Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, c-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physicalmechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB.

Аннотация: The growth of the carboxydobacterium Seliberia carboxydohydrogena Z-1062 was studied at a different concentration of mineral elements under the conditions of continuous turbidostat cultivation. The chemical composition of the bacterium was determined and the dynamics of its specific growth rate and the intracellular content of mineral elements were studied when the concentration of these elements was changed in the growth medium. The synthesis of 1 g of biomass by this strain required: N, 120 mg; P, 23 mg; S, 5.5 mg; K, 1.5 mg; Mg, 3.5 mg. Changes in the residual concentrations of the elements within a broad range had no effect on the specific rate of the carboxydobacterial growth and chemical composition.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: A mathematical model concerning the interaction of plants and rhizospheric microorganisms on complete mineral medium and under nitrogen limitation has been constructed. The model takes into account the closeness of plants and microorganisms in terms of the matter released by the plant and consumed by the microorganisms. The effect of rhizospheric microorganisms on plant growth with normal carbon dioxide and complete mineral medium has been demonstrated. Plants interacting with microorganisms have a greater biomass than plants growing without microorganisms. Wheat growth stimulation by metabolites of rhizospheric microorganisms under laboratory conditions on artificial soil has been experimentally demonstrated (Pechurkin, 1997). Under nitrogen limitation , the biomass of plants, with or without microorganisms, is identical, and is substantially reduced as compared with the medium with standard nitrogen. В© 1999 COSPAR. Аннотация: In monocultures of micro-organisms, growth is controlled by feedback mechanisms involving chemical factors such as limiting substrates and inhibitory metabolic products. The role of such feedback in the growth regulation of Escherichia coli O-124 was investigated by growing cells in batch culture using a medium containing glucose and mineral salts. In various phases of growth, portions of the native culture were diluted with culture filtrate, so that although cell density decreased, the chemical composition of the growth medium was unaltered. As the diluted cultures grew, variations in growth acceleration were calculated and compared with those of native (undiluted) cultures. Towards the end of the exponential phase and in the growth deceleration phase, the specific feedback level (FBL) was between -20 and -200 (h g l-1)-1. The feedback components resulting from changes in glucose concentration were calculated using experimentally determined values of mu(max) (0.55 +/- 0.05 h-1) and K(s) (2.5 +/- 0.7 mg l-1). Only 0.1-40% of FBL could be accounted for by changes in glucose concentration, indicating the presence of additional growth regulators. The method developed may become a new tool for determination of growth-regulating cell-density factors in microbial cultures.

Аннотация: Experiments based on an earlier developed theory with a culture of Escherichia coli 0-124 were undertaken to determine the feedback strength (FBS) in growth control. FBS is defined as the change in growth acceleration brought about by a discontinuous reduction in microbial concentration under conditions of constant chemical composition of the medium, and is an over-all measure of the regulatory influence of all chemical factors dependent on biomass concentration. FBS was determined for batch cultivation with glucose. In various growth phases the culture was diluted by its own filtrate and then the change in growth acceleration as compared to the control was measured. FBS was reliably different from zero at the end of the exponential phase and in the phase of decelerated growth and was of the order of -20 to -200 (h.g/liter)(-1). The feedback component due to glucose alone was estimated using the obtained relation between specific growth rate and glucose concentration (mu(max) = 0.55 +/- 0.05 h(-1), K-s = 2.5 +/- 0.7 mg/liter). The glucose-dependent regulatory contribution to FBS was found to be as low as 0.5 to 40%, which suggests the presence of other growth control factors. Upon accuracy improvements, this method may develop into an accepted microbiological tool for identifying major density-dependent factors involved in microbial growth control.

