Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The structure elucidation and synthesis of the luciferin from the recently discovered luminous earthworm Fridericia heliota is reported. This luciferin is a key component of a novel ATP-dependent bioluminescence system. UV, fluorescence, NMR, and HRMS spectroscopy studies were performed on 0.005 mg of the isolated substance and revealed four isomeric structures that conform to spectral data. These isomers were chemically synthesized and one of them was found to produce light when reacted with a protein extract from F. heliota. The novel luciferin was found to have an unusual extensively modified peptidic nature, thus implying an unprecedented mechanism of action.

Аннотация: The Ca2+-regulated photoprotein obelin determines the luminescence of the marine hydroid Obelia longissima. Bioluminescence is initiated by calcium and appears as a result of the oxidative decarboxylation related to the coelenterazine substrate. The luciferase of the luminescent marine coral Renilla muelleri (RM) also uses coelenterazine as a substrate. However, three proteins are involved in the in vivo bioluminescence of these animals: luciferase, green fluorescent protein, and Ca2+-regulated coelenterazine-binding protein (CBP). In fact, CBP that contains one strongly bound coelenterazine molecule is the RM luciferase substrate in the in vivo bioluminescent reaction. Coelenterazine becomes available for oxygen and the reaction with luciferase only after binding CBP with calcium ions. Unlike Ca2+-regulated photoproteins, the coelenterazine molecule is not activated by oxygen in the CBP molecule. In this work, by means of quantum chemical methods the behavior of substrates in these proteins is analyzed. It is shown that coelenterazine can form different tautomers: CLZ(2H) and CLZ(7H). The formation of 2-hydroperoxy-coelenterazine is studied. According to the obtained data, these proteins use different forms of the substrates for the reaction. In obelin, the substrate is in the CLZ(2H) form that affords hydrogen peroxide. In RM, coelenterazine is in the CLZ(7H) form, and therefore, CBP is not activated by oxygen.

Аннотация: Detecting the folding/unfolding pathways of biological macromolecules is one of the urgent problems of molecular biophysics. The unfolding of bacterial luciferase from Vibrio harveyi is well-studied, unlike that of Photobacterium leiognathi, despite the fact that both of them are actively used as a reporter system. The aim of this study was to compare the conformational transitions of these luciferases from two different protein subfamilies during equilibrium unfolding with urea. Intrinsic steady-state and time-resolved fluorescence spectra and circular dichroism spectra were used to determine the stages of the protein unfolding. Molecular dynamics methods were applied to find the differences in the surroundings of tryptophans in both luciferases. We found that the unfolding pathway is the same for the studied luciferases. However, the results obtained indicate more stable tertiary and secondary structures of P. leiognathi luciferase as compared to enzyme from V. harveyi during the last stage of denaturation, including the unfolding of individual subunits. The distinctions in fluorescence of the two proteins are associated with differences in the structure of the C-terminal domain of alpha-subunits, which causes different quenching of tryptophan emissions. The time-resolved fluorescence technique proved to be a more effective method for studying protein unfolding than steady-state methods.

Аннотация: The present study addresses the herbicidal activity and biological effects of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides used to control various weed species (Amaranthus retroflexus, Sinapis arvensis,andLeucanthemum maximum). The effects of the free herbicides and the herbicides embedded in granules of degradable polymer poly-3-hydroxybutyrate [P(3HB)] blended with birch wood flour were compared. Metribuzin, regardless of the form, caused 100% mortality of the three weeds by day 21. The herbicidal activity of tribenuron-methyl was lower than that of metribuzin, but the embedded TBM was superior to the free herbicide in the length and strength of its action on the weeds. Both metribuzin forms dramatically decreased the main parameters of fluorescence: maximum quantum yield of photosystem-II [Y(II)(max)], maximum quantum yield of non-photochemical quenching [Y(NPQ)(max)], and maximum rate of non-cyclic electron transport [ETRmax] and concentrations of chlorophyllaandb. The effect of the embedded TBM on the photosynthetic activity of the weeds was lower in the first two weeks of the growth of herbicide-treated plants but lasted longer than the effect of the free TBM and increased over time. Embedding of metribuzin in the matrix of degradable blend did not decrease its herbicidal activity.

Аннотация: Bioluminescence of a variety of marine coelenterates is determined by Ca2+-regulated photoproteins. A strong interest in these proteins is for their wide analytical potential as intracellular calcium indicators and labels for in vitro binding assays. The presently known hydromedusan Ca2+-regulated photoproteins contain three (aequorin and clytin) or five (obelin and mitrocomin) cysteine residues with one of them strictly conserved. We have constructed Cys-free aequorin and obelin by substitution of all cysteines to serine residues. Such mutants should be of interest for researchers by the possibility to avoid the incubation with dithiothreitol (or p-mercaptoethanol) required for producing an active photoprotein that is important for some prospective analytical assays in which the photoprotein is genetically fused with a target protein sensitive to the reducing agents. Cys-free mutants were expressed in Escherichia coil, purified, and characterized regarding the efficiency of photoprotein complex formation, functional activity, and conformational stability. The replacement of cysteine residues has been demonstrated to affect different properties of aequorin and obelin. Cys-free aequorin displays a two-fold lower specific bioluminescence activity but preserves similar activation properties and light emission kinetics compared to the wild -type aequorin. In contrast, Cys-free obelin retains only 10% of the bioluminescence activity of wild-type obelin as well as binding coelenterazine and forming active photoprotein much less effectively. In addition, the substitution of Cys residues drastically changes the bioluminescence kinetics of obelin completely eliminating a "fast" component from the light signal decay curve. At the same time, the replacement of Cys residues increases conformational flexibility of both aequorin and obelin molecules, but again, the effect is more prominent in the case of obelin. The values of thermal midpoints of unfolding (Tm) were determined to be 53.3 0.2 and 44.6 0.4 C for aequorin and Cys-free aequorin, and 49.1 0.1 and 28.8 0.3 C for obelin and Cys-free obelin, respectively. Thus, so far only Cys-free aequorin is suitable as a partner for fusing with a tag sensitive to reducing agents since the aequorin mutant preserves almost 50% of the bioluminescent activity and can be produced with a substantial yield.

