. A fluorescence titration indicates that luciferase hydroxyflavin and the yellow protein complex with a 1:1 stoichiometry with a K-d of 0.7 mu M (0 degrees C). These parameters account for the bioluminescence spectral shifting effects observed in these reactions.

Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, ?-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/ 3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physical-mechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB. Аннотация: Strain AIS(T), an aerobic halophilic, Gram-reaction-negative, heterotrophic bacterium isolated from the water of Shira Lake in Khakasia, southern Siberia, was characterized using a polyphasic approach. Our analysis of the 16S rRNA gene sequences showed that 'Aliidiomarina haloalkalitolerans', 'Allidiomarina sanyensis', Idiomarina maris and AIS(T) formed a distinct lineage. The sequence similarities between AIS(T) and the type strains of species of the genera Idiomarina and Aliidiomarina were 91.6-95.1 % and 94.0-96.9 %, respectively. The major isoprenoid quinone of AIS(T) was ubiquinone 8 (Q-8). Predominant cellular fatty acids were iso-C-17:0, iso-C-15:0 and summed feature 9. The genomic DNA G+C content was 45.8 mol%. It is concluded that AIS(T) represents a novel species of the genus Aliidiomarina, and the name Aliidiomarina shirensis sp. nov. is herein proposed for it. The type strain is AIST (=JCM 17761(T)=BCRC 80327(T)). Based on its fatty acid profile and our phylogenetic analysis, we propose that Idiomarina mans be transferred to the genus Aliidiomarina.

Аннотация: The yellow bioluminescence Y1 strain of Vibrio fischeri can produce a 22 kDa protein with either FMN or riboflavin as a bound fluorophore. Both forms are active for shifting the bioluminescence spectral maximum. The fluorescence spectral distribution of the two proteins differs slightly and the in vivo emission appears to be an equal mixture of the two. The bioluminescence activity of the riboflavin Y1 protein contrasts with the inactivity of the related Photobacterium type.

Аннотация: Several flavoproteins and cytochromes that occur as major components in extracts of the yellow bioluminescence Y1 strain of the murine bacterium Vibrio fischeri have been purified and characterized with respect to their mass (SDS/PAGE) and matrix-assisted laser-desorption/ionization MS), chromatographic properties, N-terminal sequence, and spectroscopy (absorption, fluorescence emission and anisotropy decay). The investigated proteins were as follows: yellow fluorescence protein (YFP) with bound riboflavin, FMN or 6,7-dimethyl-8-ribityllumazine; a blue fluorescence protein (BFP) with bound 6,7-dimethyl-8-ribityllumazine, riboflavin, or 6- methyl-7-oxo-ribityllumazine; thioredoxin reductase with FAD as ligand; and two c-type diheme cytochromes, c551 and c554. We present evidence that the riboflavin-bound YFP has an N-terminal sequence corresponding to that published for the dimeric YFP. We show that an equilibrium replacement of the riboflavin can be made with excess lumazine derivative and that lumazine- bound YFP has different bioluminescence properties to those of the lumazine protein from Photobacterium leiognathi. BFP is a different protein again, and in the bacterial lysate it occurs in multiple forms, ligated to either riboflavin, lumazine, or t he 7-oxolumazine derivative. The N-terminal sequence for BFP-shows similarities to those of the YFP proteins and to lumazine protein and riboflavin synthase from Photobacterium. BFP in any form has no bioluminescence or riboflavin-synthase activity. A 70-kDa fluorescent flavoprotein with FAD as ligand has an N-terminal sequence highly similar to those of thioredoxin reductases from Haemophilus influenza and Escherichia coli. Cytochrome contaminations in previous preparations of YFP have been removed and an identified as the two c-type cytochromes c551 and c554. Both inhibit the NADH-induced bioluminescence in the reductase/luciferase system with the luciferase from P. leiognathi and V. fischeri. The N-terminal amino acid sequence of the cytochrome (c551) corresponds to a diheme cytochrome c4. The spectral properties of c554 are similar to those of other c5 cytochromes, and both c554 and c551 have absorption spectra similar to those of the respective cytochromes from the gram-negative bacteria Pseudomonas and Azotobacter.

