Аннотация: The cladoceran Daphnia magna inhabits lakes with salinities up to 10 g L-1. We compared the effects of different salinities (up to 9 g L-1) on the survival, specific grazing rate, and size selective feeding of Daphnia females adapted to fresh or saline waters (3-4 g L-1). The freshwater population was more sensitive to high salinity (LC50 = 5.3 g L-1), while the survival of the saline water population also decreased in fresh water. Freshwater population demonstrated a higher grazing rate in fresh water, while the saline water population had a higher grazing rate at the salinity above 3 g L-1. A decrease in the grazing rate of the freshwater population was observed at the salinity above 4-5 g L-1. Populations differed in food selectivity. The saline water population consumed particles of larger sizes than the freshwater population. The average size of phytoplankton particles grazed in fresh water was larger than in saline water. This size selective salinity-dependent grazing may be related to the dependence of the feeding efficiency of cladocerans on the viscosity of water and size of phytoplankton particles. Our results indicate that Daphnia populations adapted to a certain salinity can temporarily lose the ability to control phytoplankton because of salinity fluctuations.

Аннотация: We assessed the efficiency of the transfer of essential substances (carbon, phosphorus, nitrogen, and fatty acids (FA), including polyunsaturated fatty acids (PUFAs)) from phytoplankton to planktonic crustaceans in experimental mesocosms in the presence and absence of fish. The experiments were conducted under mesotrophic conditions in 300 L mesocosms. We have found that transfer efficiencies from producers to consumers are different for different substances. In particular, FA, including PUFAs, are transferred less efficiently than carbon. In contrast, the efficiency of nutrient transfer, especially phosphorus, is higher than that of carbon. This evidences that zooplankton can accumulate nutrients, increasing their quality as a resource for higher trophic levels. Fish significantly reduced the efficiency of carbon transfer from phytoplankton to zooplankton per unit of water volume, but did not affect the transfer of substances per unit of biomass. Thus, the quality of zooplankton as a food resource for higher trophic levels did not decrease in the presence of fish, despite the decline in the efficiency of the transfer of the essential substances per unit of water volume under their influence. Since the efficiency of essential substances transfered from phytoplankton to zooplankton determines the functioning of the entire trophic web, we should seek ways to increase it.

Аннотация: The trophic transfer efficiency (TTE) is an important indicator of ecosystem functioning. However, TTE data from freshwater food webs are ambiguous due to differences in time scales and methods. We investigated the transfer of essential substances (carbon, nutrients, and polyunsaturated fatty acids) through plankton communities in 30 Polish lakes with different trophic status in the middle of summer. The results of our study revealed that different essential substances were transferred from phytoplankton to zooplankton with varying efficiencies. The average TTE of C, N, P, and the sum of ?-3 PUFA were 6.55%, 9.82%, 15.82%, and 20.90%, respectively. Our results also show a large mismatch between the elemental and biochemical compositions of zooplankton and their food during the peak of the summer stagnation, which may further promote the accumulation of essential substances. There were also large differences in TTEs between trophic conditions, with the highest efficiencies in oligotrophic lakes and the lowest in dystrophic and eutrophic lakes. Therefore, our study indicates that disturbances like eutrophication and dystrophication similarly decrease the TTE of essential substances between phytoplankton and zooplankton in freshwater food webs. © 2021 The Authors. Аннотация: The review summarizes studies on the detoxification effects of water-soluble humic substances (HS), which are products of the natural transformation of organic substances in soils and bottom sediments that serve as natural detoxifying agents in water solutions. The detoxifying effects of HS on microorganisms are quite complex: HS neutralize free pollutants (indirect bioeffects) and also stimulate the protective response of organisms (direct bioeffects). Prospects and potential problems of bioluminescent bacteria-based assay to monitor toxicity of solutions in the presence of HS are discussed. The main criterion for the bioassay application is versatility and ease of use. The detoxification efficiency of HS in different pollutant solutions was evaluated, and the detoxification mechanisms are discussed. Particular attention was paid to the direct and complex direct + indirect effects of HS. The review focuses on the protective function of HS in solutions of radionuclides and salts of stable metals, with special consideration of the antioxidant properties of HS.

