Аннотация: The database BiolumBase was designed for the collection and systematization of available information on microorganisms containing bioluminescent systems; it includes two sections: natural and transgenic luminous microorganisms. By now, logic schemes of these sections have been developed, classification of the objects has been performed, ways of presentation of characteristics and structure of fields for input of information have been elaborated, and the necessary program modules have been developed. The database is filled on the basis of published data and our own experimental results; subsequent linkage of the database to the Internet is envisaged. Users will be able to obtain not only catalogues of strains but also information concerning the properties and functions of the known species of luminous bacteria, the structure, regulatory mechanisms, and application of bioluminescent systems and genetically engineered constructions with lux genes, as well as to find references and to search strains by using any set of attributes. The database will provide information that is of interest for the development of microbial ecology and biotechnology, in particular, for the prediction of biological hazard from the application of transgenic strains. Аннотация: A mathematical model concerning the interaction of plants and rhizospheric microorganisms on complete mineral medium and under nitrogen limitation has been constructed. The model takes into account the closeness of plants and microorganisms in terms of the matter released by the plant and consumed by the microorganisms. The effect of rhizospheric microorganisms on plant growth with normal carbon dioxide and complete mineral medium has been demonstrated. Plants interacting with microorganisms have a greater biomass than plants growing without microorganisms. Wheat growth stimulation by metabolites of rhizospheric microorganisms under laboratory conditions on artificial soil has been experimentally demonstrated (Pechurkin, 1997). Under nitrogen limitation , the biomass of plants, with or without microorganisms, is identical, and is substantially reduced as compared with the medium with standard nitrogen. В© 1999 COSPAR. Аннотация: The problem of how a gradual development of ecological and morphological adaptations combines with large genome rearrangements, which have been found to occur in the phylogeny of many groups of organisms, is a matter of discussion in the literature. The objective of this work was to study the problem with the example of salmonids, whose evolution included at least six events of multiple chromosome fusions. Large karyotype rearrangements are associated with a decrease in ecological and morphological diversity in salmonids. In the above example, genome rearrangements seem to distort the function of the genetic systems that are responsible for the occurrence of certain ecological forms in salmonids. © 2017 by the authors; Licensee MDPI, Basel, Switzerland.

Аннотация: The development of inexpensive, easy-to-produce, and easy-to-use analytical tools for detection of harmful and toxic substances is a relevant research problem with direct applications in environmental monitoring and protection. In this work, we propose a novel composite material based on alumina nanofibers and detonation nanodiamonds for detection of phenol in aqueous medium. The composite material was obtained by mixing an aqueous suspension of alumina nanofibers with a diameter of 10-15 nm and a length of several microns and a hydrosol of nanodiamonds with an average cluster size of 70 nm. The mechanisms underlying the interaction of these nanomaterials are clarified and the physicochemical properties of the composite are investigated. The SEM and TEM studies show that the obtained composite has a network structure, in which clusters of nanodiamonds (10-20 nm in diameter) are distributed over the surface of nanofibers. Coupling of nanomaterials occurs due to opposite signs of their zeta potentials, which results in electrostatic attraction and subsequent chemical bonding as indicated by the X-ray photoelectron spectroscopy and simultaneous thermal analysis. The bonding apparently occurs between functional groups (mainly carboxyl) on the surface of nanodiamonds and amphoteric hydroxyl groups on the surface of alumina nanofibers. The proposed composite allows an easy-to-perform colorimetric analysis for qualitative and quantitative determination of phenol in aqueous samples with linear response over a wide range of concentrations (0.5-106 mu M). Multiple tests have shown that the composite is reusable and retains its catalytic function for at least 1 year during storage at room temperature.

