Аннотация: The heat tolerance of wheat (Triticum aestivum L.) and radish (Raphanus sativus L. var. minor) cenoses exposed to elevated and damaging air temperatures (35 degreesC for 20 h, 45 degreesC for 7 h) under photoculture conditions at various levels of photosynthetically active radiation (PAR) was assessed by measuring characteristics of the slow induction curve of chlorophyll fluorescence at 682 and 734 nm and the CO2 exchange rate. Irrespective of the illumination level, the exposure of the cenoses to 35 degreesC did not induce irreversible changes in the plant photosynthetic apparatus. The lowest extent of damage to wheat and radish cenoses exposed to 45 degreesC was observed at 150 W/m(2) of PAR, whereas the highest damage of the plants was observed at an illumination level that was close to the compensation point of the cenose photosynthesis (50-70 W/m(2) of PAR at air temperature of 24 degreesC). Viability index proved to be the most sensitive. characteristic, compared to other characteristics, which were determined by measuring the slow phase of fluorescence induction at 682 and 734 nm. In the cenoses studied, the pattern of changes in the viability index in response to a stress factor was close to the changes in the photosynthetic rate.

Аннотация: We recently introduced a new electrophoretic method, nonequilibrium, capillary electrophoresis of equilibrium mixtures (NECEEM). NECEEM provides a unique way of finding kinetic and equilibrium parameters of the formation of intermolecular complexes from a single electropherogram and allows for the use of weak affinity probes in protein quantitation. In this work, we study theoretical bases of NECEEM by developing a mathematical model for the new method. By solving a system of partial differential equations with diffusion in linear approximation, we found the analytical solution for concentrations of components involved in complex formation as functions of time from the beginning of separation and position in the capillary. The nonnumerical nature of the solution makes it a powerful tool in studying the theoretical foundations of the NECEEM method and modeling experimental results. We demonstrate the use of the model for finding binding parameters of complex formation by nonlinear regression of NECEEM electropherograms obtained experimentally.

Аннотация: For the identification of ecologically significant dominant groups of phytoplanktonic algae a polychromatic DCMU-induced fluorescence method is recommended. A special fluorometer equipped with a system of replaceable filters is used to differentiate three regions of the spectrum (410 +/- 20, 510 +/- 20 and 540 +/- 10 nm) that can excite the basic light-harvesting pigments. Total and differential (for every algal taxon studied) chlorophyll a calculated from the fluorescence signals is in good agreement with biomass estimates from direct cell counts for several different trophic types of aquatic systems. This is made possible by the vizualization of the ratios of fluorescence signal values in their own coordinates: first, to decide whether it is necessary to correct linear equations in order to eliminate negative solutions; second, to determine the possibility of nulling the negative solution if a point is situated close to a side of the triangle; and third, to reduce the number of linear algebraic equations to two if the points are situated along one of the triangle sides or to one if the points are gathered at the apex. The polychromatic DCMU-induced fluorescence method can be used for monitoring natural phytoplankton populations to detect changes in their taxonomic structure.

Аннотация: The time-resolved and steady-state fluorescence techniques,were employed to elucidate possible interactions of four aromatic compounds (anthracene, POPOP, MSB and 1,4-naphthalendiol) with bacterial luciferase. Fluorescence spectra and fluorescence anisotropy decays of these compounds were studied in ethanol, water-ethanol solutions and in the presence of bacterial luciferase. Shifts of fluorescent spectra and differences in rotational correlation times are interpreted in terms of weak (hydrophobic) interactions of the molecules with the enzyme. These interactions suggest the feasibility of intermolecular energy transfer by an exchange resonance mechanism with a collision-interaction radius as a way of excitation of these compounds in the reaction catalysed by bacterial luciferase. Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The solvation dynamics of interesting bioluminescent chromophores have been determined, using subpicosecond and wavelength-resolved fluorescence spectroscopy, in combination with global analysis of the multidimensional data sets. The systems investigated comprise the free ligands 6,7-dimethyl-(8-ribityl)-lumazine (lumazine) and riboflavin in an aqueous buffer and both ligands when noncovalently bound to two bacterial bioluminescent antenna proteins: lumazine protein (from Photobacterium leiognathi) and the blue fluorescent protein (from Vibrio fischeri Y1). Fluorescence spectral relaxation of the free ligands is complete within a few picoseconds. Subsequently, the fluorescence intensity increases by similar to7% on a time scale of 15-30 ps. Fluorescence spectral relaxation of the protein-bound ligands is largely complete within 1 ps but reveals a small red shift with a minor, but distinctly longer, relaxation time than that of the free ligands, which is tentatively assigned to the relaxation of protein-bound water in the vicinity of the excited chromophore.

