Аннотация: Abstract: A key component of the recently described bioluminescent system of higher fungi is luciferase, a new class of proteins. The properties of fungal luciferase and their relationship with its structure are interesting both for improving autoluminescent systems already created on its basis and for creating new ones. Therefore, it is extremely important to understand the spatial structure of this protein. We have performed heterologous expression and purification of Neonothopanus nambi luciferase, obtained a protein suitable for subsequent crystallization, and also determined some biochemical properties of the recombinant luciferase. © 2021, The Author(s),.

Аннотация: Mycelia of various Armillaria fungi are bioluminescent while the fruit bodies do not emit light. The presence in fruit bodies of Armillaria species of enzymes involved in the fungal bioluminescence was investigated by treating them with an exogenous analogue of the substrate for the light-emitting reaction. For this, hot extracts from nonluminous fungus Pholiota squarrosa were used. Upon spraying the pristine and transversely cut fruit bodies with the extracts, light emitting regions of different intensity were revealed. This suggests that the fruit bodies of the studied species are nonluminous due to lack of the substrate for light luminescent reaction. The prolonged incubation of the fruit bodies in water elevated the bioluminescence level. A possible mechanism which can explain this phenomenon is discussed. © 2021 Institute of Physics Publishing. Аннотация: The present study investigates, for the first time, the structure of the microbial community of cryogenic soils in the subarctic region of Siberia and the ability of the soil microbial community to metabolize degradable microbial bioplastic – poly-3-hydroxybutyrate [P(3HB)]. When the soil thawed, with the soil temperature between 5-7 and 9–11 °C, the total biomass of microorganisms at a 10-20-cm depth was 226–234 mg g?1 soil and CO2 production was 20–46 mg g?1 day?1. The total abundance of microscopic fungi varied between (7.4 ± 2.3) ? 103 and (18.3 ± 2.2) ? 103 CFU/g soil depending on temperature; the abundance of bacteria was several orders of magnitude greater: (1.6 ± 0.1) ? 106 CFU g?1 soil. The microbial community in the biofilm formed on the surface of P(3HB) films differed from the background soil in concentrations and composition of microorganisms. The activity of microorganisms caused changes in the surface microstructure of polymer films, a decrease in molecular weight, and an increase in the degree of crystallinity of P(3HB), indicating polymer biodegradation due to metabolic activity of microorganisms. The clear-zone technique – plating of isolates on the mineral agar with polymer as sole carbon source – was used to identify P(3HB)-degrading microorganisms inhabiting cryogenic soil in Evenkia. Analysis of nucleotide sequences of rRNA genes was performed to identify the following P(3HB)-degrading species: Bacillus pumilus, Paraburkholderia sp., Pseudomonas sp., Rhodococcus sp., Stenotrophomonas rhizophila, Streptomyces prunicolor, and Variovorax paradoxus bacteria and the Penicillium thomii, P. arenicola, P. lanosum, Aspergillus fumigatus, and A. niger fungi. © 2020 Elsevier Ltd

Luciferase enzyme is one of the most crucial components of this system. The luciferase from Neonothopanus nambi fungus belongs to the novel still undescribed protein family. A detailed study of the N. nambi luciferase properties is necessary for the improvement of analytical methods based on the fungal bioluminescent system. Here we present the positions of key amino acid residues and their effect on enzyme function described using bioinformatic and experimental approaches. Аннотация: The extracellular enzyme with oxidase function was extracted from the Neonothopanus nambi luminescent fungus by using mild processing of mycelium with beta-glucosidase and then isolated by gel-filtration chromatography. The extracted enzyme is found to be a FAD-containing protein, catalyzing phenol co-oxidation with 4-aminoantipyrine without addition of H2O2, which distinguishes it from peroxidases. This fact allowed us to assume that this enzyme may be a mixed-function oxidase. According to gel-filtration chromatography and SDS-PAGE, the oxidase has molecular weight of 60 kDa. The enzyme exhibits maximum activity at 55-70 degrees C and pH 5.0. Kinetic parameters K-m and V-max of the oxidase for phenol were 0.21 mM and 0.40 mu M min(-1). We suggest that the extracted enzyme can be useful to develop a simplified biosensor for colorimetric detection of phenol in aqueous media, which does not require using hydrogen peroxide.