Аннотация: Using mathematical modeling methods, the work investigates possible existence of a phytoflagellate population in the ecosystem of a stratified lake (Lake Shira, Khakasia, Russia). Until this study was carried out, no phytoflagellate population had been detected in the lake, although there had been indirect evidence of the possible presence of a cryptomonad population in this water body. To answer the question if this population may be present in this water body, as the first step of investigation, a mathematical model was developed describing the microorganism biomass dynamics and phytoflagellate relationship with the lake food web. The calculations showed that cryptophytic algae were abundant in the water column layers above the lake chemocline at the beginning of summer but showed low biomass concentrations during other periods of time. In the summer following the construction of the model, samples were collected from Lake Shira, as the next step of the study. A cryptophytic algae population was found in the water column, and the pattern of its distribution in the ecosystem was similar to that predicted by the model. In addition to that, we studied theoretically possible reasons for the early summer development of the population in the deep water layers and found that it was the result of the mixotrophic feeding strategy of cryptomonads and trophic interactions in the food web. The study shows that preliminary modeling of the locational and temporal distribution of populations whose presence in the ecosystem is only suggested by indirect evidence may be useful as a tool for searching for the species in natural ecosystems and as an approach enabling a more accurate description of the food web structure of a water body. Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers - polyhydroxyalkanoates (PHAs) - of two types - poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 +/- 0.06-0.75 +/- 0.11 mu m, while the average diameter of copolymer particles varied between 0.43 +/- 0.12 and 0.55 +/- 0.05 mu m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant.

Аннотация: The brightness spectra of the Krasnoyarsk and Dnepr reservoir systems are presented for various periods of their development, from the beginning of their filling to the final stage of their formation. The curves of the surface distribution of chlorophyll a, which were remotely obtained from the brightness coefficient spectra, are found to be monotonic when diatomaceous, green protococcus, and dinophyte algae dominate in the phytoplankton. In the presence of blue-green algae, the curve of the distribution of phytoplankton on the surface is essentially irregular. The irregularities range from tens to hundreds of meters with sharp gradients of the phytoplankton concentration. The negative values of the VI color index change for positive ones when the concentration of chlorophyll a of the phytoplankton in the subsurface layer Cchl > 250-270 mg/m3. This is associated with the change in the species composition of the phytoplankton from the dominating diatom forms to bluegreen algae. Copyright В© 2005 by Pleiades Publishing, Inc.

Аннотация: Adsorption of viral particles from the blood plasma of patients with viral hepatitis B and C on modified nanodiamonds (MNDs) was shown in the in vitro experiments. PCR method showed the treatment of plasma with MNDs leads to a decrease in the viral load by 2–3 orders of magnitude or more in both cases studied. These results make it possible to predict the applicability of MNDs for the development of new technologies of hemodialysis and plasmapheresis for binding and removal of viral particles from the blood of infected patients. © 2016, Pleiades Publishing, Ltd.

Аннотация: The results of the development and utilization of an affine magnetic sorbent with Ni2+ ions immobilized on coal ash microspheres are reported. The applicability of the material in the isolation of Histag proteins is demonstrated by examples of the recombinant green fluorescent protein from Clytia gregaria and the Ca2+ regulated photoprotein obelin from Obelia longissima. The specific sorption capacity of the sorbent was 2-7 mg/cm3 for medium-size proteins (20-30 kDa). The particles are suitable for chromatography with the presence of chaotropic agents and EDTA. They are easy to manipulate as isolation of a target protein takes 30-35 min. On the one hand, the elevated affinity of the sorbent to proteins rich in native histidines may result in a high degree of irreversible sorption; on the other hand, it allows isolation of such proteins without the introduction of artificial polyhistidine fragments. Аннотация: We have reviewed foreign and domestic literature devoted to the study of the amino acid (AA) composition of aquatic organisms representing major groups of producers (green microalgae and diatoms, and cyanobacteria) and primary consumers (zooplankton). Based on published data, we estimate the composition of essential and nonessential AAs of microalgae, cyanobacteria, and zooplankton and determine their differences. It is concluded that the AA composition of major groups of plankton is heterogeneous. The role of AAs as a limiting factor for the development of herbivorous zooplankton is discussed. The prospects and the need for further study of AA composition in order to develop a complete theory of functioning of aquatic ecosystems have been demonstrated.

Аннотация: Total dependence of aquaculture on imports of feeds can significantly compromise Russia's food supply security; hence the task of designing new domestic feeds is of primary importance. A major challenge of world aquaculture is providing fish with a balanced diet, in particular, with a well-balanced composition of amino acids. In this review, we analyze international publications on the composition of amino acids in fish feed over the last two years. We summarize the data on the amino acid composition of prospective food sources for fish aquaculture: agricultural crops, insects, annelids, higher crustaceans, animal waste, phytoplankton, zooplankton, macrophytes, and microorganisms. We performed a comparative analysis of the composition of amino acids in promising aquafeed sources and fish flour traditionally used for fish feeding. Based on the results, we conclude that each of the examined food sources can be used as a component of fish nutrition and offers realistic prospects for production of alternative feeds. To ensure high quality of fish aquaculture products, the priority should be given to development of technologies which allow to control amino acid feed composition during the production process with respect to fish species-specific needs. It is hoped that this review will be useful to researchers and practitioners involved in designing and producing new domestic aquafeed for fish. © Siberian Federal University. All rights reserved.

Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: Since almost any waste recycling requires non-zero resources, almost any technology exerts non-zero impact on the biosphere. Therefore, there is an objective of finding technological configuration providing a minimal harmful effect on the biosphere. Correct assessment of the harmful effect requires as broad a framework of consideration as possible. A comparison of ecological impacts of gasoline and electric vehicles was conducted as an example of applying this approach. The contribution of technological processes involved into production, usage, and disposal of vehicles was taken into account. Comparison of the total environmental impact of the vehicles has led to a result that differs radically from widely distributed points of view about the environmental benefits of electric vehicles. It indicates that the use of the proposed approach can significantly change perceptions about the environmental and economic efficiency of various industrial products and processes. Аннотация: Fullerenols are nanosized water-soluble polyhydroxylated derivatives of fullerenes, a specific allotropic form of carbon, bioactive compounds, and perspective basis for drug development. Our paper analyzes the antioxidant activity and toxicity of a series of fullerenols with different number of oxygen substituents. Two groups of fullerenols were under investigation: (1) C60Oy(OH)x, C60,70Oy(OH)x, where x + y = 24-28 and (2) C60,70Oy(OH)x, Fe0,5C60Oy(OH)x, Gd@C82Oy(OH)x, where x + y = 40-42. Bioluminescent cellular and enzymatic assays (luminous marine bacteria and their enzymatic reactions, respectively) were applied to monitor toxicity in the model fullerenol solutions and bioluminescence was applied as a signaling physiological parameter. The inhibiting concentrations of the fullerenols were determined, revealing the fullerenols' toxic effects. Antioxidant fullerenol' ability was studied in solutions of model oxidizer, 1,4-benzoquinone, and detoxification coefficients of general and oxidative types (DGT and DOxT) were calculated. All fullerenols produced toxic effect at high concentrations (>0.01 g L-1), while their antioxidant activity was demonstrated at low and ultralow concentrations (<0.001 g L-1). Quantitative toxic and antioxidant characteristics of the fullerenols (effective concentrations, concentration ranges, DGT, and DOxT) were found to depend on the number of oxygen substituents. Lower toxicity and higher antioxidant activity were determined in solutions of fullerenols with fewer oxygen substituents (x + y = 24-28). The differences in fullerenol properties were attributed to their catalytic activity due to reversible electron acceptance, radical trapping, and balance of reactive oxygen species in aqueous solutions. The results provide pharmaceutical sciences with a basis for selection of carbon nanoparticles with appropriate toxic and antioxidant characteristics. Based on the results, we recommend, to reduce the toxicity of prospective endohedral gadolinium-fullerenol preparations Gd@C82Oy(OH)x, decreasing the number of oxygen groups to x + y = 24-28. The potential of bioluminescence methods to compare toxic and antioxidant characteristics of carbon nanostructures were demonstrated.

Аннотация: Bioluminescence of isolated enzymes is a perspective phenomenon for biosensors development due to simplicity of registration of a physiological parameter - light intensity. Enzyme-based bioluminescent assay is widely used to evaluate a decrease in biochemical toxicities. Also the enzyme-based assay is used for the direct biochemical monitoring of oxidative toxicity. This work considers antioxidant properties of fullerenols, water-soluble polyhydroxylated derivatives of fullerenes and perspective pharmaceutical agents, in solutions of model inorganic and organic toxicants of oxidative type K-3[Fe(CN)(6)] and 1,4-benzoquinone. Two fullerenol preparations were used: C60O2-4(OH)(20-24) and mixture of two types of fullerenols C60O2-4(OH)(20-24)+C70O2-4(OH)(20-24). The enzyme-based assays showed the peculiarities of the detoxification processes: ultralow concentrations of fullerenols were active (ca 10(-17)-10(-5)g/L); no monotonic dependence of detoxification efficiency on fullerenol concentrations was observed, and detoxification of organic oxidizer solutions was more effective than that of the inorganic oxidizer. The antioxidant effects of highly diluted fullerenol solutions were attributed to hormesis phenomenon; the detoxification was concerned with stimulation of adaptive cellular response under low-dose exposures. Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.
