Аннотация: Currently, the patients with purulent wounds account for about 40% of the patients of surgical profilt. Since the treatment of septic wounds under the bandage remains the main method, as it is the most convenient and economically beneficial, but not effective enough, the development of new advanced coatings possessing simultaneously by multiple properties, is considered to be relevant. In the process of wound healing a physician faces with many problems including the issue of the selection option of a wound dressing, as they differ significantly in their design and properties. A disadvantage of many wound dressings for wound care is the adherence to wound. This results in the regenerating tissue injury and the dressings become sore. Many of the positive properties of dressings reduced as a result of damage caused by daily dressings. In recent years the biocompatible materials for injuries and wounds contribute to more effective healing and regeneration. In this regard, the paper presents the main types of current wound dressings used in the treatment of purulent wounds, their properties, indications and methods of applicatio.

Аннотация: The effects of actinic light (AL) intensity on the age dependence of nonphotochemical fluorescence quenching (q(N)) and effective quantum yield in PSII (I broken vertical bar(PSII)) were studied in continuously illuminated wheat leaves of the upper tier. Regular changes were revealed in both age dependence of q(N) at elevated AL intensities and light curves of q(N). These changes are related to alterations in strategies of redistribution and use of absorbed light energy by the photosynthetic apparatus at different stages of wheat leaf development. Unlike I broken vertical bar(PSII), q(N) as a parameter was more sensitive to the differences in the leaf age at a certain range of light intensities. At the same time, the stability of q(N) at moderate light intensities may serve as an indication of leaf maturity.

Аннотация: This paper considers an approach to the description of the age-specific and time-dependent dynamics of cancer incidence. This approach is based on a model that describes the age-specific dynamics of cancer as a phase transition of the second order and is widely used in the study of physical systems. The proposed model of cancer development as the second-order phase transitions is quite consistent with the data of medical statistics, is well described using only two free parameters, easily verified by the statistical data, and is interpreted well. The applicability of the model of second-order phase transitions to the description of the processes in the nonphysical systems is likely to be determined by the versatile nature of the processes that occur during the phase transitions. © 2015, Pleiades Publishing, Inc.

Аннотация: Biodegradable polymer poly(3-hydroxybutyrate) (P3HB) has been used as a matrix to construct slow-release formulations of the fungicide tebuconazole (TEB). P3HB/TEB systems constructed as films and pellets have been studied using differential scanning calorimetry, X-ray structure analysis, and Fourier transform infrared spectroscopy. TEB release from the experimental formulations has been studied in aqueous and soil laboratory systems. In the soil with known composition of microbial community, polymer was degraded, and TEB release after 35 days reached 60 and 36 % from films and pellets, respectively. That was 1.23 and 1.8 times more than the amount released to the water after 60 days in a sterile aqueous system. Incubation of P3HB/TEB films and pellets in the soil stimulated development of P3HB-degrading microorganisms of the genera Pseudomonas, Stenotrophomonas, Variovorax, and Streptomyces. Experiments with phytopathogenic fungi F. moniliforme and F. solani showed that the experimental P3HB/TEB formulations had antifungal activity comparable with that of free TEB.

A wide coverage of recent biosensing approaches involving aptamers, enzymes, DNA probes, fluorescent probes, interacting proteins and antibodies in vicinity to transducers such as electrochemical, optical and piezoelectric is presented. Recent advanced developments in biosensing approaches for cancer biomarker owes much credit to functionalized nanomaterials due to their unique opto-electronic properties and enhanced surface to volume ratio. Biosensing methods for a plenty of cancer biomarkers has been summarized emphasizing the key principles involved. © 2016 Elsevier B.V.

Аннотация: Adsorption of viral particles from the blood plasma of patients with viral hepatitis B and C on modified nanodiamonds (MNDs) was shown in the in vitro experiments. PCR method showed the treatment of plasma with MNDs leads to a decrease in the viral load by 2–3 orders of magnitude or more in both cases studied. These results make it possible to predict the applicability of MNDs for the development of new technologies of hemodialysis and plasmapheresis for binding and removal of viral particles from the blood of infected patients. © 2016, Pleiades Publishing, Ltd.