Аннотация: A reusable system for phenol determination in an aqueous medium was obtained by adsorption of extracellular oxidase from fungus Neonothopanus nambi onto modified nanodiamonds (MND) synthesized by detonation. It was found that the enzyme strongly binds to MND and exhibits catalytic activity in the reaction of co-oxidation of phenol with 4-aminoantipyrine without the addition of hydrogen peroxide. In the presence of the MND-oxidase complex, a significantly (by an order of magnitude) higher yield of the reaction product is recorded as compared to the yield in the presence of a free enzyme; the mechanism of the revealed effect is discussed. Model experiments have demonstrated the multiple use of the MND-oxidase complex for testing phenol in aqueous samples. The immobilized enzyme exhibits functional activity during long-term (2 months) storage of the MND-oxidase complex at 4 degrees C. The data obtained create the prerequisites for using the created system in environmental monitoring of water pollution with phenol.

Аннотация: Five permanent sample plots (SPs; 200-250 trees per plot) were established in middle-aged high-grade suburban pine stands near the industrial city of Krasnoyarsk, Siberia, Russia. Needle damage, inventory parameters of the stands, and the defense response of the stem phloem were evaluated annually for the years 2002-2019 and attributed to acute or chronic toxic exposures (creeping fire or industrial pollutants, respectively). The results form a basis for using trees as bioindicators. A newly elaborated stem lesion test was formed from a hypothesis on the upward sugar transport for the regeneration of an injured crown, based on Eschrich's model of bidirectional sugar transport in the phloem. The formation of a phloem lesion was induced by inoculation of the stem with a mycelial extract of the ophiostomatoid fungus Ceratocystis laricicola. The lesion length and its shift relative to the inoculation hole were measured. An increase in the length of needles at early stages of stand weakening by pollutants was found to correspond to the hormesis model (Selye's adaptation syndrome). A possibility of assessing the chronology of pollutant toxicity and the duration of the recovery period after creeping fire was shown.

Аннотация: Modified nanodiamonds (MNDs) produced by detonation synthesis can be used as an effective adsorbent to immobilize extracellular peroxidases of the luminous basidiomycete Neonothopanus nambi. The enzymes are firmly immobilized on MND particles and exhibit catalytic activity. The indicator system (the MND-enzyme complex) reused many times retains its ability to catalyze reaction of co-oxidation of phenol and 4-aminoantipirine in the presence of hydrogen peroxide and remains functionally active during long-term storage (for 1 month or longer) in aqueous suspensions at 4 degrees C. MNDs and enzymes of higher fungi can be effectively used to construct new reusable indicator systems for analytical applications such as monitoring contamination of aquatic environments by phenolic compounds.

Аннотация: In in vitro experiments, the possibility of using a luminescent system extracted from the luminous fungus Armillaria borealis has been shown to detect and determine the concentration of hispidin. A linear dependence of the luminescent response on the content of hispidin in solutions in the concentration range of 5.4 x 10(-5) - 1.4 x 10(-2) mu M was detected. The stability of the enzyme system and the high sensitivity of the bioluminescent reaction allows carrying out multiple measurements with the analyte detection limit of 1.3 x 10(-11) g. The obtained results show the prospects of creating a rapid bioluminescent method for the analysis of medical substances or extracts from various biological objects for the presence of hispidin.

Аннотация: A bifunctional indicating complex was created by immobilization of extracellular oxidases (glucose oxidase and peroxidases) of luminous fungus Neonothopanus nambi onto modified nanodiamonds (MNDs) synthesized by detonation. It was found that the enzymes firmly adsorb onto MND particles and exhibit their catalytic activity. Model in vitro experiments showed that the created MND-enzymes complex is suitable for repeated use for analyte (glucose and phenol) testing and retains its activity after storage at 4 degrees C in deionized water for 1 month. The data obtained offer the prospects for developing a new class of reusable multifunctional indicating and diagnostic test systems on the basis of MNDs and higher fungal enzymes for medical and ecological analytics.

Аннотация: The present study estimates the level of extracellular peroxidase activity and light emission intensity of mycelium of luminescent basidiomycete Neonothopanus nambi treated with ?-glucosidase. A hypothesis has been proposed that treatment with ?-glucosidase may trigger biochemical mechanisms of activation of ROS (primarily hydrogen peroxide) generation in N. nambi mycelium. The results obtained indicate that the enzyme causes partial disintegration of the slimy sheath of fungal hyphae and intracellular matrix, which leads to release of the extracellular peroxidases to the incubation medium. Mycelial cells treated with the enzyme reach the peak of their luminescence sooner. It has been assumed that partial loss of extracellular peroxidases, as important enzymes of antioxidant defense, may be compensated for by an increase in the level of light emission by the fungus. © 2018 Beijing Academy of Agriculture and Forestry Sciences.