Аннотация: The bioluminescent bacterial enzyme system NAD(P)H:FMN-oxidoreductase-luciferase has been used as a test system for ecological monitoring. One of the modes to quench bioluminescence is the interaction of xenobiotics with the enzymes, which inhibit their activity. The use of endogenous flavin fluorescence for investigation of the interactions of non-fluorescent compounds with the bacterial luciferase from Photobacterium leiognathi and NAD(P)H:FMN-oxidoreductase from Vibrio fischeri has been proposed. Fluorescence spectroscopy methods have been used to study characteristics of endogenous flavin fluorescence (fluorophore lifetime, the rotational correlation time). The fluorescence anisotropy behaviour of FMN has been analysed and compared to that of the enzyme-bound flavin. The fluorescence characteristics of endogenous flavin of luciferase and NAD(P)H:FMN-oxidoreductase have been shown to be applicable in studying enzymes' interactions with non-fluorescent compounds. Аннотация: Changes in absorption, fluorescence, phosphorescence, and delayed fluorescence spectra of erythrosin B are studied in the presence of bovine serum albumin at room temperature. Spectral and chronoscopic characteristics of the observed photophysical processes are defined. The binding of erythrosin B with the protein followed by spectral changes is demonstrated. Absorption and fluorescence spectra of the dye in the bound state are described, the binding mechanism is analyzed. The binding parameters of the dye-protein complex are estimated.

Аннотация: This study assesses the comparative sensitivity and possibility of obtaining fast results of various methods of biotesting for several surfactants: Tween 85, sodium dodecyl sulfate (SDS), Fairy dishwashing gel, and Mif washing powder. The following test organisms are used for the study: luminescent bacteria Photobacterium phosphoreum (Beijerinck), preparation of Ecolum luminescent bacteria, unicellular algae Scenedesmus quadricauda (G.M. Smith), infusorian Paramecium caudatum (Ehrenberg), and crustacean Daphnia magna (Straus). It has been revealed that Fairy dishwashing gel possesses the strongest toxicity against the studied test objects. Daphnia and algae are most sensitive to the effects of Fairy and SDS, protozoan and luminescent bacteria are most sensitive to SDS, and Ecolum is most sensitive to Mif washing powder. The tested aquatic organisms and Ecolum are most tolerant to the effect of Tween 85. © 2016, Pleiades Publishing, Ltd.

Аннотация: Tryptophan fluorescence lifetimes were analyzed for three proteins: human serum albumin, bovine serum albumin, and bacterial luciferase, which contain one, two, and seven tryptophan residues, respectively. For all of the proteins, the fluorescence decays were fitted by three lifetimes: ?1 = 6–7 ns, ?2 = 2.0–2.3 ns, and ?3 ? 0.1 ns (the native state), and ?1 = 4.4–4.6 ns, ?2 = 1.7–1.8 ns, and ?3 ? 0.1 ns (the denatured state). Corresponding decay-associated spectra had similar peak wavelengths and spectrum half-widths both in the native state (??1max = 342 nm, ??2max = 328 nm, and ??3max = 315 nm), and in the denatured state (??1max = 350 nm, ??2max= 343 nm, and ??3max= 317 nm). The differences in the steady-state spectra of the studied proteins were accounted for the individual ratio of the lifetime component contributions. The lifetime components were compared with a classification of tryptophan residues in the structure of these proteins within the discrete states model. © 2016, Pleiades Publishing, Inc.