Аннотация: The study addresses the effectiveness of using ion exchange substrates (IES) to optimize mineral nutrition of plants grown in the nutrient solutions containing oxidized human wastes for application in bio-technical life support systems. The study shows that the addition of IES to the root-inhabited substrate is favorable for the growth of wheat vegetative organs but causes a decrease in the grain yield. By contrast, the addition of IES to the nutrient solution does not influence the growth of vegetative organs but favors normal development of wheat reproductive organs. Thus, to choose the proper method of adjusting the solution with IES, one should take into account specific parameters of plant growth and development and the possibility of multiple recycling of IES based on the liquid products of mineralization of human wastes. © 2016 IAA. Аннотация: The question on the essence of life as phenomenon is the key one for astrobiology, since the answer to this question determines "breadth of our outlook". Taking Earth's version of life as the pattern extremely under-estimates our estimation of the probability of life origin and respectively expected probability of extraterrestrial life discovery. In the paper the hypothetical key attribute of life in general is selected on the base of comparative analyses and deductive inference. Simulation conducted on the base of neural network model shows that the same function could be realized by means of great variety of structures, which originated in the course of an evolutionary process. So multiplicity of evolutionary outcomes essentially increases the probability of final result - realization of an integrated function providing fitness to environment. Life as the integrated function can be realized via great variety of development ways and structures. A logical consequence of definitions for life as phenomenon is suggested. Final one is "Life is specific organization of informational and energetic processes coupling, enabling choice-making, and displayed as anomalies of different kinds". Anomalies of visible form, mechanical movement, chemical composition and noticeable response are considered. Presented in the paper sweeping generalization is not rigorously proven, however it can play heuristic role in increasing the level of specificity of searching for extraterrestrial life. Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types-polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV)-featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 mum thick, which became as thin as 40-60 mum after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner. Аннотация: The anthropogenic impact on the Earth's ecosysterns are leading to dramatic changes in ecosystem functioning and even to destruction of them. System analysis and the use of heuristic modeling can be an effective means to determine the main biological interactions and key factors that are of high importance for understanding the development of ecosysterns. Cycling of limiting substances, induced by the external free energy flux, and trophic links interaction is the basis of the mathematical modeling studies presented in this paper. Mathematical models describe the dynamics of simplified ecosysterns having different characteristics:1) different degrees of biotic turnover closure (from open to completely closed); 2) different numbers of trophic links (including both "top-down", "bottom-up" regulation types); 3) different intensities of input - output flows of the limiting nutrient and its total amount in the system. Adaptive values of the changes of lower hierarchical levels (populational, trophic chain level) are to be estimated by integrity indices for total system functioning (e.g. NPP, total photosynthesis). The approach developed can be used for evaluating the contributions of lower hierarchical levels to the functioning of the higher hierarchical levels of the system. Аннотация: Seasonality of burden and prevalence of phototrophic (microalgal) epibionts Characidiopsis ellipsoidea, Colacium vesiculosum and Colacium sp. on dominating crustacean zooplankton (Daphnia longispina, Cyclops vicinus and Mesocyclops leuckarti) were studied in a small reservoir Bugach with cyanobacterial bloom. The correlations between the seasonal dynamics of prevalence and the dynamics of others biotic and abiotic factors were calculated. The conclusions were as follows. The substrate species, that determined the development of the epibionts on the three studied crustacean zooplankton, was Daphnia longispina (Cladocera). Despite intensive epibiotic infestation of crustacean zooplankton, epibionts did not appear to have caused non-consumptive mortality of the crustacean zooplankton. But they could have contributed to the Daphnia summer decline by increasing mortality due to its consumption by planktivorous fishes. The phototropic epibionts may successfully coexist with cyanobacterial bloom. The possible role of the epibionts in changing nutrient fluxes in pelagic food web is discussed.