Аннотация: A comparative evaluation of the level of extracellular peroxidase activity and light-emission intensity of the mycelium of the luminescent basidiomycete Neonothopanus nambi in the presence of ?-glucosidase was performed. The enzyme activity damages the hyphae of the fungus leading to osmotic imbalance, partial degradation of the mycelium, and release of extracellular peroxidases into the incubation medium. The presence of ?-glucosidase reduces the time necessary to reach the maximum luminescence. Putative biochemical mechanisms that underlie the stimulation of reactive oxygen species formation (first and foremost, of hydrogen peroxide) in the N. nambi mycelium in the presence of ?-glucosidase are proposed. © O.A. Аннотация: The influence of various concentrations of Ag, TiO2, and SiO2 nanoparticles on conidia germination of the phytopathogenic fungus Bipolaris sorokiniana and mycelium growth of the xylotrophic fungi Pleurotus ostreatus and Neonothopanus nambi was analyzed. It is established that a decrease in conidia germination of phytopathogenic fungus and mycelium growth of the xylotrophic fungi occurs at relatively high silver nanoparticle concentrations. The appropriate ЕС50 values for Bipolaris sorokiniana, Pleurotus ostreatus, and Neonothopanus nambi are found to be 30, 14, and 31 mg/dm3, respectively. No effect of various concentrations of TiO2 and SiO2 nanoparticles on the studied cultures of fungi is highlighted. Thus, the negative impact of nanosilver on the growth of fungi at a high concentration which is untypical for objects of the environment and a lack of this influence of titanium dioxide and silica nanoparticles testifies to the high resistance of mycelial forms of organisms to engineered nanoparticles. © 2018, Pleiades Publishing, Ltd.

Аннотация: С помощью определения компонентов-участников биолюминесцентного процесса в светящихся и несветящихся органах опёнка осеннего выяснили причины неполного свечения Armillaria mellea. Полный набор ферментов и субстратов, необходимых для биолюминесценции, образуется только в мицелии, причём исключительно в условиях свободного доступа кислорода. В плодовых телах синтез предшественника люциферина (гиспидина) и 3-гидроксигиспидингидроксилазы блокирован, вследствие этого и не происходило образование люциферина - ключевого компонента биолюминесцентной системы грибов. Именно поэтому плодовое тело опёнка осеннего не светится, несмотря на наличие люциферазы - фермента, катализирующего окисление люциферина с испусканием кванта света.

Аннотация: The study estimates morphological properties and levels of extracellular peroxidase activity and light emission of mycelium of the basidiomycete Armillaria borealis IBSO 2328 treated with beta-glucosidase and chitinase. Mycelium incubated with the enzymes shows considerable morphological changes and indications of osmotic shock. Injuries observed in the cell envelope of the fungal hyphae are primarily attributed to the partial (in the beta-glucosidase treatment) or complete (in the chitinase treatment) disintegration of the melanin layer on the surface of the cell wall. Changes in the cell wall of hyphae are accompanied by release of extracellular peroxidases of the fungus into the incubation medium and an increase in light emission relative to the luminescence of the control pellets. We assume that higher level of luminescence of the enzyme-treated mycelium samples could be related to the disintegration of the surface pigment layer of the hyphae and the partial loss of extracellular peroxidases. The data obtained confirm the previously proposed hypothesis in which light producing reaction of the fungus may be an additional way to neutralize active oxygen radicals under stress.

Аннотация: Mycelium of Armillaria species exhibit bioluminescence in nature and when cultivated on artificial nutrient media. However, fruiting bodies do not emit visible light. The present study investigates biochemical changes which cause this phenomenon. Light emission was studied in experiments with mixtures of cold and hot extracts of the luminous mycelium of Armillaria borealis IBSO 2328 and nonluminous fruiting bodies of this fungus and an unidentified species of the genus (Armillaria sp.). Hot extracts of fruiting bodies of the nonluminous Pholiota squarrosa were used as the substrate analog of the luminescent reaction, as previously this fungus had been found to contain a high amount of this substance. Control experiments showed that cold extracts of A. borealis IBSO 2328 mycelium contained enzymes for the luminescent reaction, which is initiated after addition hot extracts of P. squarrosa fruiting bodies. Parallel experiments with extracts of the fruiting bodies of Armillaria showed that: (i) - cold extracts did not contain enzymes of the luminescent reaction or contain very small amounts of these enzymes and (ii) - hot extracts did not contain substrate of the luminescent reaction. Thus, the reason why fruiting bodies of Armillaria do not emit light is that they do not contain components required for visible luminescence. The study discusses possible causes why the enzymes and substrate of the luminescent reaction are not synthesized in fruiting bodies of Armillaria. © Guizhou Academy of Agricultural Sciences.