Аннотация: Application of pulse-amplitude-modulation (PAM) fluorometers for measuring slow stages of chlorophyll fluorescence induction (CFI) is considered. With an example of Triticum aestuvum L. plants grown under continuous illumination at a photon flux density of 600 μmol/(m2 s) photosynthetically active radiation (PAR), the CFI curves were analyzed with leaves of various ages as a function of actinic light intensity. The fluorometer PAM-2100 was applied for measurements of CFI curves. The characteristic peaks of CFI curves in wheat leaves were most conspicuous and had the largest amplitudes at 600–800 μmol/(m2 s) PAR, which corresponds to the middle range of actinic light intensities employed in PAM-2100 fluorometers. In plants exposed to favorable and stressful conditions, the developmental stages may proceed at different rates; thus, the comparison of fluorescence parameters for leaves of equal calendar age but having different physiological states may provide ambiguous data. Therefore, the feasibility of recording CFI curves of different types is quite important for rapid diagnostics of the age and state of plant leaves, as well as for adequate physiological conclusions. © 2016, Pleiades Publishing, Ltd.

Аннотация: Wheat (Triticum sativus L.) seedlings of various ages (2- to 16-day-old plants) were used to study age-dependent changes in the chlorophyll fluorescence induction (CFI) at various light intensities during flu- orescence measurements. Plants were raised in a growth chamber using hydroponics with expanded clay, controlled environmental conditions, and 690 A mu mol/(m(2) s) photon flux density (PFD) of photosynthetically active radiation (PAR). Parameters of CFI were determined under actinic PFD of 380, 580, 820, and 1340 A mu mol/(m(2) s) PAR. The fifth leaf from the stem base, exposed to uniform lighting, was sampled for measurements. This leaf emerged at the plant age of 16 days. Based on fluorescence data, we calculated the maximal photochemical quantum yield of photosystem II (F (v)/F (m)), the effective photochemical quantum yield of PSII (Yield), parameters of photochemical (qP) and non-photochemical (qN and NPQ) quenching of chlorophyll fluorescence, the F (p)/F (t) ratio, and the "vitality index" (fluorescence decrease ratio, R (fd)). At moderate actinic PFD, applied commonly in PAM fluorometers (about 380 A mu mol/(m(2) s)), age-dependent changes in NPQ, F (p)/F (t), and R (fd) were observed. Analysis of CFI parameters in wheat leaves of different ages at PFD increasing from 380 to 820 A mu mol/(m(2) s) revealed that R (fd), NPQ, and qN are the most sensitive markers of the leaf age among all parameters tested. These suitable indicators can be used for rapid assessment of the leaf age.

Аннотация: The effects of actinic light (AL) intensity on the age dependence of nonphotochemical fluorescence quenching (q(N)) and effective quantum yield in PSII (I broken vertical bar(PSII)) were studied in continuously illuminated wheat leaves of the upper tier. Regular changes were revealed in both age dependence of q(N) at elevated AL intensities and light curves of q(N). These changes are related to alterations in strategies of redistribution and use of absorbed light energy by the photosynthetic apparatus at different stages of wheat leaf development. Unlike I broken vertical bar(PSII), q(N) as a parameter was more sensitive to the differences in the leaf age at a certain range of light intensities. At the same time, the stability of q(N) at moderate light intensities may serve as an indication of leaf maturity.

Аннотация: A method for localization of a light source near the interface of two media is described. The method is based on an optical analog of tunnel effect when the radiation source is at the distance smaller than the wavelength from the interface. Application of the tunnel effect permits to obtain high resolution. The developed method was used to determine the localization of a bacterial luminescent system. It has been found that the sources of bioluminescence are located at thin subsurfase lager with width about 70 nm. This result is in favour of the peripheral (periplasmatic or membrane) localization of the bacterial luminescent system. This method makes it possible to investigate processes pelated to light radiation (luminescence, fluorescence and other optical processes) in a thin surface layer of various biological and physical objects.

Аннотация: Additional integral and kinetic parameters of induction curves have been introduced on the basis of analysis of currently employed parameters of slow induction of fluorescence (SIF) for a more complete quantitative description of SIF changes during ontogenesis of higher plant leaves. High sensitivity of the additional parameters to the age state of leaf tissue as compared with previously employed parameters is demonstrated on the example of cucumber leaves (Cucumis sativus L.). Analysis of the relationship between the photosynthetic rate and SIF parameters of separate cucumber leaf disks revealed the presence of a high positive correlation between the photosynthetic rate and the greatest possible average decline in fluorescence intensity during the inductive period for mature regions of the leaf and regions starting to senesce (the correlation coeffcient attained values of 0.95), which indicates the possibility of using the given SIF parameter in further elaboration of methods for estimation of chloroplast photosynthetic activity at the level of the higher plant leaf.