Аннотация: An idea of the general evolution through the long-term response of the Earth to the external flow of radiant energy from the Sun is proposed. Due to the finiteness of matter on Earth, as well as on any other planet, the continuous pumping flow of radiant energy has been shown to lead to cyclization of transformations and mass transfer along the emerging gradients. The evolution of the energy–matter interaction follows the pathway of capturing and transferring more energy by a smaller quantity of matter, i.e., the pathway of the increase in the amount of energy used by each unit mass. According to this parameter, the least effective mass transfer is a simple transfer as vortices of gases along the gradients of temperature and pressure, which took place on the primary surface of the planet. Long-term natural selection towards water accumulation on the planet has played a special role in the development of the interaction between energy and matter. Phase transitions (ice, water, and vapor) and mechanical transfers are the most common energy–matter processes. Chemical transformation of substances became possible based on water cycles, cyclic transfers, and transformations and developed with time into biological transformation. This type of energy–matter interaction is the most efficient. In particular, the energy of our star is captured during photosynthesis and utilized in the most active region of its radiation spectrum. During the biological evolution of heterotrophs, a increase in the coefficient that characterizes the energy exchange intensity from protozoa to mammals by several hundred times is most illustrative. The development and current dominance of humans as the species that is most active in the capturing of energy and meaningful organization of its new flows, in particular, based on the organic debris of former biospheres, is amazing but quite natural from the energy standpoint. During the technological evolution of humankind, the energy-exchange intensity for homoiotherms (warm-blooded animals) has increased by 20 times if it is recalculated for the technological energy that is used by the average inhabitant of the Earth. Thus, the victory of our species in planetary evolution fits well into the mainstream of the general evolution through energy–matter interactions: a multiple increase in star energy has been used to transform the matter on the surface of the irradiated planet. © 2015, Pleiades Publishing, Inc.

Аннотация: Coelenterazine-dependent copepod luciferases containing natural signal peptide for secretion are a very convenient analytical tool as they enable monitoring of intracellular events with high sensitivity, without destroying cells or tissues. This property is well suited for application in biomedical research and development of cell-based assays for high throughput screening. We report the cloning of cDNA gene encoding a novel secreted non-allelic 16.5-kDa isoform (MLuc7) of Metridia longa luciferase, which, in fact, is the smallest natural luciferase of known for today. Despite the small size, isoform contains 10 conservative Cys residues suggesting the presence of up to 5 S-S bonds. This hampers the efficient production of functionally active recombinant luciferase in bacterial expression systems. With the use of the baculovirus expression system, we produced substantial amounts of the proper folded MLuc7 luciferase with a yield of similar to 3 mg/L of a high purity protein. We demonstrate that MLuc7 produced in insect cells is highly active and extremely thermostable, and is well suited as a secreted reporter when expressed in mammalian cells ensuring higher sensitivity of detection as compared to another Metridia luciferase isoform (MLuc164) which is widely employed in real-time imaging. Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.

Аннотация: Only males (fully mature and failing to mature) and intersexes have been found among triploids of pink salmon. Possible mechanisms of genetic sex determination in pink salmon are discussed. The possibility of using triploids to regulate sizes of artificial pink salmon populations is also discussed. Triploids of pink salmon are deemed inefficient to use in aquaculture. Delayed maturation is observed in some diploid females reared in farming cages.

Аннотация: The structure and the first step of hydrolysis of hydrated Pu(IV) ion in aqueous medium were studied using relativistic density functional theory and the polarizable continuum model (PCM). According to calculations, the Pu(IV) ion was mainly coordinated with eight water molecules. The calculated distance Pu-O of 238-241 pm was in good agreement with the experimental value ((239 +/- 2) pm). It was demonstrated that for the correct replication of constants log K-1(0) of the first hydrolysis step, it was important to use scaling multipliers adequate to the charge of the hydrated complex when constructing the density that included the solvated complex in PCM model. The calculated values of log K-1(0) of -1.1...-0.2 were close to the experimentally defined range (-0.6-0.6). The correct replication of hydrolysis constants allowed considering the calculated range of the hydratation free energies of the Pu(IV) ion of 6070-6157 kJ/mol as a reasonable prediction of experimental values.