The analysis of spectral curves obtained through field ground measurements and from satellite data showed that fine spectral differences can be used to study the spatial distribution of various types of vegetation and their ecological state. Based on the created electronic spectral brightness data base, the possibilities are shown of using spectrophotometric information for determining morphophysiological changes occurring in the plants and their species composition. The determined contrasts can be effectively used to obtain necessary information while processing space images, which suffer from natural interferences (varying optical thickness of the atmosphere, cloudiness, alterations in the scanner's angle of view, varying solar height, and highly inhomogeneous underlying surface). Аннотация: Remote sensing applications have greatly enhanced ability to monitor and manage in the areas of forestry. Accurate measurements of regional and global scale vegetation dynamics (phenology) are required to improve models and understanding of inter-annual variability in terrestrial ecosystem carbon exchange and climate-biosphere interactions. Study of vegetation phenology is required for understanding of variability in ecosystem. In this paper, monitoring of vegetation dynamics using time series of satellite data is presented. Vegetation variability (vegetation rate) in different topoclimatic areas is investigated. Original software using IDL interactive language for processing of satellite long-term data series was developed. To investigate growth dynamics vegetation rate inferred from remote sensing was used. All estimations based on annual time series of Moderate Resolution Imaging Spectroradiometer (MODIS) imagery. Vegetation rate for Enhanced Vegetation Index (EVI) and Normalized Difference Vegetation Index (NDVI) was calculated using MODIS data. The time series covers spring seasons of each of 9 years, from 2000 to 2008. Comparison of EVI and NDVI derived growth rates has shown that NDVI derived rates reveal spatial structure better. Using long-term data of vegetation rates variance was estimated that helps to reveal areas with anomalous growth rate. Such estimation shows sensitivity degree of different areas to different topoclimatic conditions. Woods of heights depend on spatial topoclimatic variability unlike woods of lowlands. Principal components analysis shows vegetation with different rate conditions. Also it reveals vegetation of same type in areas with different conditions. It was demonstrated that using of methods for estimating the dynamic state of vegetation based on remote sensing data enables successful monitoring of vegetation phenology. В© 2011 COSPAR. Аннотация: This study investigates synthesis of poly(3-hydroxybutyrate)/diethylene glycol copolymers (P3HB/DEG) by Cupriavidus eutrophus B-10646 cells as related to DEG concentration in the medium and the time when it is added to the culture of cells synthesizing P3HB. The study determines the limits of physiological effect of DEG on C. eutrophus cells, showing that at DEG concentrations above 30 g/L, it inhibits cell growth, decreasing cell concentration and total P3HB/DEG yield and inducing an increase in the degree of saturation of fatty acids in lipids of cell cytoplasmic membrane. A series of copolymers containing different molar fractions of DEG (between 0.13 and 3.0 mol%) have been synthesized and their physicochemical, physical/mechanical, and biological properties have been investigated as related to the chemical composition and proportions of DEG monomers of the polymers. © 2016 American Institute of Chemical Engineers Biotechnol. Аннотация: The synthesis of polyhydroxybutyrate by the carboxydobacterium Seliberia carboxydohydrogena under various growth conditions was studied. The bacterium was able to accumulate poly-beta-hydroxybutyric acid and a heteropolymer that was a product of copolymerization of hydroxybutyric and hydroxyvaleric acids. Nitrogen and sulfur exerted the strongest effect on polymer accumulation in carboxydobacteria: The maximum polymer concentrations in cells reached 28%. Specimens of polyhydroxybutyrate films were obtained, and their degradability by soil microflora was shown.

Аннотация: Age-dependent accumulation of 137Cs in the muscles and bodies of the pike Esox lucius (aged two to seven years) inhabiting a section of the Yenisei River polluted with artificial radionuclides has been studied. The content of 137Cs in muscles varied from 0.5 to 7.0 Bq/kg of fresh weight. The maximum content of the radionuclide has been found in juveniles. The content of 137Cs in pike muscles and body decreased considerably with age. The high content of 137Cs in the muscles of juveniles is probably a consequence of their higher intensity of feeding as compared to older individuals, which is due to the intense growth of juveniles. © 2016, Pleiades Publishing, Ltd.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.

Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.
