Аннотация: Cold extract from bioluminescent earthworm Henlea sp. was studied by HPLC, 1D and 2D NMR and LC-HRMS analysis. An abundant structural analog of the luciferin was isolated and identified as ?-C-mannosyltryptophan (ManTrp), the product of unusual C2-glycosylation found earlier in humans, ascidians and other animals. Two compounds in cold extract (P300b, P300c) were characterized as C2-substituted derivatives of tryptophan. We hypothesize that a series of tryptophan-containing compounds are possible participants of bioluminescence-related metabolism in Henlea sp. © 2020 Wiley-VCH GmbH

Аннотация: The world map of anthropogenic atmospheric nitrogen deposition and its effects on natural ecosystems is not described with equal precision everywhere. In this paper, we report atmospheric nutrient, sulfate and spheroidal carbonaceous particle (SCP) deposition rates, based on snowpack analyses of a formerly unexplored Siberian mountain region. Then, we discuss their potential effects on lake phytoplankton biomass limitation. We estimate that the nutrient depositions observed in the late-season snowpack (40 +/- 16 mgNO(3)-Nm(-2) and 0.58 +/- 0.13 mg TP-Pm-2; TP for total phosphorous) would correspond to yearly depositions lower than 119 +/- 71 mgNO(3)-Nm(-2) yr(-1) and higher than 1.71 +/- 0.91 mg TP-Pm-2 yr(-1). These yearly deposition estimates would approximately fit the predictions of global deposition models and correspond to the very low nutrient deposition range, although they are still higher than world background values. In spite of the fact that such a low atmospheric nitrogen deposition rate would be enough to induce nitrogen limitation in unproductive mountain lakes, phosphorus deposition was also extremely low, and the resulting lake water N: P ratio was unaffected by atmospheric nutrient deposition. In the end, the studied lakes' phytoplankton appeared to be split between phosphorus and nitrogen limitation. We conclude that these pristine lakes are fragile sensitive systems exposed to the predicted climate warming, increased winter precipitation, enhanced forest fires and shifts in anthropogenic nitrogen emissions that could finally couple their water chemistry to that of atmospheric nutrient deposition and unlock temperature-inhibited responses of phytoplankton to nutrient shifts.

Аннотация: Two kinds of Ca(2+)-regulated photoprotein obelin with altered color of bioluminescence were obtained by active-center amino acid substitution. The mutant W92F-H22E emits violet light (lambda(max)=390 nm) and the mutant Y139F emits greenish light (lambda (max)=498 nm), with small spectral overlap, both display high activity and stability and thus may be used as reporters. For demonstration, the mutants were applied in dual-color simultaneous immunoassay of two gonadotropic hormones-follicle-stimulating hormone and luteinizing hormone. Bioluminescence of the reporters was simultaneously triggered by single injection of Ca(2+) solution, divided using band-pass optical filters and measured with a two-channel photometer. The sensitivity of simultaneous bioluminescence assay was close to that of a separate radioimmunoassay.

Аннотация: The toxicity, cytotoxicity, and genotoxicity of bulk sediments from the Yenisei River (Siberia, Russia) were estimated in laboratory bioassays based on several endpoints in the aquatic plant Elodea canadensis. The bottom sediment samples were collected in the Yenisei River upstream and downstream of the sources of chemical and radioactive contamination. Fluorescence spectral relaxation of the protein-bound ligands is largely complete within 1 ps but reveals a small red shift with a minor, but distinctly longer, relaxation time than that of the free ligands, which is tentatively assigned to the relaxation of protein-bound water in the vicinity of the excited chromophore.

Аннотация: The solvation dynamics of interesting bioluminescent chromophores have been determined, using subpicosecond and wavelength-resolved fluorescence spectroscopy, in combination with global analysis of the multidimensional data sets. The systems investigated comprise the free ligands 6,7-dimethyl-(8-ribityl)-lumazine (lumazine) and riboflavin in an aqueous buffer and both ligands when noncovalently bound to two bacterial bioluminescent antenna proteins: lumazine protein (from Photobacterium leiognathi) and the blue fluorescent protein (from Vibrio fischeri Y1). Аннотация: Triploid fish has become an important item of commercial aquaculture, but data on its fatty acid (FA) composition are still controversial, especially regarding essential polyunsaturated fatty acids, eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA). We studied FA composition and content of diploid and triploid pink salmon Oncorhynchus gorbuscha, reared in aquaculture in a bay of the White Sea (Russia). FA composition, measured as percentages of total FA of triploids and immature diploid females significantly differed from that of mature diploid fish. Specifically, mature diploids had higher percentage of EPA and DHA in their muscle tissue (filets) compared to that of triploids and immature diploid females. Nevertheless, the contents of EPA and DHA per mass of the filets in diploid and triploid specimens were similar. Thus, no special efforts are needed to improve EPA and DHA contents in filets of triploids. © 2016 Elsevier Ltd