Аннотация: The bioluminescent bacterial enzyme system NAD(P)H:FMN-oxidoreductase-luciferase has been used as a test system for ecological monitoring. One of the modes to quench bioluminescence is the interaction of xenobiotics with the enzymes, which inhibit their activity. The use of endogenous flavin fluorescence for investigation of the interactions of non-fluorescent compounds with the bacterial luciferase from Photobacterium leiognathi and NAD(P)H:FMN-oxidoreductase from Vibrio fischeri has been proposed. Fluorescence spectroscopy methods have been used to study characteristics of endogenous flavin fluorescence (fluorophore lifetime, the rotational correlation time). The fluorescence anisotropy behaviour of FMN has been analysed and compared to that of the enzyme-bound flavin. The fluorescence characteristics of endogenous flavin of luciferase and NAD(P)H:FMN-oxidoreductase have been shown to be applicable in studying enzymes' interactions with non-fluorescent compounds. Аннотация: The heat tolerance of wheat (Triticum aestivum L.) and radish (Raphanus sativus L. var. minor) cenoses exposed to elevated and damaging air temperatures (35 degreesC for 20 h, 45 degreesC for 7 h) under photoculture conditions at various levels of photosynthetically active radiation (PAR) was assessed by measuring characteristics of the slow induction curve of chlorophyll fluorescence at 682 and 734 nm and the CO2 exchange rate. Irrespective of the illumination level, the exposure of the cenoses to 35 degreesC did not induce irreversible changes in the plant photosynthetic apparatus. The lowest extent of damage to wheat and radish cenoses exposed to 45 degreesC was observed at 150 W/m(2) of PAR, whereas the highest damage of the plants was observed at an illumination level that was close to the compensation point of the cenose photosynthesis (50-70 W/m(2) of PAR at air temperature of 24 degreesC). Viability index proved to be the most sensitive. characteristic, compared to other characteristics, which were determined by measuring the slow phase of fluorescence induction at 682 and 734 nm. In the cenoses studied, the pattern of changes in the viability index in response to a stress factor was close to the changes in the photosynthetic rate.

Аннотация: Wheat (Triticum sativus L.) seedlings of various ages (2- to 16-day-old plants) were used to study age-dependent changes in the chlorophyll fluorescence induction (CFI) at various light intensities during flu- orescence measurements. Plants were raised in a growth chamber using hydroponics with expanded clay, controlled environmental conditions, and 690 A mu mol/(m(2) s) photon flux density (PFD) of photosynthetically active radiation (PAR). Parameters of CFI were determined under actinic PFD of 380, 580, 820, and 1340 A mu mol/(m(2) s) PAR. The fifth leaf from the stem base, exposed to uniform lighting, was sampled for measurements. This leaf emerged at the plant age of 16 days. Based on fluorescence data, we calculated the maximal photochemical quantum yield of photosystem II (F (v)/F (m)), the effective photochemical quantum yield of PSII (Yield), parameters of photochemical (qP) and non-photochemical (qN and NPQ) quenching of chlorophyll fluorescence, the F (p)/F (t) ratio, and the "vitality index" (fluorescence decrease ratio, R (fd)). At moderate actinic PFD, applied commonly in PAM fluorometers (about 380 A mu mol/(m(2) s)), age-dependent changes in NPQ, F (p)/F (t), and R (fd) were observed. Analysis of CFI parameters in wheat leaves of different ages at PFD increasing from 380 to 820 A mu mol/(m(2) s) revealed that R (fd), NPQ, and qN are the most sensitive markers of the leaf age among all parameters tested. These suitable indicators can be used for rapid assessment of the leaf age.

Аннотация: The usage of chlorophyll fluorescence induction (CFI) for estimating various types of plant resistance (primary, general, initial, adaptive) to stress factors is reviewed. The necessity of ontogenetic approach (considering the age-specific properties of the photosynthetic apparatus) in determining general and adaptive resistance of plants to prolonged action of stress factors by the CFI method is argued. In the plant Cucumbis sativus L., the possibility is shown of using age-specific qualitative and quantitative traits of leaf CFI (changes in the shape of chlorophyll fluorescence induction curves and in the dynamics of CFI parameters in the course of leaf ontogeny) for comparative study of differences between fully active and stressed plants. Possible criteria are suggested for estimating the effect of outer stress factors by the presence or absence of a steady-state phase in the dynamics of CFI parameters during leaf ontogeny. It is also suggested to use the duration of the steady-state phase following the termination of leaf growth (estimated by the dynamics of the slow phase of CFI as the ratio of fluorescence intensity at the peak P and the steady-state fluorescence intensity, FP/FS, or as the viability index Rfd) and the variability of CFI parameters during this period as qualitative estimates of plant resistance to prolonged action of stress factors.

Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light: 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C. quadrangula has endogenous nature and can change according to light conditions.
