Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Аннотация: Biodegradable poly-3-hydroxybutyrate [P(3HB)] and natural fillers - clay, peat, and birch wood flour – were used to prepare powdered composites to form pellets and granules. Pellets were produced by cold pressing of polymer and filler powder whereas granules were produced from the powders wetted with ethanol. Characterization techniques like IR spectroscopy, differential scanning calorimetry, X-ray analysis, mechanical analysis and electron microscopy were employed to study the properties of the initial P(3HB) and fillers and the composites. Analysis of the IR spectra of the composites showed the absence of chemical bonds between the components, i.e. the composites were physical mixtures. Young's moduli of the pellets prepared from initial materials varied considerably, and the highest value was obtained for P(3HB) pellets (350 MPa). Studies of biodegradation of composite pellets and granules in the soil for 35 days showed that the residual mass of the pellets had decreased to 68% for P(3HB); 56.4% for P(3HB)/peat; 67% for P(3HB)/wood flour, and 64% for P(3HB)/clay; granules exhibited a similar mass loss, residual mass of the granules of P(3HB) was 68.4%, P(3HB)/peat 46.4%; P(3HB)/wood flour 77%, and P(3HB)/clay 74%. This shows the significance of the material as an eco-friendly composite without sacrificing its mechanical properties. © 2018

Аннотация: Using experimental microecosystems the kinetics of phenol disappearance in small forest pond waters (Siberia, Russia) in the summer of 1995-96 were investigated. Despite of high variability of components of the ecosystem (plankton biomass and species composition) and two pronounced 'blooms' of green algae Volvox aureus the same kinetics of the disappearance took place over the investigated period. Half-lives of the pollutant depended on water temperature only. A comparison of the self-purification of the pond with that of the Krasnoyarsk reservoir, 'blooming' with blue-greens was carried out. Half-lives in the pond were significantly lower than that in the reservoir. During the periods of 'blooms' of the green algae in the pond the concentrations of inorganic nutrients were comparatively high and the phenol-degrading bacteria likely were not limited by these nutrients, in contrast to the periods of 'bloom' of the blue-green algae in the reservoir.

Аннотация: The paper considers problems of biosynthesis of higher plants' biomass and "biological incineration" of plant wastes in a working physical model of biological LSS. The plant wastes are "biologically incinerated" in a special heterotrophic block involving Califomian worms, mushrooms and straw. The block processes plant wastes (straw, haulms) to produce soil-like substrate (SLS) on which plants (wheat, radish) are grown. Gas exchange in such a system consists of respiratory gas exchange of SLS and photosynthesis and respiration of plants. Specifics of gas exchange dynamics of high plants - SLS complex has been considered. Relationship between such a gas exchange and PAR irradiance and age of plants has been established. Nitrogen and iron were found to the first to limit plants' growth on SLS when process conditions are deranged. The SLS microflora has been found to have different kinds of ammonifying and denitrifying bacteria which is indicative of intensive transformation of nitrogen-containing compounds. The number of physiological groups of microorganisms in SLS was, on the whole, steady. As a result, organic substances - products of exchange of plants and microorganisms were not accumulated in the medium, but mineralized and assimilated by the biocenosis. Experiments showed that the developed model of a man-made ecosystem realized complete utilization of plant wastes and involved them into the intrasystem turnover. Аннотация: Chronic experiments on mature Wistar rats studied the biological properties of suture threads from two types of polyoxyalkanoates (POA), such as polyoxybutyrate (POB) and the copolymer of POB and polyoxyvalerate (POV) (POB/POV) as compared to the conventional surgical material (silk and catgut). The physiological and biochemical parameters that reflect the status of the animals, including their behavior, height and development, the weight of the body and viscera, the morphology of peripheral blood, the biochemical parameters of blood, the activity of some enzymes, as well as histological findings of the viscera, and lymphoid tissue responses were assessed. POA was found to produce no negative effect on the physiological, biochemical, and functional characteristics of the animals irrespective of the duration of contact with the internal medium of the body and of the chemical composition of a material.