Аннотация: The paper deals with the study of the synthesis of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) copolymers by the bacterium Ralstonia eutropha B-5786 grown under different carbon nutrition conditions (growth on carbon dioxide, fructose, and CO2-valerate and fructose-valerate mixtures). The parameters to be analyzed included the yield of biomass, the yield, synthesis rate, and composition of copolymers, the activity of the key enzymes of polyhydroxyalkanoate (PHA) synthesis (?-ketothiolase, acetoacetyl-CoA reductase, and PHA synthase), the maximum tolerable concentration of valerate to the bacterium, and the conditions that govern the incorporation of hydroxyvalerate to copolymers. This allowed the relationship between cultivation conditions and the proportion of monomers in the copolymers to be deduced. We were able to synthesize a range of 3HB/3HV copolymers and found that the thermal characteristics and the degree of crystallinity of these copolymers depend on the molar fraction of 3HV.

Аннотация: Interactions between phytoplankton and zooplankton affect the overall functioning of lakes. Herbivores are habitually confronted with food of inferior quality, usually a result of low nutrient concentrations in plant material. Large-bodied cladocerans are better competitors for food than small-bodied species but they are more vulnerable to low food quality. Understanding the effects of food quality on zooplankton structure and competition between small - large bodied herbivorous is of considerable interest. We want to find out how differences in C:N:P ratios between phytoplankton and zooplankton communities affect their abundances in a freshwater food web. We want also to assess the role of phytoplankton and zooplankton as sinks of the phosphorus and nitrogen. Therefore, we conducted a 31-day mesocosms experiment with water from a mesotrophic and a eutrophic lake (with natural plankton communities). To simulate changes in the plankton communities large-bodied Daphnia magna and Daphnia pulicaria were added. Samples for zooplankton, phytoplankton and water chemistry were taken every 10 days. Samples for elemental analysis (C:N:P) of seston and zooplankton were collected on the first, and on the final day of the experiment. Our mesocosms experiment showed mismatch in C:P between seston (high) and zooplankton (low), which suggests that most of the phosphorus is incorporated in zooplankton biomass. This evidenced that zooplankton is an effective sink of phosphorus, while nitrogen is accumulated mainly by primary producers. Our results also indicated more stability in stoichiometry with increasing trophic levels of organisms. However, there were significant changes in the zooplankton structure. The increasing dominance of large Daphnia resulted in reduction of C:P ratio in zooplankton. Low food quality (C:P) did not limit the growth of large Daphnia in the experimental conditions, which competed effectively with small planktonic cladocerans and with Rotifera. Over time, inedible algae began to dominate resulting in increase of relative biomass of periphyton grazers, which suggests that plankton community is transformed into littoral system in mesocosms for about 30 days. © 2019

Аннотация: Light-sensitive Ca2+-regulated photoprotein berovin is responsible for the bioluminescence of the ctenophore Beroe abyssicola. It shares many properties of hydromedusan photoproteins although the degree of identity of its amino acid sequence with those of photoproteins is low. There is a hydrogen bond between C-terminal Pro and Arg situated in the N-terminal ?-helix of hydromedusan photoproteins that supports a closed conformation of the internal cavity of the photoprotein molecule with bound 2-hydroperoxycoelenterazine. The C- and N-terminal hydrogen bond network is necessary to properly isolate the photoprotein active site from the solvent and consequently to provide a high quantum yield of the bioluminescence reaction. In order to find out which berovin residues perform the same function we modified the N- and C-termini of the protein by replacing or deleting various amino acid residues. The studies on berovin mutants showed that the interaction between C-terminal Tyr208 and Tyr13 localized in the first ?-helix of the photoprotein is important for the stabilization and proper orientation of the oxygenated coelenterazine adduct within the internal cavity as well as for supporting the closed photoprotein conformation. We also suggest that the interplay between Tyr residues in ctenophore photoproteins occurs rather through the ?-? interaction of their phenyl rings than through hydrogen bonds as in hydromedusan photoproteins.