Аннотация: The efficiency of the transfer of carbon, polyunsaturated fatty acids (PUFA), nitrogen and phosphorus from phytoplankton to zooplankton determines the functioning of the entire ecosystem. However, this parameter depends on environmental conditions. Fish as a very important factor to regulate planktonic communities are very likely to affect the efficiency of the transfer of basic elements and substances from phytoplankton to zoo plankton. In experimental mesocosms filled with water from an eutrophic lake and containing phyto- and zooplankton, we investigated how fish affect both primary and secondary production and the efficiency of transfer of carbon, nitrogen, phosphorus, PUFA and fatty acids (FA) from phytoplankton to zooplankton. Two treatments (control and fish treatment) were repeated in three replicates. The transfer efficiency of substances from phytoplankton to zooplankton was measured as the ratio of secondary to primary production, expressed in liters and per biomass unit in percent. The efficiency expressed per liter characterizes the productivity of the water body, while the efficiency expressed per biomass unit indicates the effectiveness of aquatic species to transfer biologically valuable substances from one trophic level to another. We found that phytoplankton-zooplankton-fish interactions are determined not only by predator-prey relationships, but also are affected by the quality of both phytoplankton and zooplankton, measured as the contents of phosphorus, nitrogen, PUFA and FA in their biomass. We showed that, in the presence of fish, the transfer efficiency of carbon, phosphorus, nitrogen, eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), FA per biomass unit from phytoplankton to zooplankton was increased two-fold, 12.4-fold, 2,5-fold, 12.4-fold, 7.4-fold, and 10-fold, respectively, relative to control. This provides the sustainability of the functioning of the ecosystem under fish pressure. Such a mechanism prevents zooplankton over-exploitation by fish and enables to prolong the food chain.

Аннотация: The transfer pathways of organic matter and elements from phytoplankton to zooplankton in freshwater ecosystems are important for understanding how aquatic ecosystems function. We conducted a mesocosm experiment to determine how fish and zebra mussels altered the transfer efficiencies of essential substances including carbon (C), polyunsaturated fatty acids (PUFAs), total fatty acids (FAs), phosphorus (P), and nitrogen (N) from phytoplankton to zooplankton. We assessed the transfer efficiencies of the essential substances from phytoplankton to zooplankton as the ratio of their zooplankton production (P) per unit of biomass (B) to that of phytoplankton to exclude grazing or predation effects. We hypothesized that zebra mussels and fish would affect the transfer of materials from phytoplankton to zooplankton by altering the contents of essential elements and FAs in phytoplankton and zooplankton communities and/or due to shifts in the planktonic community structure mediated by grazing and/or predation. Fish increased the transfer efficiencies of eicosapentaenoic acid 20:5 omega-3 (EPA), docosahexaenoic acid 22:6 omega-3 (DHA), and P relative to the control. We speculated that fish weakened the control of zooplankton over algal assemblage by selectively feeding on larger cladocerans such as Daphnia. Therefore, fish can increase the relative proportion of high-quality food for zooplankton, improving food conditions for the available zooplankton. In contrast, zebra mussels reduced the transfer efficiencies of EPA and DHA relative to the control treatment likely due to competition with zooplankton for PUFA-rich food particles. However, zebra mussels did not have any impact on the transfer efficiencies of C, total FAs, N, and P. EPA, DHA, and P were transferred more efficiently than C from phytoplankton to zooplankton, while total FAs, which are commonly used as an energetic source, were transferred as efficiently as C. The enrichment of consumers with the most important substances relative to their basal food sources creates the potential for the successful transport of these substances across aquatic trophic webs./p

Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers - polyhydroxyalkanoates (PHAs) - of two types - poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 +/- 0.06-0.75 +/- 0.11 mu m, while the average diameter of copolymer particles varied between 0.43 +/- 0.12 and 0.55 +/- 0.05 mu m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant.