Аннотация: Biodegradation of polyhydroxyalkanoates (PHA) - polymers of hydroxylated fatty acids - differing in chemical composition, crystallinity, melting temperature, and molecular weight was studied in soil and water environments under conditions of varying ambient temperature and pH. Under similar conditions, homogeneous films of polyhydroxybutyrate (PHB) with different initial properties were degraded at virtually equal rates. Hydroxybutyrate-hydroxyvalerate copolymers with a lower crystallinity and different microstructure in comparison to PHB were degraded at a 20 to 30% higher rate on average. The ambient temperature significantly influenced the rate of PHA biodegradation.

    Specific character of bacterial biodegradation of polyhydroxyalkanoates with different chemical structure in soil / S. V. Prudnikova, O. N. Vinogradova, M. Y. Trusova // Doklad. Biochem. Biophys. - 2017. - Vol. 473, Is. 1. - P94-97, DOI 10.1134/S1607672917010185 . - ISSN 1607-6729
Аннотация: The study addresses the influence of the physicochemical properties of the reserve cellular macromolecules (polyhydroxyalkanoates, PHA) with different chemical composition on their biodegradation in the agro-transformed field soil of the Siberian region (Krasnoyarsk Territory, Russia). It was shown that the degradation of the PHA samples depends on the degree of polymer crystallinity (Cx). For the first time, it was shown that the range of PHA-degrading microorganisms differs for each of PHA types. The study defines the primary degraders specific to each PHA type and common to all types of examined polymers. © 2017, Pleiades Publishing, Ltd.

Аннотация: Bioregenerative life-support systems (BLSS) are studied for developing the technology for a future biological life-support system for long-term manned space missions. Ways to utilize human liquid and solid wastes to increase the closure degree of BLSS were investigated. First, urine and faeces underwent oxidation by Kudenko's physicochemical method. The products were then used for root nutrition of wheat grown by the soil-like substrate culture method. Two means of eliminating sodium chloride, introduced into the irrigation solution together with the products of urine oxidation, were investigated. The first was based on routine electrodialysis of irrigation water at the end of wheat vegetation. Dialysis eliminated about 50% of Na from the solution. This desalinization was performed for nine vegetations. The second method was new: after wheat cultivation, the irrigation solution and the solution obtained by washing the substrate containing mineral elements not absorbed by the plants were used to grow salt-tolerant Salicornia europaea L. plants (saltwort). The above-ground biomass of this plant can be used as a food, and roots can be added to the soil-like substrate. Four consecutive wheat and Salicornia vegetations were cultivated. As a result of this wheat and Salicornia cultivation process, the soil-like substrate salinization by NaCl were considerably decreased. В© 2008 Humana Press.

Аннотация: Studying material transformations and biotic cycling in artificial ecosystems (AES), we need to know the principles of biological adaptation of active organisms to change in the environment. Microorganisms in AES for water purification are the most active transforming organisms and consumers of the organic substances contained in wastes. Utilization of organic substances is directly connected with the energy fluxes used by AES. According to energy criteria, the energy fluxes used by a biological system tend to reach maximum values under stable conditions. Unutilized substrate concentration decreases as a result of biological adaptations. After a dramatic change in environmental factors, for example, after a sharp increase in the flow rate of organic substances, the biological system is not able to react quickly. The concentration of unutilized substrate increases and the energy flux used by the biological system decreases. The structure of the microbial community also changes, with a decrease in biological diversity. The efficiency of energy use by simple terrestrial ecosystems depends on the energetic intensity and interactions between plants and rhizospheric microorganisms. В© 2005 COSPAR. Аннотация: As intracellular carbon and energy storage materials, polyhydroxyalkanoates (PHA) are a diverse biopolyesters synthesized by many bacteria. PHA have been produced in large quantity for various application research including medical implants for approximately 30 years. Many studies demonstrated that PHA are promising implant materials due to their diverse and ascendant mechanical, biodegradable and tissue compatible properties. Importantly, common PHA biodegradation products including oligomers and monomers are also not toxic to the cells and tissues. Pharmaceutical applications of some PHA degradation products also have been reported. So far, no study has been reported to have any carcinogenesis result induced by any PHA or their biodegradation products. All results suggest that PHA could be developed into various bio-implant products.