Аннотация: Two chemolithoautotrophs - the hydrogen-oxidizing bacterium Alcaligenes eutrophus and the carboxydobacterium Seliberia carboxydohydrogena - were investigated with respect to their ability to produce heteropolymeric polyhydroxyalkanoates (PHA) during mixotrophic growth on CO2 and medium-chain-length (C6 to C9) alkanoic acids. Hydroxy derivatives of the alkanoic acids were incorporated into PHA synthesized by the bacteria studied in proportions dependent on the time of cultivation. Irrespective of the nature of the acid added to the medium, PHAs were found to contain hydroxybutyrate as the main constituent and also hydroxyvalerate and hydroxyhexanoate, whose maximal incorporation was observed 4 to 6 h after the addition of an alkanoic acid to the medium. During further cultivation, the fractions of hydroxyvalerate and hydroxyhexanoate in PHA decreased, and the fraction of hydroxybutyrate and the total intracellular content of the polymer increased. Three-component polyhydroxyalkanoates with different proportions of hydroxybutyrate, hydroxyvalerate, and hydroxyhexanoate were obtained, and some of their properties were analyzed. Аннотация: -Growth of the hydrogen-oxidizing bacterium Alcaligenes eutrophus under nonoptimum conditions (acidic and alkaline pH, elevated and lowered temperatures, absence of exogenous sources of carbon and energy) was studied in relation to the initial intracellular concentration of poly-?-hydroxybutyrate (PHB), which varied from virtually zero, to 60-70% of the dry biomass. An initial PHB pool of greater than 30-40% provided for a 2- to 2.5-fold higher biomass yield in acidic and alkaline media or at nonoptimum temperatures as compared with cultures initially containing no pommer. The intracellular PHB pool also provides for the growth of the culture in the absence of exogenous carbon sources and its survival (without growth) with no exogenous energy supply.

Аннотация: The intracellular degradation of poly-?-hydroxybutyrate (PHB) in Alcaligenes eutrophus culture was found to occur at different rates, depending on growth conditions. The degradation of PHB probably allows protein synthesis and cell growth under nonoptimal environmental conditions. The isolated native granules of poly-?-hydroxybutyrate were stable, unless they were isolated from biomass subjected to freezing-thawing treatment. Degradation of isolated granules could also be stimulated by increasing the ionic strength of the medium or adding cell-free extracts. It is anticipated that bacterial cells contain soluble factors that either stimulate or inhibit the degradation of PHB granules.

Аннотация: Some morphological, physiological, and biochemical manifestations of heterogeneity in the population of the hydrogen-oxidizing bacterium Alcaligenes eutrophus Z1 were studied. The population dissociated into R and S variants differing in the size, appearance, and architectonics of colonies, cell ultrastructure, growth rate, enzyme activity, and chemical composition. Аннотация: The inhibition of bacterial luminescence has been used in testing industrial enterprises sewage. The toxicity of the sewage is less than the total toxicity of separate components due to neutralization of quinone products of polyphenol oxidation in the reactions with the other phenol components of sewage. Toxicity increase is due to their influence on the cell membrane. Studies of cell ultrastructure confirm this fact. The studied mechanism of the complex effect allowed a more accurate forecast of the ecological situation during the discharge of phenol compounds and metals. It also showed the necessity of taking into account the complex effect of sewage components on contaminant discharge into water reservoirs.

Аннотация: The growth of the carboxydobacterium Seliberia carboxydohydrogena Z-1062 was studied at a different concentration of mineral elements under the conditions of continuous turbidostat cultivation. The chemical composition of the bacterium was determined and the dynamics of its specific growth rate and the intracellular content of mineral elements were studied when the concentration of these elements was changed in the growth medium. The synthesis of 1 g of biomass by this strain required: N, 120 mg; P, 23 mg; S, 5.5 mg; K, 1.5 mg; Mg, 3.5 mg. Changes in the residual concentrations of the elements within a broad range had no effect on the specific rate of the carboxydobacterial growth and chemical composition.