Аннотация: Bioluminescence of isolated enzymes is a perspective phenomenon for biosensors development due to simplicity of registration of a physiological parameter - light intensity. Enzyme-based bioluminescent assay is widely used to evaluate a decrease in biochemical toxicities. Also the enzyme-based assay is used for the direct biochemical monitoring of oxidative toxicity. This work considers antioxidant properties of fullerenols, water-soluble polyhydroxylated derivatives of fullerenes and perspective pharmaceutical agents, in solutions of model inorganic and organic toxicants of oxidative type K-3[Fe(CN)(6)] and 1,4-benzoquinone. Two fullerenol preparations were used: C60O2-4(OH)(20-24) and mixture of two types of fullerenols C60O2-4(OH)(20-24)+C70O2-4(OH)(20-24). The enzyme-based assays showed the peculiarities of the detoxification processes: ultralow concentrations of fullerenols were active (ca 10(-17)-10(-5)g/L); no monotonic dependence of detoxification efficiency on fullerenol concentrations was observed, and detoxification of organic oxidizer solutions was more effective than that of the inorganic oxidizer. The antioxidant effects of highly diluted fullerenol solutions were attributed to hormesis phenomenon; the detoxification was concerned with stimulation of adaptive cellular response under low-dose exposures. Аннотация: SNP genotyping based on the reaction of specific primer extension with the following bioluminescent detection of its products was shown to be potentially applicable for biomedical exploration. The paper describes its elaboration and first application in extensive biomedical research concerning MC1R gene variants’ frequency and associations with clinical characteristics in melanoma patients of Eastern Siberia (Krasnoyarsk region, Russia). Polymorphisms rs 1805007 (R151C), rs 1805008 (R160W), and rs 1805009 (D294H) were detected in 174 DNA samples from patients with histologically proved diagnosis of cutaneous melanoma and in 200 samples from healthy individuals. All the results on bioluminescent SNP genotyping were confirmed by Sanger sequencing. Some features characteristic of the population were found, i.e. melanoma is mostly associated with R160W or R151C while variant D294H is extremely rare; simultaneous carriage of any two investigated variants is also strongly associated with melanoma; R151C is associated with ulceration and consequently the disease course is more aggressive, etc. The design of the technique allows fast evaluation of any known diagnostically important SNP frequencies and associations across population. © 2018 Elsevier B.V.

Аннотация: The production of poly-3-hydroxybutyrate (P3HB) and poly-3-hydroxybutyrate/polyethylene glycol (PEG)-based microparticles, loaded with antitumor drugs paclitaxel (PTX) and 5-Fluorouracil (5-FU) by spray-drying technique, was investigated. The average diameter of microparticles was found to be 3.4 +/- 0.5 pm and zeta potential was about -44 mV. The addition of surfactant PEG did not show any effect on the morphological characteristics of the particles. But the chemical structure of drug influenced on the properties. Microparticles had heterogeneous pores on the surface when the hydrophobic PTX was encapsulated. It was established that the addition of surfactant positively influenced on the properties of particles and led to the loading of 5-FU directly into the matrix. This is confirmed by the results of electron microscopy and dynamics of drug release in vitro. As a whole, the release profiles of PTX and 5-FU from composite P3HB/PEG microparticles were less than from P3HB microparticles. The results of the morphological evaluation of Hela cells demonstrated that the use of cytostatic drugs loaded in P3HB microparticles induces morphological changes associated with apoptosis (chromatin condensation, core fragmentation, margination of nucleus). Thus, the obtained results can serve as the basis for the development of new antitumor drugs of prolonged action, intended for various modes of administration.

Аннотация: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) belong to an important therapeutic class for treatment of type 2 diabetes. Six GLP-1 RAs, each utilizing a unique drug delivery strategy, are now approved by the Food and Drug Administration (FDA) and additional, novel GLP-1 RAs are still under development, making for a crowded marketplace and fierce competition among the manufacturers of these products. As rapid elimination is a major challenge for clinical application of GLP-1 RAs, various half-life extension strategies have been successfully employed including sequential modification, attachment of fatty-acid to peptide, fusion with human serum albumin, fusion with the fragment crystallizable (Fc) region of a monoclonal antibody, sustained drug delivery systems, and PEGylation. In this review, we discuss the scientific rationale of the various half-life extension strategies used for GLP-1 RA development. By analyzing and comparing different approved GLP-1 RAs and those in development, we focus on assessing how half-life extending strategies impact the pharmacokinetics, pharmacodynamics, safety, patient usability and ultimately, the commercial success of GLP-1 RA products. We also anticipate future GLP-1 RA development trends. Since similar drug delivery strategies are also applied for developing other therapeutic peptides, we expect this case study of GLP-1 RAs will provide generalizable concepts for the rational design of therapeutic peptides products with extended duration of action. © 2018 Elsevier B.V.