Аннотация: Composite films and nonwoven mats of the poly-3-hydroxybutyrate and chitosan natural polymers were prepared and investigated. DSC and FTIR techniques were used to confirm that P(3HB) blending with chitosan resulted in a decrease in P(3HB) crystallinity to 47% and 62% in the films and nonwoven mats, respectively. Scanning electron microscopy showed that addition of chitosan induced changes in the surface morphology of the composite films and a reduction in the diameter of ultrafine fibers in the nonwoven mats from 800 nm to 460 nm. The values of water contact angle for films (53°) and nonwoven mats (50.6°) suggested that chitosan enhanced hydrophilic properties and moisture absorption capacity of the composite materials. On the other hand, P(3HB) showed its reinforcing ability and improved the physical/mechanical properties of chitosan. The work included studies of in vitro biodegradation of the composite specimens and their ability to maintain cell growth and attachment in NIH 3T3 fibroblast culture. © 2020

Аннотация: The integrated study addressing biodegradation of microbial linear polyesters of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) in tropical conditions by microbial communities of Vietnamese soils was performed in locations close to Hanoi and Nha Trang, which differed in their weather conditions and microbial communities. It shows that PHA degradation in tropical soils is influenced by polymer chemical composition, specimen shape, and microbial characteristics. The homopolymer of 3-hydroxybutyric acid is degraded at higher rates than the copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids. The average rates of mass loss were 0.04-0.33% per day for films and 0.02-0.18% for compact pellets. PHA degradation was accompanied by a decrease in the polymer molecular mass and, usually, an increase in the degree of crystallinity, suggesting preferential degradation of the amorphous phase. Under the study conditions, representatives of the bacterial genera Burkholderia, Bacillus, Cupriavidus, Mycobacterium, and Nocardiopsis and such micromycetes as Acremonium, Gongronella, Paecilomyces, and Penicillium, Trichoderma have been identified as major PHA degraders. Аннотация: The present study investigates, for the first time, the structure of the microbial community of cryogenic soils in the subarctic region of Siberia and the ability of the soil microbial community to metabolize degradable microbial bioplastic – poly-3-hydroxybutyrate [P(3HB)]. When the soil thawed, with the soil temperature between 5-7 and 9–11 °C, the total biomass of microorganisms at a 10-20-cm depth was 226–234 mg g?1 soil and CO2 production was 20–46 mg g?1 day?1. The total abundance of microscopic fungi varied between (7.4 ± 2.3) ? 103 and (18.3 ± 2.2) ? 103 CFU/g soil depending on temperature; the abundance of bacteria was several orders of magnitude greater: (1.6 ± 0.1) ? 106 CFU g?1 soil. The microbial community in the biofilm formed on the surface of P(3HB) films differed from the background soil in concentrations and composition of microorganisms. The activity of microorganisms caused changes in the surface microstructure of polymer films, a decrease in molecular weight, and an increase in the degree of crystallinity of P(3HB), indicating polymer biodegradation due to metabolic activity of microorganisms. The clear-zone technique – plating of isolates on the mineral agar with polymer as sole carbon source – was used to identify P(3HB)-degrading microorganisms inhabiting cryogenic soil in Evenkia. Analysis of nucleotide sequences of rRNA genes was performed to identify the following P(3HB)-degrading species: Bacillus pumilus, Paraburkholderia sp., Pseudomonas sp., Rhodococcus sp., Stenotrophomonas rhizophila, Streptomyces prunicolor, and Variovorax paradoxus bacteria and the Penicillium thomii, P. arenicola, P. lanosum, Aspergillus fumigatus, and A. niger fungi. © 2020 Elsevier Ltd