PHOTOLUMINESCENCE QUANTUM YIELD
Аннотация: Coelenterazine-v (CTZ-v), a synthetic derivative with an additional benzyl ring, yields a bright bioluminescence of Renilla luciferase and its "yellow" mutant with a significant shift in the emission spectrum toward longer wavelengths, which makes it the substrate of choice for deep tissue imaging. Although Ca2+-regulated photoproteins activated with CTZ-v also display red-shifted light emission, in contrast to Renilla luciferase their bioluminescence activities are very low, which makes photoproteins activated by CTZ-v unusable for calcium imaging. Here, we report the crystal structure of Ca2+-regulated photoprotein obelin with 2-hydroperoxycoelenterazine-v (obelin-v) at 1.80 angstrom resolution. The structures of obelin-v and obelin bound with native CTZ revealed almost no difference; only the minor rearrangement in hydrogen-bond pattern and slightly increased distances between key active site residues and some atoms of 2-hydroperoxycoelenterazine-v were found. The fluorescence quantum yield (phi(FL)) of obelin bound with coelenteramide-v (0.24) turned out to be even higher than that of obelin with native coelenteramide (0.19). Since both obelins are in effect the enzyme-substrate complexes containing the 2-hydroperoxy adduct of CTZ-v or CTZ, we reasonably assume the chemical reaction mechanisms and the yields of the reaction products (phi(R)) to be similar for both obelins. Based on these findings we suggest that low bioluminescence activity of obelin-v is caused by the low efficiency of generating an electronic excited state (phi(S)). In turn, the low phi(S) value as compared to that of native CTZ might be the result of small changes in the substrate microenvironment in the obelin-v active site.

Аннотация: The efficiency of the transfer of physiologically valuable substances including polyunsaturated fatty acids (PUFA), nitrogen, and phosphorus from phytoplankton to zooplankton is extremely important for the functioning of the whole food web. The bivalve mollusk Dreissena polymorpha has a great impact on planktonic communities. However, its effects on the efficiency of transfer of physiologically valuable substances from phytoplankton to zooplankton have not been assessed so far. The goal of our study was to assess the effect of zebra mussels on the transfer efficiency of carbon, fatty acids including PUFA, nitrogen, and phosphorus from phytoplankton to zooplankton under mesotrophic conditions. In the experimental mesocosms filled with water from a mesotrophic lake containing phyto- and zooplankton, we manipulated the presence/absence of zebra mussels. The experiments showed that zebra mussels decreased the efficiency of the transfer of carbon, PUFA, nitrogen, and phosphorus from phytoplankton to zooplankton, thus reducing the quality of zooplankton as a food resource for higher trophic levels. However, zooplankton revealed the ability to increase the content of physiologically valuable substances in their tissues relative to their contents in the diet. We believe that the mechanism of accumulation of physiologically valuable substances by zooplankton enables to at least partially resist the negative influence of zebra mussels on the quality of zooplankton as a food resource.

Аннотация: The review is devoted to the study of the internal radiation regime in the canopies cultivated under controlled environmental conditions. The expediency of using canopies as an object of research for evaluating the photobiological efficiency of radiation in light culture conditions is justified. The appropriateness of light measurements in multi-tiered canopies is shown, taking into account the role of leaves of different tiers in the formation of an economically useful crop. The main requirements for light devices for their use in measuring artificial radiation in light culture conditions are considered, and a brief analysis of the existing instrument base for performing these studies is given. A number of examples show the complexity and ambiguity of the internal structure of the light field that is forming within canopies in light culture conditions. Conceptual approaches to the choice of spectral and energy characteristics of artificial irradiation for plant light culture are proposed and justified. The necessity of taking into account the light conditions of leaves of different tiers when choosing the spectral and energy characteristics of light sources for the cultivation of multi-tiered canopies is justified. Techniques, methods, and light sources used for additional intracanopy lighting are analysed. The efficiency of using side illumination of plant canopies and conditions for its implementation are considered. The advantages of the volume distribution of canopies on the most common multi-tiered lighting installations are discussed. Based on the presented mate- rial, we consider ways to improve methodological approaches for evaluating the photobiological effectiveness of artificial radiation in light culture conditions for canopies of cultivated plants, taking into account the features of their architectonics and internal radiation regime.

Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers – polyhydroxyalkanoates (PHAs) – of two types – poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 ± 0.06–0.75 ± 0.11 ?m, while the average diameter of copolymer particles varied between 0.43 ± 0.12 and 0.55 ± 0.05 ?m. © 2021 Wiley Periodicals LLC.

Аннотация: In the present investigation, the role of dietary intervention in male Wistar rats (n = 8, 3 groups) was studied to observe absolute sirtuin 1 (SIRT1) levels (expressed as ng mg(-1) total protein) in serum, stomach, liver, and kidney. Dietary buckwheat at 30% (w/w) level of incorporation in the standard diet (Buckwheat Enriched Diet, BED) improved SIRT1 with values 0.933 +/- 0.05, 210 +/- 7, 63.26 +/- 4, and 69.89 +/- 3 in serum, stomach, liver, and kidney respectively when compared to the respective control values of 0.536 +/- 0.03, 156 +/- 23.3, 31.07 +/- 2 and 47.11 +/- 4. Moreover, BED though isocaloric to CR diet, led to weight gain (g) by 63.11 +/- 3.8, ca. 10%, and 40% higher than control (56.27 +/- 5.6) and CR (45.05 +/- 4.1) diet groups. A marked rise in Feed Efficiency Ratio (FER) by ca. 37% while a 30% decrease in Feed Conversion Ratio (FCR) was observed for the BED group which supports unexpected weight gain in rats post-dietary intervention. The results justify the superior nutritional profile of buckwheat laden with essential nutrients, essential proteins, and bioactives. In contrast, Calorie Restriction (CR) resulted in a decline of the total protein content in circulation by 19%, while reduction of total protein in stomach, liver, and kidney was estimated to be 95%, 35.2%, and 27% respectively though SIRT1 values were comparatively the highest in all the samples studied. A 30-fold enhancement of SIRT1 in stomach post CR is presumed to counter enhanced stress in gastric tissues. Therefore, mild to moderate expression of SIRT1 may confer beneficial effects such as delayed aging and stress resistance but exceedingly high SIRT1 may evoke increased oxidative stress.

Аннотация: Accumulation of essential/beneficial and non-essential chemical elements by submerged (Stuckenia pectinata (L.) Borner) and emergent (Phragmites australis (Cav.) Trin. ex Steud.) rooted macrophytes in three water bodies was studied as dependent on water salinity (specific conductivity) and main environmental factors (temperature, pH of water, growing period, element contents in water and sediments). Inductively coupled plasma optical emission spectrometry (ICP - OES) was used for the element detection in the plants, sediments, and water. Multivariate statistics redundancy analysis (RDA) showed that investigated factors together explained 37.8 - 44.7% and 29.9 - 42.6% of the essential/beneficial and non-essential elements variability, respectively, whereas salinity was one of the main factors determining the accumulation of a number of elements (Mg, S, Na, Li, Sr, Sb) by the macrophytes. A decrease in the essential element contents (K, P, N, Cu, Zn) was observed in both species of the macrophytes during the growing season. Environmental factors (salinity, growing period, and pH) were more significant than the element contents in water and the sediments for the essential/beneficial element accumulation by two species of the macrophytes. The element contents in the sediments to a greater extent determined non-essential element accumulation by P. australis than by S. pectinata. The study showed that the efficiency of using P. australis and S. pectinata for remediation of aquatic ecosystems contaminated by heavy metals should not decrease with a salinity increase. © 2020 Elsevier Masson SAS

METABARCODING APPROACH
RAPID ASSESSMENT
Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.
